OBJECTIVETo investigate the infection of Helicobacter pylori (Hp) in the liver tissue of patients with chronic liver disease and the association between Hp and chronic liver disease.
METHODSLiver tissue samples were obtained by liver biopsy and surgical resection from 30 healthy subjects, 30 patients with chronic hepatitis, 30 with cirrhosis and 30 with liver cancer. All the samples were confirmed by pathological examination. The gene fragment coding for 16SrRNA were amplified by PCR with sequence analysis.
RESULTSThe PCR product of the 16SrRNA gene was 109 bp in length. Hp 16SrRNA was detected in 18 out of 30 liver biopsy samples from patients with primary cancer (60.0%), in 14 samples from patients with liver cirrhosis (47.0%), and in none of the samples from normal subjects or patients with chronic hepatitis. Sequencing analysis of Hp 16SrRNA gene in the liver tissue showed a 98.8% homology with the gene fragment encoding Hp 16SrRNA.
CONCLUSIONHp is identified in the liver tissue of patients with chronic liver disease, suggesting the possible correlation between Hp infection and hepatocellular carcinoma.

Journal of Southern Medical University 2011;31(2):341-343
OBJECTIVETo establish standard serum proteomic pattern for early liver cancer and explore its value in early liver cancer diagnosis.
METHODSThe serum samples were obtained from 12 healthy subjects and 12 patients with early liver cancer. After removal of serum albumin and IgG, 300 µg serum sample was mixed with the hydrating solution for two-dimensional gel electrophoresis. Each experiment was repeated for 3 times. With silver staining and image analysis, the differentially expressed proteins were identified to establish the standard proteomic pattern for early liver cancer. The proteomics pattern was then tested in the diagnosis of 13 cases of chronic liver disease and 10 healthy individuals in a double-blind manner.
RESULTSThirty-eight differentially expressed protein spots were identified. Eleven protein spots with differential expressions by over 5 folds were analyzed to establish the standard proteomics pattern for early liver cancer, which showed a sensitivity of 92% and specificity of 90% in early diagnosis of liver cancer.
CONCLUSIONThe standard serum 2-DE proteomic pattern for early liver cancer shows distinct differences from that of healthy subjects. The differentially expressed proteins and their specific combination pattern may have the potential for diagnosis and prognostic evaluation of early liver cancer.

PURPOSE: The purpose of this study was to study the protective effect and influence of sodium hyaluronate (Na-HA) on mRNA expression of peroxisome proliferators-activated receptor gamma (PPAR-gamma) in cartilage of rabbit osteoarthritis (OA) model. MATERIALS AND METHODS: Forty eight white rabbits were randomly divided into A, B, and C groups. Group A was normal control group, B and C groups underwent unilateral anterior cruciate ligament transection (ACLT). The rabbits in group B were injected normal saline after ACLT; and Group C received intraarticular1% sodium hyaluronate (HA) injection 5 weeks after surgery, 0.3 mL once a week. At 11th week after surgery, all the rabbits were sacrificed. The cartilage changes on the medial femoral condyles were graded separately. Cartilage sections were stained with safranin-O and HE, and messenger RNA (mRNA) expression of PPAR-gamma was detected by using real time polymerase chain reaction (Real Time-PCR). RESULTS: Cartilage degeneration in group B was significantly more severe than in A and C injection group. The grey value of Safranin-O of B group was higher than A and C groups. Expression of PPAR-gamma mRNA in group B was higher than group A and C. CONCLUSION: This study shows that Na-HA has a protective effect on articular cartilage degeneration, and the inhibitory effect on the PPAR-gamma mRNA expression may be one of therapeutic mechanism of Na-HA.

