Objective To study the spatial distribution character of brain evoked potential(BEP)in primary depressive illness(PDI)patients and the effect of Yi'nao Jieyu Formula.Methods Case control study and self-control study in Chinese herbal treatment group were conducted.Brain-stem auditory evoked potential(BAEP)and visual evoked potential(VEP)of 60 PDI patients and 45 healthy controllers were recorded by BEP apparatus made in Italy.BAEP and VEP of 30 patients of PDI group were detected again after taking Yi'nao Jieyu Formula for 6 weeks.Results Latencies of whole waves in BAEP and VEP prolonged significantly in PDI group(P

3.Hyperthermia treatment of dose-response relationship study in human cervical carcinoma Hela cells

Objective To investigate the dose-response relationship of the treatment temperatures and heating time on human cervical carcinoma hela cells,aiming at providing experimental evidences for clinical hy-perthermia. Methods Hela cells were heated at 37 ~ 70 ℃ in temperature-controlled water baths, the tempera-ture was divided into nine groups,each time was divided into eight subgroups (1 ~ 30 min). The morphology changes of cells after hyperthermia were detected by inverted microscope. Proliferation rates were measured by MTT colorimetric assay. The apoptesis rates were determined by flow eytometric analyse. The levels of prolifera-ring cell nuclear antigen (PCNA) were measured with immunohistochemistry. Results lnereaseing the heating time at the same temperature, or increaseing heating temperature at the same time, the cell proliferation, survival rates and PCNA expression decreased. There was no significant morphological change about cells ,but have small amount of apoptosis and a direct role of the suppression and destruction at 41 ℃ and 43 ℃ group. A large num-ber of cells shrinked to round and a major role for apoptosis at 46℃ group. Cell necrosis was major role at 50 ℃and 55 ℃ group. More than 55 ℃ for necrotic cells. Conclusion With the increase of heating temperature and heating time, its treatment of Hela cells gradually enhance. So combining dose-effect relationship of hyperthermia temperature and time can reach the best therapeutic effects.
4.Effects of Isopimaric Acid on cognitive handicap and synaptic plasticity in APP/PS1 mice

Objective To investigate the protective effects of isopimaric acid ( ISO), the BKCa channel activator, on cognitive function and synaptic plasticity in APP/PS1 mice. Methods Alzet osmotic pump was loaded with ISO or DMSO only and assembled with ALZET Brain Infusion Kit III. The cannula was implanted into the lateral ventricle of 4-month-old male APP/PS1 mice or matched wild type ( WT) mice. Two weeks later, open field test and Morris water maze were conducted. Paired-pulse facilitation ( PPF) and TBS-induced long-term potentiation ( LTP ) were recorded in CA1 region of hippocampus. Results The open field test showed that there was no significant difference among the four groups in spontaneous activities and vertical plane movement distance within 30 minutes. Floor plane movement distance was significantly greater in APP/PS1+DMSO group than that in WT+DMSO group(P<0.05) . Compared with the WT+DMSO group, APP/PS1+DMSO group had significantly longer escape latency from the third to fifth day and lower percentage of time spent in the target quadrant ((43.27±3.24)% vs (34.19±2.56)%) and the number of crossing through the platform ((4.25±0.66)times vs (1.93±0.33)times)(P<0.05). Compared with the APP/PS1+DMSO group, the APP/PS1+ISO group had significantly shorter escape latency from the fourth to fifth day and higher percentage of time spent in the target quadrant ((46.16±3.51)%) and the number of crossing through the platform ((3.41±0.34) times) (P<0.05). PPF in APP/PS1+DMSO group significantly reduced compared with that in WT+DMSO group at 30-50ms interstimulus interval(P<0.05). PPF in APP/PS1+ISO group((224.50±13.79)%) was significantly augment compared with APP/PS1+DMSO ((174.99 ±6.68)%) group at 40 ms interstimulus interval (P<0.05). The LTP at 60 min post-TBS was significantly smaller in the APP/PS1+DMSO group ((135.19±1.32)%) than that in the WT+DMSO group ((172.17± 4.15)%)(P<0.001). The LTP of the APP/PS1+ISO group((160.48±1.19)%) became significantly in-creased compared with that in the APP/PS1+DMSO group(P<0.001).Conclusion BKCa channel activator ISO improve the learning and memory function of APP/PS1 mice by promoting PPF and increasing LTP to recover synaptic plasticity in the hippocampus.
5.Effects of Yinao Jieyu Prescription on the Behaviors and Damages of Pathology in Hippocampal CA1 Area of Rats with Post-stroke Depression

