Background To construct and analyze the differentially expressed gene library in the SD rat's brain of Parkinson's disease. Methods The lateralization Parkinson's disease model of SD rat was established by stereotaxis injection of 6-OHDA in lateralization corpora striatum. Suppression subtractive hybridization was utilized to isolate the cDNA fragments in both control and experimental groups. Then the cDNA fragments were directly bound to pMD18-T to set up the subtractive library. Amplification of the library was carried out by transformation of E. coli with high voltage electroperforation. Ninety five positive bacteria clones were randomly picked up and identified by colony PCR.Results The amplified library contained more than 1500 positive bacteria clones. Ninety-five clones were analyzed randomly by colony PCR. And there were 81% clones contained 100 ~600 bp DNA fragments. They might be the cDNA fragments of differentially expressed genes of Parkinson's disease. Conclusions A differentially expressed genes subtracted cDNA library of Parkinson's disease of SD rat was constructed successfully with SSH and T/A cloning techniques. The library is efficient and it was useful in screening and cloning new and specific differential expression genes of Parkinson's disease.

Chinese Pharmacological Bulletin 1987;0(01):-
In the study we determined the content of malondialdehyde(MDA) , which is the metabolite of lipid peroxide(LPO) , in plasm and lung homogenate by thiobarbituric acid(TBA) colorimetry, and the content of phosphatidyl-choline(PC), which is the major functional composition of pulmonary surfactant (PS).and total phospholipid (TPL) by thin-layer chromatogra-phy and inorganic phosphorus quantitative analysis in rabbits with lung injury induced by oleic acid. The results showed that the MDA level wasincreased in plasm and lung homogenate; PC and TPL were decreased in lung homogenate. Administrating of 654-2 before and after (0. 5 h)lung injury could both inhibit the production LPO and prevent the decrease of PS. The findings suggest that the decrease of PS may be due to the increase of LPO and that G54-2 could modify the process.

Progress in Biochemistry and Biophysics 2006;0(09):-
The effects of glutamate transporters on synaptic plasticity in rat models of pilocarpine-induced status epilepticus were investigated. Male Wista rats ((304.06?13.79) g) were randomly divided into 5 groups, short-term seizures (SE) and its control (SC), long-term seizures (LE) and its control(LC), normal control (Sham) groups. Epilepsy rat models were induced by injection of pilocarpine(25 mg/kg, i.d.). Glutamate transporter inhibitor, DL-threo-benzyloxyaspartate (TBOA, 7.5 nmol,1 ?l) was microinjected into right side of hippocampus after 14 days of initial status epilepticus in SE and LE groups. The same volumes of artificial cerebrospinal fluid were injected into same side of hippocampus in SC and LC groups. Electroencephalographys (EEG) were detected in SE and SC groups after 2 h of drug injection. Long term potential (LTP) at perforant pathway and dentate gyrus(PP-DG) and EEG were recorded in LE and LC groups after two weeks of drug injection. Example of Fluoro-Jade-B staining in the rat brain was made at the end of electrophysiological experiment. The results showed that there was a significant decrease in theta band power of EEG in SE group compared with that of SC group (P 0.05). The slope of excitatory postsynaptic potential (EPSP) was significantly increased in LE group compared with that of LC group (P
