3.The effect of Connexin43 downregulation on biological functions of HUVEC.

Connexin43 has been shown to play a pivotal role in wound healing process. Wound repair is enhanced by acute downregulation of connexin43, by increasing proliferation and migration of keratinocyte and fibroblast. Angiogenesis is also a central feature of wound repair, but little is known about the effects of connexin43 modulation on functions of endothelial cells. We used connexin43 specific small interference RNA (siRNA) to reduce the expression of connexin43 in human umbilical vein endothelial cell (HUVEC), and investigated the effects of connexin43 downregulation on intercellular communication, viability, proliferation, migration and angiogenic activity of HUVEC. Treatment of siRNA markedly reduced the expression of connexin43 by -80% in HUVEC (P < 0.05), and decreased the intercellular communication by -65% (P < 0.05). The viability, proliferation, migration and angiogenic activity of HUVEC decreased significantly (P < 0.05), compared with that of the normal cells. The results suggest that temporally downregulation of connexin43 expression at early stage of wound to inhibit the abnormal angiogenesis characterized with leaky and inflamed blood vessels, maybe a prerequisite for coordinated normal healing process.

4.STUDY ON CONDITIONS OF SWINE BLOOD MEAL FERMENTATION WITH COMPOUND STRAINS

Two high protease producing strains, whic h are desi gnated Aspergillus oryzae AS100 and AS102, were screened and applied as main fermentation strains of swine blood meal. A Saccharomyces cerevisiae Y113 and a Bacillus Asp007 were also used as assistant fermentation strains. The enzyme production abilities we re studied,and a series of parameters of the fermentation technology were deter mined. Through swine blood meal fermentation, a high-protein fermented feed wa s produced, which is strong soy flavor and was rich in protein(69%), free amino acid, vitamin such as VitD 3 and niacin and organic elements such as Fe. It is a kind of high-protein feed for animals and it could be used as feedstuff addi tive.
5.Relation between Injury Time and the Expression of COX6C mRNA in Skeletal Muscle of Rats after Contusion.

OBJECTIVE:
To investigate the relation between injury time and the expression of cytochrome c oxidase subunit VIc (COX6C) mRNA in skeletal muscle of rat after contusion.
METHODS:
A total of fifty-four SD rats were divided into the control group and the contusion groups (0.5, 1, 6, 12, 18, 24, 30, and 36 h after contusion), randomly. The contusion model was established by free fall drop of gravity hammer. At corresponding time point after contusion, the regular histology was examined and expression level of COX6C mRNA was tested by real-time PCR after extraction of total RNA from the tissues.
RESULTS:
The main pathological features of 6 h after injury included edema and hemorrhage in myocytes with no inflammatory cells found. After 6 hours, the findings included myocyte degeneration and necrosis, inflammatory cells infiltration, and fibrous connective tissue proliferation in the contused zone. The expression level of COX6C mRNA was higher than that of the control group within 6 h after contusion. The expression level was lower than that of the control group from 6-36 h after contusion.
CONCLUSION
The level of COX6C mRNA expresses in a regular way after contusion. It may be useful for estimating wound age in combination with the results of pathological features.

6.Effects of chrysotile fibres lipid peroxides reaction in exposed female workers and in rat lung macrophages

Objective To investigate the mechanism of lipid peroxidation by which chrysotile fibres exert their effect in human in vivo and in vitro.Methods Plasma malondialdehyde (MDA, an indicator of lipid peroxidation) and activities of superoxide dismutase (SOD) and glutathion peroxidase (GSH-Px) were determined in 47 female workers, who were exposed to high airborne concentration of chrysotile fibres (mean 18.40±3.23, ranging from 5.0~186.3 mg/m3), 12 female asbestosis patients with simple asbestosis and controls. Meanwhile MDA level and SOD activity in cell membrane were detected in macrophages treated by chrysotile fibres.Results Plasma MDA level and activities of SOD and GSH-Px of exposed female workers were 3.23±0.59 μmol/L, 17.39±4.63 U/ml, 181.61±12.49 U/L, respectively. Likewise those of non-exposed controls were 2.15±0.75 μmol/L, 11.99±3.99 U/L, 170.13±15.12 U/L respectively. Plasma MDA level and activities of SOD and GSH-Px of exposed female workers were significantly higher than those of the non-exposed controls (P<0.001). While, mean plasma MDA and SOD, GSH-Px of the asbestosis patients (mean 3.03±0.71 μmol/L,18.23±6.97 U/ml, 180.04±7.73 U/L, respectively) and those of chrysotile-exposed controls (mean 2.77±1.54 μmol/L, 17.44±4.47 U/ml, 182.52±6.61 U/L, respectively) were not significantly different (P>0.05). Correlations between plasma MDA and durations of exposure among asbestos exposed workers were statistically insignificant: γ=-0.1934, P>0.05. Moreover, it existed a balance between MDA level and SOD activity with MDA levles increasing and SOD activity decreasing. While SOD was feedbackly increasing and MDA decreasing, the MDA was then increasing gradually and SOD decreasing. The process occurred progressively. At the same time, chrysotile fibres could elevate MDA level and decrease SOD activity in the cell membrane of macrophages at the concentrations of 100 μg/ml in vitro.Conclusions The results suggest the involvement of lipid peroxidation on exposure to chrysotile fibres and development of antioxidant mechanisms in exposed workers and asbestosis patients, and support the opinion that LPO plays an important role in the development of asbestosis, and MDA and antioxidase, such as SOD, porbably constitute sensitive features of prediction and trending observation.
7.Cloning and analysis of a novel human CD44 variant

