1.Role of RANTES in evolution of alloreactive CD4+ memory T cells adoptive transfer-induced cardiac allograft acute rejection

Objective To observe the intluence ot chemokine RANTES influence on cardiac allograft acute rejection caused by alloreactive memory CD4+ T cells (Tm) adoptive transfer.Methods Heterotopic heart transplantation (HTx) from Balb/c donors to C57BL/6 recipients was performed by anastomosis of the vessels of the neck.Mice undergoing heterotopic heart transplantation received either adoptive transfer of 1 × 106 CD4+ Tm from the spleen of alloantigen-primed C57BL/6 mice or no cells (control group).After the cardiac transplantation,the mean survival time (MST),mean histologic rank of rejection,relative gene expression and serum concentration of RANTES in the cardiac grafts.Results (1) The percentage of CD4+ Tm was 26.83% at the spleen of alloantigenprimed mice; (2) The MST was 5.17 ± 0.17 days in the CD4+ Tm+ HTx group versus 7.76 ± 0.21 days at the HTx group (control group) (P<0.01); (3) The histological tests revealed that mean histologic rank of rejection activity in the sections of cardiac allografts on the day 5 post grafting was grade 3.92 ± 0.08 in the HTx+ CD4+Tm group versus grade 2.67 ± 0.14 in HTx group (P<0.01) ;(4) The relative gene expression level of RANTES was 2.6 ± 0.21 in the CD4+ Tm + HTx group,significantly higher than in the control group (P<0.01) ; (5) The serum concentration of RANTES in the CD4+ Tm+ HTx group was 223.6 ± 16.79 pg/mL,higher than in the control group (120.7 ±9.47 pg/mL,P<0.01).Conclusion Alloreactive CD4+ Tm contribute to the increased expression and secretion of RANTES,and cardiac allograft acute rejection was more extensive in the CD4 + Tm + HTx group.
2.Comparison of Nutritional Status before and after Treatment with Pre diasure on Children with Anorexia

Objective To evaluate the clinical effect of the enteral nutrient(Pediasure) on making improvements in nutritional status of children with anorexia.Methods Thirty children were treated with pediasure for 2 months, twice a day, and height(Ht),weight(Wt),AG,TSF,SSF,ASF,RBC,Hb were measured before and after treatment and their appetites,alimentary canal reactions were recorded.Results Compared with the effects after 2 months, the average Ht,Wt,AG,SF all increased, and there was significant difference between pretreatment and after treatment(P0.05). All of the 30 children had no adverse reactions during treatment.Twenty-two children (73.33%) had better appetites and increased the amount of eating after treatment.Conclusion The study suggests that pediasure may be safely used in the treatment of children′s anorexia and effectively improve patients′ nutrition without adverse reactions.

4.Changes of Serum Zinc Level of Rotaviral Enteritis in Children and Therapeutic Effect of Zinc

Objective To explore the change of serum zinc level of children with rotavirus(RV) enteritis and efficacy of zinc supplementation on children with rotaviral enteritis.Methods One thousand one hundred and eighty-three healthy children were enrolled in this study,who were divided into different groups,and the zinc levels of children in every different age group were detected.These levels were used as normal levels of children in Dongguan district.One hundred and forty-three diarrhea patients in this 2 years were enrolled.Confirmed dia-gnosis was depended on the RV test in patients′ stool.The children with diarrhea were randomly divided into zinc treatment group (n=72) and conventional therapy group (n=71).The serum zinc levels in RV enteritis was analyzed.The patients in conventional therapy group were received diarrhea conventional therapy inchuded smectite and fluid infusion and so on;in addition to diarrhea conventional therapy,zinc gluconate 20 mg/d (10 mg/d below 6 months of age) for successive 14 days were given the patientsin zinc treatment group.The different efficacy of the 2 groups and the changes of serum zinc in treatment group were analyzed.Results Compared with normal children,the ones suffering from RV enteritis had lower serum zinc level(P0.05).Conclusions The ones suffering from RV enteritis have lower serum zinc level.In addition to diarrhea conventional therapy,giving zinc treatment has better effect on enhancing curative effect of RV enteritis.On the other hand,the suggestion of zinc supplement made by WHO is also suitable for Chinese children.
5.Study on synergic or antagonistic effects of different acupoint groups on vertebro-basilar insufficiency.

OBJECTIVETo observe the synergic or antagonistic effect of needling acupoints Fengchi (GB 20) and Tianzhu (BL 10), and Jiaji C4-C6 (EX-B2) on vertebro-basilar insufficiency (VBI).
METHODSSelf-control method was used and 20 cases of VBI were respectively treated with acupuncture at Fengchi (GB 20) and Tianzhu (BL 10), Jiaji (EX-B2). Their combination and the changes of vertebro-basilar artery's (VBA) systolic velocity of blood flow was detected.
RESULTSThe VBA's systolic velocity of blood flow after acupuncture were increased in all the 3 groups (P < 0.05 or P < 0.01), with no significant difference among the 3 groups (P > 0.05).
CONCLUSIONAcupuncture at Fengchi (GB 20) and Tianzhu (BL 10) or Jiaji (C4-C6 ) or their combination can increase VBA's systolic velocity of blood flow, improving blood supply of vertebro-basilar artery, but they have no synergic or antagonistic effects.

6.A blind technique used in randomized controlled trials of treatment based on changes of syndromes.

