1.Observation on Legionella pneumophila surviving and multipling in Acanthamoeba polyphaga in vitro

Objective To observe the survival and multiplication of Legionella pneumophila cocultured with Acanthamoeba polyphaga for setting up the laboratory model of such infections. Methods With the ratio of 1∶1000, Acanthamoeba polyphaga and Legionella pneumophila were cocultured under laboratory condition. We investigated the intracellular growth, survival and multiplication of Legionella pneumophila in Acanthamoeba polyphaga in morphological way by using inverted microscopy,electron microscopy and Giemsa staining of the cover glass picking up from the flask. Results Giemsa staining of the cover glass from the culture flask showed that the vacuoles appeared after 8 h coculture, and became more and bigger with the time. After 24 h, Giemsa staining showed that the bacteria started to accumulate within amoeba. In the anaphase of the coculture, the infected amoeba lacked defined organelle structures and was filled with legionella. The electron microscope revealed the survival of legionella in vacuoles of both trophozoites and cysts, showing different phases of binary fission. Conclusions Legionella pneumophila can survive and multiply in Acanthamoeba polyphaga . The results indicated that live amoeba could be a reservoir host for Legionella pneumophila.
2.Application of reminder software for management of indwelling catheter in daily management of indwelling catheter

Objective To assess the application effect of reminder software for the management of indwelling catheter in catheter associated with urinary tract infection in emergency intensive care unit (EICU). Methods We conducted a comparative study among patients with catheter-associated urinary tract infection from January 2014 to December 2015 in emergency intensive care unit (EICU). By applying with reminder software, we assessed the omission rate of indwelling urethral catheterization management, the incidence rate of catheter associated urinary tract infection (CAUTI), the utilization rate of urinary tract intubation and the average time of indwelling urethral catheterization.Results By the application of reminder software, the omission rate of indwelling urethral catheterization management was decreased. The incidence rate of CAUTI declined from 9.16‰ to 1.90‰ (x2=4.843,P<0.05),while the utilization rate of urinary tract intubation declined from 73.60% to 60.67% (P<0.01), and the average time of catheterization also shortened from (10.21±2.28) days to (8.20±1.67) days (t=2.47,P<0.05).Conclusions Reminder software applied in the control of indwelling catheter related infections has achieved good effectiveness and it can be used widely in clinical work.
3.Clinical evaluation on idiopathic pulmonary fibrosis.

Idiopathic pulmonary fibrosis (IPF) is a chronic fatal pulmonary disease characterized by complex illness condition. There is no effective treatment at present except lung transplantation. The comprehensive evaluation is helpful for the management of patients with IPF in hierarchical stages. Therefore, it is very important to evaluate IPF by various independent factors. At present, the commonly used methods for clinical evaluation on IPF include assessment of health-related quality of life, assessment of physiological function, assessment of imaging, assessment of laboratory examination, and multi-dimensional assessment system. However, there are different advantages and disadvantages on diverse evaluation methods for the evaluation of IPF.

4.Effect of auricular point press needle on improving sleep quality of postoperative patients with gastric cancer

Objective:To observe the effect of auricular point press needle on sleep quality of patients with gastric cancer after operation.Methods:From October 2017 to March 2019, a total of 80 patients underwent radical gastrectomy for gastric cancer in the First People's Hospital of Huzhou, Zhejiang Province were selected by using convenience sampling method, and divided into observation group and control group by random number table method, with 40 cases in each group. The control group received routine nursing, while the observation group received auricular point press needle on the basis of routine nursing. Sleep quality was monitored in both groups using sleep monitoring bracelets. In the course of the study, 2 patients in the observation group had surgical complications, and finally 35 patients completed the experiment; 2 patients in the control group had surgical complications, and a total of 33 patients completed the experiment.Results:From the 6th day of intervention (the 4th day after operation) , the sleep duration of the observation group was longer than that of the control group, and the difference was statistically significant ( P<0.05) , the group effect, time effect and interaction effect were statistically significant ( P<0.05) . From the 6th day of intervention (the 4th day after operation) , the proportion of deep sleep of the observation group was higher than that of the control group, and the difference was statistically significant ( P<0.05) , the group effect, time effect and interaction effect were statistically significant ( P<0.05) . Conclusions:Auricular point press needle can effectively improve the sleep quality of patients with gastric cancer after operation.