AIM: To detect unknown CD44 variants(CD44v) in nasopharyngeal cancer by using reverse transcription-polymerase chain reaction(RT-PCR) to analyze the expression of cell adhesion protein CD44 gene in nasopharyngeal cancer tissue and cell lines.METHODS: Specific primers at up start code,down terminal code of CD44 and primers at the middle,splicing points of variable splicing exon v10 of CD44 were designed.cDNA of nasopharyngeal cancer tissues,5-8F and HNE1 cell lines were analyzed by RT-PCR.Products of RT-PCR were sequenced and further analyzed by bioinformatics.RESULTS: The new CD44v sequence possessed 1634 bp with a completed open reading frame.The start code was at 12 bp site and terminal code at 1301bp site.It was predicted to code 429 amino acids,and only variable splicing exon 10 existed in the flexible region.It was given an accessible number EF581837 by GenBank.CONCLUSION: A new CD44 variant predicted to code 429 amino acids exists in the studied nasopharyngeal cancer tissues and cell lines.
8.Expression of CD44v3 and CD44v6 in patients with non small cell lung cancer and its relationship with lymph node metastasis

objective To study the expression of CD44v3 and CD44v6 and its relationship with lymph node metastasis in non small cell lung cancer (NSCLC). Methods Specimens (lung tissues) from 52 c ases of NSCLS and 12 normal lung tissue were used to detect the expression of CD 44v3 and CD44v6 by immunohistochemical method (SP method) and flow cytometry, co rrelation was analysed between the expression of CD44v3 or CD44v6 a nd lymph node metastasis of the lung cancer. Results CD44v3 and CD44v6 were not, or weakly expressed in all normal lung tissues from 12 cases. In contrast, the expression levels of CD44v3 and CD44v6 were obviously higher in lung cancer than that in normal tissue(P<0.05). The expression of CD44v 3 and CD44v6 were much higher in patients with lymph node metastasis than those without lymph node metastasis(P<0.05). Conclusion ① CD44v3 and CD44v6 are expressed with different degree in NSCLC. ②There is a close rel ationship between high expression of CD44v6 and lymph node metastasis, and CD44v 6 may be a co-marker for predicting the potentiality of lymph node metastasis i n lung cancer.
9.The value of 5-HTT gene polymorphism for the assessment and prediction of male adolescence violence.

OBJECTIVETo establish an adolescent violence crime prediction model, and to assess the value of serotonin transporter (5-HTT) gene polymorphism for the assessment and prediction of violent crime.
METHODSInvestigative tools were used to analyze the difference in personality dimensions, social support, coping styles, aggressiveness, impulsivity, and family condition scale between 223 adolescents with violence behavior and 148 adolescents without violence behavior. The distribution of 5-HTT gene polymorphisms (5-HTTLPR and 5-HTTVNTR) was compared between the two groups. The role of 5-HTT gene polymorphism on adolescent personality, impulsion and aggression scale also was also analyzed. Stepwise logistic regression was used to establish a predictive model for adolescent violent crime.
RESULTSSignificant difference was found between the violence group and the control group on multiple dimensions of psychology and environment scales. However, no statistical difference was found with regard to the 5-HTT genotypes and alleles between adolescents with violent behaviors and normal controls. The rate of prediction accuracy was not significantly improved when 5-HTT gene polymorphism was taken into the model.
CONCLUSIONThe violent crime of adolescents was closely related with social and environmental factors. No association was found between 5-HTT polymorphisms and adolescent violence criminal behavior.

10.Relative Expression of Indicators for Wound Age Estimation in Forensic Pathology.

OBJECTIVE:
In order to understand which kind of function genes play an important role for estimating wound age, the variation of difference genes' mRNA expression were compared after injury.
METHODS:
The mRNA expression levels of seven candidate genes (ICAM-1, NF-κB, MX2, MT1, MT2, sTnI, and Cox6c) were analyzed in contused rat skeletal muscle at different time points using real-time fluorescent quantitative PCR (RT-qPCR). The raw Ct values were normalized relative to that of RPL32 mRNA, and converted to standard Ct values. At each time point after injury, the standard deviations (SD) of the standard Ct values were calculated by SPSS.
RESULTS:
The expression trends of the seven genes were all found to be related to wound age, but there were lower variation coefficients and greater reliability of s TnI and Cox6c when compared with other genes.
CONCLUSION
The genes encoding structural proteins or proteins that perform basic functions can be suitable for wound age estimation.
