AIM To investigate the role of Trp3 in the Ca 2+ influx induced by ? 1B AR in HEK293 cells and the effect of tyrosine kinase on it. METHODS With lipofect AMINE2000 reagent, hTrp3 cDNA was transfected to HEK293 cells and ? 1B HEK293 cells respectively. The expression of Trp3 was examined by Western blot. With Fura 2/AM spectrophoto fluorometry, Ca 2+ influx was determined. RESULTS HTrp3 was expressed endogenously in HEK293 cells, and the expression increased in hTrp3 transfected cells. Compared with untransfected cells, transfection of hTrp3 cDNA increased Ca 2+ influx induced by ? 1B AR ( P 0 05). 5~30 ?mol?L -1 genistein inhibited Ca 2+ influx induced by ? 1B AR in hTrp3 cDNA transfected cells and the maximum inhibitory rate was (75 2?12 6)% . CONCLUSION Transfection of hTrp3 cDNA increased Ca 2+ influx induced by ? 1B AR in HEK293 cells. This process was regulated by tyrosine kinase.

Aluminum salts are the most popular adjuvants applied in human vaccines currently. However, they can′t achieve satisfying results in the development of novel vaccines because of the cellular immune responses induced by them are weak and their adjuvant activities for some novel vaccines are poor, especially in vaccination against peptide antigens with small molecular weight. cGAMP (cyclic guanosine monophosphate-adenosine monophosphate) has recently been known as a mammalian second messenger, which plays an important role in the innate immune signaling pathway and is capable of boosting the immuno-genicity of vaccines, activating antigen-presenting cells and enhancing specific T cell responses. cGAMP is expected to become a new generation of vaccine adjuvants against infectious diseases and cancer. In this re-view,we summarize the application and current situation of vaccine adjuvants, describe the discovery of cGAMP and its mechanism as a vaccine adjuvant, and focus on the advances in using cGAMP in the fields of vaccination against infectious diseases, intradermal immunization and tumor immunotherapy. Finally, it is also pointed out that cGAMP,as a novel vaccine adjuvant, will have a broad prospect of application in areas such as anti-tumor,anti-virus,anti-inflammatory and vaccines.

Objective To investigate the current status of the beliefs and attitudes about sleep and to discuss its influencing factors in patients with hepatocellular carcinoma. Methods A total of 206 patients with hepatocellular carcinoma were investigated with (beliefs and attitudes about sleep scale, BASS). Single factor analysis was used to compare the difference of beliefs and attitudes about sleep among hepatocellular carcinoma patients with different basic data. Multiple linear stepwise regression was adopted to explore the influencing factors of the beliefs and attitudes about sleep in patients with hepatocellular carcinoma. Results The total score of BASS in patients with hepatocellular carcinoma was (85. 15 ± 13. 36). There were statistically significant differences in the beliefs and attitudes about sleep in patients with hepatocellular carcinoma among different degrees of education, family incomes, occupations, duration of disease, the Karnofsky score and knowledge of the facts (P<0. 05). The multiple linear stepwise regression analysis results showed that the influencing factors the beliefs and attitudes about sleep in patients with hepatocellular carcinoma were the Karnofsky score, informed situation, duration of disease, occupation, family income (P<0. 05). Conclusions The beliefs and attitudes about sleep in patients with hepatocellular carcinoma were in low level and were influenced by various factors. Targeted interventions should be taken to strengthen their beliefs and attitudes about sleep.

Objective To explore the correlation of hope level and mental health in patients with primary liver cancer. Methods A total of 146 patients with primary liver cancer were investigated by the Herth Hope Scale and the Symptom Checklist 90 ( SCL-90 ) . Pearson correlation analysis and hierarchical regression analysis were used to explore the correlation between hope level and mental health in patients. Results Of the 146 patients, 85 cases were metal ill -health. In the mental health group, positive attitude of current and future, positive behavior, intimate relationship with others, and total score of hope were (12. 22 ± 1. 38), (12. 83 ± 1. 58), (13. 42 ± 1. 71), (38. 47 ± 4. 43),which were higher than those of the metal ill-health group (t=5. 977, 4. 228, 4. 116, 4. 779,respectively; P <0. 01). Pearson correlation analysis showed that the positive attitude of current and future, positive behavior, intimate relationship with others, and total score of hope had negative correlation with SCL-90 ( r = -0. 586, -0. 537, -0. 485, -0. 528, respectively;P <0. 01). Hierarchical regression analysis showed that each dimensions of hope level could unite to explain the 29. 1% change status of mental health for patients. Conclusions Patients with primary liver cancer have a higher hope level leading to higher metal health.