Treatment based on syndrome differentiation is an essential feature of traditional Chinese medical diagnosis. The interventions based on changes of syndrome types in randomized controlled trials are complicated, leading to the difficulty of blind method enforcement. This article described a double-blind method. It could be used in randomized controlled trials under the condition of different syndrome types and different medications. It numbered drugs in two stages, and in two phases to achieve double-blind. This method not only guaranteed investigators and subjects to be in blinded conditions, but also achieved using different medications for patients of different syndromes. It also caused no drug waste. It was scientific and feasible.

7.Study on antisense oligoncleotides as inhibitor of human acute promyelocytic leukemia proliferation and protein expression

AIM: To study on antisense oligoncleotides as inhibitor of human acute promyelocytic leukemia (HL-60) proliferation and C-myc protein expression. METHODS: Oligonucleotides with different lengths (18-21 mer) complementary to the definite regions of C-myc mRNA, modified groups (with S replaced O in internucleotide phosphate linkage) and unmodified ones (with natural internucleotide phosphate linkage) were designed and synthesized. These olignucleotides were tested for their activity on HL-60 cell and also for their toxicity on normal lymphatic cells of human. RESULTS: It was found that two of the oligonucleotides complementary to 5′-44-61 and 5′-556-576 the regions of C-myc mRNA exhibited great inhibitory effects (59.5 % and 62.7 %) on growth of HL-60 cells for a definite time. And no toxicity of the two antisense oligonucleotides was found on normal lymphatic cells of human. CONCLUSION: The sequence of antisense oligonucleotides complementary to 5′-44-61 of C-myc mRNA was designed newly by us may be turned into inhibitory medicine of HL-60 cells.
8.Observation on therapeutic effect of electroacupuncture combined with functional training for treatment of peripheral nerve incomplete injury of upper limbs.

OBJECTIVETo search for the best program for treatment of peripheral nerve incomplete injury.
METHODSNinety cases were randomly divided into a treatment group, a control group I and a control group II, 30 cases in each group. The treatment group were treated with electroacupuncture at Jianyu (LI 15), Hegu (LI 4), Quchi (LI 11), etc. plus functional training, and the control group I with electroacupuncture and the control group H with functional training. After treatment for 3 months, basic function, practical function, EMG, nerve conduction velocity were compared among the 3 groups.
RESULTSThe good rate of basic function of 50.0%, the curemarkedly effective rate of practical function of 50.0% and the total effective rate of neurophysiology of 64.3% in the treatment group were better than 20.7%, 17.2%, 41.4% in the control group I (P < 0.05) and 23.3%, 20.0% and 36.O7% in the control group II (P 0.05).
CONCLUSIONElectroacupuncture combined with functional training can accelerate nervous repair, promote functional recovery of the denervated muscles, so as to shorten the restoring time of nerve-muscle and increase life quality of the patient.

9.Observation on the therapeutic effect of ear point combined therapy on common migraine at the remission stage and effects on plasma CGRP and ET in the patient.

OBJECTIVETo probe into the therapeutic effect of ear point combined therapy on common migraine at the remission stage and the mechanism.
METHODSThirty-two cases of common migraine at the remission stage were treated with ear point combined therapy, including blood-letting at ear back, injection of autoblood into Fengchi (GB 20), Yanglingquan (GB 34), and pricking at ear points Nie, Yi-dan (pancreas and gallbladder), Shenmen, etc.. Clinical therapeutic effect was evaluated at the end of one therapeutic course, and calcitonin gene-related peptide (CGRP) and endotheline (ET) were detected before and after treatment, and the CGRP and ET contents were compared with those in 22 normal persons.
RESULTSThe effective rate was 78.1% (P<0.001), CGRP and ET contents had very significant changes after treatment (P<0.01), and CGRP and ET showed negative correlativity (P<0.05).
CONCLUSIONThe ear point combined therapy can regulates the balance between the plasma CGRP and ET in the patient of common migraine and improve vasodilative, vasoconstrictive and endothelial fuctions in the brain, hence stopping pain.

10.Clinical observation on treatment of cervicogenic headache with turtle-probing needling at Tianzhu (BL 10).

OBJECTIVETo observe therapeutic effect of turtle-probing needling at Tianzhu (BL 10) on cervicogenic headache.
METHODSSeventy cases were randomly divided into a treatment group (n=36) and a control group (n= 34). The treatment group were treated with turtle-probing needling at Tianzhu (BL 10) on the affected side, first perpendicularly needling the point and then penetration-needling homolateral Fengchi (GB 20), Fengfu (GV 16), below cervical Jiaji points (EX-B2), and occipital tuberosity in order. And the control group were treated with acupuncture at Fengchi (GB 20),Tianzhu (BL 10), Shuaigu (GB 8), Touwei (ST 8) and corresponding cervical Jiaji points (EX-B2), Houxi (SI 3), Ashi points at the affected side with routine needling manipulation. Treatment was given once each day, 6 sessions constituting one course.
RESULTSThere was no significant difference between the two groups in therapeutic effect (P > 0.05), but the cured and markedly effective rates for analgesic effect in the observation group at the first three sessions were higher than those in the control group (P < 0.05).
CONCLUSIONThe turtle-probing needling method and the routine needling have a similar therapeutic effect, but the turtle-probing needling method more rapidly produces the analgesic effect.
