Objective To evaluate the clinical effect and safety of Huangzi Zhike granule for acute bronchitis with the syndrome of wind-cold attacking the lung.Methods A multi-center,randomized,double-blinded,double-simulation and positive drug parallel controlled trial was adopted.Three hundred and nine cases in the treatment group were treated with Huangzi Zhike granule,and 103 cases in the control group were treated with Tongxuan Lifei oral liquid.Results Huangzi Zhike granule had a total effective rate of 95.15 %and significantly relieved the symptoms of cough,spitting thin clear white sputum and throat itching.Compared with Tongxuan Lifei liquid,Huangzi Zhike granule showed a better clinical effect(P

4.Protective effect of intravenous infusion injection of safflor yellow and hydroxyl safflor yellow A on acute myocardial ischemia injury in rats

Objective The comparison research of protective effect between intravenous infusion injection of safflor yellow and hydroxyl safflor yellow A on acute myocardial ischemia injury in rats. Methods Qualified 60 male Wistar rats were divided into 5 groups at random (12 in each group):Blank control group, AMI model (treated with normal saline) group, intravenous infusion injection of safflor yellow (90 mg/kg) group, HSYA low dosage (20 mg/kg) group and high dosage (40 mg/kg) group. The acute myocardial ischemia injury in rats was induced by ligating the anterior descending coronary artery in Wistar rats and administered different dosage of safflor yellow and hydroxyl safflor yellow A with intraperitoneal injection. Myocardial infarction degree (MID) was calculated by detecting myocardial infarction area with nitroblue tetrazolium(NBT) assay. The changes of ST-elevation, CK-MB, cTnT, SOD activities and MDA contents were detected and analyzed. Results The intravenous injection of safflor yellow and HSYA low dosage group can significantly decreased ST-elevation [difference is (0.087?.022)mv,(0.091?.014)mv],MID[difference is (20.13?.17)%,(18.36+9.38)%], CK-MB [difference is (1460.70+219.73)U/L, (1472.72?85.61)U/L], cTnT[difference is (2.345?.883)ng/ml, (2.358?.843)ng/ml], and MDA [difference is(5.71 ?.67) mmol/ml, (5.76?.84)mmol/ml] contents in serum, increased SOD[difference is(58.27?.99)U/ml,(56.49+5.19)U/ml]activities in serum.Conclusion It showed that intra venous injection of safflor yellow and hydroxyl safflor yellow A had the same protective effect on acute myocardial ischemia injury in rats. Hydroxyl safflor yellow A is an important portion of safflor yellow.
5.Detection p16INK4a protein in cervical tissues with time-resolved fluoroimmunoassay

Purpose To establish a new method for detecting p16INK4a in cervical tissues with time-resolved fluoroimmunoassay (TRFIA).Methods 126 cases of paraffin imbedding tissues of cervix were selected for immunohistochemistry (IHC) of EnVision two-step and TRFIA.Results There were 20 cases of no intraepithelial lesion or malignancy,24 cases of low-grade squamous intraepithelial lesion (LSIL),53 cases of high-grade squamous intraepithelial lesion (HSIL) and 29 cases of squamous cell carcinoma (SCC).In the groups of no intraepithelial lesion or malignancy,LSIL,HSIL and SCC,p161NK4a positive was seen in 1,19,53 and 28,respectively.TRFIA test results displayed p16INK4a positive in 3,17,50 and 27 cases,respectively.Positive of p16 using by TRFIA in no intraepithelial lesion or malignancy,LSIL,above HSIL was 15.00%,70.83% and 93.90%,respectively (P < 0.01).Conclusion TRFIA is suitable for detecting of p16INK4a protein and demand low detection equipment,p16INK4a expression detected by TRFIA may helpful for large scale detection in various clinical institution.
6.Adsorption Performance in Binding of Methylene Blue in Aqueous Solution by Orange Peel

Orange peel was used as lowcost adsorbent in binding of Methylene Blue.The effects of equilibrium time,pH,dye concentration have been studied.Carboxyl,amine and phosphonate functional groups were present in the orange peel.The equilibrium time was 1 hour,the maximum adsorption capacities of the orange peel was 370.3?31.0 mg/g at pH 10.The Langmuir and Freundlich isotherms were well fitted in this biosorption system.Results showed relatively higher rate constant and biosorption capacities.These adsorption performance indicate the orange peel as a potentially economical adsorbent for dye removal.
7. Effects of cistanche phenylethanol glycosides on the cardiac hypertrophy and PI3K/PKB signaling pathway in rats with pressure overload

