1.Evaluation value of miRNA-21 and miRNA-431 in the development and prognosis of pulmonary tuberculosis

Objective To evaluate the value of bronchoscopy specimens' microRNA-21 (miRNA-21) and microRNA-431 (miRNA-431) in the development and prognosis of pulmonary tuberculosis. Methods A total of 43 confirmed pulmonary tuberculosis patients admitted to Shaanxi Provincial Tuberculosis Prevention Hospital from May 2022 to July 2023 were included as the study group, and 43 healthy volunteers during the same period were included as the control group. In the study group, the severity of lung lesions was classified as mild in 14 cases, moderate in 19 cases, and severe in 10 cases. According to the prognosis, patients were divided into poor prognosis and good prognosis groups. The miRNA-21 and miRNA-431 levels of patients with different disease severity and prognosis in the study group and the control group were compared, and the factors affecting the development and prognosis of tuberculosis were analyzed by logistic multivariate analysis, and a prediction model for the development and prognosis of tuberculosis was established. Receiver operating characteristic (ROC) curve analysis was performed to evaluate the predictive value of the model. Results The levels of miRNA-21 and miRNA-431 in the study group were significantly higher than those in the control group, and the difference between groups was statistically significant (P<0.05). In the study group, the levels of miRNA-21 and miRNA-431 were positively correlated with the severity of the disease, and the levels in severe pulmonary tuberculosis patients were significantly higher than those in mild and moderate pulmonary tuberculosis patients, with statistically significant differences between the groups (P<0.05). In the study group, the levels of miRNA-21 and miRNA-431 in patients with pulmonary tuberculosis with poor prognosis were significantly higher than those in patients with pulmonary tuberculosis with good prognosis, with statistically significant differences between the groups (P<0.05). Using miRNA-21 and miRNA-431 expression as independent variables, and the development and prognosis of pulmonary tuberculosis as dependent variables, logistic regression analysis was performed. The results showed that the high expression of miRNA-21 and miRNA-431 were both influential factors for the development and prognosis of pulmonary tuberculosis (P<0.05). The ROC curve analysis showed that the optimal cutoff value of the prediction model was 0.0895, and the area under the curve (AUC) of miRNA-21, miRNA-431 and combined were 0.614 (0.651-0.732), 0.671 (0.686-0.811), and 0.891 (0.787-0.993). Conclusions The high expression levels of miRNA-21 and miRNA-431 in patients with tuberculosis are closely related to the development of the disease and can predict the prognosis of patients with tuberculosis to a certain extent. The comprehensive application value of combined diagnosis is high, and it is recommended to promote clinical use.

3.Establishment of a rabbit model of oral soft tissue defects

BACKGROUND: In clinical, the research on the method and the material of the soft tissue defect of the operation area has been in depth. It wil have a positive impact on our exploration and research in this field through the establishment of reliable experimental animal oral soft tissue defect model. OBJECTIVE: To establish a rabbit model of oral soft tissue defects for oral treatment of soft tissue defects in-depth study. METHODS: Eighteen 3-month-old male New Zealand rabbits were enrol ed. A tissue ring cutting dril of 5 mm diameter was used to prepare round ful -thickness soft tissue defects in the front part of the hard palate and, respectively, from the back of the maxil ary incisors, about 2 mm from the hard palate mucosal edge. Morphology and histology were observed at 3, 7, 14, 21, 28 and 56 days after model establishment. RESULTS AND CONCLUSION: (1) Morphological observation: After 3 and 7 days, the wound’s inflammatory reaction was obvious. As the time goes, the inflammatory response subsided, the wound gradual y healed. Scar formation was observed at 21, 28 and 56 days after surgery. (2) Histological observation: 3 and 7 days after injury, many inflammatory cel s were infiltrating, and tissue necrosis area was larger. At 7 days after surgery, with the extensive formation of connective tissue proliferation and new blood capil aries, the wound gradual y shaping completely. (3) Results indicated that a rabbit model of oral soft tissue defects was successful y established, which was in line with the physical laws of wound healing and the characteristics of human oral soft tissue defects healing.
4.Research progress of immunoglobulin receptors as new therapeutic target

Immunoglobulins ( Ig ) , also called antibodies, are important components in humoral -mediated immunity. Ig can bind with their receptors, called immunoglobulin receptors ( IgR ) , trigger biologic activities respectively. Different sub-types of Igs show different function. And IgRs have been treated as therapeutic targets in inflammation and immunity related dis-eases for many years. This article reviewed the recent progresses in the study of IgR function and its therapeutic role in inflamma-tion and immunity related diseases.
5.Platelet-rich fibrinversus collagen membrane in the repair of gingival defects

BACKGROUND:Previous studies have found that platelet-rich fibrin has a good ability to induce gingival soft tissue repair and regeneration.
OBJECTIVE:To observe the effects of platelet-rich fibrinversus colagen membrane on gingival soft tissue healing, and to evaluate the ability of platelet-rich fibrin to repair gingival defects.
METHODS:Twenty-two patients (2 premolar teeth and 20 molars) scheduled for premolar or molar removal or ridge preservation due to various reasons were selected and randomized into two groups. Bio-Oss was implanted into the extraction socket folowed by covering with platelet-rich fibrin or colagen membrane. Healing time and healing rate of gingival defects were detected to evaluate the ability of platelet-rich fibrin to promote gingival tissue healing at 1-2 weeks after Bio-Oss implantation.
RESULTS AND CONCLUSION:The healing time was (12.17±2.25) days in the platelet-rich fibrin group and (17.30±2.58) days in the colagen group. The healing rate of the platelet-rich fibrin group was notably higher than that in the colagen membrane group at 1 and 2 weeks after Bio-Oss implantation. These findings indicate that platelet-rich fibrin is better than colagen membrane to improve the healing of gingival soft tissues with a shorter healing time.
6.Effect of Huangshao Yinxue Granula on psoriasis-like animal models of animals

