1.Differentiation of bone marrow mesenchymal stem cells induced by continuous cell culture

[Abstract ] Objective Bone marrow mesenchymal stem cells(BMSCs) can be induced to the differentiation into vascular smooth muscle cells in many induction conditions.We sought to explore the possibility of the differentiation of mesenchymal stem cells into vascular smooth muscle cells by continuous cell culture in vitro. Methods Rat BMSCs were isolated from the bilateral tibial and femoral bones by the method of whole bone marrow adherence, followed by ex vivo expansion.BMSCs were identified by flow cytometry and three-lineage differentiation.After continuous five days'cell culture of BMSCs, the specific surface antigens of VSMCs were detec-ted by immunofluorescence, western blot and real-time PCR. Results BMSCs expressed CD29、90, in contrast, they did not express CD45、34、49d.After induction of osteogenesis, adipogenesis and chondrogenesis, alizarin red、oil red and alcian blue staining pro-duced a strong reaction in cells.The expressions ofα-SMA、Calponin1、SM-MHC and SM22 in the cells of experimental group were no-tably increased, which indicated that BMSCs were differentiating towards VSMCs. Conclusion In the absence of exogenous stimula-tion, BMSCs can be successfully induced to differentiate into VSMCs by continuous cell culture.
2.Cytotoxic effects of the extractions of three gingival retraction cords with different extraction time

Objective:To evaluate the toxic effect of 3 different gingival retraction cords.Methods:DMEMextraction of DL-adren-aline HCl,aluminium sulphate and non-drug retraction cords with the extraction time of 5,10,15 and 30 min were respectively pre-pared and were used to culture human gingival fibroblasts(HGFs)in vitro respectively.Cell proliferation was tested by MTT assay. Cell apoptosis was examined by Annexin/PI method.Results:The 3 gingival retraction cord extractions inhibited the roliferation,pro-moted the apoptosis of HGFs(P <0.05),the effects were related to the extraction time.Conclusion:The 3 retraction cords have time-dependant cytotoxity.
3.Clinical Efficacy Observation of Humai Powder for Chemotherapy Phlebitis

OBJECTIVE:To observe the clinical efficacy of the Humai powder for chemotherapy phlebitis. METHODS:80 pa-tients with chemotherapy phlebitis were randomly divided into treatment group and control group with 40 cases in each group. Treat-ment group was given Humai powder for external use,1 h/time,2 times/d;control group was given Hirudoid cream for external use,2 times/day. Venous pain,red and swollen disappearance time,and overall clinical efficacy were compared between 2 groups after treatment 48 hours. RESULTS:The time of red and swollen disappearance and pain disappearance in treatment group were shorter than in control group,with statistical significance (P<0.05);there was no statistical significance in clinical efficacy (cure rate + significant efficiency)between 2 groups(P>0.05),while cure rate of treatment group was significantly higher than that of control group (P<0.05). CONCLUSIONS:Humai powder can relieve clinical symptom of patients with chemotherapy phlebitis and has high cure rate.
4.Pharmacokinetics and relative bioavailablity of domestic penicillin V dispersion tablet

Aim To study pharmacokinetics and bioavailablity of domestic penicillin V dispersion tablet in healthy volunteers. Methods According to the crossover design, each volunteer in two groups was orally given a single dose ( 0.75 g ) of domestic penicillin V dispersion tablet or imported penicillinV tablet alternately and the plasma concentrations were determined by RP HPLC. The pharmacokinetic parameters were obtained by using ATPK program and calculated on the basis of open single compartment model. Results After a single oral dose( 0.75 g ), the t 1/2(ke) was ( 0.75 ? 0.10 ) h and ( 0.70 ? 0.14 ) h ,the c max was( 8.44 ? 2.40 ) mg?L -1 and ( 8.75 ? 3.04 ) mg?L -1 at ( 0.56 ? 0.11 ) h and ( 0.63 ? 0.17 ) h and AUC 0~4 was( 8.44 ? 2.40 ) mg?h?L -1 and ( 8.75 ? 3.04 ) mg?h?L -1 for two formulations, respectively. Relative bioavailability of domestic penicillin V dispersion tablet was ( 90.50 ? 8.84 )%. Conclusion The result shows that the two formulations are bioequivalent.
5.Detection and significance of phosphatidylinositol 3-kinase in adipose tissue of polycystic ovary syndrome patients with insulin resistance