Aim To investigate the inhibitory effect of cistanche phenylethanol glycosides (CPhGs) on cardiac hypertrophy in rats caused by pressure overload and its related mechanism. Methods Male SD rats(n =70) were randomly divided into control group (Con), sham operation group (Sham), model group (Mod), positive control group (Vst), and different CPhGs dosage (125, 250, 500 mg • kg-1) groups. Cardiac ultrasound indexes, heart-weight to body-weight index (HWI), cardiac histopathological changes, and the area of myocardical cells (AMC) were detected. Plasma ET-1 and BNP levels were detected by Elisa, and protein expressions of phosphorylated PI3K(p-PI3K), PI3K, phosphorylated PKB (p-pKB) and PKB were detected by Western blot. Results Compared with Mod group, LVPWT, HWI, plasma ET-1, BNP and AMC decreased to different degrees. LVEDD, LVEF, LVFS, the protein expressions of myocardial tissues pPI3K and p-PKB increased to different degrees in CPhGs groups. Moreover, the indexes of CPhGs 250 and 500 mg • kg-1 groups were significantly improved compared to those of Mod group (P < 0. 05 or 0. 01). Compared with Vst group, there were no significant difference in CPhGs 500 mg • kg-1 group. Conclusions CPhGs could inhibit cardiac hypertrophy in rats induced by pressure overload, which might be related to the activation of PI3K/PKB signaling pathway.

9.Effects of α7nAChR on CD11b and inflammatory cytokines in mice with acute respiratory distress syndrome

Objective To investigate the effects of α7 nicotinic acetylcholine receptor (α7nAChR) on CD11b, IL-1β, IL-18 and TNF-α in acute respiratory distress syndrome (ARDS) mice. Methods A total of 40 healthy and clean male Balb/C mice (6 weeks old) were randomly divided into normal group (N group), normal saline control group (NS group), ARDS group (A group), and ARDS mice treated with nicotinic acetylcholine receptor agonist after bilateral cervical vagotomy group (J group), with 10 mice in each group. The right lung structure of mice in each group was observed by hematoxylin-eosin (HE) staining, the lung tissue wet weight/body weight ratio (LWW/DW ratio) was detected, and the percentage of CD11b in the alveolar lavage fluid of mice was detected by flow cytometry. Real-time fluorescence quantitative polymerase chain reaction (RT-qPCR) was used to detect the expression of IL-1β mRNA, IL-18 mRNA and TNF-α mRNA in left lung tissue. Serum IL-18 was determined by enzyme-linked immunosorbent assay (ELISA) and double antibody sandwich method. Results HE staining of the right lung of mice in group N and NS showed normal structure, while the lung interstitial of mice in group A showed a large number of inflammatory cells infiltrated, alveolar wall thickened, alveolar structure destroyed and alveolar cavity fused. The alveolar structure of mice in group J was intact, with a little damage and alveolar cavity. The percentage of CD11b in alveolar lavage fluid in group A was higher than that in the other three groups, and the difference was statistically significant compared with group N, NS and J, respectively (P<0.05). The expressions of IL-1β mRNA, IL-18 mRNA and TNF-α mRNA in the left lung of mice in group J were statistically significant compared with those in group N, NS and A (P<0.05), and the serum IL-18 level of mice in group A was higher than that in the other three groups, and the differences were statistically significant compared with groups N, NS and J, respectively (P<0.05). Conclusions Activation of α7nAChR can directly inhibit the release of CD11b in lung tissue and reduce the accumulation of inflammatory factors. Simultaneously, it can also directly inhibit the expression of IL-β1 mRNA, IL-18 mRNA and TNF-α mRNA in lung tissue and the release of IL-18, thus inhibiting the inflammatory response of ARDS and alleviating the pathological changes of ARDS.
10.The mechanism of rosiglitazone compound based on network pharmacology.

Applications of network pharmacology are increasingly widespread and methods abound in the field of drug development and pharmacological research. In this study, we choose rosiglitazone compound as the object to predict the targets and to discuss the mechanism based on three kinds of prediction methods of network pharmacology. Comparison of the prediction result has identified that the three kinds of prediction methods had their own characteristics: targets and pathways predicted were not in accordance with each other. However, the calcium signaling pathway could be predicted in the three kinds of methods, which associated with diabetes and cognitive impairment caused by diabetes by bioinformatics analysis. The above conclusion indicates that the calcium signaling pathway is important in signal pathway regulation of rosiglitazone compound, which provides a clue to further explain the mechanism of the compound and also provides a reference for the selection and application of methods of network pharmacology in the actual research.
