1.The Renoprotective Mechanism of Mobic in Experimental Type 2 Diabetic Rat

Objective To observe the renal expression of cyclooxygenase-2 (COX-2) in rats with type 2 diabetes, and explore the effect of selective COX-2 inhibitor Mobic on the expression of renal COX-2, matrix metalloproteinase-9(MMP-9), tissue inhibitor of matrix metalloproteinase-1(TIMP-1), TXB 2 and 6-Ket-PGF1?, as well as renal structure and function. Methods All rats were divided into control group, diabetes mellitus group and treatment group. Type 2 diabetic rats were treated with Mobic and vehicle respectively. Immunohistochemistry was used to detect the expression of COX-2,MMP-9 and TIMP-1 in renal tissues. The urinary TXB 2 and 6-Ket-PGF1? concentration was determined by radioimmunoassay at 6th week. Results There were an increasing expression of COX-2, TIMP-1 and decreasing MMP-9 expression in the renal tissue of type 2 diabetic rats. Mobic could increase MMP-9 expression and depress TIMP-1 expression througth inhibiting the expression of COX-2 in the renal tissues of type 2 diabetic rats. Conclusion COX-2 was involved in the pathogenesis of type 2 diabetic nephropathy. Selective COX-2 inhibitor Mobic might exert its renoprotective effects through inhibiting COX-2 activity, decreasing prostagladins systhesis, and modulating MMP-9 and TIMP-1 expression.
2.Pathogenesis,diagnosis and treatment of recurrent spontaneous abortion with immune type

Recurrent spontaneous abortion (RSA),which affects 1% to 5% of women of reproductive age,is difficult to treat in the clinical setting. In the investigations of immunopathogenesis,diagnosis and treatment of RSA since the late 1980s,it was found that RSA was associated with abnormal maternal local or systemic immune response,the pathogenesis of autoimmune RSA was mainly associated with antiphosphlipid antibody (APA),while that of alloimmune RSA was due to the disturbance of maternofetal immunological tolerance. Systemic etiological screening process and diagnosis systems of RSA with immune type were developed,and anticardiolipin (ACL)+? 2-GP1 combining multiple assay for effective diagnosis of RSA with immune type was initially established. According to dynamic monitoring clinical parameters before and during gestation,low-dose,short-course and individual immunosuppressive therapy and lymphocyte immunotherapy for RSA with immune type were initiated. The outcomes of the offsprings of patients with RSA were followed up,and the safety and validity of the therapies were confirmed. The research achievement leads to great progress in the diagnosis and treatment of RSA in China.
4.The Study of DNA in Middle Ear Effusions

Objective To determine whether or not the bacterial DNA which was detected by PCR comes from viable bacteria.Methods Observe the effection of middle ear effusion(MEE) on DNA viscosity and enzymatic digestion of DNA.Results The middle ear effusion and DNA are stable and DNase 1 rapidly digests DNA,The effusion does not seem to degrade DNA.The middle ear effusion signifcantly inhibits DNase 1.Conclusions Middle ear effusion provides an inhibition of the enzymatic digestion of purified DNA.Thus any DNA found in effusion by PCR techniques could well be fossilized remains and chronic otitis media with effusion may not be the bacterial infection that recent studies have suggested.

6.The expression of CD14 on Kupffer cells during the course of carbon tetrachloride-induced liver injury

Objective To investigate the expression of CD14, the receptor of lipopolysaccharide (LPS) on Kupffer cell membrane during the course of CCl4-induced liver injury and its role in activation of Kupffer cells. Methods The experimental rats were hypodermically administered CCl4 twice weekly for up to 8 weeks. Kupffer cells were isolated from the liver of normal and CCl4-treated rats by the combined ‘collagenase-pronase’ perfusion method and discontinuous density gradient centrifugation. On the following day after isolation, the cells were incubated with RPMI-1640 containing various doses of LPS for 6 h. The tumor necrosis factor (TNF-?) in the supernatants was measured by ELISA. The expression of CD14 mRNA on Kupffer cells was determined by RT-PCR. The plasma levels of endotoxin were determined by chromogenic substrate limulus amebocyte lysate assay. 8.Efficacy of Lamivudine Combined with Thymosin ?_1 on Young Patients with Chronic Hepatitis B

Objective To investigate the efficacy of lamivudine( LAM) combined with thymosin ?1 (T?1) on young patients with chronic hepatitis B. Methods Sixty six chronic hepatitis B (CHB) patients aged from 12 to 16 years and weight greater than 33 kg were divided into the LAM + T?1 group and the LAM group; in LAM + T?1 group,T?1 was administered 1.6 mg hypodermic once daily and after 5 days it was changed to 1 6 mg hypodermic, twice in two weeks, and the drug was stopped after 26 weeks, while LAM had been administered 100 mg each time once daily for 52 weeks. In LAM group, only LAM had been administered 100 mg once daily for 52 weeks. The serum level of HBV DNA, HBV markers and the ALT were measured before and after treatment Results After the treatment, the rate of serum HBV UNA turned to negative in LAM + T?1 group was 100 % , and that in LAM group was 93.7 % ; recovery ratios at 52 weeks of LAM+ T?1 group was 87.1% ,and that of LAM group was 78. 1% .After 52 weeks, the HBeAg serum conversion rate of LAM + T?1 group was 58. 1 % , and that of the LAM group was 21.9%. Comparing the results of the two groups, the difference was remarkable. Conclusion The efficacy of T?1 combined with LAM on young patients with chronic hepatitis B is superior to that use of single LAM