OBJECTIVE:
Utilize high-resolution chromosome analysis and microarray detection to determine the genetic etiology of infertility of a 32-year old female patient.
METHODS:
The peripheral blood of the patient was cultured for high-resolution chromosome G and C banding karyotype analysis, and then 750K SNP-Array chip detection was performed.
RESULTS:
Karyotype analysis results showed that the patient's karyotype was 45,XX,-13 [7]/46,XX,r(13) (p13q34) [185]/46,XX,dic r(13;13)(p13q34;p13q34) [14]/ 47,XX,+der(13;13;13;13) (p13q34;p13q34;p13q34; p13q34), dic r(13;13) [1]/ 46,XX [3]. The microarray results showed that the patient had a 3.3 Mb deletion in the 13q34 segment of chromosome 13, which may be related to infertility.
CONCLUSION
Infertility of the patient reported in this article may be related to the deletion of chromosome segment (13q34-qter).

Objective:To understand the impact of intimate social networks and sexual networks on HIV-infection in men aged ≥50 years in Pengzhou, Sichuan Province, and provide reference for the formulation of HIV/AIDS prevention and control strategies in middle-aged and elderly people. Methods:A case-control study was conducted by an interview-style questionnaire survey in a case group consisted of 114 men aged ≥50 years and a control group consisted of 423 healthy men matched by age frequency in Pengzhou, Sichuan Province, from April 2019 to October 2020, and the influencing factors of HIV infection were identified by unconditional stepwise logistic regression analysis. Results:Among the 114 cases, 63.16% (72/114) and 96.49% (110/114) of men had intimate social network size and sexual network size ≥4 people, which were higher than the control group of 23.40% (99/423) and 11.58% (49/423) (all P<0.05).The overlap of intimate social network and sexual network was 49.12% (56/114) in the case group, which was lower than that in the control group (82.27%, 348/423)( P<0.05). Logistic regression analysis showed that the size of the intimate social network ≥4 people ( OR=8.66, 95 %CI: 2.01-37.30), the size of the sexual network ≥4 people ( OR=1 121.11, 95% CI: 28.38-4 429.61), the material support from the first intimate network member ( OR=6.39, 95% CI: 1.03-39.75), and multiple sexual partners ( OR=55.50, 95% CI: 6.22-494.96) were the risk factors of HIV-infection in men aged ≥50 years. Receiving health education about AIDS ( OR=0.07, 95% CI: 0.01-0.37), high sexual satisfaction with the first sexual partner and good personal relationship ( OR=0.02, 95% CI: 0.01-0.59; OR=0.01, 95% CI: 0.01-0.26), and the overlap between intimate social network and sexual network ( OR=0.02, 95% CI: 0.01-0.38) were the protective factors of HIV-infection in men aged ≥50 years. Conclusions:The overlap of intimate social network and sexual network can reduce the risk of HIV-infection in men aged ≥50 years. It is important to help middle-aged and elderly men to build good and stable intimate social and sexual networks to meet their social communication and sex demands and prevent HIV infection by reducing high riak sexual behaviors.

A 54-year-old man was admitted to respiratory department with chief complaints of recurrent cough and dyspnea. Chest imaging showed multiple patchy shadows and interstitial changes. Evidence of infectious diseases was not definite, and antibiotic treatments were not effective. In the meantime, myelodysplasia syndrome was diagnosed with pancytopenia. The pathologic findings of transbronchoscopic lung biopsyshowed chronic inflammatory interstitial changes, suggesting a clinical diagnosis of organizing pneumonia. After glucocorticoids treatment, his condition aggravated. The second percutaneous lung biopsy showed the infiltration of a large number of neutrophils. Therefore, the final diagnosis of myelodysplasia syndrome with Sweet syndrome was made. Then glucocorticoids and supportive treatment were given This case may improve physicians' understanding of myelodysplasia syndrome complicated with Sweet syndrome.
