1.CT features and clinicopathological analysis of xanthogranulomatous cholecystitis

Objective To investigate CT features of XGC in order to improve pre-operative diagnosis of the disease.Methods The CT features of 6 cases with XGC confirmed by histopathology were analyzed retrospectively.Results The gallbladder walls thick-ened in all 6 cases,diffusely thickened in 4 cases and focally thickened in 2 cases.Hypodensity nodules or bands in thickened walls were seen in all 6 cases.The continuous mucosal lines were observed in 4 cases.After contrast administration,the hypodensity band sign was seen in most cases in artery phase.Stones were found in all 6 gallbladders and in 4 common bile ducts,dilatation of biliary tract was seen in 3 cases.There were hepatic involvement in 5 cases,duodenal,hepatic flexure of the colon and antrum of the stom-ach adhesion with gallbladder was seen in 1 case respectively.Conclusion Hypodensity nodules or bands in thickened gallbladder walls and continuous mucosal line are characteristics of XGC,which are helpful in diagnosis and differential diagnosis.
2.Trimester-Specific Gestational Weight Gain in Twin Pregnancies:A Retrospective Cohort Study

Objective:To determine the relationship between trimester-specific gestational weight gain and pregnancy outcome among women with twin pregnancies.Methods:A retrospective cohort study was conducted among Chinese women with twin pregnancies delivered from January 2013 to October 2015 in the International Peace Maternity & Child Health Hospital of China Welfare Institute.Gestational weight gain was compared between women with different progestational BMI.After the adjustment for age and progestational BMI,binary Logistic regression analysis was conducted to explore the relationship between gestational weight gain rate and gestational complications,premature birth,premature rupture of membrane and neonatal birth weight.Results:①A total of 472 subjects met the inclusion criteria.Rate of gestational weight gain were(0.41 ±0.15),(0.64 ±0.30) and (0.49 ±0.15) kg/week in Early,Mid-and Late-,and Whole of pregnancy respectively.②With regards to trimester-specific gestational weight gain,statistically significant associations were found between rate of weight gain and gestational hypertension and preterm birth (P < 0.05).Those with faster gestational weight gain had higher rate of premature rupture of fetal membrane(P < 0.05).And those with lower gestational weight gain had lower neonatal birth weight(P<0.05).③After adjusting maternal age,progestational BMI and gestational age at delivery,the sum of twins' birth weight increased 25.21g,30.89g and 21.46g respectively when the maternal body weight increased 1kg during the whole pregnancy,in Early and Mid-pregnancy (P < 0.001),and during in Late-pregnancy(P =0.001).Conclusions:Gestational weight gain among women with twin pregnancy are closely related with adverse pregnancy outcomes.The gestational weight gain rate in different gestation are of some predictive value for pregnancy outcomes.More research with long follow-up are needed to determine proper gestational weight gain for twin pregnancy suitable for Chinese women.
3.Expression of EZH2 mRNA and protein in human prostate carcinoma and its relationship with cell proliferation

Purpose To investigate the expression of EZH2 mRNA and protein, and its relationship with tumor cell proliferation in human prostate carcinoma (PCa).Methods A tissue microarray was constructed, which contained 68 dots of formalin-fixed,paraffin-embedded tissue samples, including 48 cases of human PCa.Immunohistochemical markers, including EZH2 and Ki-67,were used on the tissue microarray sections by the immunohistochemical staining method.In situ hybridization (ISH) using an EZH2 oligonucleotide probe was also performed on the tissue microarray sections.Additional 15 cases of benign prostate hyperplasia (BPH) and 12 cases of high grade prostate intraepithelial neoplasia (HGPIN) were used as controls.Results The positive rates of EZH2 protein and mRNA expression were 87.50% and 81.25% in the PCa, 16.67% and 16.67% in the HGPIN,13.33% and 6.67% in the BPH,respectively. There were statistical difference between PCa, HGPIN and BPH, respectively (P<0.05).The expression of EZH2 protein and mRNA had no statistical difference (P>0.05).The expression of EZH2 protein was related to Gleason score, TNM stage (P<0.05), but not to age and serum prostate-specific antigen (PSA) level (P>0.05). The expression of EZH2 mRNA was related to TNM stage (P<0.05),but not to age, PSA and Gleason score (P>0.05).The staining intensity of EZH2 was positively correlated with the Ki-67 indexes (r=0.746,P<0.05).Conclusions Over-expression of EZH2 which is involved in accelerating proliferation may play an important role in the pathogenesis and progression of human prostate carcinoma. As a molecular marker of prostate carcinoma, EZH2 may serve as a new index for estimating the level of malignancy and progression of the prostate cancer.
4.Research progress in strategy of lateral femoral offset reconstruction in total hip arthroplasty.

