1.Qualitative research of post-traumatic growth in caregivers of dementia patients

Objective To analyze post-traumatic growth of Chinese caregivers of dementia patients and to explore the caregivers′positive and potential energy, in order to formulate more perfect nursing intervention measures. Method Useing phenomenological method of qualitative research in selecting 6 dementia caregivers in-depth interviews to understand possible posttraumatic growth. Result Five major themes were found: changes in individual cognitive, changes in relations with others, promote their children′s education, reflections and concerns on status of pension-related changes in philosophy of life. Conclusions Dementia caregivers also have the positive performance of posttraumatic growth, in addition to the negative experience. Clinical nurses should actively explore the potential and power from the growth perspective. This is conducive to the caregivers with more health caring for life, so as to improve the quality of care, and ultimately improve the quality of life.
3.Expression of Dkk-3 and Cyclin D1 protein in hepatocellular carcinoma and their clinical significance

Purpose To investigate the expression of Dkk3 and Cyclin D1 protein in human hepatocellular carcinoma (HCC) and clinical significance.Methods Immunohistochemistry was used to detect Dkk-3 and Cyclin D1 protein expression level in 80 cases of hepatocellular carcinoma and corresponding para-cancer tissue.Results The expression of Dkk-3 in hepatocellular carcinoma was significantly lower than those in corresponding para-cancer tissue (P < 0.05) and the expression of Cyclin D1 in hepatocellular carcinoma was significantly higher than those in CoTesponding para-cancer tissue (P < 0.05).The up-regulation of Cyclin D1 and the down-regulation of Dkk-3 proteins were correlated with pathologic differentiation degree (P <0.05).There was a significant inverse correlation between Dkk3 and Cyclin D1 expression (P =0.044,rs =-0.226).Conclusion The abnormal expression of Dkk-3 and Cyclin D1 gene in human hepatocellular carcinoma suggest that Dkk-3 and Cyclin D1 gene may play an important role in the development and progression of the cancer.The combination deteetion of the two biomarkers may provide valuable data for diagnosis and prognosis estimation of HCC.
4.Minimally invasive treatment for multiple calculi in homolateral kidney and ureter

Objective To investigate surgical techniques of minimally invasive management for multiple calculi in the kidney and ureter. Methods Combined use of ureteroscopic lithotripsy(URSL) and minimally invasive percutaneous nephrolithotomy(MPCNL) was carried out in 21 patients with multiple calculi in homolateral kidney and ureter between January and October 2005 in this hospital.Results Percutaneous nephrostomy was required before the stone removal in 2 patients with pyonephrosis.Of the remaining 19 patients,all the ureteral calculi were completely removed on one session.And the renal calculi were removed by single session in 13 patients,by secondary percutaneous nephrolithotomy in 3 patients,and by tertiary nephrolithotomy in 2 patients.The combination treatment failed in 1 patient because of stricture of uretero-vesical orifice and a conversion to open surgery was needed.Conclusions Combination use of ureteroscopic lithotripsy and minimally invasive percutaneous nephrolithotomy is effective for the treatment of multiple calculi in homolateral kidney and ureter,offering advantages of minimal invasion,rapid recovery,and few complications.
5.Clinical analysis of nasal cavity and paranasal sinus diseases with symptoms of eyes as primary presentation in 39 cases

AlM: To improve the diagnosis and treatment of diseases associated with noses and eyes.
METHODS: To retrospectively analyze 39 nasosinusitis patients with the symptoms of eyes as the primary presentation during 2006. 3-2013. 6.
RESULTS:Among the 39 cases, 37 recovered, 2 cases with malignant tumor, which were confirmed by orbital CT scan, sinus CT scan and endoscopic examination.
CONCLUSlON: Some nasal disease without nasal symptoms in the early stage, so patients consult doctors for symptoms of eyes. lf oculists know little about these diseases, it may lead to a high misdiagnosis rate with those patients. Oculists should realize it and get more knowledge to afford the patients timely diagnosis and proper treatments. Please for otorhinolaryngologists' consultation if the patients receiving ineffective treatment in Department of Ophthalmology.
6.The application of subepithelial connective tissue graft in the treatment of gingival recession

