Objective To investigate the genetic diversity of five populations of cultivated Morinda officinalis in Guangdong Province using random amplified polymorphic DNA(RAPD).Methods Sixty-four individuals in five populations of cultivated M.officinalis were analyzed by RAPD markers to determine the genetic variations among the populations. The data of genetic diversity were analyzed with Popgen32 software. Results The levels of genetic variation and patterns of population structure in M.officinalis were investigated using RAPD markers.Of the 100 primers screened,15 primers produced highly reproducible RAPD bands, using these primers,224 discernible DNA fragments were generated with 112(50.00%) polymorphic fragments, indicating considerable genetic variation at the species level. In contrast, there were relatively high levels of polymorphism at the population level with the percentage of polymorphic bands ranging from 37.05% to 53.13%,and the mean percentage of polymorphic loic(P=45.86%).Genetic variation among populations was detected based on Nei's genetic diversity analysis(0.175 6),Shannon's diversity index(0.287 6) for every population with Shannon's index 0.103 3—0.236 2 and Nei's indexes 0.074 5—0.154 0.Conclusion There is a little genetic differentiation among populations of cultivated M.officinalis and result in different cultivated types, the genetic diversity with M.officinalis(POP2) is higher than that of other populations. It may be the main reason of the difference of cultivated M.officinalis quality.

Maintaining genetic diversity is a major issue in conservation biology. In this study, we demonstrate the differences of genetic diversity levels between wild and captive individuals of Elliot's Pheasant Syrmaticus ellioti. Wild individuals showed a higher genetic diversity level than that of the captive individuals. Nucleotide diversity and haplotype diversity of wild individuals were 0.00628 and 0.993, while those of captive individuals were 0.00150 and 0.584 respectively. Only 3 haplotypes of mtDNA control region sequence were identified among 36 captive individuals, while 16 unique haplotypes were identified among the 17 wild individuals in this study. One captive haplotype was shared by a wild individual from Anhui Province. It is concluded that a low number of founders was the likely reason for the lower level genetic diversity of the captive group. Careful genetic management is suggested for captive populations, particularly of such an endangered species, to maintain genetic variability levels.

Chinese Journal of Analytical Chemistry 2017;45(12):1915-1920
A label-free method for sensitive and selective detection of thrombin ( Tb) was constructed based on rare earth ion mediated fluorescence switch of graphene quantum dots ( GQDs) . Rare earth ion ( Er3+) can assemble onto the surface of GQDs through the coordination interaction between Er3+ ions and the carboxylate groups located on the surface or edge of the GQDs, resulting in the aggregation of the GQDs and thereby decrease of the fluorescence of the GQDs. In the presence of Tb, the oxygen and nitrogen-donor atoms in Tb can coordinate with Er3+ ions and compete with the carboxylate groups on GQDs to coordinate Er3+ ions, thus the interaction between GQDs and Er3+ ions is reduced, which lead to the restoration of the GQDs fluorescence. In this study, the sensing mechanism was demonstrated by transmission electron microscopy, atomic force microscope, Fourier transform infrared spectroscopy, UV-Vis absorption and fluorescence spectroscopy. The limit of detection for Tb assay was as low as 0. 049 nmol/L. Moreover, this assay was successfully applied to the selective determination of Tb in real samples.

Acta Pharmaceutica Sinica 2012;47(4):535-540
PCR sequencing ITS genes methods were used to assess the genetic diversity of Morinda officinalis How different populations. The sequence of Morinda officinalis ITS gene was 567 bp in length, and the content of G/C was 64.5%. In this study, 17 haplotypes were obtained, which were at a high level of branching, and the haplotypes of Guangdong population showed to be the expansion origin. The result of the analysis of molecular variance (AMOVA) also showed that the percentage of variation among populations (56.65%) was greater than that within a population (43.35%). The F(ST) value was 0.566 5, and the genetic divergence among populations was significant. Mantel test results also indicated that the level of geneflow was positively correlated with geographic distances (R2 = 0.721 1). The result showed a good correlation between genotype and geographic distribution of Morinda officinalis, and ITS gene sequencing could be useful molecular method for the genuineness and phylogeography of Morinda officinalis.

Objective To observe the state of endemic flurosis in Qinghai province in 2008 and to provide scientific basis for endemic flurosis control and prevention. Methods All the endemic fluorosis villages in Qinghai province was divided into slight, medium and heavy degree according to the water fluoride content before improving water, and 2,4,4 villages were collected from each degree, respectively. Water samples were collected from each direction of east, west, south, north and centre, and fluoride concentration were determined from each surveyed villages with unimproved-water. At the same time,3 copies of the peripheral water samples and a sample of source water were collected to determine fluoride concentration. In all the village children aged 8 to 12 were tested for dental fluorosis by Dean method. Six copies of the urinary fluoride were randomly sampled in different age groups. The fluorine content in urine was determined with F-ion selective electrode. The situation of clinical skeletal fluorosis of adults over 16 years of age was examined, and 20 adults in the villages of medium and heavy levels were filmed for skeletal fluorosis. Results The detectable rate of dental fluorosis of children aged 8-12 years old was 43.59% (245/562),that of skeletal flurosis of adults aged above 16 years was 50.32% (1582/3144) and detectable rate of X-ray was 29.45% (48/163). Fluoride content in urine was 1.52 mg/L. Conclusions Endemic fluorosis is still very serious in Qiaghai province in a rising trend,so the control measures need to be strengthen.
