1.Determination of sinomenine and tanshinnone Ⅱ_A of Shuanghuoqufengzhentong Wine by HPLC

AIM: A determination was created of sinomenine and tanshinnone Ⅱ A in Shuanghuoqufengzhentong Wine (Caulis Sinomenii, Radix Salviae Miltiorrhizae, etc.) at the same time by HPLC. METHODS: The method contained a Waters ? PORASIL TM P/N27477 column with methanol water ethylene diamine (90∶10∶0.125) as mobile phase, UV detection wavelength was set up at 262nm. RESULTS: The average recovery of sinomenine was 97.9%, the RSD was 2.83%. The average recovery of Tanshinnone Ⅱ A was 99.0%, The RSD was 1.96%. CONCLUSION: The method is rapid, accurate, sensitive. This study provides a testing method for the quality control of similar products.
2.The early postoperative fluid management experience in infant living-donor liver transplantation

Objective To discuss the effective early postoperative fluid management in infant living-donor liver transplantation. Methods From January 2008 to March 2009, 17 cases of infant living-donor liver transplantation were carried out. According to infant postoperative physiological and hemodynamic characteristics, rehydration was properly controlled on the surgery day, the negative fluid balance was achieved as soon as possible in three days. The infant condition changes, vital signs and urine output were closely monitored. At the same time, the nature and volume of fluid infusion was timely adjusted to maintain the infant in a stable environment in accordance with the laboratory tests. Results The early postopera-donortive hemodynamic stability was effectively maintained in 17 cases of infants, no case appeared with the capacity-related complications. Conclusions It is the key to reduce postoperative complications and mortality with effective circulating blood volume and hemodynamic stability and the negative fluid balance state in the postop-erative 3 days.
3.The characteristic of gene polymorphism of Kell blood group in Zhuang population from Guangxi region in China

Objective To investigate gene polymorphism of Kell blood group in different Zhuang population from Guangxi region .Methods The genotypes of Kell blood group of 1025 non‐related individuals in different areas of Guangxi Zhuang popula‐tion were analyzed by PCR‐SSP .Results The Kell antigen in all individuals was homozygous ,the gene frequency of K and Jsa was 0 ,while that of k and Jsb was 1 .000 .Conclusion The distribution characteristic of Kell blood group in Guangxi Zhuang population was monomorphism ,which was similar to other Chinese population reported by literatures .
4.Observation and analysis of STR loci mutation among 1 786 cases of paternity test in Guangxi area

Objective To observe and analyze the mutation characteristics of 17 STR loci among the paternity test cases in Guangxi area .Methods Among 1 786 cases of non—exclusion parentage ,1 430 cases were parental triplet and 356 cases were uniparental diad ,1 001 persons were Han people ,2 102 persons were Zhuang people and 113 persons were other ethnic group in the parents .The genome DNA was extracted by Chelex-100 method .17 short tandem repeat (STR) loci were detected by Power Plex ? 18D System Kit .The paternity testing containing mutant STR loci were screened out from 1786 cases .The locus-specific ,specificity of paternal and maternal ,and allele-specific mutation rates were observed and analyzed ,respectively .The characteristics of the muta-tions were studied .Results In total ,75 mutations events were observed at 16 of the 17 loci .Among them ,73 (97 .34% ) times were one step mutation ,onece(1 .33% ) was two—step mutation ,and once(1 .33% ) was three—step mutation ,no mutation was found at the TPOX locus .The mutation rates ranged 0 .031 1% —0 .404 2% ,and the mean mutation rate was 0 .145 8% .The proportion of the paternal mutations and the maternal mutations was 5 .4:1 .0 ,the difference had statistical significance(P<0 .01) .and the mu-tation difference between Han people and Zhuang people had no statistical significance(P>0 .05) .Conclusion STR loci mutation is common phenomenon in paternity test .The data of STR loci mutations should be constantly accumulated for selecting the genetic characteristics in line with the Guangxi population and the genetic markers of STR loci with high identification ability to ensure ac-curate and reliable identification results .
5.Investigation and analysis on ABO and Rh blood groups distribution among Zhuang polulation in Nanning area

Objective To investigate the distribution situation of ABO and Rh blood groups among Zhuang population in Nanning area to guide the scientific voluntary blood donor recruitment and rational storage of blood in this area.Methods 2 052 blood samples from Zhuang population in Nanning area were performed the identification of ABO and Rh blood groups.Whether the gene distributions conforming to the Hardy-Weinberg equilibrium principle was assessed by using the Chi-square test.The data was compared with the distribution characteristics in other areas or other minorities.Results The distribution characteristics of ABO blood groups in Nanning area were O > B> A > AB.Blood group O was highest (46.78%)and blood group AB was lowest (4.34%).The gene frequencies of A,B and O were 0.131 9,0.181 5 and 0.686 6,respectively.The blood groups distribution con-formed to the Hardy-Weinberg equilibrium principle;the RhD positive proportion(RhD+ )in Zhuang population was 99.90%,while the RhD negative (RhD- )proportion was 0.10%,which was lower than that in Han population.No phenotypes of ccdE,CcdE, CCdee,CCdE,CCDEE and ccDee were observed in this study.Conclusion The distribution of the ABO and Rh blood groups among Zhuang population in Nanning still maintains their own exclusive characteristics.The extremely low proportion of RhD- will bring about serious difficulties to the patients with long term blood transfusion.
6.Influence of Exterior-interior Meridian Acupoint Selection on Hippocampal Cell Apoptosis and Expression of c-jun N-terminal Kinase in Rats with Ischemia-reperfusion Injury

