1.Effects of inhibitors of glycosphingolipid biosynthesis D-PDMP on proliferation and differentiation of neural stem cells.

Aim To explore the effects of inhibitors of glycosphingolipid biosynthesis D-PDMP on the proliferation and differentiation of neural stem cells. Methods Neural stem cells cultured in vitro were treated with different concentration of D-PDMP. The effects of D-PDMP on proliferation and differentiation of neural stem cells were evaluated by counting of neurospheres size and cellular number, MTT assaying and the experiment of inducing differentiation.Results Neurospheres were smaller in size after treated with D-PDMP. The data of cellular counting and MTT assay suggested that D-PDMP could inhibit proliferation of neural stem cells and lead cells death in higher concentration, D-PDMP also reduced the differentiation ability of neural stem cells. Although differentiated cells were fewer, they could be induced into neurons and astrocytes.Conclusion The inhibitors of glycosphingolipid biosynthesis D-PDMP could inhibit the proliferation of neural stem cells, even lead cells death. It also reduced the differentiation ability of neural stem cells.
2.Analysis of factors influencing the health-related quality of life in patients with cervical cancer based on FACT-Cx(V4.0)

Objective To evaluate the health-related quality of life (HRQOL) and its influencing factors in pa tients with cervical cancer.Methods 120 inpatients with cervical cancer at Yunnan Tumor Hospital were investigated by functional assessment of cancer therapy-cervix (FACT-Cx),t-test,analysis of variance and multiple linear regression were used to analyze factors influencing the HRQOL.Results The total HRQOL score of patients was (104.88 ± 19.51),and the domain scores of physical well-being (PWB),social/family well-being (SWB),emotional well-being (EWB),functional well-being (FWB),cervical cancer subscale (CxS) were (20.41 ± 5.46),(17.17 ± 6.10),(15.73 ± 5.06),(13.34 ±5.83),(38.23 ±6.26),respectively.Based on multiple linear regression,the influencing factors of PWB were marital status and clinical stages,the influencing factor of SWB was marital status,the influencing factors of EWB were education levels and clinical stages,the influencing factors of FWB,CxS and total score were all marital status,clinical stages and education levels.Conclusions The HRQOL scores were influenced by a number of factors and it is significant to improve HRQOL in patients with cervical cancer by relieving symptoms and reducing treatment side effects according to patients' specialty,also psychological support and encouragement was essential.
3.Application of the Glazer pelvic floor muscle evaluation protocol to patients with spinal cord injury

Objective To identify differences in the anal sphincter surface electromyography (sEMG)variables in spinal cord injury (SCI) subjects with different severities. Methods One hundred and ten SCI patients' impairments were classified as ASIA A,B,C or D using the International Standards for the Neurological Classification of Spinal Cord Injury.The evaluation was pedormed using sEMG equipment with an inserted anal sensor electrode and the Glazer pelvic floor muscle sEMG protocol.The sEMG variables were recorded and compared. Results There was no significant difference in sEMG variables between groups A and B,but the mean and maximum sEMG values of groups C and D in flick contractions ( 1 s),tonic contractions ( 10 s),and endurance contractions (60 s) were significantly higher than those of group A.Compared with group C,the mean and maximum sEMG values of group D were all significantly higher. Conclusions The sEMG data from the anal sphincter during contractions decreases significantly after SCI.Glazer's pelvic floor muscle sEMG protocol is a noninvasive and convenient real-time assessment.It is a useful complementary tool for quantitative assessment of the pelvic floor muscles of SCI patients.
4.Relationship of Glazer Pelvic Floor Muscle Surface Electromyography and Items of Bowel Function for Patients with Spinal CordInjury

