Objective:To discuss the depressed expression of BUB1 gene in trophoblastic cells by microRNA.Methods:Endogenous BUB1 mRNA level was compared by using quantitative real-time PCR before and after transfected with microRNA plasmid; and the Bub1 protein level was compared by western blot.Results:Ratio of BUB1 mRNA/GAPDH mRNA of control group was 0.1960?0.0688,and after transfected with microRNA recombinant plasmid was 0.1968?0.0689 and 0.1962?0.0670 respectively.Statistical study of samples showed they have no instinct different;otherwise the protein level was significantly decreased after transfected with microRNA plamid.Conclusion:This study established microRNA can control gene expression in mammalian cells at translation level,it is a good foundation to study on the control of gene expression.
3.Relationship between hepatic membrane protein MRP3 and nuclear receptor RXR?

Objective To investigate the relationship between the hepatic membrane protein MRP3(multidrug resistance-associated protein 3,MRP3) expression and the nuclear hormone receptor RXR?(retinoid-X receptor alpha,RXR?)expression in both cultured hepatoma cell HepG2 and bile duct ligated(BDL) rat liver.Methods Total cellular protein and nuclear protein were isolated from HepG2 cells induced by chenodeoxycholic acid(CDCA) or phenobarbital(PB),as well as from the liver tissue of BDL rats.The protein expressions of MRP3 and RXR? were determined by Western blotting.Results The membrane protein MRP3 expression was significantly enhanced and the nuclear receptor RXR? expression was suppressed by CDCA or PB in HepG2 cells.Similarly up-regulation of MRP3 and down-regulation of RXR? were also observed in BDL rat liver.Conclusion The up-regulation of hepatic membrane protein MRP3 may be associated with down-regulation of nuclear receptor RXR?.
4.THE STATUS AND RISK FACTORS OF COGNITIVE DISORDER IN HEMODIALYSIS PA-TIENTS

To study the status and risk factors of cognitive disorder in hemodialysis patients.Methods We re-cruited the hemodialysis patients in our hospital during March, 2013 to February, 2014 and assessed their cognitive function.The case -control method was used to test the risk factors of cognitive disorder in these patients.Result-sThe occurrence rate of cognitive disorder in these hemodialysis patients was 59.4%.Education level and chronic in-somnia significantly influenced the risk of cognitive disorder.The higher education level, the lower the incidence of cognitive dysfunction (OR =0.51; 95%CI =0.30 -0.86); The patients with a history of chronic insomnia were at a 1.98 times higher risk of had (OR =1.98; 95%CI =1.05 -3.75) of cognitive disorder compared to those without chronic insomnia.Conclusion There is a high prevalence of cognitive disorder in hemodialysis patients.Education lev-el and chronic insomnia are independent risk factors for cognitive disorder.
5.Relationship between Mycoplasma pneumonia infection and sex, age and season

Objective To assess the relationship between Mycoplasma pneumonia (MP) and sex,age and season,and to provide a basis for the epidemiology of MP.Methods The serum samples were tested for Mycoplasma pneumonia antibody IgM (MP-IgM) by enzyme-linked immunosorbent assay for 782 children with respiratory tract infection.Results Among the 782 children with respiratory tract infection,221 (28.3%,221/782) were MP-IgM positive,and no difference was seen between male and female (P> 0.05).The incidence was in all ages,but it in 3-5 years old was the highest (50.0%,78/156),and < 1 year old was the lowest (9.6%,19/198),there was significant difference compared with the others (P< 0.05).The total MP-IgM positive rate in children less than 6 years old was 32.4% (193/595),which was higher than that in children over 6-14 years old [15.0% (28/187)] (P < 0.05).MP was epidemic in all year round,but the positive rate of MP-IgM was highest in spring (42.6%,107/251),the positive rate in summer,autumn and winter was 27.4%(57/208),10.8%(19/176) and 25.9%(38/147),respectively,and there was significant difference (P < 0.05).Conclusions MP infections in children demonstrate a definite seasonality,and the positive rate may differ by age.The higher rate of MP infection is higher in the children less than 6 years old without sexual difference.MP is epidemic in spring.

