Objective To investigate the effects of regional infusion of hypothermic normal saline contained captopril on free radicals level and myeloperoxidase(MPO) activity in rabbits during spinal cord ischemia-reperfusion(I/R).Methods The 80 rabbits were randomly divided into four equal groups,group A(sham operated,no I-R injury,n=8),group B(control,only I/R,n=24),group C(I/R + intraaortic hypothermic normal saline,n=24),and group D(I/R + N.S plus 4mg/kg captopril,n=24).The model of spinal cord ischemia in the rabbit was induced with the aorta clamped.Hypothermic solution was infused into the isolated aortic segments.The content of spinal cord MDA and the activities of superoxide dismutase(SOD),MPO were measured at 8h,24h and 72 h after the reperfusion respectively.Results The level of spinal cord MDA increased,the activity of SOD decreased and the activity of MPO increased markedly after the reperfusion.Compared with group B,the MDA content decreased significantly at the three observation time points and the MPO activity decreased significantly in groups C and D(all P
2.A discussion of the combined application value of FMEA and RCA in the IV infusion risk management

Objective:To explore the combined application effect of failure mode and effect analysis (FMEA) and root cause analysis (RCA) in IV infusion risk management of pediatric outpatient so as to enhance the quality of care and ensure the safety of outpatients.Methods: For this exploration, a special analysis team was established and the flowchart was drawn up, and the potential failure modes were listed by using FMEA. And the RCA was used to analyze and evaluate the key mode in these potential failure modes so as to find the root cause might affect the safety of IV infusion, and then the improved measures were implemented and the effects of new measurement were evaluated. Results:After implemented the improved measures, the risk of top 4 risk priority number (RPN) in intravenous infusion flow decreased obviously. For the transfusion management, patients' satisfaction increased from 78.09% to 95.03% (P<0.05), and nurses' satisfaction increased from 83.79% to 97.80%(P<0.05).Conclusion: The combined application of FMEA and RCA in the IV infusion risk management of pediatric outpatient can effectively reduce the occurrence of risk events, reduce nursing complaints and disputes, and enhance the quality of nursing care.
3.Protective effect of regional captopril infusion on ischemia-reperfusion injury of spinal cord

Objective To investigate the effect of regional captopril infusion on ischemia-reperfusion (I-R) injury of spinal cord in rabbits. Methods 32 rabbits were divided randomly into four groups, the group A (sham operated, no I-R injury), the group B (control group, only with I-R injury), the group C (I-R injury + intra-aortic infusion of hypothermic normal saline), and the group D (I-R injury + infusion of normal saline plus 4mg/kg captopril). Spinal cord ischemia was induced by clamping the aorta both below the left renal artery and above the aortic bifurcation. Hemodynamic data were measured in every group. The spinal cord function was assessed 24, 48 and 96 hours after aortic declamping according to Tarlov's Scale. Spinal cords were harvested for histological analysis. Results There were no significant differences in blood pressure and heart rate among four groups. The neurological status in the groups C and D was significantly superior to that of the group B (P<0.01). In the group B, all animals were paraplegic with Tarlov's Scale of 0 or 1. Three of 8 animals in the group C were paraplegic with Tarlov's Scale of 0 or 1, and only one animal in the group D. Compared to the group B, there were more normal neurons in the anterior horn of spinal cords in the groups C and D (P<0.01). Conclusion Regional infusion of hypothermic normal saline contained captopril can provide sufficient spinal cord protection against ischemia.

5.Electrochemistry and Electrocatalysis of Electropolymerized Vitamin B1 Film Electrode

A polymer film of vitamin B1 was obtained at a graphite carbon electrode by cyclic voltammetric method in phosphate buffer solution (PBS). Cyclic voltammograms of the film electrode in PBS (pH 5) exhibited one pair redox waves, Epa=350 mV,Epc=325 mV,different from monomer vitamin B1. The peak currents of poly-VB1 films was proportional to (scan rate)1/2, which showed that the charge transfer was controlled by diffusion in the polymer film. The Poly-VB1 modified electrode showed a good catalytic ability to dopamine and epinephrine. At the modified electrod, the peak potential for oxidation of dopamine occurred at 250 mV and 200 mV for epinephrine, while they were shifted towards the negative direction 100 mV and 300 mV respectively at the bare glassy carbon electrode . And their peak current had a linear relationship to the concentrations in the range of 8.0×10-7~1.0×10-4 mol/L for dopamine and 1.0×10-6~1.0×10-4 mol/L for epinephrine.
6.Summary of the 2008 national autoantibody testing quality control

