1.Association between the Level of LDH in Gastric Cancer Patient’s Serum, the Relative Expression of LDH in Tissues and the Patient’s Prognosis

Objective To explore the association between the level of lactate dehydrogenase (LDH)in gastric cancer patients’ serum or the relative expression of LDH in their tissues and the patients’prognosis.Methods 60 specimens of gastric canc-er patients who confirmed by pathological diagnosis were collected from 2012 to 2013,include the serum specimens and tis-sue specimens.Their serum level of LDH were detected,and the relative expressions level of LDH in the sample of normal tissues,gastric cancer tissues and metastatic lymph nodes were quantify by real-time fluorescence quantitative reverse tran-scription-PCR (Realtime-PCR).Finally,the relationship between the expression of LDH and clinical pathologic features were analyzed by independent t-test or anova statistics.Results The serum level of LDH was highly increased than control group (340.89±10.67 IU/ml,t=24.7,P<0.01);the relative gene expression of LDH in normal tissue,human gastric cancer and metastatic lymph node were 1.0,3.39 and 2.35.The result suggests the serum level of LDH were associated with pTNM stage and lymph node metastases (t=5.2,4.8,P<0.01).The relative gene expression of LDH in gastric canc-er tissues were associated with tumor sizes,pTNM stage and lymph node metastases (t=18.2,15.3,P<0.01 and F=7.2, P<0.01).Conclusion The serum LDH level and the expression of LDH in cancer tissue were significantly increased,and their expression were correlated with the patient’s prognosis.The serum level of LDH and the expression of LDH in gastric cancer tissue may be potential indicator to evaluated the prognosis of the patient with gastric cancer.
2.Review of HPV Infection Status of the Women in Shantou Region

Objective To review the HPV infection status of the females in Shantou in the past years.Methods 3 664 speci-mens of cervical exfoliated cells of the female patients with gynecological disease from 2013 to 2014 were selected to ampli-fied HPV gene and conducted genetypes detection.The infection rate and various subtypes of HPV were analyzed.All these patients were divided into 5 groups according to their ages and the infection status of each group were also discussed.Results 805 patients were infected with HPV (22%,805/3 664).The single infection type and the mixed infection type were both visible,HPV-16 was the most common single infection type,followed by subtypes HPV-52,43,58 and 56.HPV16+52 was the most common mixed infection type,followed by HPV16+33 and HPV43+52.The infection rates of the each group were differences (χ2 =27.08,P < 0.01),No.1 (> 25),No.2 (25 ~ 34)and No.5 group (< 55)were the high-risk groups of HPV infection.Conclusion The main genotypes of these women whose infection with HPV were HPV-16,52,43,58 and 56 in Shantou region,respectively.The females aged less than 35 years old and more than 55 years old both had high-risk for HPV infection.It has important significance to developed the vaccines which can efficiently prevent HPV-16,52,43,58,56 and reduce the cervical cancer incidence.
3.The expression of leptin and soluble leptin receptor in breast cancer patients′serum and the clinical significance

Objective To explore the levels of leptin(LEP) and soluble leptin receptor (sOB‐R) in breast cancer patients′ser‐um ,and to discuss the relationship between LEP ,sOB‐R ,free leptin index(FLI) and the patients′clinical pathologic features .Meth‐ods 70 serum specimens of breast cancer patients who confirmed by pathological diagnosis were collected before the surgery ,the serum of benign breast disease group(n=50) and normal group(n=50) were also collected as control .The levels of LEP and sOB‐Rinserumweredetected,andtheresultafterbodymassindex(BMI)correctionwereanalyzed.Finally,therelationshipbetweenthe expression of LEP ,sOB‐R and clinical pathologic features were analyzed .Results The serum level of LEP in breast cancer group was highly increased than the other two groups (P<0 .05) ,and sOB‐R was reduced(P<0 .05) .After BMI correction ,LEP was still highly increased(P<0 .05) ,but sOB‐R was no significant difference (P>0 .05) .The level of LEP in the postmenopausal breast cancer patients were highly increased than menopause patients (P>0 .05) ,and the expression of LEP in cancer group were signifi‐cant higher than two control groups by both postmenopausal and menopause (P>0 .05) .The result suggested the serum level of LEP and FLI are associated with pTNM stages and lymph node metastases (P<0 .05) ,and elevated level of LEP in postmenopausal patients had differences in pTNM stages and lymph node metastases (P< 0 .05) .Conclusion The serum LEP level and the FLI may be potential indicators to evaluate the prognosis of the patient with breast cancer .The serum LEP may promoted the process of the postmenopausal patients of breast cancer .

