1.Effect of Mutouhui Extracts on Hemorheology of U14 Cervical Cancer Mice

Objective To study the influence of Mutouhui extracts on hemorheology of U14 cervical cancer mice.Methods The U14 cervical cancer mice were divided into Mutouhui extracts high,middle and low dose group,Cisplatin Injection group,model group and blank comparing group.The influence on hemorheology change was observed after being given the medicine.Results The high,middle dose group of Mutouhui extracts and Cisplatin injection group could reduce the blood viscosity of U14 cervical cancer mouse in different shear,and hemotcrit of the low did not obviously change,groups of Mutouhui extracts plasma viscosity changed slightly.Cisplatin Injection group could reduce the blood viscosity,but did not obviously affect to the plasma viscosity.Conclusion Mutouhui extracts can reduce the U14 cervical cancer mouse blood viscosity,and improve the indexes of hemorheology.
2.Different concentrations of glycyrrhizin on hepatic stellate cell proliferation and its control mechanism

Objective To explore the different concentrations of glycyrrhizin on hepatic stellate cell proliferation and its regulation mechanism.MethodsHepatic stellate cell proliferation rate of three different concentrations of glycyrrhizin was detected by MTT assay.The expression of cyclin E was detected by immunocytochemistry.ResultsMTT assay was used to detect the cell proliferation inhabitation in different concentration groups,from control group 0.8 mg/L glycyrrhizin group to 1.2 mg/L glycyrrhizin group and 1.6 mg/L glycyrrhizin group.The cell proliferation inhibited rates were 0,2.73%,14.75 % and 25.96%.The difference was significant (P<0.05).The numbers of Cyclin E positive cells in different concentrations of glycyrrhizin group were significantly different compared with control group,and it was concentration dependently decreased (P<0.01).ConclusionsGlycyrrhizin concentration dependently inhibited rat hepatic stellate cells proliferation and it could reduce the expression of cyclin E and inhibit the proliferation of rat hepatic stellate cells.

Objective To investigate the effects of acupoint massage on labor analgesia efficacy and it's related clinical factors,so to definite the analgesia mechanism and the relationship between the neurotransmitter dopamine and analgesia mechanism.Methods We choosed patients who have been hospitalized in No.1 hospital from March 2009 to September 2009,and divided them into two groups randomly: observation group and control group.Patients in the observation group were treated with acupuncture massage when the production process went into the active phase.Control group indicated that childbirth was naturally without any treatment.We observed the analgesic effect of point massage and the impact of pressure on the uterine contractions.We tested the dopamine level in the blood by fluorescent spectrophotometry before and afte the acupoint massage.We explored the effects of the point massage on the dopamine level in the puerpera.Results The observation group′s pain decreased more than that of the control group.The intensity of contractions in observation group was decreased more obvious than that of the the control group.The serum dopamine levels was significantly lower than that pre-massage(P
4.Detection of Escherichia coli O157:H7 by multiplex PCR-DHPLC

Objective To develop the PCR-denaturing high performance liquid chromatography (DHPLC) for detection of E. Coli O157: H7. Methods The virulence genes of Shiga-like toxin(SLT) and rfbE were specifically amplified by 2 sets of primers. The target gene fragments of the PCR assay were 224 bp and 499 bp, respectively. Results Analysis of 37 strains demonstrated that this PCR system was specific. The detection limit of the PCR was 4 CFU/ml. Conclusion These results indicated that the multiplex PCR-DHPLC assay can be used for specific and sensitive detection of E. Coli O157:H7.
5.Experimental studies of the effect of erythropoietin on fas-associated death domain protein and caslmse-8 protein in rat with intracerebral hemorrhage

