1.Analysis of the death cause of children less than 5 years old in Huishan district of Wuxi

Objective To analyze the death cause of children less than 5 years old in Huishan district of Wuxi,in order to reduce the mortality of children under 5 years of age and put forward effective intervention measures.Methods According to “monitoring project of children below 5 years old died in Jiangsu Province”,the death surveillance data of this area of children below 5 years old were analyzed.Results The mortality of children under 5 years of age in Huishan district from 2001 to 2010 was 6.87‰,infant mortality was 5.06‰,the neonatal mortality was 3.75‰ and early neonatal mortality rate was 2.66‰.Death cause of children under 5 years old,the 5 cis-position were accident,premature and low birth weight,congenital malformation,congenital heart disease,birth asphyxia.Conclusion The death of children under 5 years old in Huishan area decreased,accidental injury,premature and low birth weight were the main causes of death.To reduce the mortality of children under 5 years old,should strengthen the safety education,reduce the accident,vigorously promote the pre-marital medical examination,strengthen health care and supervision in pregnancy,reduce the birth defect,improve obstetric,pediatric medical quality and medical technology.
2.Correlation between fatty acid receptor GPR120 and glucose transporter 4

Objective:To investigate the correlation between G protein-coupled receptor 120 ( GPR120 ) and glucose transporter 4 (GLUT4) in 3T3-L1 cells.Methods:3T3-l1 cells were induced for differentiation,GRP120 mRNA was detected by RT-PCR and Oil red O was used to determine fat expression.GPR120 expression was knocked down by a specific siRNA in 3T3-L1cells.3T3-L1 cell that were tranfected with GPR120 siRNA were incubated with palmitic acid for 24 hours.Then,real-time PCR and Western blot were used to detect the expression of GLUT4 mRNA and protein.Results: Induced differentiation led to GPR120 overexpression in 3T3-L1 cells ( P<0.05 ).GPR120 knockdown resulted in reduction of both lipid volume and number in 3T3-L1 cells.Furthermore,GPR120 knockdown decreased GLUT4 mRNA and protein levels in 3T3-L1 cells (P<0.05,respectively).Conclusion:GPR120 affects the expression level of GLUT4 in insulin signaling pathway,suggesting its involvement in the initiation of insulin resist-ance.
3.Preliminary study on the biological characters of Ornidazole Slow-Release Membrane

Objective To evaluate the biocompatibility and antibacterial activities of the Ornidazole Slow-Release Membrane.Methods 1.The lower lips of 12 rats were sewed into 12 pockets and the pockets were immited with extracting solution of the ornidazole membrane, formaldehyde and normal saline respectively once per day.The specimens were examined histologically 7 days later.2.The dorsal muscles of 16 rats were implanted with the membranes or silk threads,and examined histologically 1 week and 2,4,6 weeks later respectively.3. The antibacterial activity against Streptococcus mutans and Fusobacterium nucleatum was observed on solid culture medium in vitro.Results The animal experiments showed the membranes were not irritative to the oral mucosa.It was found that the tissue reaction of the membranes was similar to that of the silk threads after implanted into dorsal muscles and the membranes had been degraded in the second week.And the membranes had effective antibacterial action against Streptococcus mutans and Fusobacterium nucleatum.Conclusion The Ornidazole Slow-Release Membrane possesses favorable biocompatibility and antibacterial activities.
4.Hydrogen attenuates the inhibition of P.g-LPS on osteogenic capacity of huma periodental ligament cells

Objective:To observe the effects of hydrogen on osteogenic capacity of human periodontal ligament cells(hPDLCs)stim-ulated with P.g-LPS.Methods:hPDLCs were cultured and divided into 4 groups:control(C)group,osteogenic induction(OI) group,OI +1 00 ng/ml LPS(OILPS)group and OIPLS +3%H2 (H2 OIPLS)group,and treated respectively.Alizarin red staining (ARS)was carried out 3 weeks after treatment.ALP and OC mRNA expression of the cells was examined by RT-PCR after 7-d treat-ment.Results:LPS decreased A value of ARS(P <0.01 ),ALP mRNA expression(P <0.001 )and OC mRNA expression(P <0.001 )of the cells.H2 increased the A value(P <0.05),ALP mRNA expression(P <0.01 )and OC mRNA(P <0.01 )of the cells treated by LPS.Conclusion:High concentration of P.g-LPS can inhibit osteogenic capacity of hPDLCs,while hydrogen can impair the P.g-LPS induced suppression of hPDLC's osteogenesis.
5.Determination of emodin and chrysophanol in Fuyanling Effervescent Tablets by HPLC

