1.Expression of α1-AT and VIEGF-C in human bronchoalveolar carcinoma

Objective To study the expressions of α1-AT and VEGF-C in human bronchoalveolarcarcinorrm, and the relation of the expression to the patholo~cM differentiation and clinical stage. Methods All 49 Darffin embedding samples of patients with bronchoalveolar carcinoma were studied. α1-AT and VEGF-C were detected by immunohistochemical SP method.Automated image analyzer was used to quantify α1-AT and VEGF-C expressions.Results The immunohistochemical positive stainings of α1-AT and VEGF-C in brown or dark brown were located in cytopla8m.The expression levels of α1-AT and VEGF-C were not related with the gender,age,tumor position and size,and histology subtypos(P>0.05).It Was found that the expression of α1-AT in patients with local lymph node metastasis was significantly lower than those without node metastasis(P<0.001).It was found that the expression of VEGF-C in patients with local node metastasis significantly higher than th08e without node metastasis(P<0.001).There Was a negative correlation between the expression level of α1-AT and the expression level of VEGF-C in bronchoalveolar carcinoma(r=-0.324,P<0.05).Conclusion α1-AT and VEGF-C could be secreted by bronehoalveolar carcinoma.Bronehoalveolar carcinoma with lower α1-AT expression and higher VEGF-C expression is more likely to have lymph node metastasis.Lower α1-AT expression and higher VEGF-C expression can participate in the mechanism of lymph node metastasis in carcinoma together.
2.The analysis of common mutation in deafness-associated gene in 111 neonates who failed to pass newborn hearing screening

Objective To explore the genetic mutation in neonates who failed to pass hearing screening.Methods A total of 111 cases of neonates who failed to pass hearing screening and were conifrmed sensorineural deafness by auditory brainstem evoked potential (ABR) were randomly selected. The heel blood was collected and DNA was extracted.GJB2, SLC26A4, and 11 mutation hotspots in mitochondria gene12SrRNA were tested. The relationship between degree of hearing loss and gene mutation was analyzed.Results In 111 neonates, mutation in deafness gene were found in 24 cases (21.6%) . Among them 14 cases (12.6%) hadGJB2 gene mutation including 5 cases of 235delC single heterozygous mutation, 5 cases of 235delC, and 1 case each of 299_300delAT compound heterozygous mutation, 235delC homozygous mutation, 299_300delAT single heterozygous mutation, 176_191del16 and 235delC compound heterozygous mutation, and 299_300delAT and 508_511dupAACG compound heterozygous mutation respectively. Ten cases (9.0%) hadSLC26A4 gene mutation including 2 cases of IVS7-2A>G single heterozygous mutation, 3 cases of 1226G>A single heterozygous mutation, 2 cases of 2168A>G single heterozygous mutation, and 3 cases of IVS7-2A>G and 2168A>G compound heterozygous mutation. Mitochondrial gene mutations were not detected. Conclusions Deafness gene mutation is detected in more than 1/5 neonates who failed to pass newborn hearing screening. GJB2 gene mutation is the most commons. The implementation of hotspots deafness gene detection can improve the diagnostic rate of deafness.
3.Relationship between cariogenic microbe, salivary buffer capacity and early childhood caries.

OBJECTIVETo investigate the relationship between Mutans streptococcus (MS), Lactobacilli (LB), pH value and buffer capacity of saliva and early childhood caries (ECC).
METHODSA total of 178 children aged from 42 to 54 months were recruited from 14 urban kindergartens in Beijing. The ECC group contained 87 children with more than 5 decayed teeth, and the control group was composed of 91 caries-free children. Unstimulated (UWS, 2 ml) and stimulated (SWS, 2 ml) whole saliva were collected in each subject. The pH value and buffer capacity of saliva were measured using an electro-acidimeter (+/- 0.01pH).
RESULTSMS and LB were isolated from 96.6% and 79.3% of children with ECC, which were significantly higher than those (63.7%, 27.5%) of caries-free children (P < 0.05) respectively. The counts of MS and LB in children with ECC were approximately 10 times higher than that in caries-free children. Initial pH value and buffer capacity of SWS were significantly higher than that of UWS (P < 0.001) in both groups. The pH value and buffer capacity of both UWS and SWS in ECC children were significantly higher than caries-free children (P < 0.05). There were no significant correlations between MS, LB and pH value and buffer capacity of saliva in caries-free children. Significant correlation (r = 0.249, P < 0.05) was found between the numbers of MS and buffer capacity of stimulated saliva in ECC children.
CONCLUSIONSMS and LB were important pathogens for ECC. Lower initial pH value and buffer capacity of saliva may be an important factor of ECC.

