1.Relationship of hypothyroxinemia to gestational diabetes mellitus during early pregnancy

Objective To investigate the relationship of hypothyroxinemia to gestational diabetes mellitus during early pregnancy. Methods A total of 11 365 cases of women with early singleton pregnancies were collected from Chongqing Health Center for Women and Children. The screening of thyroid function was performed. The postload glucose concentrations and the risks of gestational diabetes mellitus in hypothyroxinemia were investigated. The relationship of thyroid hormones to postload glucose concentration was evaluated. The association between thyroid peroxidase antibodies(TPOAb) and gestational diabetes mellitus was analyzed. Results Early pregnancy women with hypothyroxinemia had a higher postload glucose concentration. Comparing to normal pregnancy women, the prevalence of gestational diabetes mellitus in hypothyroxinemia was higher(42.2%vs18.4%, P<0.05). Free thyroxine(FT4) was significantly negatively correlated with postload glucose concentration. FT4 was associated with gestational diabetes mellitus. The risk of gestational diabetes mellitus was greater when FT4 was lower. The prevalence of gestational diabetes mellitus in early pregnancy women with TPOAb levels ≥500 IU/ml was significantly higher than that in normal pregnancy women (31.2%vs 18.4%, P<0.05). Conclusion Early pregnancy women with hypothyroxinemia and TPOAb≥500 IU/ml were related to the risk of gestational diabetes mellitus. Lower FT4 was the risk factor of gestational diabetes mellitus.
2.Relationship of subclinical hypothyroidism with gestational diabetes mellitus during early pregnancy

A screening of thyroid function was performed in 2 320 pregnant women whose gestational age was less than 12 weeks.The incidence of subclinical thyroid disease and its relationship with gestational diabetes mellitus were analyzed.The results showed that there were 8 patients (0.34%) with clinical hypothyroidism,226 (9.74%) with subclinical hypothyroidism,and 14 patients (0.6%) with hyperthyroidism among 2 320 pregnant women.The prevalence of gestational diabetes mellitus in pregnant women with subclinical hypothyroidism was apparently higher than that in normal pregnant women (33.3% vs 12.9%,P<0.05),suggesting that the increased incidence of gestational diabetes mellitus seems to be related to subclinical hypothyroidism during early pregnancy.Therefore,it is important to screen the function of thyroid during early pregnancy
3.The Development of Managing Information System in Initial Response to Environmental Incidents of Public Health

To prevent and control these incidents effectively, governments explored managing information system, which provides technology and decision-making supports for dealing with the incidents. The development, application, services and characteristics of these information systems were summarized in this review, the impendence was also analyzed to develop a management information system in public health field in China.
4.Comparison of occupational exposure limits of hexavalent chromium compounds in China and foreign workplaces

Objective To compare and analyze the occupational exposure limit(OEL)of hexavalent chromium compounds in
China and foreign countries. Methods The OEL and background information of hexavalent chromium compounds released by
nine official and unofficial organizations in seven countries/regions were collected and sorted. The classification,limit levels,
formulation principles and compound characteristics labeling of OEL were compared. Results The OEL values published by
nine organizations ranged from 0.000 02 to 0.050 00 mg/m3
. Among them,the limits of seven organizations(including China)
did not clearly distinguish the soluble types of hexavalent chromium compounds;the limits of eight organizations(including
China)were calculated in terms of chromium;seven organizations(including China)have only developed OEL for long-term
exposure. The OEL of hexavalent chromium in China was 0.050 00 mg/m3 which is relatively loose. Its carcinogenicity label was
consistent with international standards,its sensitization label was consistent with some countries,and its percutaneous
absorption label had not been included in the characteristic labeling of limit values. Conclusion It is suggested that the OEL of
hexavalent chromium in China should be revised timely,and the systemic adverse effects caused by percutaneous absorption of
hexavalent chromium compounds should be focused on. At the same time,it is recommended to systematically adjust the
management concepts and principles of all carcinogens.
5.Quality criteria of xiaozhining tablet

Objective: To establish the quality criteria for xiaozhining tablet. Methods: TLC method was applicated to determine and differentiate the Scutellaria , the Radix salviae miltiorrhizae, the Rheum, the Leonurus of the tablet. HPLC method was used for detecting the content of emodin in xiaozhining Tablet. Results: Four main components of the tablet was determined well. The regression equation by HPLC was A=2.43?10 7C+6.06?10 4(r=0.999 7). Curcumin had good linear relation within the range of 0.5~0.05 mg/ml. The average recovery rate was 96.70%(RSD=1.06%). The average value of emodin was 0.021 4 mg. Conclusion: The method of determination is reliable. It can be used as the basis for the quality control of xiaozhining Tablet.
6.Rational Use of Traditional Chinese Medicine Guided by Pharmacoeconomic Theory

OBJECTIVE:To promote rational use of traditional Chinese medicines. METHODS:The practical importance of applying basic pharmacoeconomic theory to the rational use of traditional Chinese medicines was analyzed and the measures regarding how to apply the pharmacoeconomic theory to guide rational use of traditional Chinese medicines were put forward. RESULT & CONCLUSION: It is quite necessary and practical to apply the basic pharmacoeconomic theory to the rational use of traditional Chinese medicine, which plays a positive role in promoting rational use of traditional Chinese medicines.
7.Endoplasmic reticulum molecular chaperone involved in the impairment of inner ear consistented with the mimetic aging rats