Objective To investigate the expression of phosphatidylinositol 3-kinase(PI-3K)in adipose tissue of polycystic ovary syndrome patients(PCOS),and explore molecular mechanisms of insulin resistance(IR)in PCOS.Methods Samples from patients with PCOS with IR(n=19),PCOS without IR (n=10)and controls(n=15)were collected.Serum fasting insulin(FIN)and fasting plasma glucose (FPG)were measure& Insulin resistance index was calculated using homeostasis model assessment (HOMA)to analyze the relationship between these markers and IR.Western blot technique was used to detect the PI-3K p85 subunit.Gene expression of PI-3K p85 subunit was detected by reverse transcription polymerase chain reaction(RT-PCR)method.Kinase activity was detected by immunoprecipitation,thin- layer chromatography and gamma scintillation counting.Results(1)The levels of FIN[(25.2?3.8) mU/L]and HOMA-IR(1.6?0.3)in PCOS with IR were significantly higher than those in PCOS without IR[(13.4 +3.8)mU/L,0.9?0.3]and controls[(9.5 +2.6)mU/L,0.5?0.3;all P<0.05).(2) There was no significant difference in the protein(0.65?0.10)and gene expression(0.92?0.12)of PI-3 K p85 subunit in PCOS with IR compared with PCOS without IR(0.72?0.10,1.01?0.10)and control groups(0.73?0.14,1.00?0.12;P>0.05).(3)PI-3K activity in PCOS with IR(81%)and PCOS without IR(89%)was significantly decreased(P<0.01,P<0.05)and negatively correlated with HOMA- IR(r=-0.69,P<0.01;r=-0.62,P<0.05).Conclusions No significant difference in the protein and gene expression of PI-3K p85 subunit in PCOS with IR is found.The decreased PI-3K activity may lead to IR of PCOS.

The First Affiliated Hospital of Zhejiang University founded an online hospital campus leveraging the Internet-plus environment. Supported by the offline hospital, this campus made full use of Internet features and advantages in its attempts to optimize the distribution of medical resources,to improve the accessibility of medical service, to reduce medical costs, and to expand the service coverage among others. Meanwhile,this online system, via interconnection with the hospital′s HIS system, achieved online medical insurance payment,prescriptions checking and drug delivery service.
7.Construction and evaluation of reference standards for detection and quantification of Klebsiella pneumoniae using real-time PCR

Objective To construct reference standards for detection and quantification of Klebsiella pneumoniae (K.pneumoniae) with SYBR Green I-based real-time PCR assay. Methods Primers were designed based on the published sequence of the phoE gene of K.pneumoniae. The standard was prepared by cell culture, PCR and T-A clone methods, and was identified by colony PCR and DNA sequencing. Results The standard curve showed a very good linear negative regression between threshold cycle (Ct) and Log starting quantity of copy number. The detection range was from 5.2 to 5.2×106 copies per reaction, and the detection limit was 6 copies per reaction. The coefficients of variance (CVs) of three parallel experiments were in the range of 0.05%-0.91%. Conclusion The reference standards have high stability and reproducibility. They can be used in the quantitative detection of K.pneumoniae.
8.Clinical Study on Gubenzhike Granule in Treating Cough Variant Asthma