Objective To study the protein expressions of Fas-associated death domain protein (FADD) and caspase-8 in rats with intracerebral hemorrhage ,and the effects of erythropoietin tp reveal the mechanism of neu-m-protection by EPO. Methods 126 male SD rats were randomly divided into three groups: Sham-operated group, intracerebral hemorrhage group, and EPO group. Each group was divided into seven subgroups according to the differ-ent time points (3,6,12,24,48,72 h and 7 d). The model of intracerebral hemorrage was established in rats by in-tracerebral injection of autogenous blood. The protein expressions of FADD and caspas-8 in rats tissue around the hemorrhagic and the normal brain tissue were detected by immunohistochemistry. Results The protein expressions of FADD and caspase-8 were increased [(4.66±0.46 ) and ( 15.89±1.81)] at 3 h after intracerebral hemorrhage, and peaked at 48 h [ (35.88±4.24 ) and (45.04±3.99)], the expressions of FADD and caspas-8 in the region around hematoma in EPO group significantly decreased compared with model group[ (3.92±0.64) and (28.24±1.90), (13.32±2.01 ) and (35.08±2.82)] at 3 h and 48 h. Conclusion The protein expressions of FADD and easpase-8 are markedly increased after intracerebral hemorrhage. EPO can protect the neurons by signifi-cantly reducing the expressions of FADD and caspase-8.
6.The dignostic value of serum vitamin B12 in predicting rebleeding in patients with liver cirrhosis of esophagogastric varices

Objective:The aim of this study was to investigate the prognostic value of vitamin B12 as the non-invasive biomarker to predict long-term rebleeding rate in cirrhotic patients with esophagogastric varices.Methods:From Dec 1, 2016 to Dec 31, 2017, cirrhotic patients with esophagogastric varices who had been admitted to Zhongshan Hospital affiliated to Fudan University were enrolled. All these patients received endoscopic treatment to prevent variceal rebleeding. The serum vitamin B12 and folic acid levels were measured in all of them. The receiver operating characteristic (ROC) analysis, Kaplan-Meier analysis, univariate and multivariate cox regression analysis were conducted to explore the value of vitamin B12 in predicting 3-year variceal rebleeding in cirrhotic patients with esophagogastric varices after endoscopic treatment.Results:115 patients were included. The ROC curve analysis indicated that the optimal cutoff value of vitamin B12 for 3-year variceal rebleeding was 567.25 pg/ml. According to the cut-off value, the patients were divided into high-level vitamin B12 group ( n=49) and low-level vitamin B12 group ( n=66). Compared with the low vitamin B12 group, the high vitamin B12 group had lower albumin level, less male (63.3% vs 80.3%), and higher 3-year rebleeding rate ( P<0.05). Cox analysis showed that vitamin B12 and platelet were independent prognostic factors for 3-year rebleeding in patients with variceal bleeding. Conclusions:Elevated peripheral blood vitamin B12 predicts a higher risk of long-term rebleeding in patients with liver cirrhosis and esophagogastric varices.
8.Effect of down-regulation of GAT-1 on analgesic action of oxysophoridine.

OBJECTIVETo study the effect of GABA transporter (GAT-1) on the analgesic action of oxysophoridine (OSR) in the central nervous system of mice.
METHODHot plate test was used to observe and analyze the effect of gamma aminobutyric acid and the inhibitor of GAT-1 (NO-711) on the analgesic action of oxysophoridine. Real time RT-PCR was used to investigate the influence of OSR on the expression of GAT-1 mRNA induced by formalin in spinal cord and brain of mice.
RESULTBoth GABA (2.0 mg x kg(-1), icv) and NO-711(0.125 mg x kg(-1), icv) enhanced the analgesic action of OSR (32.0 mg x kg(-1), iv) in the hot plate test, and the latencies was markedly increased (P < 0.05, P < 0.01). OSR (500.0 mg x kg(-1), iv) significantly inhibited the expression of GAT-1 mRNA induced by formalin (P < 0.05).
CONCLUSIONGAT-1 was involved in the analgesia effect of OSR and the down-regulation of GAT-1 mRNA enhanced the analgesic effect.