Objective To study the expression of B7-H1 in gastric mucosa of patients with gastric carcinoma,and to identify its relationship with neoplasm metastasis and prognosis.Methods Expression of B7-H1 in gastric carcinoma cell line SGC-7901 and in freshly-resected gastric mucosa including gastric carcinoma,adjacent tumor tissue and distal normal gastric mucosa were examined by flow cytometric analysis,immunochemical staining,immunofluorescence staining and Western blot.The correlated data was analyzed statistically.And so the correlation among expression level of B7-H1 and the patients,clinicopathological parameters was established.Results B7-H1 expression was detected in SGC-7901 cell line.B7-H1 was found in cell membrane and little cytoplasm.The positive rate of B7-H1 expression in gastric carcinoma was(13/21)62%,and it was(7/21)33% in adjacent tumor tissue,Whereas B7-H1 was absent in distal normal gastric mucosa.Statistical analysis demonstrated a positive correlations of B7-H1 expression in gastric carcinoma with the depth of carcinoma infiltration,lymph node metastasis and pTNM stage(P
5.Hsa-miR-218 inhibits growth of cervical cancer HeLa cells by targeting to LASP1

AIM:To study the effect of hsa-miR-218 on cervical cancer HeLa cell growth and the underlying molecular mechanism .METHODS:The lentivirus expression vector pmiR-218 targeting to hsa-miR-218 was constructed . pmiR-218 was transfected into HeLa cells .The number of viable HeLa cells was counted by the method of Trypan blue ex-clusion.The inhibitory rate of cell activity was detected by WST-8 assay.The expression of LIM and SH3 protein 1 (LASP1) at mRNA and protein levels was determined by real-time PCR and Western blot.The interaction between miR-218 and LASP1 was examined using a luciferase reporter assay .RESULTS:The lentivirus expression vector pmiR-218 tar-geting to hsa-miR-218 was constructed successfully and confirmed by DNA sequencing .Over-expression of miR-218 inhibi-ted the activity of HeLa cells with the inhibitory rates of 15%, 26%and 65%at 24 h, 48 h and 72 h, respectively .The difference between transfection group and blank control /negative control group was statistically significant .The luciferase activity was reduced when co-transfection with miR-218 mimics and LASP1-3’ UTR plasmid.The relative expression of miR-218 was increased after transfection with pmiR-218.Over-expression of miR-218 down-regulated the LASP1 expression at mRNA and protein levels by 25%and 75%respectively.Compared with blank control group and negative control group , the difference was statistically significant (P<0.05).CONCLUSION:pmiR-218 effectively inhibits the growth of HeLa cells in a time-dependent manner.miR-218 targets to the 3’UTR of LASP1, thus down-regulating the expression of LASP1 in HeLa cells .
6.Influence of psychological nursing intervention on postoperative mental status and quality of life