Objective To observe the influence of exterior-interior meridian acupoint selection on hippocampal cell apoptosis and c-Jun N-terminal kinase ( JNK) in ischemia-reperfusion injury rats, and to explore the possible protective mechanism for ischemia-reperfusion injury. Methods One hundred and twenty SD rats were randomly divided into sham-operation group (SH), model group (I/R), electroacupuncture (EA) group and JNK inhibitor SP600125 ( SP) group. The ischemia-reperfusion model was established by occlusion of middle cerebral artery with intraluminal thread. EA group was given EA on acupoints of Zusanli ( ST 36) , Sanyinjiao ( SP 6) , Chize ( LU 5) , Hegu ( LI 4) , once a day, based on the method of exterior-interior meridian acupoint selection. We observed the neurological behavioral changes by Longa neurological function standard, detected the activation of phosphorylated JNK ( p-JNK) by immunohistochemical method and examined the hippocampal apoptotic index by TUNEL method. Results The neurologic impairment scores of EA group and SP group were lower than I/R group (P<0.05) . Apoptotic index of I/R group was higher than SH group (P<0.01) and that of EA group and SP group was lower than I/R group (P <0.05) . The level of p-JNK in I/R group was higher than SH group ( P <0.01) and that of EA group and SP group was lower than I/R group ( P <0.05). Conclusion Exterior-interior meridian acupoint selection therapy has certain effect on alleviating neurologic impairment and reducing apoptosis in ischemia- reperfusion injury rats, and the mechanism might be associated with the inhibition of the activation of JNK signaling pathway.
7.IgA deficiency allergic transfusion reaction and its prevention and treatment strategies

IgA deficiency allergic transfusion reaction often occurs in patients with anti-IgA. When blood products containing IgA are transfused into these patients, it may lead to severe allergic transfusion reactions, which can be fatal in severe cases. This review summarizes the clinical manifestations and influencing factors of immunoglobulin A deficiency (IgAD), as well as the pathogenesis, laboratory test and prevention strategies of allergic transfusion reactions caused by IgA deficiency, so as to enhance prevention awareness of IgA deficiency allergic transfusion reaction in medical staff.
8.Advances of pathogen inactivation technology in blood components and quality evaluation

Through the screening of blood donors, detection of pathogen antigen and antibody, full coverage of nucleic acid detection, the risk of infectious blood transfusion has been reduced to a very low level. Especially, pathogen inactivation technology (PRT) has played an irreplaceable role in ensuring blood safety. The best way to inactivate pathogens is not only to effectively remove the target pathogens in the blood, but also to maintain the activity of active ingredients in the blood to the maximum extent, and it doesn′t affect the effect of blood therapy. In this paper, the development of pathogen inactivation technology is summarized, and the influence of pathogen inactivation treatment on the quality of blood components is discussed. It provides references for improving or developing new processing technology.
9.Blood security and management of transfusion-dependent thalassaemia (TDT) patients in Nanning: analysis of " collection-based-supply" blood management mode

【Objective】 To retrospectively analyze the blood use of transfusion-dependent thalassemia (TDT) patients in 9 designated transfusion medical institutions from 2018 to 2023 in Nanning, and to evaluate the effect of " three designated " blood transfusion mode (hereby means TDT patients undergoing blood transfusion in designated transfusion medical institutions regularly) and " collection-based-supply" blood management mode on blood security of TDT patients. 【Methods】 The " three designated" blood transfusion mode was implemented to ensure that TDT patients registered in the local household registration (referred to as the " register" ) obtain the rights and interests of outpatient transfusion and blood security of designated medical institutions. The " collection-based-supply" blood management mode was implemented to assess the blood needs of "register" TDT patients and meet their needs to the maximum extent according to the blood inventory (collection). 【Results】 From 2018 to 2023, the total blood supply of "register" TDT patients was 10.37% of the total red blood supply of all medical institutions (138 509.5 U /1 335 788.0 U), with the highest proportion of type O blood as 46.34% (64 181.0 U/138 509.5 U) and the lowest proportion of type AB blood as 3.85% (5 331.0 U/138 509.5 U). In 2018, 9 transfusion medical institutions were designated for TDT patients.There were a total of 766 TDT patients in the register, with the per capita annual blood transfusion volume increased from 20.28 U (15 531.0 U/766 patients) in 2018 to 36.01 U (27 586.0 U/766 patients) in 2023, maintaining a positive growth every year(30.26%, 4.94%, 11.71%, 8.61%, 4.94% and 7.10%). 【Conclusion】 The " three designated" blood transfusion mode and the " collection-based-supply " blood management mode can effectively guarantee the blood supply of TDT patients.