Objective To investigate the intervention effects ofYinao Jieyu Prescription on the behaviors and damages in hippocampal CA1 area of the rats with post-stroke depression (PSD).Methods Totally 168 SPF male SD rats were randomly divided into normal group, sham-operation group, stroke group, PSD group, Western medicine group and TCM group. There were 24 rats in the normal group and sham-operation group, and 30 rats in the other groups. Rats in the normal group received no intervention. Rats in the sham-operation group received no suture. Rats in the stroke group were given middle cerebral artery occlusion operation and normally fed after operation. Rats in the PSD group, Western medicinal group and TCM group were made into PSD models by chronic immobilization stress for one week and individual battery to the end. At the inception of modeling, Western medicine group received fluoxetine hydrochloride for gavage; TCM group receivedYinao Jieyu Prescription for gavage; other groups received distilled water for gavage, once a day. At the end of week 2, 4, and 8, the morphology of the hippocampal CA1 area in each rat was observed by microscope after HE stained.Results Except for the week 2, at the same time point, the behavior scores of the rats in the TCM group were higher than those in the PSD group. At the same time point, the CA1 region of the hippocampus in the TCM group was more complete than the PSD group, and the cells were arranged neatly and in normal morphology.ConclusionYinao JieyuPrescription can improve the symptoms of PSD rats, and has protective effects on hippocampal CA1 area.
6.Optimization of Extraction Technology of Shen Qi Jie Yu Prescription with Orthogonal Test

Objective: To optimize the extraction technology of Shen Qi Jie Yu prescription. Methods: High performance liquid chromatography (HPLC) method was selected to determine the content of hesperidin of Shen Qi Jie Yu prescription of traditional Tang and granules. Decoction time, times and add water were used as the examining factors. L9 (34) orthogonal test was used as the best condition of extraction. Results: The hesperidin content in traditional Tang of Shen Qi Jie Yu prescription was significantly higher than granules. The order of the effect of hesperidin content is decoction time> decoction times>add water. There is a significant difference in the number of boiling times. The best decoction condition of Shen Qi Jie Yu Prescription was 3 times for decoction, with 0.5 hours of decoction and 7 times of water addition. Conclusion: The high performance liquid chromatography of Shen Qi Jie Yu Prescription is the quantitative method and the preferred decoction technique of Shen Qi Jie Yu prescription is stable and reliable.
7.Clinical efficacy of laparoscopic pyeloplasty with extracorporeal ureteral tailor

Objective:To evaluate the availability and safety of laparoscopic pyeloplasty with extracorporeal ureteral tailor.Methods:Clinical data of 26 patients with ureteropelvic junction obstruction (UPJO)who were treated by laparoscopic combined with extracorporeal ureteral tailor pyeloplasty in our hospital from March 2016 to August 2019 were retrospectively analyzed. There were 19 males and 7 females. 22 cases had unilateral lesion, including 6 cases on the right side and 16 cases on the left; 2 cases were bilateral. The average age was 22.3 years old (6-54 years). 19 cases felt discomfort in the renal region and 7 cases were asymptomatic. The mean body mass index was 21.7 kg/m 2 (17.2- 26.4 kg/m 2). 5 cases had mild hydronephrosis, 17 cases had moderate hydronephrosis and 4 cases had severe hydronephrosis. 6 cases combined with cross vessels. In all the 26 cases, the ureter was pulled out of the abdomen through a laparoscopic incision, cut lengthways in vitro, sutured at the lowest point, and then returned it to the abdomen. Then, double J tube implantation and ureteropelvic anastomosis were performed under the laparoscopy. Results:26 cases were completed successfully without conversion, with the average operation time of 99 minutes (50-158 minutes), the average blood loss of 19.4 ml (10-50 ml), the average hospital stay of 6.5 days (5-11 days), and the average drainage indwelling time of 5.3 days (4-10 days). For complications, urine leakage occurred on the 3rd day after the operation in 1 case, and the daily drainage fluid was more than 500 ml, which decreased suddenly after 4 days. Postoperative average follow-up was 10.8 months (6-24 months). Renal region pain disappeared in all patients. CT reexamination 3 months after the operation showed that hydronephrosis was alleviated or disappeared in 24 cases, and there was no significant change in 2 cases compared with the preoperative images, but no progress was found in the subsequent reexamination, so we didn't deal with the hydronephrosis again.Conclusions:Laparoscopic pyeloplasty combined with extracorporeal ureteral tailor is minimally invasive and flexible, which greatly reduces the difficulty and time of operation, and has a high success rate.
8.Effects of kidney-tonifying qi-regulating formula on time-course expression of bFGF in rat limbic system with cerebral ischemia