Objective To explore the effect of psychological nursing intervention on postoperativemental status and quality of life. Methods The 74 cases of patients with breast cancer after radical operation from January 2012 to January 2015 in our hospital,all married patients,all patients were randomly divided into intervention group(37 cases)and con-trol group(37 cases),were collected. All clinical data of patients with complete and signed the informed consent,vol-untary acceptance of various scale survey. Exclude the presence of severe somatic diseases associated with distant metastasis of the tumor. Results The psychological intervention group SAS score and SDS score were significantly lower than those before intervention, and psychological intervention group SAS score and SDS score were significantly lower than those of the control group,with significant difference(P<0.05),psychological intervention group do prognostic score function field the score and overall health status were compared with that before the intervention significantly elevated scores and psychological intervention group of the function field of the score and overall health status were significantly higher than those in control group,with statistically significant difference(P<0.05),psychological interven-tion group the symptoms score and domain specific symptoms scores were significantly decreased than before interven-tion,and psychological intervention group symptoms score and specificity of the field symptom scores were significantly lower than those in control group,with significant difference(P<0.05). Conclusion The psychological nursing interven-tion in breast cancer patients is conducive to improve the psychological status of the patients after operation,improve the quality of life of patients.
7.Optimization of the Microwave Processing Technology of Yellow Wine-processed Corydalis yanhusuo

OBJECTIVE:To optimize the microwave processing technology of yellow wine-processed Corydalis yanhusuo . METHODS:The contents of opioid alkaloid ,berberine hydrochloride and tetrahydropalmatine in C. yanhusuo processed with yellow wine were determined by HPLC. The contents of the extracts were determined by hot dipping method. Based on the single factor tests ,using the appearance of yellow wine-processed C. yanhusuo with microwave processing technology ,the contents of extract,opioid alkaloid ,berberine hydrochloride and tetrahydropalmatine as indexes ,with the amount of yellow wine ,wetting time,microwave power and microwaving time as factors ,the processing technology was optimized with orthogonal test combined with comprehensive weighted scoring method ,and then validated and compared with traditional yellow wine-processed C. yanhusuo . RESULTS:The linear ranges of opioid alkaloid ,berberine hydrochloride and tetrahydropalmatine were 0.100-1.500 μg(R2=0.999 6), 0.012-0.188 μg(R2=0.999 5),0.050-0.750 μg(R2=0.999 8). RSDs of precision ,stability(12 h)and repeatability tests were all less than 2% . The recoveries were 99.15% -100.34%(RSD=0.54% ,n=6),99.52% -100.78%(RSD=0.69% ,n=6), 99.26%-99.79%(RSD=0.28%,n=6). The optimum microwave processing technology included that the amount of yellow wine was 4 g(about 20% of medicinal material amount ),microwave power was 40%,wetting time was 3 hour,processing time was 3 min. The results of three verification tests showed that the contents of extract ,opioid alkaloid ,berberine hydrochloride and tetrahydropalmatine were 15.7%-16.1%,0.061%-0.063%,0.003%-0.004% and 0.061%-0.063%. The comprehensive scores were 97.916,94.730 and 97.217,and RSD were 0.42%,0.38%,0.46%(n=3),respectively. Compared with traditional yellow wine processing technology ,there was no significant difference in the contents of opioid alkaloid and other components ,but no scorched spot and crumbs was found in yellow wine-processed C. yanhusuo with microwave processing technology. CONCLUSIONS:Established method for content determination is simple ,accurate,reliable and reproducible ,and can be used for quantitative analysis of active components in yellow wine-processed C. yanhusuo . Optimized microw ave processing technology is stable and feasible ,and can be used for the processing of yellow wine-processed C. yanhusuo .
8.Effect of Modified Gualou Zhishitang Combined with Piperacillin Sodium and Tazobactam Sodium on Immune Function and Serum Levels of Inflammatory Cytokines in Patients with Stroke-associated Pneumonia with Syndrome of Phlegm-heat Accumulation in Lung