AIM: To develop a method to determine the content of emodin and chrysophanol in Fuyanling Effervescent Tablets(Radix et Rhizoma Rhei, Flos Lonicerae, Radix Sophorae Tonkinensis, etc.). METHODS: HPLC was used to determine the content of emodin and chrysophanol in Fuyanling Effervescent Tablets. The separation was performed on YWG ODS column with methanol (0.1%) phosphoric acid(85∶15) mixture as a mobile phase and the wavelength of UV detector was at 254nm. RESULTS: The resolution and the linearity of this method was good. in the range of emodin was 50.2~401.6ng( r =0.9999); chrysophanol was 49.9~ 399.2 ng( r =0.9994); and with the average recovery of emodin: 99.4%( RSD =2.2%); chrysophanol:100%( RSD =1.1%). CONCLUSION: The method is simple, rapid and satisfactory and suitable for quality control of Fuyanling Effervescent Tablets.
6.Outcomes and obstacles of “treatment before payment” reform as viewed by medical managers

Objective To analyze implementation outcomes and obstacles of “treatment before payment” reform seen by medical institutions' managers,along with an optimal strategy proposed.Methods Quantitative survey and qualitative interviews were called into play,and the data so acquired are subject to evaluation of the radar chart and fishbone diagram analysis to learn the implementation outcomes and obstacles.Results The reform proves workable at hospitals of county and lower levels,achieving win-win for the people,hospitals and government.Obstacles found include poor allocation of designated funds for risk coverage,insufficient circulating funds in advance,poor risk aversion mechanism,and poor preparation for tertiary hospitals.Conclusion It is recommended to promote awareness of the reform,enhance inter-department cooperation,improve hospital information systems,clarify the screening standards of “purposely evasion offenders”,standardize the issuance and use of the NRCMS certificates,and better build the credit system of the society.
7.Association between nitrotyrosine and coronary heart disease in type 2 diabetes

Objective To investigate the association of nitrotyrosine with coronary heart disease (CHD) in type 2 diabetes mellitus.Methods The nitrotyrosine levels were determined in 109 patients of type 2 diabetes mellitus without CHD (T2DM).One hundred and fifty-two patients of type 2 diabetes mellitus with CHD (T2DM-CHD) and 103 healthy control subjects by ELISA.Results T2DM-CHD patients had significantly increased nitrotyrosine compared with T2DM group and the control group [ ( 78.17±10.68 )nmol/L,(70.50 ± 9.13) nmol/L vs ( 63.23 ± 11.55 ) nmol/L,Ps < 0.01 ].Nitrotyrosine was correlated with total cholesterol,triglyceride,fasting glucose and Gensini Score (r=0.361,P=0.009;r =0.206,P=0.001 ;r=0.347,P=0.026; r=0.466,P < 0.001 ).Multivariable logistic regression showed nitrotyrosine was independently associated with CHD combined with type 2 diabetes mellitus ( OR=1.094,95% CI:1.053-1.137 ; P < 0.01 ).Conclusion Nitrotyrosine plays an important role in the formation and development of cardiovascular disease in tvoe 2 diabetes.
8.Preparation of tissue-engineered esophageal scaffolds using electrospinning technology

