Objective To explore the prenatal genetic diagnosis for classic phenylketonuria (PKU) families.Methods Probands and their family members from three classic PKU families were analyzed by combining linkage analysis through short tandem repeats (STR) polymorphism and PCR-sequencing for the exons within mutation hot spot of phenylalanine hydroxylase gene.Results Linkage analysis found uninformative for Family 1,while 100 % confirmative information was obtained from Family 2 and 3.Sequencing showed compound heterozygous mutations of phenylalanine hydroxylase gene for all of the three probands.Five mutations were detected,namely Y166X,R243Q,R413P,EX6-96A ＞ G and IVS11-1G＞ C,and IVS11-1G ＞ C was a novel identified muntation.Information from linkage analysis and mutation screening showed clearly that the fetus of Family 1 and 2 were affected,while normal for Family 3.Conclusions For those PKU families,reliable service of prenatal genetic diagnosis could be provided by combining linkage analysis with mutation screening of phenylalanine hydroxylase gene.

Objective To investigate the optimal time to remove the urinary catheter in patients undergoing laparoscopic adrenalectomy. MethodsPatients undergoing laparoscopic adrenalectomy were assigned to group A (6 h), group B ( 12 h), group C ( normal control group) with 20 cases in each. Subject feeling, adverse reactions, postoperative bed time and days of absolute diet were compared. Results The incidence of bacteriuria in group A (35％) and B (30％) were significantly lower than that of group C (85％)(F=14. 80, P =0. 000). The incidence of hematouria in group A (25％) and B (10％) were also significantly lower than that of group C ( 80％ ) ( F =22. 98, P =0. 000). Conclusions 6 - 12 hours after laparoscopic adrenalectomy are the optimal time to remove the catheter. In this manner, it can reduce the incidence of bacteriuria and hematouria, promote the postoperative recovery, thus to reduce the cost of hospitalization and improve the nursing work efficiency.

OBJECTIVETo investigate the clinical efficacy of Jianpi Yiqi Huoxue Decoction (JYHD) on children bronchial asthma complicated with recurrent respiratory infection (RRI) that could not be effectively controlled by regular inhalation of corticosteroid (ICS).

METHODSFifty-eight patients were divided into 2 groups: both were treated with anti-infective and symptomatic therapy, but JYHD was given additionally to patients in the treated group. Changes in clinical symptoms of RRI and asthma as well as the serum contents of IgA, IgG and IgM were observed.

RESULTSCompared with the control group, the frequency of respiratory infection occurred, duration of fever, cough and asthma sustained and time of antibiotics used were all lower significantly in the treated group (P < 0.05). The serum contents of IgA and IgG increased in the treated group after treatment, as compared with those before treatment and in the control group, all showed significant difference (P < 0.05).

CONCLUSIONFor children suffered from intractable bronchial asthma complicated RRI, application of JYHD in addition with the conventional treatment could prevent RRI effectively, so as th reduce the frequency of asthma attack and alleviate the severity of the disease.

Objective To investigate the apoptosis of T lymphocytes,the expression of Bcl-2, Fas on the peripheral blood T lymphocytes in end stage renal disease patients;and to explore the characteristics of Th1 /Th2 profile and the influence of dialysis membranes with different permeability on the apoptosis of T lymphocytes of maintenance hemodialysis patients.Methods The study included 10 non-dialyszed (ND)patients,45 maintenance hemodialysis patients with cellulose acetate (CA) membranes(13),low-flux polusulfone(PS-LF) membranes(16),high-flux polusulfone (PS-HF) membranes (16) and 8 healthy volunteers (C).The apoptosis of T lymphocytes,expression of Bcl-2,Fas on peripheral blood T lymphocytes cultured with phytohemagglutinin (PHA) stimulation for 24 hours were measured by flow cytometry and immunohistochemical.ELISA was performed for detecting the levels of IFN-?and IL-4 in culture supematants.Results In ESRD patients,the apoptosis of T lymphocytes was greater than that of group C.Group CA was greater than group PS-HF and group PS-LF (P＜0.05).The expression of Bcl-2 on T lymphocytes in ESRD patients was lower than that of group C (P＜0.05).There was negative correlation between the T lymphocytes apoptosis and Bcl-2. The expression of Fas on T lymphocytes in ESRD patients was greater than that of group C (P＜0.05), and it was positive correlated with T lymphocytes apoptosis.The level of IFN-?of ESRD patients was decreased significantly compared with that in group C (P＜0.05),and there was negative correlation between T lymphocytes apoptosis and IFN-?.IL-4 was increased in ESRD patients (P＜0.05) and it was positive correlated with T lymphocytes apoptosis.Conclusions The accelerated apoptosis of T lymphocytes in ESRD patients may be related to the expression of Bcl-2 and Fas of T lymphocytes.ESRD patients show a suppressed secretion of IFN-?and an increased secretion of IL-4. T lymphocytes apoptosis of maintenance hemodialysis patients is influenced not only by the biocompatibility but also by the permeability of the dialysis membrane.

