OBJECTIVE:To determine the retative bioavailability of Diclofenac sodium eye-ointment in rabbit eyes and to evaluate its pharmacokinetics following topical application METHODS:48 rabbits were divided into two groups 0 1% Diclofenac sodium eye-ointment and Diclofenac sodium eyedrops were applied topically to rabbit eyes and aqueous humor and iridic tissue were collected at given times The concentration of Diclofenac sodium in tissue were measured by HPLC RESULTS:Diclofenac sodium eye-ointment group had higher concentration and longer half-time than Diclofenac sodium eyedrops group,however,the peak time were the same in two groups The relative bioavailabilities of Diclofenac sodium ointment in aqueous humor and iridic tissue were 183 33% and 205 68% respectively,compared with those of Diclofenac sodium eyedrops CONCLUSION:Diclofenac sodium eye-ointment can be absorbed well and released slowly,which can reduce the frequency of application of drug It is especially suitable for operation patient or application at night
4.Effects of environmental enrichment on learning and memory ability, and the expression of brain-derived neurotrophic factor and synaptophysin in hippocampus of neonatal rats with hypoxic-ischemic brain damage

Objective To explore the effects of environmental enrichment (EE) on learning and memory ability and the expression of brain-derived neurotrophic factor (BDNF) and synaptophysin in hippocampus of neonatal rats with hypoxic-ischemic brain damage(HIBD).Methods Forty Wistar neonatal male rats aged 7 days were randomly divided into EE intervention for 6 hours(6 h EE) group (n =10),EE intervention for 12 hours (12 h EE) group (n =10),model group (n =10) and sham group (n =10).The first 3 groups were performed with HIBD.The 6 h EE and 12 h EE group received EE stimuli for 6 h and 12 h respectively,once a day for 14 days.Learning and memory of the rats were tested by using Morris water maze.The expression levels of BDNF and synaptophysin in hippocampus were determined with Western blot.Results The escape latency of all groups gradually reduced with the increase of training days,but there was no significant difference in the escape latency among the 4 groups (F =0.237,P > 0.05).The rats in the 6 h EE group,12 h EE group and model group spent less time in the target quadrant and showed a significant reduction of BDNF and synaptophysin(6 h EE group:0.529 ± 0.038,0.889 ± 0.027;12 h EE group:0.660 ± 0.034,1.114 ± 0.037;model group:0.225 ± 0.015,0.672 ± 0.057) in the hippocampus compared with the sham group (0.803 ± 0.026,1.347 ± 0.092) (all P < 0.01).In the 6 h EE group and 12 h EE group,the rats significantly increased the time spent in target quadrant and aggrandized the expression of BDNF and synaptophysin in hippocampus compared with the model group.Moreover,the 12 h EE group had a better performance than the 6 h EE group in the space exploration and the expression of BDNF and synaptophysin.Conclusion EE is helpful for improving learning and memory ability in neonatal rats with HIBD,which may be associated with up-regulating the expression of BDNF and synaptophysin in hippocampus.
5.Epidemiology and clinical characteristics of Mycoplasma pneumoniae pneumonia in hospitalized children younger than three year-old