Objective:To explore the involvement of endoplasmic reticulum molecular chaperone GRP78 in the impairment of inner ear consistented with the mimetic aging model.Method:Twenty-four Wistar rats were randomly divided into two groups. model group was in duced by daily hypodermic injection of 10% D-galactose (800 mg·kg~(-1)·d~(-1)) for 8 weeks and the control group was given saline accordingly. Spatial learning and memory was measured by Morris-Water-Maze. Colorimetry was used to analyze superoxide dismutase (SOD) and malondialdehyde (MDA) extracted from inner ear tissue. Hearing threshold of rats were detected with Auditory brainstem response (ABR).In addition, expression of GRP78 in the inner ear was detected by immunohistochemistry,RT-PCR and Western blot. The control group was studied parallel.Result:The escape latency in the model group injected with D-galactose was markedly longer than that in the control group.accordingly ,the changes of SOD and MDA were more significant in the model group, the difference between two groups was significant(t-test,P<0.01). the variation of ABR in two groups was observed, There was no statistically difference of the hearing in the model group compared with the control group(P>0.05). The expression of GRP78 was significantly different between two groups ,which is increased in the inner ear tissue of model group(P<0.01).Conclusion:The impairment of inner ear tissue partly dued to the oxidative stress in the model, which was induced by D-galactose.and endoplasmic reticulum molecular chaperone was thought to contribute to the impairment mechanism of inner ear in mimetic aging model.
8.Different distribution and expression of mammalian target of rapamycin complex in the kidney of diabetic nephropathy mice

Objective To investigate the different distribution and expression of mammalian target of rapamycin complex (mTORC) in the kidney of diabetic nephropathy (DN) mice.Methods Fourteen eight-week-old male C57BL/6 mice were assigned to 2 groups: the control group ( n=7 ) and the streptozotocin ( STZ )-induced DN group ( n=7 ) . Blood and urinary variables including glucose , albumin, creatinine and albumin/creatinine ratio were assessed 2 weeks after STZ injection.Hematoxylin-eosin staining was performed for renal pathological analyses .The distributions of mTOR , phosph-ser2448-mTOR(p-mTOR), mTORC1(Raptor), mTORC2(Rictor) and phosph-ser240/244-S6K1 (p-S6K1) were determined by immunofluorescence.The expression levels of mTOR, p-mTOR, mTORC1(Raptor), mTORC2(Rictor), S6K1 and p-S6K1 were detected by Western blotting .Results Two weeks after STZ injection , the diabetic mice developed albuminuria (P<0.01) and renal hypertrophy (P<0.05).The immunofluorescence positive staining for mTOR , Raptor, and Rictor was distributed in the epithelial cells of proximal tubules , glomerular mesangium and capillary loops as well as the medullary collecting ducts of the control mouse kidney .These positive signals increased in the DN mouse kidney ( P<0.05).However, pS6K1 was not detected in the inner medulla of control mouse and p-mTOR was not found in the glomeruli of both control and DN mice .Conclusion mTORC is widely expessed in the mouse kidney and participates in the development of DN , whereas the 2448 serine phosphorylation of mTOR may be not implicated in the hyperglycemia mediated glomerular injury .
9.Pharmacokinetic interaction between scutellarin and valsartan in rats.

Scutellarin is the main effective constituent of breviscapine, a flavonoid mixture isolated from the dried whole plant of Erigeron breviscapus (Vant.) Hand-Mazz, and valsartan is used as an antihypertensive drug. These two drugs have already been clinically used together to treat diabetic nephropathy (DN) in China, and the combined medications showed some enhanced protection against DN. The aim of this study is to investigate the potential pharmacokinetic interaction between scutellarin and valsartan in rats. Breviscapine injection (20 mg x kg(-1), i.v.) and valsartan (15 mg x kg-, i.g.), either alone or together were given to 18 male Sprague-Dawley rats. Concentrations of scutellarin and valsartan were quantified by HPLC, and pharmacokinetic parameters were calculated by non-compartmental methods. We found that the pharmacokinetic parameters of scutellarin altered significantly after co-administration of oral valsartan. The plasma clearance (CL(p)) and the bile clearance (CL(b)) of scutellarin were reduced significantly in the presence of valsartan. After oral administration of valsartan with or without intravenous scutellarin, however, the pharmacokinetic parameters of valsartan were comparable. In conclusion, our data suggests that the concurrent use of valsartan reduces the biliary excretion of scutellarin, and this may be due to the inhibitory effect of valsartan on the biliary excretion of scutellarin mediated by Mrp2 (Multidrug resistance-associated protein 2).
10.Recombinant human glucagon-like peptide-1 (7-36) enhances insulin release and insulin mRNA expression in INS-1 cells

In INS-1 cells, the insulin secretion was investigated by radioimmunoassay (RIA) after 4 h incubation in medium containing different concentrations of glucose and recombinant human glucagon-like peptide-1 (rhGLP-1) (7-36). Insulin mRNA level in INS-1 cells was assessed by a semi-quantitative RT-PCR method. rhGLP-1 (7-36) is not only a powerful insulin secretagogue, but also can increase insulin gene expression in INS-1 cells.