Objective:To study the effect of subepithelial connective tissue grafting in covering an exposed root surface.Methods:Eight cases with gingival recession on upper and lower anterior teeth and premolars receited connective tissue grafting received connective tissue grafting were presented with 12 months of follow-up.Results:Initial gingival recession averaged 4.3 mm,with a mean probing pocket depth of 2.3 mm.At the 12-month follow-up,a mean of 3.0 mm of root coverage was observed(ie,71% root coverage,range 60% to 80%).Probing pocket depth averaged 1.6 mm,indicating a 3.6 mm gain of clinical attachment(range 2 to 5 mm).Conclusion:The application of connective tissue graft results in predictable root coverage and clinical attachment while maintaining shallow pockets.
7.Mechanism of the mesenchymal stem cells in treating the multiple organ dysfunction syndrome

Multiple organ dysfunction syndrome MODS (MODS) is the one of the major factors,which causing the death of the severe patients.More and more research proved that there are some relatonship be-tween MODS with the cellular apoptosis and necrosis.In recent years mesenchymal stem cells has been ap-plied to a variety of diseases as seed cells in the field of rehabilitation,and shows the initial effect in treat-ment and research.Through analysis the distribution and division of mesenehymal stem cells in the body,we found it may play an important role in the treatment in the MODS.We reviewed the mechanism of the MSC dealing with the MODS.
8.Construction and Identification of NBS1-targeting microRNA Expressing Eukaryotic Vector

Objective:To construct NBS1 microRNA expressing eukaryotic recombinants,and identify biological activity of recombinants in Hela cell after transfection.Methods:According to sequence of NBS1mRNA,the NBS1 pre-microRNA was designed and synthesized,then cloned into the GFP reporter pcDNA6.2-GW/EmGFP-miR vector and transfected into Hela cell line.To detect integrity of inset fragment through colony PCR and sequencing analysis.The biological activity of recombinants through identify interference efficiency of NBS1 microRNA recombinants by way of Real-Time PCR and Western blot were determined.Results:Sequences of inset fragment in four microRNA expressing recombinants were correct.NBS1 mRNA and protein expression of four microRNA recombinants were decrease,which is the lowest in the NBS1mi-2 group.Conclusion:Four NBS1-targeting microRNA expressing recombinants all have biological activity in Hela cell line,and NBS1mi-2 recombinant has the most interference efficiency.The microRNA expressing plasmid which were successfully constructed and lay foundation for the studies on the tumor gene therapy of microrna targeting NBS1.
9.Relationship between glucokinase gene 6 tag single nucleotide polymorphism sites and type 2 diabetes mellitus

