1.Clinical and pathological analysis of seven patients with limb-girdle muscular dystrophy 2B

Objective To analyze the clinical and pathological features of limb-girdle muscular dystrophy2B(LGMD2B).Methods The clinical and pathological features of 7 patients with LGMD2B were analyzed retrospectively.Results Seven patients had a slow onset,and progressive proximal muscle weakness,muscle atrophy,progressive,and incresed serum creatine phosphokinase; muscle biopsy showed different degree of muscle fiber degeneration,necrosis; stromal and inflammatory cell infiltration in muscle fiber; monoclonal antibody immunohistoehemical staining:showed expression of Dysferlin protein was not found in muscle cell membrane,Dystrophin,Sarcoglycans protein expression was normal.Monoclonal antibody immunohistochemical staining the proteins were expressed in normal cell membrane.Conclusions LGMD2B is a slow onset,progressive proximal muscle weakness,muscle atrophy.Histochemical staining on the basis of further immunohistochemical staining,to detect the membrane protein and Dysferlin protein expression,that is a necessary means to diagnose LGMD2B and inflammatory myopathies.
2.Transmembrane Segment Analysis of Transient Receptor Potential Channel

Objective To investigate the membrane topology of transient receptor potential(TRP)channel. Methods Glycosylation method was used to investigate the membrane integrations of each hydrophobic segment of canonical TRP(TRPC5). Results In TRPC5 channel,S4?S8 segments were integrated into membrane with Ncyt/Cexo and Nexo/Ccyt orientations sequentially ,and C?terminus was intracellular. S1?S3 segments were integrated into membrane with two possible types. One was that S1 and S3 were integrated into membrane,whereas S2 was left out of the membrane on the cytosolic side;and the other was a mixed type that S1 and S3 were exposed to cytoplasm respectively. Both of them,the N?termi?nus was intracellular. Conclusion S4?S8 segments of TRPC5 are transmembrane segments. The integrations of S1?S3 segments into membrane need to be further investigated.

4.Effects of individual differences of constructed muscle bridge on neural regeneration

BACKGROUND: Studies on the reparation of peripheral nervous system are not only a key focus in medical research, but also a major topic for biologists, biochemical engineers and materialogy specialists. OBJECTIVE: To investigate the individual difference of ideal muscle bridging neurologic defect configuration on nerve regeneration. DESIGN, TIME AND SETTING: The control experiment was performed at the Shenyang Medical College from June 2004 to September 2005. MATERIALS: Ten common dogs weighing 20-30 kg were used in this study. METHODS: After intravenous anesthesia, muscle strips in the caudal right sartorius and middle musculus flexor digitorum superficialis of dogs were sliced into two segments. One was observed by dissection under a stereoscope. Another received Haematoxylin & Eosin (HE) Staining and transverse area of muscle fiber of distal and proximal ends was analyzed by image analysis. The same muscle strip was sutured at 2 cm neurologic defect region of autologous left and right limbs. Number of medullated nerve fibers was measured after osmic acid staining. MAIN OUTCOME MEASURES: Morphological difference of muscle bridge, transverse area of muscle fiber of distal and proximal ends, and number of medullated nerve fibers at bridging femoral nerve defect of left and right limbs. RESULTS: The muscle fiber in sartorius paralleled the longitudinal axis of muscle strip. The angle between muscle fiber in musculus flexor digitorum superficialis and the longitudinal axis of muscle strip was big. 80% muscle fiber in sartorius was equa1 to the length of the muscle bridge. Muscle fiber in the musculus flexor digitorum superficialis was shorter than the length of muscle strip. Muscle fiber in the musculus flexor digitorum superficialis was thick, whereas muscle fiber in sartorius was thin. 5.Investigation and analysis of dietary status for diabetic patients before admission

Objective To provide scientific evidence for nutrition therapy and health education for patients with type 2 diabetes mellitus (T2DM).Methods Based on body mass index (BMI),75 newly admitted T2DM patients were assigned to the normal body-weight group and the over-weight group.Three days before admission,their dietary status was investigated.The average energy intake was compared with the average standard supply.The energy from protein,fat or carbohydrate and the intake of fiber were compared with the recommendations from the American Diabetes Association (ADA).The intake of other nutrient was compared with the recommended nutrient intakes (RNI) or adequate intakes (AI).Results Compared with the standard supply,the average energy intake of the normal body-weight group was significantly decreased (t =2.61,P < 0.05 ),however the average energy intake of the overweight group was significantly increased (t =3.91,P<0.05).The percentage of energy from protein of the two groups was significantly higher than target levels ( t values were 13.23,21.13 respectively; all P < 0.05 ) ; the percentage of energy from protein of the two groups was significantly higher than target levels (t values were 5.36,10.66 respectively ; all P < 0.05 ) ; however,the percentage of energy from carbohydrate was lower in both groups ( t values were 6.94,15.76 respectively; all P < 0.05 ) ; the average intake of fiber were also lower in both groups (t values were 26.54,40.80 respectively ; all P < 0.05 ).ConclusionThe participants showed insufficient knowledge on healthy diet.Health education could play a role in balanced diet and energy intake.

