1.Studies on esterase isozyme and peroxidase isozyme of Polyporus umbellatus

OBJECTIVE:To study the change regular of esterase isozyme and peroxidase isozyme of Polyporus umbellatus. METHOD:Using polypropylene amide circle electrophoresis.RESULTS:The pattern and activity of isozyme in same strain had some difference at different culture days,but some of isozyme pattern bands of the Rf value were visible in whole culture stage.Every strain had its peculiar pattern bands.CONCLUSION:The number of isozyme pattern bands and enzyme activity were proportional to the growing rate of myceliun,as well as its metabolic activity.
2.Analysis of Shenzhen Universal Infant Hepatitis B Immunization Strategy:Cost-effectiveness Analysis Based on Markov Model

Objective: To optimize the Shenzhen universal infant hepatitis B immunization method. Methods: To analyze the long effectiveness of different immunization strategies and disease transition after hepatitis B virus infection by using cost-effectiveness method through decision-tree and Markov models among the Shenzhen newborns cohort in 2010 based on the local parameters. Results:Through the current strategies, the program of screening all pregnant women for HBsAg and vaccinating newborns of single-positive mothers combing with HBIG and vaccine would not only prevent more new cases, hepatic carcinoma and related deaths; but also gain more living years and QALYs. From the social prospect, it will save about RMB 40 million compared with the current program. Conclusion: It is suggested that Shenzhen Government should amend the current universal infant hepatitis B immunization program to gain more economic and social benefits.
3.Therapeutic effect of Shexiang Baoxin Pill combined with treatment based on syndrome differentiation for acute ischemic stroke

AIM: To investigate the effect of treatment based on syndrome differentiation only and combined with Shexiang Baoxin Pill for acute ischemic stroke(AIS). METHODS: Fifty-eight cases of AIS were randomly (allocated) to two groups.Thirty cases of group A were treated with treatment based on syndrome differentiation combined with Shexiang Baoxin Pill and twenty-eight cases of group B with the treatment based on syndrone differentiation. [Objective] To investigate the effect of aromatic herbs for resuscitation such as Moschus and Borneol on nitric oxide (NO) content and expression of endothelial NO synthetase (eNOS) and inducible NO synthetase (iNOS) in brain tissue of rats with cerebral ischemia and reperfusion injury. [Methods] Forty rats were allocated to 4 groups: mimic operation group (Group A), model group (Group B), herbs treatment group (Group C) and nimodipine treatment group (Group D). Rat model of cerebral ischemia and reperfusion injury was established by Pulsinelli 4VO method. NO content in brain tissue was determined by nitrate reductase method. Expression of eNOS and iNOS in micrangium and iNOS in neurons was measured by semi-quantitative analysis. [Results] NO content was decreased and approached normal level in Group C and Group D (P 0.05 as compared with Group A). Objective To investigate the psychological experiences of young uremic patients during different periods waiting for renal donor .Methods The phenomenology study method was adopted to conduct the semi-structure interviews in young uremic patients waiting for renal donor .Results The four themes were summarized:(1) having hope at the early stage (0—6 months);(2) ineffective coping at the middle stage(7—12 months);(3)uncertainty at the late stage(13—24 months);(4)serious negative emo-tion(more than 2 years) .Conclusion The young uremic patients patients waiting for renal donors have complex and changeable psychological experience .Medical staff should provide the pertinent professional support for promoting the living quality of patients by aiming at the psychological characteristics in young uremic patients .
8.Self-efficacy training on fluid intake in maintenance hemodialysis patients

Objective To examine the effectiveness of self-efficacy training on fluid intake in maintenance hemodialysis patients.Methods Fifty-four hemodialysis patients participated in the study.The intervention was based on Bandura's self-efficacy theory.The outcome measure was self-efficacy for managing chronic disease,the mean interdialysis weight gain,dialysis-related complications and patient satisfaction.Data were collected at baseline and 3 months after the intervention.Results The mean score of selfefficacy,the mean interdialysis weight gain,dialysis-related complications and patient satisfaction after the intervention were all statistically significant compared with those bcfore the intervention.Conclusions The study supports the effectiveness of the self-efficacy training in improving the fluid intake compliance in maintenance hemodialysis patients.So it is worth being popularized in clinic.
9.Construction and characterization of a novel bispecific antibody against both IL-1β and IL-17A

Objective To construct bispecific antibody BsAb1/17 against both IL-1β and IL-17A,express and purify the biologically active BsAbl/17 protein in prokaryotic system for further studies and applications. Methods VH1VL17-CL and VL1VH17-CH1 gene segments were constructed by overlap-PCR.Restriction enzyme sites Nco Ⅰ and BamH Ⅰ were designed at the 5'and 3' end primers respectively. The products of overlap-PCR were ligated to the Nco Ⅰ/BamH Ⅰ -prepared pET-27b vector. The recombinant plasmids pET-27b-VH1 VL17-CL(petA) and pET-27b-VL1 VH17-CH1 ( petB ) were transformed into E. coliRosetta separately. The expressing products were analyzed by SDS-PAGE and Western blot. Neutralization activity of the bispecific antibody for blocking the induction of IL-18 expression by IL-1β in human T cells was determined by real-time PCR. Neutralization activity of the bispecific antibody for blocking the induction of IL-6 expression by IL-17A in HeLa cells was determined by ELISA assay. Results The structure of the plasmids pET-27b-VH1 VL17-CL(petA) and pET-27b-VL1 VH17-CH1 (petB)was confirmed by DNA sequencing. After induction, the fusion proteins were expressed mainly as inclusion bodies. The purity of the both proteins exceeded 90%. SDS-PAGE analysis suggests the relative molecular mass of both products expressed by petA and petB were approximately 38× 103, which is in accordance with the theoretical value. The results of Western blot and ELISA test demonstrated that BsAb1/17 molecule had binding ability to both IL-1β and IL-17A. The BsAb1/17 could block IL-1β to stimulate human T cell to express IL-18 and block IL-17A to stimulate HeLa cell to express IL-6. Conclusion We successfully constructed a novel bispecific antibody BsAb1/17 against both IL-1 β and IL-17A, and expressed biologically active BsAb1/17 protein in prokaryotic system.
10.Radiation barrier for cervical cancer treated with external radiation

Objective To study more reasonable method of radiation barrier for cervical cancer treated with external radiation. Methods The distance from cervical canals to tumor's margin in x axis was measured by B ultrasonic, around uterus were evaluated by physical examination.and the dose around uterus for patient who used fixed lead brick for radiation barrier when treated with external radiation were calculationed by TPS, in 39 case of cervical cancer treated with external radiation combine with intracavitary irradiation. Pay attention to the lower dose area around uterus. Results It might bring about lower dose area around uterus who used fixed lead brick for radiation barrier when treated with external radiation,and mass might be in above area. Conclusion Cervical cancer treated with external radiation with source axial distance (SAD), and radiation barrier with lead brick individuate may help for to avoid the lower dose area around uterus.