Chinese Journal of Hematology 2012;33(1):34-37
OBJECTIVETo screen and identify differentially expressed proteins of mesenchymal stem cells (MSC) during endothelial differentiation.
METHODSMSCs were induced to endothelial differentiation with vascular endothelial growth factor (VEGF) and epithelial growth factor (EGF) mixture. The whole cell proteins were extracted and isolated by two-dimensional gel electrophoresis. After gel was analyzed by Imagemaster 5.0 software, differentially expressed proteins were partially selected and identified by MALDI-TOF-MS.
RESULTSThe differentiated MSC highly expressed endothelial cells related markers, CD31, CD34 and FVIIIAg were 56.8%, 38.8% and 14.5% respectively by flow cytometer. Compared with the primary cultured MSC, the differentiated cells differentially expressed 91 proteins. Among the 19 identified proteins, 11 up-regulated and 8 down-regulated, which include cytoskeletal proteins, such as myosin, filamin, vimentin and vinculin; cell metabolism enzymes, such as ORP-150, ERO1-α, Delta(3,5)-Delta(2,4)-dienoyl-CoA isomerase, protein disulfide-isomerase A3, FAS and enolase 3; nuclear factors, such as TAR DNA binding protein, guanine nucleotide binding protein and hypoxia up-regulated protein 1; VEGF receptors, such as KDR and so on.
CONCLUSIONSCytoskeletal proteins, metabolism enzymes and KDR were all involved in endothelial differentiation of MSC. These proteins may be the regulatory targets for endothelial differentiation of MSC.

OBJECTIVETo study the influential factors on psychosocial health of the migrant workers in Guangzhou.
METHODSThe Symptom Checklist 90 (SCL-90) and Eysenck Personality Questionnaire (EPQ) were used to investigate 518 migrant workers in Guangzhou.
RESULTSThe rate of migrant workers with psychosocial problems was 36.5%. The scores of SCL-90 and positive rates in migrant workers with the different personality types had significant difference (P < 0.01). The results of binary logistic regression analysis indicated that the working years, drinking, sex, P scores, E scores and N scores of EPQ were main predictors of the poor physical fitness status. The vocations, working years, P scores and N scores of EPQ were strong predictors of the somatization. he vocations, P scores and N scores of EPQ were strong predictors of the obsessive compulsive symptom. The smoking, P scores and N scores of EPQ were strong predictors of the interpersonal sensitivity. The working years, P scores of EPQ were strong predictors of the depression. P scores of EPQ was strong predictors of the anxiety. P scores, E scores and N scores of EPQ were strong predictors of the hostility. The working years, smoking, P scores, E scores and N scores of EPQ were strong predictors of the phobic anxiety. The working years, P scores of EPQ were strong predictors of the paranoid ideation. The working years, P scores and N scores of EPQ were strong predictors of the psychosis.
CONCLUSIONThe level of mental health of the migrant workers was significantly associated with the personality. The results of present study indicated that different vocation, sex, working years, smoking and drinking might interfere with the psychological states. The migrant workers with the personality of psychoticism, neuroticism and introversion may have unhealthy mental reaction.

Objective To understand the risk behavioral networks of newly reported HIV infections in Taizhou prefecture, Zhejiang province. Methods Newly reported HIV infections from May 2008 through March 2010 in Taizhou prefecture were invited to participate in a cross-sectional survey which requested numbers and contact information of individuals with whom they had had high risk contacts as well as risk behavioral acts with them. Persons having had risky contacts with HIV-infected cases were thereof approached to participate in this survey and to receive HIV testing. Those who tested positive for HIV were subject to further round of the surveys. Results A total of 267 HIV cases were newly reported during this study period. Among them, 191 participated in the survey and served as 'index cases', including 158 cases identified from routine HIV surveillance programs and 33 cases identified from the present survey. Heterosexual transmission was the primary transmission route (74.9%, or 143/191 ), followed by homosexual transmission ( 19.4%, or 37/191 )and injection drug use (5.8%, or 11/191 ). These 191 HIV cases reported a total of 1152 individuals with whom they had had risky contacts. They were able to provide contact information of 461 risk contacts. Of them, 129 received HIV testing and 61 (47.3%) tested positive for HIV. HIV prevalence was the highest among spouses or long-term sex partners of HIV cases (45.6%, or 47/103) and malesex partners of HIV-infected men having sex with men (MSM) (60.0%, or 12/20). Condom use wasvery low among them, with only 33.9% consistently using condoms for sex. Conclusion Newlyreported HIV infections in Taizhou prefecture reported a large and complicated risk behavioral networks and low condom use, suggesting a potential risk of HIV among these connected people, especially among MSM. Much efforts are needed to intervene these high risk subgroups and high risk behavioral networks.