Objective To consecutively understand the national clinical testing status and to reassure the quality-control of autoantibody detection. Methods Letter or telephone notification were conducted to the participating hospitals or departments. Autoantibodies for quality control survey included anti-nuclear anti-body (ANA), anti-double-stranded DNA (anti-dsDNA), anti-extractable nuclear antigens (A-ENA), anti-mitochondria antibody (AMA)/anti-smooth muscle antibody (ASMA), and anti-citrulline antibody (anti-CCP). Each had 3 control samples, and altogether 15 samples for testing. Sample distribution and data analysis were double-blinded. Results One hundred and two hospitals/departments participated in the national quality-control survey. The accurate rate for this survey was 70%, 88%, 93%, 85%, 79% respectively for ANA, anti-dsDNA, AMA, ASMA and anti-CCP. Anti-ENAs were further divided into anti-RNP, Sm, SSA, SSB and Scl-70 subgroups, and the accurate rate was 88%, 77%, 92%, 93% and 87% respectively. Conclu-sion Compared to the previous 4 national surveys, the accurate rates of ANA, anti-dsDNA, anti-ENAs, AMA in our country's autoantibody testing is improved, and the detection rate of ASMA and anti-CCP antibody is also increased. More hospitals and testing items can test these autoantibodies, which implies that autoantibody testing status has been improving in our country.
7.Expression and clinical significance of long chain non-coding RNA LOC285194 in human breast cancer tissue

Objective To investigate the expression of long chain non-coding(lnc) RNA LOC285194 in breast cancer tissue and paracancerous tissue and its clinical significance.Methods Forty-two samples of paraffin embedded breast cancer tissue and 16 samples of paraffin embedded paracancerous tissue were selected.The expression of lncRNA LOC285194 in these tissue were detected by using quantitative real-time polymerase chain reaction(PCR).Then its correlation with clinicopathological features was analyzed.Results The expression level of lncRNA LOC285194 in breast cancer tissue was significantly lower that in the paracancerous tissue (P<0.01);the level of lncRNA LOC285194 in human epidermal growth factor receptor-2(HER2)overexpression tissues was up-regulated compared with HER2 negative breast cancer tissue(P =0.013),there was a positive correlation between them(r=0.385,P=0.012).Conclusion lncRNA LOC285194 may play the role of cancer suppressor gene and may be involved in the generation of breast cancer by HER2 association,which may become a target gene of breast cancer treatment.
8.Knowledge and attitude of nursing interns towards needle stick injury be-fore clinical practice

Objective To investigate nursing interns’knowledge and attitude towards needle stick injury before clinical practice.Methods In 20-23 June,2015,nursing interns who were about to start clinical practice in a hospi-tal were investigated,nursing interns’knowledge and attitude towards needle stick injury were surveyed through questionnaire.Results A total of 350 questionnaires were distributed,324 (92.57%)responded questionnaires were available.40(12.35%)questionnaires were responded by male interns,and 284(87.65%)were by female in-terns;34(10.49%)interns had bachelor degree;the mean age of interns were (20.83 + 1 .24)years old.The correct answer rates about questions related to injury occurring during needle recapping and wearing gloves were low (about 60%).About 70% of the interns gave the correct answers to questions about hepatitis B infection due to needle stick injury following hepatitis B vaccination,as well as medication after injury.Score for individual question about attitude towards needle stick injury was ≤3,the major related problems were susceptibility of blood borne diseases and recapping needles.Conclusion Needle stick injury-related knowledge and attitude among nursing interns is inadequate,including recapping needles,timely report,susceptible to infectious diseases,and so on.It is necessary for schools and teaching hospitals to strengthen the education about occupational protection among nursing interns, so as to improve the attitude and ability of professional protection.
9.The endoplasmic reticulum stress pathway involving protein kinase R-like ER kinase-activating transcription factor 4-CCAAT/enhancer binding protein homologous protein implicated in apoptosis in lungs of rats with bronchopulmonary dysplasis