Objective To investigate the relationships between Glucokinase (GCK) gene 6 (tag single-nucleotide polymorphisms,tagSNPs)sites which named rs12702070,rs2971672,rs2268569,rs2268573,rs2300587 and rs1476891 polymorphisms and type 2 diabetes in Chinese Southern Han Population.Methods This study was designed as a case-control.499 type 2 diabetes patients and 499 healthy controls were chosen.All subjects were from August 2013 to December 2014 in Zhongshan Affiliated Hospital of Sun Yat-sen University.6 GCK tagSNPs sites were analyzed by improved multiple ligase detection reaction (iMLDR),and genotype and allele frequency between T2D group and healthy controls could be determined by chi-square test,logistic regression analysis,and tagSNPs were further analyzed under three genetic modes(dominant,recessive and additive).What's more,Haploview software was used to construct the haplotype of 6 GCK tagSNPs and the linkage disequilibrium (LD) and relationship between various GCK haplotype and T2D susceptibility could be analyzed.Results Genotype distribution of rs2268573,rs2300587,rs2268569 and rs1476891 (x2 were 3.361,2.076,0.582 and 0.918 respectively,all P >0.05) and allele frequency (x2 were 0.222,1.980,0.590 and 0.851 respectively,all P > 0.05) in T2D group were no significant differences with health controls.Significant differences in genotype distribution of rs2971672 and rs12702070 (x2 were 6.896 and 7.990 respectively,all P < 0.01) and allele frequency (x2 were 4.708 and 5.979,P < 0.05 and P < 0.01 respectively) were observed between T2D group and health controls.Under dominant model (rs2971672:OR =1.74,95% CI =1.17-2.57,P < 0.01;rs12702070:OR =1.54,95 % CI =1.17-2.04,P < 0.01) and additive model (rs2971672:OR =1.51,95 % CI =1.06-2.14,P < 0.05;rs12702070:OR =1.26,95% CI =1.04-1.52,P < 0.05),Genotype distribution of rs2971672 and rs2971672 in T2D were significantly different from health controls.There are two LD domains in 5 tagSNPs among those 6 sites of GCK gene.There are three main haplotypes(TC,TA,CA)in rs2971672 and rs2300587,and four main haplotypes (TAG,TGG,TAT,CGG) in Rs2268569,rs12702070 and rs1476891.Although TAG,TGG,TAT and CGG have no relevance to the individual risk of T2D (P > 0.05),haplotype TA and CA reduce the individual risk of T2D with OR 0.81 (95% CI:0.66-1.00,P<0.05) and0.78 (95% CI:0.62-0.98,P <0.01)respectively.Conclusions The results indicated that GCK gene 2 tagSNPs sites included rs2971672 and rs12702070 imparts susceptibility to T2D in Han Chinese,but not rs2268573,rs2300587,rs2268569 and rs1476891.Haplotype TA and CA in rs2971672 and rs2300587 reduce the individual risk of T2D and four main haplotypes (TAG,TGG,TAT,CGG) in rs2268569,rs12702070 and rs1476891 have no relevance to T2D.
10.Characteristics of oral glucose tolerance test in 9 803 pregnant women of different pre-pregnancy body mass index and its relationship with the incidence of gestational diabetes mellitus

Objective To investigate the values and characteristics of 75 g oral glucose tolerance test (OGTT) in women with different pre-pregnancy body mass index (BMI) and to evaluate the risk of gestational diabetes mellitus (GDM).Methods Medical records of 9 803 pregnant women attending the Peking University First Hospital and delivered between July 1, 2011 and December 31, 2013 were retrospectively analyzed.The frequency of adverse pregnancy outcomes across different degrees pre-pregnancy BMI was calculated.We divided participants into 4 groups based on pre-pregnancy BMI, underweight: BMI<18.5 kg/m2 (1 221 cases), normal weight: 18.5-23.9 kg/m2 (6 594 cases), overweight: 24-27.9 kg/m2 (1 549 cases), obesity: ≥28.0 kg/m2 (439 cases).The diagnosis of GDM was made when any one of the values was met or exceeded in 75 g OGTT.The characteristics of 75 g OGTT and the incidence of GDM were analyzed.Results (1) The average age, pre-pregnancy weight, height and pre-pregnancy BMI of the participants was (30.5±3.7) years, (57.7±9.0) kg, (162.8±4.9) cm and (21.8±3.2) kg/m2, respectively.All the values of 75 g OGTT were presented normal distribution.(2)There was statistical difference in the glucose levels among women with different pre-pregnancy BMI.The fasting, 1-hour, 2-hour glucose were (4.55± 0.34), (7.31 ± 1.54), (6.38± 1.23) mmol/L in underweight women, (4.65 ±0.38), (7.70± 1.59), (6.70± 1.27) mmol/L in normal weight women, (4.82±0.47),(8.29±1.67),(7.04±1.29) mmol/L in overweight women and (4.94±0.48), (8.56 ± 1.64), (7.10 ± 1.35) mmol/L in obesity women (P<0.01).(3) The incidence of GDM was 21.76%(2 133/9 803) in our study.There were 1 374 cases (64.42%, 1 374/2 133)with only one abnormal OGTT value while 759 cases (35.58%, 759/2 133)with two or more abnormal values.The incidence of GDM in women with underweight, normal weight, overweight and obesity was 12.53%(153/1 221),19.71%(1 300/ 6 594),32.73% (507/1 549) and 39.41% (173/439), respectively (P<0.05).Meanwhile, the incidence of women with two and more abnormal OGTT value in GDM was increased as the pre-pregnancy increasing.Conclusion The risk of GDM is increased as pre-pregnancy BMI increasing, and the risk of GDM increases significantly in women with pre-pregnancy overweight or obesity.