As an important indicator of total hip arthroplasty (THA) the rate and degree of offset reconstruction play an important role in improving the prognosis and life quality of patients. The reconstruction of femoral offset is closely related to reserved length of calcar femorale, the head and neck length of prosthesis, angle degree of neck shaft and whether lower limb is isometric. Reconstruction strategy includes making a meticulous and standard measurement before the surgery, predicting the reserved length of calcar femorale, selecting a prosthesis with approximate anatomical neck-shaft angle and reconstructing offset by adjusting the head and neck length of the prosthetic during the operation. The aim of this article was to introduce the research progress and influence of offset on hip function, prosthetic wear and postoperative complications such as pain, limp and unequal leg length, and to discuss the reconstruction strategy.

5.Pathogenesis and early drugs therapy of symptoms after concussion

Objective To study the pathogenesis and the effect of early drugs therapy of symptoms after concussion.Methods One-hundred and thirteen patients with symptoms after suffered concussion enrolled in the study and divided randomly into three groups.The patients of different groups were treated respectively with drugs,i.e.erigeron breviscapus,nimodipine,energy mixture.Symptom Check-List 90 ( SCL-90 ) and electroencephalogram(EEG) and cerebral blood flow of trancranial Doppler(TCD) were examined in the early and late stage of post-concussion.Normal population were recruited as control.All examination results and the effect of treatment were compared between treatment group and control group.Results In the scores of SCL-90,the factor scores of somatization symptom,compulsion,depressive disorder,anxiety disorders,and hostile symptom in 113 post-concussion patients in early stage were all higher than those in the normal( t =10.78,2.08,9.53,11.09 and 2.52,P <0.01or P <0.05),and as well as that of somatization symptom,depressive disorder,anxiety disorders,terror and mental disorder symptom in 12 post-concussion patients in late stage compared to normal( t =3.21,6.85,3.07,3.14 and 4.73,Ps < 0.01 ).The test of EEG was abnormal in 57.4 % of patients in early stage and in 33.3% in the late stage.Of all TCD results,67.0 % was abnormal in the early stage and 25.0% in the late stage,The disappearance rate of post-concussion symptoms in the 14th day was significantly higher in the erigeron breviscapus group ( 45.0% ) than in the nimodipine group ( 23.7% ) ( x2 =3.91,P <0.05 ) and in the energy mixture group ( 20.0% ) ( x2 =5.25,P < 0.05 ).The disappearance rate of postconcussion symptoms in the 30th day in the energy mixture group(40.0% ) was significantly lower than those in nimodipine group( 68.4% )( x2 =5.94,P < 0.05 ) and in erigeron breviscapus group( 75.0% )( x2 =9.43,P < 0.01 ).The occurrence rate of post-concussion sequelae was higher in the energy mixture group ( 22.9% )than in the nimodipine group ( 5.2% ) ( x2 =4.77,P < 0.05 ) and in the erigeron breviscapus group (5.0% )( x2 =5.15,P < 0.05 ) three month after injuries.Conclusion The occurrence of symptoms originated from the organic damage of brain tissue.However,its changes were influenced by psychologic factors after concussion.Early vasodilative drugs therapy could improve the recovery of patients with post-concussion symptoms in time and decrease the occurrence of post-concussion sequelae.
7.Effect of Numb gene on tubular epithelial-to-mesenchymal transition in rat proximal epithelial cells