Objective To investigate the role of endoplasmic reticulum stress (ERS) pathway involving protein kinase R-like ER kinase (PERK)-activating transcription factor 4 (ATF4)-CCAAT/enhancer binding protein homologous protein (CHOP) in apoptosis in lungs of rats with bronchopulmonary dysplasis (BPD).Methods Forty eight premature SD rats were divided into BPD group and control group according to random number table.Rats in BPD group were continually exposed to O2 with volumetric concentration factor of 850 mL/L,while rats in control group were exposed to air.Lung tissues in each group were obtained in 7,14 and 21 days respectively.The apoptosis in lung cells was evaluated by terminal dexynucleotifyl transferase-mediated dUTP nick end labeling (TUNEL) assay.The mRNA levels of glucose regulated protein 78 (GRP78),PERK,ATF4 and CHOP were detected by real-time quantitative reverse transcription-polymerase chain reaction (RT-PCR).The protein levels of GRP78,phosphorylated PERK (pho-PERK),ATF4 and CHOP were detected by using Western blot.Results Compared with control group,the lung cells of the rats in BPD group developed more serious apoptosis.Furthermore,the apoptosis index (AI) in lung cells of the rats increased rapidly with the hyperoxia exposure time.This had been statistically verified by comparison with the control group at different timing(7 d:15.50 ± 0.58 vs 1.25 ± 0.50,14 d:27.75 ± 1.71 vs 3.25 ± 0.96,21 d:50.50 ±3.70 vs 4.00 ± 1.15 ;t =57.00,20.58,25.16,all P <0.01).The mRNA levels of GRP78,PERK,ATF4 and CHOP in BPD group increased significantly compared to the control group [GRP78:7 d (33.88 ± 3.73) vs (11.65 ± 1.00),14 d (54.50 ±2.18)vs(12.84 ± 1.41),21 d (95.34 ± 7.61)vs(12.43 ±0.59) ;PERK:7 d (5.23 ±0.92)vs (1.45 ±0.46),14 d (7.60 ± 1.56)vs(2.18 ±0.97),21 d (16.55 ±0.50)vs(2.90 ± 1.18) ;ATF4:7 d (23.04 ± 2.45)vs(12.56 ±2.81),14 d (28.66 ±2.66)vs(15.18 ±2.92),21 d (36.63 ±2.99)vs(15.14 ±2.09) ;CHOP:7 d (2.21 ±0.19)vs(0.81 ±0.02),14 d (4.19 ±0.17)vs(0.90 ±0.08),21 d (6.08 ±0.38)vs(0.88 ±0.10) ;all P < 0.05].The protein levels of GRP78,pho-PERK,ATF4 and CHOP in BPD group increased significantly as well [GRP78:7 d (1.33 ±0.03)vs(0.85 ±0.04),14 d (1.31 ±0.02)vs(0.92 ±0.01),21 d (1.82 ±0.28)vs(0.87 ± 0.01);pho-PERK:7 d (0.68±0.02)vs(0.54±0.01),14 d (1.04±0.01)vs(0.65±0.01),21 d (1.29± 0.02)vs(0.73 ±0.01) ;ATF4:7 d (1.26 ±0.01) vs(0.83 ±0.01),14 d (1.39 ±0.02) vs (0.87 ±0.02),21 d (1.67 ±0.02)vs(0.94 ±0.02) ;CHOP:7 d (1.37 ±0.01)vs(0.47 ±0.06),14 d (1.50 ±0.04)vs(0.74 ±0.05),21 d (1.61 ± 0.03) vs (0.55 ± 0.02) ; all P < 0.05].Positive correlation was demonstrated between the expression levels of CHOP protein and AI,PERK,ATF4 in the BPD group (r =0.87,0,92,0.93 respectively,all P < 0.05).Conclusion PERK-ATF4-CHOP mediated ERS may participate in and contribute to the apoptosis mechanism in lungs of rats with BPD.
10.Effect of Erythropoietin on Expression of Nuclear Factor-κB in Rabbit Spinal Cord Tissue after Ischemia-Reperfusion

Objective To explore the effects of erythropoietin (EPO) on the neural function recovery and the expressions of nuclear factor-κB (NF-κB) in rabbit spinal cord after ischemia-reperfusion injury. Methods 24 rabbits were randomized into 3 groups: EPO treatment group, ischemia-reperfusion (I/R) group and sham group with 8 rabbits in each group. The rabbit model of spinal cord ischemia-reperfusion injury was established with Tetik's method. The changes of neural functional recovery of rabbits of 3 groups were observed through Tarlov's scale at different time points post-operation. The expressions of NF-κB were tested with immunohistochemistry. Results The grade of nerve function improved distinctly in EPO treatment group (P<0.01).The expression of NF-κB in EPO group was lower than that of I/R group (P<0.01).Conclusion EPO can significantly improve the motor function of hind limbs in rabbits after spinal cord ischemia-reperfusion injury, which might be related with inhibition of the expressions of NF-κB.