Objective To identify the relationship of pelvic floor muscle surface electromyography (sEMG) variables and the items ofthe International Bowel Function Basic Spinal Cord Injury (SCI) Data Set in patients with SCI. Methods 180 SCI patients were divided intoGroup A, B, C and D according to the International Standards for Neurological Classification of Spinal Cord Injury (American Spinal InjuryAssociation, ASIA). Testi was performed on 180 SCI subjects utilizing sEMG equipment, an inserted anal sensor electrode, and the Glazerprotocol. Questionnaire of the items in the International Bowel Function Basic SCI Data Set were conducted. Results The values of pelvicmuscle sEMG variables were significantly correlated with awareness of the need to defecate, major defecation method, average time requiredfor defecation, frequency of fecal incontinence, need to wear pad or plug (P<0.05), as well as the impairment scale of ASIA (P<0.01).Conclusion Glazer pelvic floor muscle sEMG protocol and questionnaire of the International Bowel Function Basic SCI Data Set are usefulcomplementary methods for the choice of hydrotherapy types in patients with SCI.
5.Treatment of Post-stroke Paralytic Ileus by Combining Lai's Tongyuan Needling Treatment with Fuzi Lizhong Decoction

Paralytic ileus is one of the post-stroke familiar complications, belonging to the field of TCM"Changjie".Tongyuan Needling is invented by Professor LAI Xin-sheng,including two laws of promoting governor vessel to nourish spirit and leading qi to its origin, which focuses on regulating original qi and views governor and concept vessels as the key of regulating yin and yang, and then to regulate qi activity and hold the essence through leading qi to its origin,and finally to achieve balance between yin and yang.Fuzi Lizhong Decoction mainly consists of Lizhong Pills and Aconiti Lareralis Radix Praeparata, which is composed of processed Aconiti Lareralis Radix Praeparata, Codonopsis Radix, Atractylodis Macrocephalae Rhizoma, Zingiberis Rhizoma and Glycyrrhizae Radix et Rhizoma, which is an important prescription for warming yang and tonifying kidney, invigorating qi and strengthening the spleen. This article introduced one case adopting the therapy combination of Lai's Tongyuan Needling and Fuzi Lizhong Decoction to treat post-stroke paralytic ileus, and then summarized the experience of clinical acupoint selection and prescription.
6.Effects of high-frequency electromagnetic field on morphology of hippocampal cells in female rats.

OBJECTIVETo analyze the effects of high-frequency electromagnetic field (HF-EMF, 30 MHz, 0-1600 V/m) on the apoptosis and ultramicrostructure of the hippocamp and demonstrate the cytotoxicity of hippocamp.
METHODS120 Wistar female adult rats were randomly divided into ten groups based on body weight with different levels of 30 MHz electromagnetic field (0, 25, 100, 400, 1600 V/m) for eight hours daily. Five group rats were irradiated for three days. The other five group rats were irradiated for fifty-six days. Weekly the rats were continuously exposed five days. The apoptotic rate of the hippocamp was detected with TUNEL System. Meanwhile, the ultramicrostructure was observed with the transmission electron microscope.
RESULTS(1) There was no significant difference on the apoptotic rate and pathological change of the hippocamp cell between the exposure and the control groups through short term experiment (P > 0.05). (2) The apoptotic rate of the granulocyte on the DG campus of the hippocamp in the 400 V/m group and the 1600 V/m group (0.165% +/- 0.049%, 0.189% +/- 0.049% respectively) were increased significantly (P < 0.01) through inferior chronic experiment compared with the control group (0.052% +/- 0.016%). Along with the increase of radiation dose, the ultramicrostructure of the neuron cell appeared more abnormal cells. Especially there were marked change on the neuron in the 1600 V/m group.
CONCLUSIONSThere is no association between cell apoptotic rate of the hippocamp and short period exposure to HF-EMF (30 MHz, 25-1600 V/m). However inferior chronic exposures to HF-EMF might induce the cytotoxicity, especially in the high dose exposure (1600 V/m) under our experiment.