Objective To explore the effect of Numb on tubular epithelial-to-mesenchymal transition (EMT) in rat proximal epithelial cells. Methods NRK52E cells were treated with different concentrations of recombinant human transforming growth factor-β1 (TGF-β1) (0, 1, 5, 10, 15, 20 μg/L) for 48 h or 10 μg/L TGF-β1 for different times (0, 24, 48, 72 h) in vitro. The expressions of E-cadherin, a-smooth muscle actin(α-SMA) and Numb in NRK 52E cells were detected by RT-PCR, Western blot and immunofluorescence staining. Meanwhile Numb siRNA oligo was transfected into NRK 52E cells with lipofectamine before TGF-β1 treatment, then Western blot was applied to detect the protein expression of E-cadherin, α-SMA and Numb in NRK52E cells. Results TGF-β1 could induce EMT in NRK52E cells in dose- and time-dependent manner. During the progress of TGF-β1-induced EMT, the protein expression of Numb in 5, 10, 15, 20 μg/L group was 1.33 folds (P=0.024), 1.39 folds (P=0.035), 1.45 folds (P=0.025), 1.51 folds (P=0.000) respectively as compared to 0 μg/L group. Likewise, the protein and mRNA expression of Numb in 24 h, 48 h, 72 h group was 1.48 folds (P=0.046) and 1.56 folds (P=0.012), 1.54 folds (P=0.011) and 1.82 folds (P=0.008), 1.79 folds (P=0.028) and 1.82 folds (P=0.002) respectively as compared to 0 h group. Moreover, large amount of Numb was accumulated in the cytoplasm. Down-regulation of Numb expression by siRNA transfection did not influence the basal expression of E-cadherin and α-SMA in NRK 52E cells, but attenuated the progression of EMT in NRK52E cells induced by TGF-β1. The up-regulation of α-SMA protein was reduced to 18.1% (P=0.004) while the down-regulation of E-cadherin protein was reversed to 2.19 folds (P=0.004). Conclusion Numb can promote EMT in rat proximal epithelial cells.
8.Expression of CD38 in T cells of chronic lymphocytic leukemia

Objective To explore the CD38 expression in T-cens(CD38-T)of chronic lymphocytic leukemia(CLL).Methods Multi-parameter flow cytometry was used to detect the expression of CD38,ZAP-70 and CD4/CD8 in 83 CLL patients.Results CD38 was positively expressed in 49.4%(41/83)T-cells(CD38-T)and 50.6%(42/83)tumor cells(CD38-B)of all CLL patients.The expression of the CD38-T was highly correlated with CD38-B(r=0.553,P<0.01).ZAP-70+CD38+-T and ZAP-70-CD38-T constituted 67.5%(56/83)of CLL patients.There was significant correlation between CD38-T and ZAP-70 (r=0.349,P<0.01).There was 33.3%(14/42)patients with CD38-T in Binet A patients,and 65.9% (27/41)in Binet B+C patients.There was also significant correlation between CD38-T and Binet B+C(r=0.312,P<0.05),and CD4/CD8(r=0.453,P<0.05).Conclusions Aberrant expression of CD38 in T-cells might be of prognostic relevance in CLL Most of the patients with high level CD38-T expression are accompanied with disorder or imbalance of immune function.CD38 expression in T cells might be a new prognostic factor in CLL.

Objective:To study the influence of Yizhiping Capsules on hemorrheology of experimental rats with hyperlipoidemia and its embryonic toxicity, teratogenicity. Methods:① The rat experimental hyperlipoidemia models were established by feeding with high lipid diet for 10 days. Meanwhile Yizhiping was taken orally at dosages of 75、150mg?kg -1 , some indices of hemorrheology markers were observed. ②According to the procedure of toxicology, the rat in the experimental groups were administered with Yizhiping capsules by gavage at a dose of 250、500、1000mg?kg -1 respectively, the embryonic toxicity and teratogenicity were observed. Objective To investigate the effect of β-elemene on SGC7901 gastric cancer cell line and the potential proteins involved. Methods Human SGC7901 gastric cancer cells were treated with different concentrations ofβ-elemene.Cell viability was assessed.A proteomic method,isobaric tags for relative and absolute quantitation (iTRAQ),was employed to detect the proteins altered by β-elemene.Protein expression was validated by Western blot.Results β-elemene inhibited the viability of SGC7901 gastric cancer cells in a dose-dependent manner.Altogether,147 upregulated proteins and 86 downregulated proteins were identified in response to β-elemene treatment in SGC7901 gastric cancer cell line.Among them,the expressions of p21-activated protein kinase-interacting protein 1 (PAK1IP1 ),Bcl-2-associated transcription factor 1 (BTF)and topoisomerase 2-alpha (TOPIIα)were validated by Western blot and the trends were consistent with iTRAQ results.Top pathways involved inβ-elemene treatment in SGC7901 gastric cancer cell line included ribosome signaling,peroxisome proliferator-activated receptors (PPARs)signaling pathway,regulation of actin cytoskeleton,phagosome,biosynthesis and metabolism of some amino acids.Conclusion Our results suggest a promising therapeutic role of β-elemene for gastric cancer.The differentially expressed proteins give us better insights into the potential mechanisms involved in gastric cancer treatment using β-elemene.