4.The Protective Effect of Sodium Hyaluronate on the Cartilage of Rabbit Osteoarthritis by Inhibiting Peroxisome Proliferator-Activated Receptor-Gamma Messenger RNA Expression.

PURPOSE: The purpose of this study was to study the protective effect and influence of sodium hyaluronate (Na-HA) on mRNA expression of peroxisome proliferators-activated receptor gamma (PPAR-gamma) in cartilage of rabbit osteoarthritis (OA) model. MATERIALS AND METHODS: Forty eight white rabbits were randomly divided into A, B, and C groups. Group A was normal control group, B and C groups underwent unilateral anterior cruciate ligament transection (ACLT). The rabbits in group B were injected normal saline after ACLT; and Group C received intraarticular1% sodium hyaluronate (HA) injection 5 weeks after surgery, 0.3 mL once a week. At 11th week after surgery, all the rabbits were sacrificed. The cartilage changes on the medial femoral condyles were graded separately. Cartilage sections were stained with safranin-O and HE, and messenger RNA (mRNA) expression of PPAR-gamma was detected by using real time polymerase chain reaction (Real Time-PCR). RESULTS: Cartilage degeneration in group B was significantly more severe than in A and C injection group. The grey value of Safranin-O of B group was higher than A and C groups. Expression of PPAR-gamma mRNA in group B was higher than group A and C. CONCLUSION: This study shows that Na-HA has a protective effect on articular cartilage degeneration, and the inhibitory effect on the PPAR-gamma mRNA expression may be one of therapeutic mechanism of Na-HA.

6.Health inspection and monitoring results analysis on secondary water supply in Shanghai

[Objective] To investigate the quality of secondary water supply in Shanghai , in order to provide the basis for efficient management measures . [ Methods] Secondary water supply data were collected and analyzed from Shanghai drinking water health inspection and monitoring information system . [ Results] Cleaning and disinfection of secondary water supply facilities and water quality self -check and others were found to be low in pass rate .The drinking water quality of secondary water supply was lower . The main unqualified monitoring indexes were oxygen consumption , total number of bacteria and residual chlorine. [ Conclusion] Several problems exist in secondary water supply .By using Shanghai drinking water health inspection and monitoring information system , we can take effective measures to achieve sec-ondary water supply scientific supervision , then ensuring water safety .
7.Observation on clinical therapeutic effect of acupuncture on upper limb spasticity in the patient of poststroke.

OBJECTIVETo observe therapeutic effect of acupuncture at acupoints selected according to rehabilitation medical theory on upper limb spasticity in the patient of poststroke.
METHODSSixty cases were randomly divided into an acupuncture group and an electro-stimulation group, 30 cases in each group. The acupuncture group were treated by acupuncture at the contralateral scalp motor region of the affected limb, Jiquan (HT 1), Chize (LU 5), Daling (PC 7) on the flexor side and Jianyu (LI 15), Tianjing (TE 10), Yangchi (TE 4) on the extensor muscle side of the affected limb; the electro-stimulation group were treated by electric stimulation. The two groups also were treated with necessary medical treatment and anti-spasm rehabilitation motor training. The course was 3 weeks. Modified Ashworth Scale for muscle spasm (MAS), modified Fugl-Meyer Assessment (FMA) for upper limb motor function, and Modified Barthel Index (MBI) for ability of daily living were used for assessment of the therapeutic effect.
RESULTSAfter treatment, the spasm was significantly alleviated, the motor function of the upper limb and daily living ability were significantly increased (P<0.01) in the two groups; after treatment, BMI scores in the acupuncture group was very significantly superior to that in the electro-stimulation group. The total effective rate was 93.3% in the acupuncture group and 86.7% in the electro-stimulation group, with no significant difference between the two groups.
CONCLUSIONProper acupuncture is an effective method for upper limb spasm in the patient of poststroke, and the therapeutic effect is better for mild-moderate spasm of the upper limb.