7.The clinical study of continuous veno-venous hemofiltration in treatment of severe acute pancreatitis

Objective:To explore the efficacy and mechanism of continuous veno-venous hemofiltration in treatment of severe acute pancreatitis(SAP).Methods:21 consecutive patients with SAP were included in this study.They were administrated with central venous catheteration.AV600 hemofilter(1.4m~2) was applied for CVVH.CVVH was started 2.8?1.4 days after onset of disease and sustained for 44.6?30.7 hours.We monitored the changes of symptorm,physical sign,laboratory examination and cytokines during CVVH.Results:The hemodynamic state in all patients was stable during CVVH.The overall surviral rate in ICU reached to 76.2%.After implement of the CVVH treatment,abdominal pain and abdominal distension were considerably improved.Temperature,heart rate,breath rate decreased significantly,as well as serum concentrations of BUN,Cr,ALT,ALP,AMY,LPS declined.There was a considerable reduction in cytokines including TNF-?,IL-6,?-INF at the end of CVVH treatment.Conclusion:CVVH is an effective therapeutic option in treatment of SAP.The early clearance of inflammatory mediators including TNF-?,IL-6,and ?-INF is thought to be an important mechanism for improving outcome.
8.Comparative study on use therapeutic efficacy of single of Morphine and drug combined medication

Objective; To study the efficacy, side effect and economic value of morphine controlled - release tablets plus diclofenac sodium and antidepressive drugs in the treatment of pain caused by metastatic bone carcinoma. Methods: 83 patients with pain of meta-static bone carcinoma were randomly divided into two groups. Group I was treated only with morphine controlled - release tablets, Group II with morphine controlled - release tablets, diclofenac sodium and amitriptyline. The dosage of morphine controlled - release tablets, the adverse reaction and the medical cost were observed. Results: The dosage of morphine controlled - release tablets and the medical cost in Group II were obviously lower than those in Group I. The side effects were not significantly different between the two groups. Conclusions: Morphine controlled - release tablets plus diclofenac sodium and antidepressive drugs can treat the pain of metastatic bone carcinoma effectively, relieve the economic burden of the patient while not to increase the side effects.
9.Influence of biotic and abiotic elicitors on production of tanshinones in Salvia miltiorrhiza hairy root culture

Objective To compare influence of several biotic elicitors(yeast extract,(oligogalactu-)ronides,and fungal elicitor) and abiotic elicitors(Ag~+,Co~(2+),and ?-amino isobutyric acid) on tanshinone production in the hairy root culture of Salvia miltiorrhiza.Methods The contents of three tanshinones were(analyzed) by high performance liquid chromatography.Results Biotic elicitors increased effectively the yield of tanshinones and there were difference among different elicitors' effects.The highest content of tanshinones was achieved by fungal elicitor at 100 ?g/mL,which was(0.42 mg/g),4.7-fold of the control.The elicitation effects of abiotic elicitors were relatively lower than those of biotic elicitors,with Co~(2+) at 50 ?mol/L giving the highest content of tanshinone(1.75 mg/g,4.1-fold of the control) among abiotic elicitors.Biotic and abiotic elicitors showed the selectivity of elicitation.All biotic elicitors notably increased the crypotanshinone content and all abiotic elicitors selectively enhanced the tanshinone I content.And in most cases,the elicitors added did not cause any obviously inhibitory effect on the hairy root growth. Conclusion Adding different biotic and abiotic elicitors may selectively promote tanshinone accumulation and not affect the hairy root growth in the hairy root cultures of S.miltiorrhiza.
10.Construction of the plasmid expressing gfp and MAD2 gene controlling separation of cell and its expression in HepG2 cell

Objective: To establish an in vitro screening system for testing effects of MAD2 gene of embryonic cell genome.Methods: In order to construct the plasmid expressing MAD2-GFP fusion plasmid,the target gene fragment was obtained by RT-PCR amplification and inserted into pEGFP-N1 reporter vector.The structure of constructed plasmid was confirmed by electrophoresis analysis and DNA sequencing.The function of construct was confirmed by lipofectamine-mediated transient expression in HepG2 cells.Results: DNA sequencing showed that the inserted fragment of the constructed plasmid was the same as the template MAD2 genome.The HepG2 cells transfected with the constructed plasmid could express reporter gene of gfp.Conclusions: the plasmid expressing gfp gene controlled by MAD2 gene is successfully constructed and an in vitro testing system for evaluating founction of MAD2 gene in separation of cell is established.