9.Extracardial rhabdomyoma:a clinicopathologic analysis of 9 cases

Objective To investigate the clinicopathologic characteristics , differential diagnosis and biological behavior of extracardiac rhabdomyoma.Methods Nine cases of extracardiac rhabdomyoma diagnosed between January of 1997 and July of 2014 were reviewed.The clinical , pathologic and immunohistochemical profiles were evaluated.Results There were 5 males and 4 females at diagnosis with age ranging from 2 years and three months to 59 years (mean, 37.6 years).Sites included the head and neck region (7 cases), chest (1 case ) and vagina wall (1 case).Clinically, most cases manifested as a subcutaneous nodule or as a submucosal polypoid lesion with a mean diameter of 3.2 cm.Histologically , 4 were adult-type rhabdomyoma characterized by tightly packed large round or polygonal rhabdomyoblasts with abundant eosinophilic to clear cytoplasm; 3 were myxoid variant of fetal rhabdomyoma composed of immature myofibrils, spindled and primitive mesenchymal cells embedded in a myxoid background , 1 was an intermediate form of fetal rhabdomyoma consisting of densely arranged differentiated myoblasts with little myxoid stroma;1 was a genital rhabdomyoma composed of elongated or strap-like myoblasts scattered in loose fibrous connective tissue.By immunohistochemistry , they showed diffuse and strong positivity for desmin , MSA and myoglobin with variable expression of myogenin.A case of intermediate type also stained forα-smooth muscle actin.Follow up data ( 2 months ~17 years ) showed local recurrence in one patient 6 months after surgery.Conclusions Rhabdomyoma is a distinctively rare benign mesenchymal tumor showing skeletal muscle differentiation , which may occassionally recur if incompletely excised.Familiarity with its clinical and morphological variants is essential to avoid misdiagnosing this benign lesion as embryonal rhabdomyosarcoma.
10.The effect of maternal anxiety during pregnancy on the social emotional development among toddlers

Objective:
To explore the impact of maternal anxiety during pregnancy on social emotional development of toddlers aged 1-3 year old, so as to provide references for scientific early parenting and early intervention for toddlers with social emotional difficulties.
Methods:
From September 2022 to March 2023, a total of 815 toddlers aged 1-3 who underwent physical examinations and their mothers at Nantong Maternal and Child Health Hospital were enrolled. The Chinese Infant Toddler Social and Emotional Assessment (CITSEA) was used to evaluate the social emotional ability among toddlers. Maternal anxiety evaluated using the Self rating Anxiety Scale (SAS) during prenatal visit was collected.
Results:
The average scores on the externalizing, internalizing, dysregulation and competence domains of the CITSEA were (49.40±9.48,47.42±9.60,48.67± 10.15 , 50.07± 10.20), respectively. Among boys, the score of externalizing domain (50.89±9.45) was higher than that of girls (48.76± 9.50 ), while the score of competence domain (49.22±10.30) was lower than that of girls (51.17±9.84), and the differences were statistically significant( t =2.10, -3.03, P <0.05). The detection rates of abnormalities in the externalizing, internalizing, dysregulation, and competence domains were 7.36%, 7.12%, 7.61%, and 7.24%, respectively. Among them, boys (8.43%,6.32%, 7.96 %,7.49%) and girls (6.19%, 7.99 %,7.22%,6.96%) showed no statistical differences ( χ 2=1.50, 0.85, 0.16, 0.09, P >0.05). There were significant differences in externalizing domain scores(47.77±9.52,49.56±8.95,52.51±9.77) and competence domain scores(51.70±10.38,49.65±10.05,46.68±10.03) among toddlers of different maternal anxiety(normal, mild, moderate to severe) ( F =7.05,7.10, P <0.01). There were significant differences in the abnormal detection rate of externalizing domain (4.81%,7.54%,11.17%) and competence domain(4.81%,6.96%,11.73%)( χ 2=6.60,7.98, P <0.05).
Conclusion
Maternal anxiety during pregnancy has a negative impact on the social emotional development among toddlers. In order to improve social emotional development of toddlers, multidimensional social support and education during pregnancy should be carried out.