Objective To investigate the time-course expressions of the basic fibroblast growth factor ( bFGF) in limbic system of cerebral ischemic rats and explore the effects of Yishen Tiaoqi ( kidney-tonifying qi-regulating) Formula.Methods Male Wistar rats were randomly divided into normal group , sham-operation group, MCI group, treatment group and control group according to the scores of Open-Field test. The normal group received no intervention. The sham-operation group received sham operation, and intragastric administration of distilled water.Homogenous microthrombus were injected into internal carotid artery through the external carotid artery of those rats in other groups to duplicate MCI model first.The treatment group and control group received intragastric administration of Yishen Tiaoqi Formula (6.2g/kg) and Nimodipine (9.45mg/kg) respectively.The changes of limbic bFGF and intervention efficacy of Chinese medicinal were observed by using immunohistochemistry technique in the 1st Week,2nd Week and 4th Week.Results Immunohistochemistry test showed that positive cells of bFGF increased significantly in MCI group.The expression of bFGF was significantly higher in the first week than in other time points in MCI group, reached peak at day 7 and then decreased gradually.The positive cells of bFGF increased significantly in Chinese medicinal group compared with MCI group and normal group in the 2nd Week and 4th Week.Conclusion There are changes of limbic bFGF in rats after cerebrovascular disease, and Yishen Tiaoqi Formula can block mental disorder after stroke with improved vascular repair and regeneration.
9.Effects of method of nourishing heart and spleen on estrogen,progestin and TCM syndrome score in patients with postpartum depression

Objective To evaluate the efficacy and safety of Shenqi Jieyu formula (SQJYF),a Chinese medicine compound formulated by the method of nourishing heart and spleen,on patients with postpartum depression (PD),so as to provide more evidence for Chinese medication treatment no depression. Methods Altogether 60 PD patients with the syndrome of deficiency of both heart and spleen from the third affiliated hospital and three communities were included,who were randomly divided into treatment group and placebo group (n =30 each).The patients of the treatment group were taken orally SQJYF one dose daily,while patients of the placebo group were taken starch particles at the same dose for consecu-tive six weeks.The efficacy was assessed by using TCMDepression Scale as well as measuring serum es-trogen,progestin before treatment,at the first week,second week,fourth week and sixth week of treat-ment.Results The total effective rate of treatment group was 86.67%,while 73.33% of the placebo group;the clinical control rate of treatment group was 26.67% to 10.00% in the placebo group.There was significant difference in decreasing rate of TCM score between the two groups at the second week, fourth week and sixth week after treatment (P <0.05).The serum levels of estrogen both increased in two groups.There were significant difference between before and after treatment of each group,and the serum estrogen in treatment group was higher than that in placebo group(P <0.05).The serum levels of progestin both decreased in two groups.After six-week-treatment,the levels showed significant difference between before and after treat(P <0.05),while no difference was proved between the two groups(P >0.05).Conclusion SQJYF had the advantages of rapid onset and less side effects by decreasing TCM syndrome scores and the serum level of estrogen,and increasing the serum level of progestin,on PD depression patients with the syndrome of deficiency of both heart and spleen.
10.Lentivirus-mediated NDRG2 gene overexpression inhibits radioresistance of bladder cancer cells

Objective:The purpose of this study was to investigate the expression and role of N-myc downstream regulatory gene 2 (NDRG2) in radiation resistance of bladder cancer cells.Methods:T24 cells were cultured in vitro and irradiated with different doses of X-ray (0, 2, 4, 8, 10 and 20 Gy). The best dose of X-ray was selected for subsequent treatment. The radioresistant BCa cell line T24/R was established. The cytotoxicity of T24/R cells was detected by counting kit-8 (CCK-8) method. The proliferation and invasion ability of T24/R cells and T24 cells were detected by flow cytometry and transwell, respectively. Western blot was used to detect the expression of epithelial mesenchymal transition (EMT) related proteins. The survival rate of T24/R group (control group) and T24/R-NDRG 2 group was detected, and the migration ability of T24/R-NDRG 2 cells was detected after 2 Gy treatment. Results:The cell viability was inhibited significantly when the dose of X-ray was ≥2 Gy X-ray, so 2 Gy X-ray irradiation was chosen as the best condition for BCa cytotoxicity and T24/R radiation resistance cell line was successfully established; Apoptosis test showed that the number of S-phase cells was increased in T24/R group, and the proportion of S-phase cells in T24/R vs T24 was (26.49±4.5)% vs (14±2.6)% ( P<0.05); Transwell test showed that T24/R cells showed stronger migration ability than control group ( P<0.05), but there was no significant difference in EMT related protein expression between the two groups ( P>0.05). Overexpression of NDRG2 can significantly decreased the activity and migration ability of radiation-resistant T24/R cells ( P<0.05) when the radiation dose was gradually increasing in both groups. Conclusions:The radiation resistance of BCa cells is one of the causes of local tumor recurrence. Up-regulation of NDRG2 expression can inhibit the radiation resistance of T24 cells, so it can be used as a candidate for treatment of radiation-resistant BCa patients.