Objective To summarize the epidemiology and evaluate possible age-related differences in the presenting clinical features in three year-old children with Mycoplasma pneumoniae pneumonia(MPP)during 2009 to 2014.Methods The medical records of 17855 children with community-acquired pneumonia enrolled by Children′s Hospital of Soochow University during 2009 to 2014.Totally 1145 younger than three year-old children with MPP were enrolled,and they were classified into three groups of 1 month to 1 year-old group (n=512),1 to 2 year-old group (n=393) and 2 to 3 year-old group (n=240).The epidemiology and possible age-related differences in the presenting clinical features,main laboratory and imaging results in three year-old children with MPP were summarized.Results (1) The highest infection rate of different reasons in Suzhou was in autumn(10.46%),and the lowest was in spring(6.95%),The highest infection rate of different ages was 2 to 3 year-old group(11.61%),and the difference was statistically significant (P<0.05).(2) Compared with 1 month to 1 year-old group(n=512,4.31%) and 1 to 2 year-old group (n=39,10.09%),2 to 3 year-old group (n=240,11.61%) had higher infection rate,more patients with fever(53.9% vs. 77.1% vs. 85.4%) and high fever(16.8% vs. 30.5% vs. 41.4%),longer time to normalization of temperature(3d vs. 5d vs. 6d),but less patients with wheezing(61.3% vs. 52.4% vs. 42.9%) and dyspnea(7.6% vs. 4.6% vs. 3.8%).(3) The differences were statistically significant in the percentage of neutrophils,lymphocytes,CRP and platelet count between different ages(P<0.001).(4) The incidences of large area of lesions in chest X-ray examination among different ages had significant differences(P<0.001).Conclusions MP is one of the important pathogens of respiratory tract infection in younger than 3 year-old children.It can occur in every season,and the highest infection rate of MP is in autumn and spring.In younger than 3 year-old,older patients are more vulnerable to infection of MP,the number of fever and high fever are more,fever duration is longer,but younger patients are more vulnerable to virus infection and prone to dyspnea.Chest X-ray examination shows small patchy shadow in most cases,the younger children are more easily to have large area of lesions and pleural effusion.
6.Experimental studies of the effect of erythropoietin on fas-associated death domain protein and caslmse-8 protein in rat with intracerebral hemorrhage

Objective To study the protein expressions of Fas-associated death domain protein (FADD) and caspase-8 in rats with intracerebral hemorrhage ,and the effects of erythropoietin tp reveal the mechanism of neu-m-protection by EPO. Methods 126 male SD rats were randomly divided into three groups: Sham-operated group, intracerebral hemorrhage group, and EPO group. Each group was divided into seven subgroups according to the differ-ent time points (3,6,12,24,48,72 h and 7 d). The model of intracerebral hemorrage was established in rats by in-tracerebral injection of autogenous blood. The protein expressions of FADD and caspas-8 in rats tissue around the hemorrhagic and the normal brain tissue were detected by immunohistochemistry. Results The protein expressions of FADD and caspase-8 were increased [(4.66±0.46 ) and ( 15.89±1.81)] at 3 h after intracerebral hemorrhage, and peaked at 48 h [ (35.88±4.24 ) and (45.04±3.99)], the expressions of FADD and caspas-8 in the region around hematoma in EPO group significantly decreased compared with model group[ (3.92±0.64) and (28.24±1.90), (13.32±2.01 ) and (35.08±2.82)] at 3 h and 48 h. Conclusion The protein expressions of FADD and easpase-8 are markedly increased after intracerebral hemorrhage. EPO can protect the neurons by signifi-cantly reducing the expressions of FADD and caspase-8.
7.Aerosol challenge model in murine by Bordetella pertussis

Objective To evaluate the BALB/c murine infective effects in different concentrations and different aerosol challenge times by Bordetella pertussis.Methods Four experiment groups according to different concentrations and different aerosol challenge times were designed.BALB/c murines were challenged by aerosol way.Group 1: 1010cfu/mL Bordetella pertussis challenge 15 min, group 2: 1010cfu/mL challenge 30 min, group 3: 109cfu/mL challenge 30 min, group 4: 1011cfu/mL challenge 30 min, using the normal saline challenge 30 min as control.At 0d,3d,7d,14d and 21d after challenge, the WBCs of all groups were measured and lung tissues were homogenized to calculate the bordetella pertussis clone in lung.Results After 3 days of challenge, WBCs in all groups were slightly increased.The WBCs of group 1, group 2, group 3 and group 4 were significantly increased after 7 days, with the average numbers of 8.52×109 per/L, 1.74×1010per/L, 1.15×1010per/L and 5×1010per/L, respectively.After 14 days, they were 1.77×1010per/L, 1.67×1010per/L, 1.27×1010per/L and 3.84×1010per/L respectively.WBCs in all groups were dramatically declined after 21 days.The WBC of negative control group had no obvious change during the whole process with the stable number of 3.4~7.0×109per/L.Bordetella pertussis were detected in lung of all experimental groups in each sampling point.The CFU in lung wase at peak at 7d or 14d after challenge, which was obviously decreased at 21d.Conclusion This aerosol challenge method can establish a bordetella pertussis infection mouse model successfully.
8.Identification of a non-essential region for replication of fowl adenovirus QU strain.