AIM:To study whether or not Huangshao Yinxue Granula(HYG) has the therapeutic effect on experimental psoriasis. METHODS: The mice were treated with HYG 3.125,6.25,12.5,and 25 g/kg(i.g.) for 1 or 2 weeks.We studied the effects of HYG on mitosis in mice vagina epithelium,the formation of granular layer in scale epidermis of mice tails,the changes in CD3, CD4 and CD8 factors of the serum in psoriasis-like mice and guinea-pigs. RESULTS: HYG could inhibit the mitosis in mice vagina epithelium,promote the formation of granular layer in scale epidermis of mice tails.It could also diminish the value of serum CD4/CD8 of mice to regulate abnormal immune reaction.HYG had evident therapeutic effect on the ear skin damaged with 5% propanolol in psoriasis-like guinea-pigs. CONCLUSION: HYG has therapeutic effect on psoriasis-like animal models.
7.Resting-state fMRI study of adolescents with conduct disorder: an ALFF analysis

Objective To investigate the functional characteristic of adolescents with conduct disorder (CD) in the resting state.Methods Resting-sate fMRI (rs-fMRI) scans were administered to 28 male adolescents with CD and 28 age-,gender-and IQ-matched healthy controls (HCs).The rs-fMRI data were subjected to amplitude of low-frequency fluctuation (ALFF) analysis and ALFF values were compared between the two groups.Additionally,the region ROIs showed significant difference and correlation analysis was conducted between ALFF in each ROI and Barratt's scores.Results ① Compared to HCs,the CD group showed decreased ALFF (P<0.005) bilaterally in the superior frontal gyrus,right middle frontal gyrus,right posterior cingulate gyrus,right inferior parietal lobule (MNI coordinates:-21,48,27;21,24,48;36,24,33 6,-36,39;48,-45,57 respectively) as well as increased ALFF (P<O.005) in the left thalamus and left lingual gyrus (MNI coordinates:-15,-27,-3;-18,-87,-12 respectively).② There was no significant correlation between ALFF and Barratt's scores in ROIs.Conclusion The results suggest that CD is associated with abnormal intrinsic brain activity,mainly in frontal-parietal-occipital-limbic cortices,which is related to emotional and cognitive processing and behavioral functions.
9.Effect of different scan parameters and algorithm on detection of iron deposition coexiting in fat by dual source CT

Objective To explore the effect of different scan parameters and algorithm on detection of iron deposition with coexsiting fat by dual soure CT.Methods One ml homogenate of normal rat liver tissues and 1 ml iron dextran solution with different concentrations (50.000,25.000,12.500,6.250,3.125,and 1.560 mg/ml) were fully mixed respectively.The 6 test tubes were scanned by dual source CT in the mode of dual energy to gain the iron specific slope.Different concentrations of dextran iron(50,40,30,20,10,and 5 mg/ml),triglyceride(volume percent were 60%,30%,and 10%,respectively),and homogenate of normal rat liver tissues were mixed.All 18 tubes were scanned by dual source CT with 80-140 kVp and 100-140 kVp,and all these images were reconstructed by filtered back projection (FBP) and singogram affirmed iterative reconstruction(SAFIRE).Raw data were reconstructed by filtering and iterative method.△H value (△H =CT value low kVp-CT valuehigh kVp) and overlay value of three substances decomposition algorithm were measured and analyzed by t test.The Spearmen correlation was applied in iron concentration and measured values.Results Iron specific slope was found to be 1.9392.When 80-140 kVp and 100-140 kVp were applied,△H values were (198 ± 28) and (152 ± 18) HU by using FBP reconstruction,and △H values were (191 ± 23) and (149 ± 17) HU by using SAFIRE reconstruction.The differences were statistically significant(t =-2.934 and-3.492,P =0.032 and 0.017).Overlay values were (381 ±54) and (236 ±31) HU by using FBP reconstruction,and overlay values were (399 ±53) and (276 ± 43) HU by using SAFIRE reconstruction.The differences were also statistically significant (t =-5.739 and-7.949,P =0.002 and 0.001).Under the same energy portfolio but different reconstruction,both △H and overlay value showed no differences (P > 0.05).Except for the conditions with 100-140 kVp tube voltage,△H had no correlation with iron solution concentration (P > 0.05) under either FBP or SAFIRE reconstruction.When other reconstruction methods and tube voltage combination were used,overlay or △H value showed correlation with iron solution concentrations (r value ranged from 0.959 to 0.997,P values < 0.01).Conclusions Different energy portfolios of dual source CT will act on the measured value of iron deposition.The measured values will not be affected by ways of reconstruction.For the detection of iron coexisting in fat,overlay values of material decomposition algorithm is better.
10.Expression of mieroRNA-224 in HepG2 cells and analysis of its predicted target genes

Objective To investigate the expression of microRNA-224 in HepG2 cells and analyze its target genes to reveal its role in the carcinogenesis of hepatoma. Methods The genes with differential expression in HepG2 cells and LO2 cells were obtained by gene expression microarray analysis. The up-regulated target genes of microRNA-224 were predicted by bioinformatics method, and their functions were analyzed. Results Compared with LO2 cells, microRNA-224 was highly expressed in HepG2 cells. A total of 264 target genes of microRNA-224 were predicted, including genes involved in cell cycle, signal transduction, cell differentiation, proliferation and apoptosis. Conclusions MicroRNA-224 is highly expressed in HepG2 cells. MicroRNA-224 plays an important role in the carcinogenesis of hepatoma via regulating the expression of its target genes directly or indirectly.