8.Comparison of salivary proteins between children with early childhood caries and children without caries.

OBJECTIVETo compare the concentrations of IgA, lactate dehydrogenase, lysozyme and alkaline phosphatase (ALP) in unstimulated (UWS) and stimulated (SWS) whole saliva between children with severe early childhood caries (S-ECC) and children without caries.
METHODSOne hundred and ninety-two children aged from 42 to 54 months were recruited from 11 urban kindergartens in Beijing. The S-ECC group contained 98 children with more than 5 decayed teeth, and the control group contained 94 caries-free children. The age and sex were matched in the two groups. Two milliliter UWS and 2 ml SWS was collected between 9 and 11 a.m. The salivary IgA was measured by immunoturbidimetric technique. The concentrations of lactate dehydrogenase and ALP were measured by continuous monitoring method, while lysozyme was detected by turbidimetric technique. All results for paired observations between unstimulated and stimulated whole saliva were analysed by paired-samples t test.
RESULTSIn both UWS and SWS, the concentrations of IgA, lactate dehydrogenase and lysozyme in S-ECC children were higher than those in caries-free children (P < 0.01), but the concentration of ALP showed no significant difference in SWS between S-ECC children and caries-free children (P > 0.05).
CONCLUSIONSThe presence of early childhood caries may be associated with an increase of IgA, lactate dehydrogenase and lysozyme in unstimulated and stimulated whole saliva.

9.Immune Regulation of Interleukin-27 in Malignant Pleural Effusion.

BACKGROUNDInterleukin (IL)-27 has been reported to have anti-proliferate and anti-angiogenic activities on cancer cells. However, the involvement of IL-27 in malignant pleural effusion (MPE) remains unknown. Thus, in this research, we compared the immune functions of IL-27, interferon (IFN)-γ, and IL-17 on lung cancer cells and revealed the regulatory mechanism of IL-27 in MPE.
METHODSThe distribution of IL-27 in both MPE and blood was evaluated by enzyme-linked immunosorbent assay and flow cytometry. The expressions of cytokine receptors and the levels of the phosphorylated signal transducer and activator of transcription (STAT) signalings were detected by flow cytometry. As well as the effects of proliferation, apoptosis, migration, and adherent activity of IL-27, IFN-γ, and IL-17 on lung cancer cells were also explored.
RESULTSThe expression of IL-27 was increased in MPE when compared with blood (147.3 ± 25.1 pg/ml vs. 100.3 ± 13.9 pg/ml, P = 0.04). IL-27 was noted to suppress both proliferation (18.33 ± 0.21 vs. 27.77 ± 0.88, P = 0.0005) and migration (1.82 ± 0.44 vs. 3.13 ± 0.07, P = 0.04) of A549 cells, but obviously promoted apoptosis of A549 cells (9.47 ± 1.14 vs. 4.96 ± 0.17, P = 0.02) by activating STAT1 signaling. Interestingly, IL-27 played totally opposite effects on A549 cells by activating STAT3 pathway. Moreover, IL-27 exerted different intercellular adherent activities of A549 cells to pleural mesothelial cell monolayer by activating different STAT signalings.
CONCLUSIONSIL-27 might exert an important immune regulation on lung cancer cells in human pleural malignant environment.

10.Synthesis and antitumor activity of selenophosphocholine analogues containing tegafur.

AIMTo synthesize the selenophosphocholine analogues containing tegafur and test their antitumor activities.
METHODSThe cyclic glyceroselenophospholopid conjugate of tegafur was synthesized by the reaction of hexaethylphosphorous triamide with N1-(2-furanidyl)-N3-(hydroxyalkyl)-5-fluyorouracil and 1-O-hexadecyl glycerol as well as selenium in one-pot. Cyclic glyceroselenophospholopid conjugate of tegafur reacted with triethylamine to give title compounds.
RESULTSSix new compounds have been synthesized. Their structures were confirmed by 1H NMR, 13P NMR and elemental analysis. Antitumor activity of the title compounds against PGA1 was tested.
CONCLUSIONThe reaction of triethylamine with cyclic glyceroselenophospholopid conjugate of tegafur very readily occurred, which was finished within 2 h at room temperature. The opening-ring products of trans isomers showed antimutor activity against human uriaryl bladder cancer cell more effective than that of the tegafur.