The avirulent QU strain of fowl adenovirus, a member of duck adenovirus type 1, could be a potential vector in recombinant vaccine development. To identify a non-essential region for replication of QU virus, a 3.4 kb fragment near the E4 region of QU virus genome was amplified by PCR to construct a plasmid pADGFP, in which ORF1, ORF8 and ORF9 was replaced with a system expressing enhanced green fluorescence protein. Further, a recombinant virus rQUGFP was constructed by homologous recombination after pADGFP and QU virus were co-transfected into chick embryo fibroblast. The one step growth curve of the rQUGFP was found to be identical with that of parent QU virus and the TCID50 titers of different generation recombinants maintained stable. These findings suggest that the region including ORF1, ORF8 and ORF9 of QU virus genome is dispensable for virus replication, and the foreign gene inserted into virus genome can be efficiently and stably expressed. The work lays the foundation for further studies of developing this virus as a vector of recombinant vaccine.

9.In vivo study on the potential differention capabilities of human peripheral blood-derived mononuclear cells transplanted into the nude mice with myocardial infarction.

The purpose of this study was to observe whether human peripheral dervied monouncleas cells (hMNCs) could participate in the regeneration process of the ischemic hearts in the way of differentiating into cardiomyocytes, vascular endothelial cells and smooth muscle cells. hMNCs were transplanted into the bodies of the mice with myocardial infarction through the tail vein injection. Hearts were harvested 2-12 weeks after injection then sliced up into frozen sections of 5 micron thickness. Double immunofluorescence staining was used to test the differentiation of the grafted cells into cardiomyocytes, smooth muscle cells and vascular endothelial cells which revealed that cells expressing both HLA and TNT, HLA and alpha-SMA, HLA and vWF existed in the hearts of the mice. According to the study, it is probable that hMNCs could participate in the regeneration process of the infarcted hearts in the way of differentiation.

10.Characteristics and prognosis of interrupted inferior vena cava with azygous continuation

Objective To explore the clinical characteristics of interrupted of the inferior vena cava with azygous continuation and the prognosis.Methods Retrospective analysis of 21 fetuses diagnosed with interrupted inferior vena cava with azygous continuation among 28 567 pregnant women who underwent routine ultrasound scan.The clinical data,ultrasonographic features,genetic information and prognosis were collected. Results Interrupted of the inferior vena cava with azygous continuation occurred in 21(0.07%, 21/28 567)of 28 567 patients.Three fetuses(14%,3/21)complicated with heart and extracardiac malformations, including endocardiac cushion defect,single atrium and single ventricle,double superior vena cava,dextrocardia, asplenia syndrome,visceral heterotaxy,duodenal atresia;six fetuses(29%,6/21)were associated with cardiac anomalies, such as hypoplastic left heart syndrome, double outlet right ventricle, pulmonary stenosis, ventricular septal defect,persistent left superior vena cava,endocardiac cushion defect and transposition of the great arteries;six cases(29%,6/21)were only combined with extracardiac malformations,includingasplenia syndrome, visceral heterotaxy, duodenal atresia. Three fetuses (14%,3/21) were nonorganic abnormalities included thickening of the right ventricle wall, fetal bradycardia, pericardial effusion, hydrops abdominis, increased peak systolic velocity/end diastolic velocity and single umbilical artery.Three fetuses(14%,3/21) were isolated interrupted inferior vena cava with azygous continuation,but without other anomalies and 2 of them had normal fetal karyotype.Five cases(24%,5/21)were successfully vaginal delivery,1 case(5%,1/21) had cesarean section. After 12-40 months follow-up, we didn′t obeserve obviously abnormality, nor any chromosomal abnormality.Ten patients(48%,10/21)opted for termination of the pregnancy and the autopsies were not done.Five cases(24%,5/21)were lost to follow up.Conclusions Interrupted inferior vena cava with azygous continuation are associated with cardiovascular and extracardiac anomalies, cardiac malformation and visceral heterotaxy are the most common anomalies. Visceral heterotaxy should be considered and fetal karyotype should be suggested. In the cases of isolated interrupted inferior vena cava with azygous continuation and normal karyotype,the outcome is favorable.