Metastasis of renal cell carcinoma to mammary gland is rare clinically. Based on the retrospective analysis of 3 cases of metastatic clear cell renal cell carcinoma of the breast identified pathologically from January 2010 to June 2017 in Tianjin Medical University Cancer Institute and Hospital, we discussed the clinical manifestation, pathological characteristics, diagnosis, treatment and prognosis of metastatic breast cancer of renal cell carcinoma.
8.Correlation studies on three water testing methods-comparison of test results of turbidity and total chlorine

[Objective] To study the correlation between onsite rapid examination , online monito-ring and laboratory examination results for drinking water turbidity and total chlorine , and to obtain a basis for rational disposition of the three methods . [ Methods] A total of 87 sets of onsite rapid examination and online monitoring results were compared with laboratory examination results respectively by using paired t tests.Linear correlation coefficients between onsite rapid examination results and laboratory examination results , between online monitoring results and laboratory examination results were calculated .Corresponding linear regression equations were set up . [ Results] There were no significant differences found either between onsite rapid examination results and online monitoring results or between online monitoring results and laboratory examination results .Linear correlation coefficients showed that the degree of correlation be-tween onsite rapid examination results and laboratory examination results was higher than that between on -line monitoring results and laboratory examination results for both turbidity and total chlorine . [ Conclu-sion] Onsite rapid examination and online monitoring are both reliable water examination methods in health inspection .The characteristics of the three methods should be considered when making disposition decisions .One or more methods should be used to maximize working effects and efficiency .
9.Simvastatin inhibits apoptosis of endothelial cells induced by sepsis through upregulating the expression of Bcl-2 and downregulating Bax

BACKGROUND: Many studies have showed that apoptosis of endothelial cells plays a curial role in the progress of sepsis. But the role of simvastatin in apoptosis of endothelial cells induced by sepsis is not clear. The present study aimed to investigate the role of simvastatin in apoptosis of endothelial cells induced by sepsis and its mechanism. METHODS: Human umbilical vein endothelial cells (HUVECs) were randomly divided into three groups: control group, sepsis serum intervention group (sepsis group) and simvastatin+sepsis serum intervention group (simvastatin group). After 24-hour incubation with corresponding culture medium, the relative growth rate of HUVECS in different groups was detected by MTT assay; the apoptosis of HUVECs was detected by Hoechst33258 assay and flow cytometry; and the expression of the Bcl-2 and Bax genes of HUVECs was detected by PCR. RESULTS: Compared with the sepsis group, HUVECs in the simvastatin group had a higher relative growth rate. Apoptotic HUVECs decreased significantly in the simvastatin group in comparison with the sepsis group. Expression of the Bcl-2 gene in HUVECs decreased obviously, but the expression of the Bax gene increased obviously after 24-hour incubation with sepsis serum;however, the expression of the Bcl-2 and Bax genes was just the opposite in the simvastatin group. CONCLUSIONS: Our study suggests that simvastatin can inhibit apoptosis of endothelial cells induced by sepsis through upregulating the expression of Bcl-2 and downregulating Bax. It may be one of the mechanisms for simvastatin to treat sepsis.
10.miR-124 regulates radiosensitivity of colorectal cancer cells by targeting PRRX1.

OBJECTIVETo detect the expression of miR-124 in colorectal carcinoma (CRC) cells and tissue specimens and analyze its association with the radiosensitivity of the cells.
METHODSThe expression of miR-124 in CRC cell lines and tissues were detected using qRT-PCR. The effect of miR-124 in modulating cell radiosensitivity was assessed in CRC cells with miRNA-124 overexpression and miRNA-124 knockdown, and bioinformatics prediction and dual luciferase reporter system were employed to identify the direct target of miR-124.
RESULTSs miR-124 expression was down-regulated in CRC cell lines and tissues. CRC cells over-expressing miR-124 showed an obviously enhanced radiosensitivity, whereas miR-124 knockdown resulted in a reduced radiosensitivity of the cells. Bioinformatics prediction and dual luciferase reporter system verified PRRX1 as a direct target of miR-124, which regulated the radiosensitivity of CRC cells by directly inhibiting PRRX1.
CONCLUSIONmiR-124 can enhance the radiosensitivity of CRC cells by directly targeting PRRX1, which provides a target for improving the therapeutic effect of radiotherapy of CRC.