ObjectiveTo explore the effects of modified Gualou Zhishitang combined with piperacillin sodium and tazobactam sodium on the immune function and serum levels of inflammatory cytokines in the patients with stroke-associated pneumonia (SAP, syndrome of phlegm-heat accumulation in lung). MethodEighty SAP patients with the syndrome of phlegm-heat accumulation in lung were randomized into a control group (40 cases) and a study group (40 cases). The SAP patients in the control group were treated with piperacillin sodium and tazobtam sodium, while those in the study group were treated with modified Gualou Zhishitang on the basis of the treatment in the control group for 2 consecutive weeks. The clinical therapeutic effects, immune function indexes, inflammation indexes, and lung function of SAP patients in the two groups before and after treatment were determined and compared. ResultAfter treatment, the scores of lesion, pulmonary rales, cough, fever, phlegm color, and constipation in both groups decreased (P<0.05). After treatment, the ratio of forced expiratory volume in the first second to forced expiratory volume (FEV1/FVC) and forced expiratory volume in the first second as percentage of predicted value(FEV1%) in both groups improved (P<0.05), and the study group outperformed the control group (P<0.05). The treatment decreased the neutrophil to lymphocyte ratio (NLR) in the two groups (P<0.05), and the study group had lower NLR than the control group after treatment (P<0.05). The serum levels of procalcitonin (PCT) and hypersensitive C-reactive protein (hs-CRP) in both groups declined after treatment (P<0.05), and the declines were more significant in the study group than in the control group (P<0.05). After treatment, the study group was better than the control group (P<0.05). The treatment in both groups elevated the levels of CD3+, CD4+, and CD4+/CD8+ in the peripheral blood and lowered the level of CD8+ (P<0.05), and the changes were more significant in the study group than in the control group (P<0.05). The total response rate of the study group was 95.00% (38/40), which was higher than that (80.00%, 32/40) of the control group (χ2=4.114,P<0.05). There was no significant difference in the incidence of adverse reactions between the two groups. ConclusionModified Gualhou Zhishitang combined with piperacillin sodium and tazobactam sodium demonstrates a significant therapeutic effect on the SAP patients with the syndrome of phlegm-heat accumulation in lung. This therapy can mitigate the clinical symptoms, improve the lung function, lower the serum levels of inflammatory cytokines, and improve the immune capacity, with high safety.
9.Effect of wrist-ankle acupuncture on pharmacodynamics of remimazolam for loss of consciousness in patients of different genders and ages undergoing painless gastroscopy

Objective:To evaluate the effect of wrist-ankle acupuncture on the pharmacodynamics of remimazolam for loss of consciousness in the patients of different genders and ages undergoing painless gastroscopy.Methods:American Society of Anesthesiologists Physical Status classificationⅠor Ⅱ patients, undergoing elective painless gastroscopy, were divided into 4 groups according to gender and age: agedmale (age≥65 yr) group (group AM), agedfemale (age≥65 yr) group (group AF), young male (18 yr≤age <65 yr) group (group YM), and young female (18 yr≤age <65 yr) group (group YF). After 10 min of wrist-ankle acupuncture on 1st and 2nd areas of the bilateralupper limbs of the patient, modified Dixon′s up-and-down sequential experiment was used for the test.The initial dose of intravenous remimazolam was 0.20 mg/kg. Each time the dose increased/decreased by 0.05 mg/kg. If the patient lost consciousness, the next patient received a lower dose of remimazolam, otherwise a higher dose was given in the next patient. Loss of consciousness was defined as MOAA/S score was 0 or 1, and the process was repeated until 7 turning points appeared.The median effective dose (ED 50)and 95% confidence interval of remimazolam were calculated by probit method. Results:The ED 50 and 95% confidence interval of remimazolam were 0.296(0.233-0.376), 0.319(0.262-0.388), 0.323(0.278-0.375) and 0.344(0.285-0.415)mg/kg in AM, AF, YM and YF groups, respectively. There was no significant difference in ED 50 among the four groups ( P>0.05). Conclusions:Under the action of wrist-ankle acupuncture, the pharmacodynamics of remazolam is comparable when used for the patients of different genders and ages undergoing painless gastroscopy.