Objective To observe the clinical curative effect of Gubenzhike granule on cough variant asthma (CVA), and explore its mechanism. Methods A total of 140 cases of CVA were randomly divided into two groups, 70 cases for each group. The treatment group took Gubenzhike granule orally, and the control group inhaled Budesonide Powder for Inhalation and Salbutamol Sulphate Aerosol, respectively for 8 weeks, with 8 weeks follow-up after treatment. The same nursing intervention was implemented in two groups. Cough symptom scores of the two groups were observed after treatment and at the end of follow-up, the number of eosnophils (EOS) and content of immunoglobulin E (IgE) in peripheral blood were also observed. Results The treatment group completed 67 cases and the control group completed 66 cases. After treatment, the cough symptom scores, EOS and IgE in two groups were significantly reduced (P<0.01, P<0.05), and the cough symptom score of treatment group decreased more significantly than that of control group (P<0.01). The total effective rate and recurrence rate were 91.04%and 9.84%in treatment group, and 83.33% and 30.91% in control group. The total effective rate of treatment group was better than the control group (P<0.01), and the recurrence rate was lower than the control group (P<0.01). Conclusion Gubenzhike granule showed significant effect and low recurrence rate on CVA. Good anti-inflammatory and antianaphylaxis effects may be one of its mechanisms.
9.Contribution of depression to instrumental ability of daily living among the elderly with mild cognitive impairment

Objective To examine the independent effect of depression on activities of daily living of patients with mild cognitive impairment (MCI). Methods Data were obtained through the face-to-face interview. Totally 154 elderly patients that were screened from 552 old people by the Montreal Cognitive Assessment (MoCA) were diagnosed as MCI. They were investigated with the Geriatric Depression Scale, and instrumental ability of daily living (IADL) were measured by the Functional Activities Questionnaire. The effect of depression on IADL of patients with MCI was analyzed by logistic regression analysis. Results The prevalence of depressive symptoms among Chinese elders with MCI was 31.82%. The scores of Medical Outcomes Study Social Support Survey, and Functional Activity Questionnaire of depression group and non-depression group were significantly different (P<0.05) . Logistic regression analysis showed that scores of Functional Activity Questionnaire was positively associated with the scores of Memory Inventory for the Chinese (OR=1.36, 95% CI: 1.15-1.61) and depression (OR=6.19, 95% CI:2.07-18.55) . Conclusion The prevalence of depression in elderly patients with MCI was high. Depression would significantly reduce the IADL of the old patients with MCI. Medical workers should be aware of the extent and impact of depressive symptoms in MCI and take active measures in evaluating and treating the depressive symptoms.
10.18F-FDG uptake changes in the brain functional loop in patients with refractory obsessive compulsive disorder

Objective To investigate the glucose metabolic pattern of brain functional loop in patients with refractory obsessive compulsive disorder (OCD) using 18 F-FDG PET.Methods Eight patients with refractory OCD and 8 age- and gender-matched healthy volunteers underwent 18F-FDG PET brain imaging.SPM software was used for image post-processing and quantitative analysis.Correlation analysis between 18F-FDG uptake and Yale-Brown obsessive compulsive scale(Y-BOCS) score was performed.Results Compared with the controls,the glucose metabolism of bilateral frontal cortices ( including the rectal gyrus,orbital gyrus and cingulate gyrus),left thalamus,right temporal lobe and bilateral cerebellum in refractory OCD patients increased significantly ( Zmax =3.45 - 5.80,all P < 0.001 ).Bilateral motor cortices and bilateral parietal lobes (BA7),however,showed decreased glucose metabolism (Zmax =3.44 - 4.46,all P <0.001 ).Y-BOCS score was positively correlated with the glucose metabolism of the bilateral anterior cingulate cortex (Zmax =3.77,3.48 and 2.97,all P < 0.01 ).Conclusions There is a characteristic metabolic pattern of increased glucose utilization in the fronto-striato-thalamic loop and decreased glucose utilization in bilateral motor cortices and parietal lobes in patients with OCD.The glucose metabolism in the anterior cingulate cortex might serve as a quantitative parameter for the assessment of the severity of OCD.