Purpose To study the radioactive purity and activity of 131I labeled human single chain variable fragments antibodies (scFv) against anaplastic thyroid carcinoma (ATC),and to explore its distribution and radioimmunoimaging characteristics in tumor bearing nude mice model in vivo so as to provide a new method for anaplastic thyroid carcinoma diagnosis and treatment.Materials and Methods The nude mice model bearing human anaplastic thyroid carcinoma was constructed.The chloramine T method was used to label scFv with 131I and the Sephadex G25M was used for purification of labeled scFv.Labeling rate was determined by trichloroacetic acid method;radiochemical purity,room temperature stability and serum stability were examined using paper chromatography.131I-scFv was injected via tail vein in mice,and the distribution of 131I-scFv in body tissues and organs was analyzed at 12,24,48,72 h after injection.Static SPECT imaging was performed at 12,24,48,72 h after injection to observe the intratumoral accumulation of radioactivity.The SPECT/CT image fusion was performed when the tumor tissues were clearly visible.Results 131I-scFv was purified,and the labeling rate was 91.64％;the radiochemical purity was (93.3 ±0.3)％.The radiochemical purity of 131l-scFv placed at room temperature and the serum for 1,6,12,24 h were all ＞90％.The radioactive distribution of 131I-scFv in tumor,liver,kidney,intestine and blood was high.SPECT imaging showed 131I-scFv was selectively concentrated in tumor tissue;the target/non-target ratio was the highest at 48 h,and the imaging was most satisfactory.Conclusion 131I-scFv can be successfully prepared.SPECT imaging of 131I-scFv in nude mice model is satisfactory,which lays the foundation for further research in ATC diagnosis and treatment.

Objective To explore the clinical application value of early bundle therapy in patients with septic shock after per‐cutaneous nephrolithotomy(PCNL) .Methods The retrospective analysis was conducted patients with septic shock after PCNL ad‐mitted to the central ICU of the First Affiliated Hospital ,University of South China from January 1st ,2011 to september 30 ,2013 . The patients were divided into non‐bundle therapy group and bundle therapy group according to whether treated by early bundle therapy .the APACHE‐Ⅱscore and SOFA score in the before and 1 ,3 ,7 d after treatment ,mortality rate within 28 d and length of ICU were compared with both groups .Results 54 patients were enrolled in the study ,there were 28 and 26 patients in non‐bundle therapy group and bundle therapy group ,respectively .The clinical data of patients in both groups had no significant difference be‐tween the groups ,all P>0 .05 .Compared with the patients in non‐bundle therapy group ,the APACHE‐Ⅱscore and SOFA score in 1 ,3 ,7 d after treatment significantly decreased in bundle therapy group ,all P<0 .05 .mortality rate in bundle therapy group and non‐bundle therapy group were 15 .38% and 35 .71% ,respectively ,P<0 .05 ;and length of ICU were(9 .04 ± 4 .48)d and(7 .00 ± 2 .32)d ,respectively ,P<0 .05 .Conclusion Early bundle therapy can effectively alleviate the severity of the disease and reduce mor‐tality of patients with septic shock after PCNL .

Objective: To study the influence of inflammatory cytokine TNF-? on the expression of adhesion molecules and specific markers of rat MSCs, and to study the optimal stimulation of MSCs with inflammatory factors in inducing adhesion molecule expression which promotes migration of MSCs to the ischemic area in liver. Methods: The MSCs stimulated with different concentration of TNF-? were detected for adhesion molecules and stem cells markers on cell surface with the method of flow-cytometry, MSCs which were stimulated with the optimal concentration of TNF-? and labeled with 1, 1-Dioctadecyl-3, 3, 3, 3-tetramethylindocarbocyanine Iodade(DiI)were delivered intravenously to the rats whose liver was injured by ischemia, the liver function of the experimented animals were tested, and liver samples in the ischemic area were obtained, the number of MSCs was counted under a fluorescent microscope. Results: Stimulated with TNF-?, MSC ex-pression of VCAM-1 increased, while that of stem cell markers did not change markedly. Exposed to lower concentration of TNF-?, the adhesion ability of MSCs obviously increased and more MSCs rested in the ischemic areas in the rat liver, compared to the control groups. Conclusions: TNF-? can increase the expression of VCAM-1 on rat MSCs while exert little effect on the stem cell character of MSCs. Suitable concentration of TNF-? can promote MSCs migration to the damaged tissue, which provides rationale for the treatment of liver disease.