BACKGROUND:We have found that oriented fibers can guide the alignment of smooth muscle cells in our previous experiments. Thus, we designed the experiment to prepare wel aligned polymeric fibers using electrospinning technology, aiming at guiding the growth of esophageal smooth muscle cells to maintain cellmorphology and biological function. OBJECTIVE:Using electrospinning technology, to fabricate isotropic and directed nano-fibrous scaffolds made of polycaprolacton, gelatin and silk fibroin. METHODS:Polycaprolacton/silk fibroin fibers at a ratio of 4:1 were prepared with proper parameters, including solution concentration, voltage and injection speed, under the self-made spinning system. The polycaprolacton/gelatin sheets with mass ratio of 2:1, 1:1 and 1:2, respectively, were also fabricated under suitable process parameters. Using the rol er col ector instead of the metal plate, polycaprolacton/gelatin nano-fibrous scaffold with good alignment of fibers was manufactured. RESULTS AND CONCLUSION:The isotropic polycaprolacton/silk fibroin scaffold with fiber diameter of (535.9±126.7) nm was prepared under conditions of solution concentration (0.08 g/mL), injection speed (1.6 mL/h) and voltage (22.5 kV), and these fibers were uniform with no beads. The isotropic polycaprolacton/gelatin scaffold with fiber diameter of (257.9±117.8) nm was prepared under conditions of solution concentration (0.10 g/mL), injection speed (0.8 mL/h) and voltage (22.5 kV). Using the rol er col ector instead of the previous metal plate, polycaprolacton/gelatin (w:w, 1:2) nano-fibrous scaffold with good alignment of fibers was manufactured. The process parameters were 3 000 r/min of rol ing speed, 0.8 mL/h of injection speed and 15 kV of voltage.
9.Investigation on undergraduates′cognition,attitude and behavior about low-carbon life

Objective To investigate the undergraduates′cognition ,attitude and behavior about low‐carbon life and then to provide a relatively scientifically strategy .and provide scientific basis to formulate countermeasures about undergraduates′low‐car‐bon lifestyle .Methods By multistage sampling ,382 students′knowledge ,attitude and practice about low‐carbon life in Wuhan Uni‐versity of science and technology were analyzed .Results The students′cognition about low‐carbon life was good ,the awareness rate of girl was higher than that of boy ;Their attitude was positive ,grade and knowledge awareness were the two influence factors ;the behavior situation was unsatisfactory ,the origin of students and attitude enthusiasm were the elements affect rationality of be‐havior .Conclusion Undergraduates should strengthen the cognition of low‐carbon life ,improve attitude enthusiasm ,and form a good behavioral habit of low‐carbon life .
10.Peripheral blood biological markers for early screening in children with autism spectrum disorder

Objective:
To investigate the specifity of amyloid precursor protein(APP), brain derived neurotrophic factor (BDNF)and gamma aminobutyric acid(GABA) in peripheral blood in children with autism spectrum disorder, so as to explore the biomarkers for early screening of ASD and its relationship with the severity of ASD.
Methods:
A total of 41 children diagnosed with autism from January to December 2019 were enrolled in the ASD group. Meanwhile, 41 healthy children with normal growth and development who were examined in the same period were selected as control group. And the sera total sAPP, sAPP α, sAPP β, BDNF and GABA of all participants were tested by sensitive enzyme linked immunosorbent assay method, and were compared between the two groups.
Results:
The serum sAPP level in ASD group(2 132.98±333.28 ng/mL) was higher than control group(1 734.76 ±357.97 ng/mL),the serum sAPP α level(335.11±33.87 pg/mL) was higher than control group(274.84±32.12 pg/mL) and the serum GABA level(4.17±0.95 μmol/L)was lower than control group(6.35±0.84 μmol/L). GABA level (4.17±0.95 μmol/L) was lower than that of control group (6.35±0.84 μmol/L), the differences were statistically significant ( t =3.92, 4.25, -7.27, P < 0.05 ). In addition, the serum GABA level in children with severe ASD (3.48±0.77 μmol/L)was lower than children with mild to moderate (4.94±0.98 μmol/L).The difference was significant ( t =-3.31, P <0.05). ROC curve showed that total sAPP( AUC = 0.77 ,95% CI =0.66-0.87), sAPP α( AUC =0.77,95%CI=0.67-0.87), and GABA ( AUC =0.95,95% CI =0.90-0.93)had diagnostic efficacy for ASD( P <0.05), among which the AUC of GABA was the largest (0.95)and its sensitivity(85.65%) and specificity(80.76%) were the highest. The results of binary Logistic regression showed that the abnormal expression of sAPPα ( OR =1.04,95% CI = 1.00- 1.07) and GABA( OR =0.02,95% CI =0.00-0.32) were associated with risk for ASD( P <0.05).
Conclusion
Considering the specific change of total sAPP, sAPPα andGABA in peripheral blood in ASD children, total sAPP, sAPP α and GABA can be considered as promising biomarkers in the early diagnosis of ASD, among which GABA has the highest sensitivity and specificity.