8.Simultaneous Determination of 4 Components in Compound Paracetamol and Amantadine Hydrochloride Tablet by GC

OBJECTIVE:To establish a method for the simultaneous determination of paracetamol,amantadine hydrochloride, caffeine,chlorphenamine maleate in Compound paracetamol and amantadine hydrochloride tablet. METHODS:GC was performed on the column of HP-5 sillica capillary,temperature programmed,detector was FID detector,with the temperature of 300 ℃,car-rier gas was nitrogen gas,the flow rate is 1.5 mL/min,the split ratio was 20:1 and injection volume was 1μL. RESULTS:The lin-ear range was 156.0-4990.4 μg/mL for paracetamol,125.7-4023.2 μg/mL for amantadine hydrochloride,19.14-612.4 μg/mL for caffeine and 2.515-80.48 μg/mL for chlorphenamine maleate(all r=0.9999);the limits of quantification were 1.4,0.5,1.1,0.9 ng,limits of detection were 0.4,0.2,0.3,0.3 ng;RSDs of precision,stability and reproducibility tests were lower than 2.0%;re-coveries were 99.59%-101.77%(RSD=0.8%,n=9),99.56%-101.80%(RSD=0.7%,n=9),98.44%-100.83%(RSD=0.7%,n=9) and 100.05%-101.91%(RSD=0.6%,n=9),respectively. CONCLUSIONS:This method is simple,rapid,accurate and reli-able,and suitable for the simultaneous determination of paracetamol,amantadine hydrochloride,caffeine,chlorphenamine maleate in Compound paracetamol and amantadine hydrochloride tablet.
9.Effect of lycopene on immunity in rats with acute lung injury

Objective: To investigate the effects of lycopene on T lymphocyte subpopulations and pulmonary alveolar macrophagic (PAM) functions in rats with acute lung injury (ALI). Methods: Rats were randomly divided into the following groups. (1) Control group, (2) ALI model group, (3) Low dose group, (4) Mid dose group and (5) High dose group. Control group and ALI model group were treated with solvent of lycopene, and the other groups were gastrically incubated with lycopene. Thirty-five days later, control group were given physiological saline, ALI model group and lycopene administrated groups were injected with lipopolysaccharide (LPS) (6.0 mg/kg) to induce ALI. One hour, four hours or six hours after LPS or physiological saline challenged, abdominal aorta blood for measuring lymphocyte subpopulations and bronchoalveolar lavage fluid for measuring function of PAM were gathered respectively. Results: (1) At h 1, the percentages of CD3+,CD4+ and CD8+ of lycopene administrated groups compared with control group were not significantly different. At h 4, the percentage of CD4+ was similar to that at h 1. As for the percentages of CD3+, except high dose group [(28.8?9.9)%] was significantly lower, low dose, mid dose and ALI model group showed no significant difference compared with control group[(39.5?4.5)%]. The percentages of CD8+ of ALI model and lycopene administrated rats, separately (10.2?3.9)%,(10.3?2.8)%,(9.8?2.8)%,(10.1?3.5)%, had been significantly reduced compared with control group[(15.1?2.5)%]; between ALI model and lycopene ad-ministrated groups there was no significant difference. The instance at h 6 was the same as that at h 4. The percentage ratios of CD4+ T-lymphocyte to CD8+ T-lymphocyte of ALI model rats were not significantly different compared with control group or lycopene administrated groups at h 1 and h 6. At h 4, the ratio of the CD4 + and CD8 + in Low dose and Mid dose groups had significant difference and ALI model, high dose hadn’t when they were compared with control group. Objective To analyze the PCR and molecular characters of the first serogroup W135 meningo-coccal death case in Shanxi province on April,2013.Methods Epidemiological survey of suspected epidemic cere-brospinal meningitis case was conducted,blood serum and petechia tissue fluid samples were identified by PCR for crgA gene and siaD gene of W 135.Multilocus sequence typing(MLST)was performed for determining the sequence types(STs).Results The patient in the case died of serogroup W135 Neisseria meningitides,which belonged to ST-11.Conclusion This is the first case died of serogroup W135 Neisseria meningitidis in Shanxi province,which prompts that the surveillance of meningococcal pathogeny should be strengthened.
