1.Clinical analysis of purulent meningitis caused by streptococcus pneumoniae in 12 children

Objective To investigate the clinical characteristics of purulent meningitis caused by streptococcus pneumoniae.Methods We studied clinical features of 12 children with purulent meningitis caused by streptococcus pneumoniae who were hospitalized from Jan 2007 to Oct 2011 in our hospital.Results Twelve children were penicillin-resistant streptococcus pneumoniae.The ages ranged from 2 months to 10 years.Nine cases(75%) were aged less than 5 years,and 5 cases(41.6%) were aged less than 2 years.All cases were with fever,and with the nervous system symptoms involvement.All cases were with complications:9 cases(75%) with septicemia,8 cases (66.7%) with pneumonia.The white blood cells,blood C-reactive protein,peripheral blood procalcitonin,erythrocyte sedimentation rate,white cells and protein in cerebrospinal fluid were mostly significantly high,sugar in cerebrospinal fluid were low significantly.Eleven cases were eventually treatmented by vancomycin plus 3rd to 4rd generations of cephalosporins antibiotics or other antibiotics.Eight cases were cured,2 cases with serious sequela,and 2 cases died.Conclusion We should attach importance to the meningitis caused by streptococcus pneumoniae.For clinical suspects pneumonia streptococcus meningitis,to given the powerful therapy by vancomycin plus the other antibiotics,as early as possible.
2. Transcriptome analysis of Polygala tenuifolia seedlings induced by methyl jasmonate and key genes mining for triterpenoid biosynthetic pathway

Objective: To obtain the transcriptome sequence database induced by methyl jasmonate (MeJA) and identify the genes related to the biosynthesis of triterpenoid saponin in Polygala tenuifolia. Methods: The seedlings grown for 30 d were respectively treated with sterile water, 50 μmol/L MeJA and 100 μmol/L MeJA for 24 h. The transcriptome data of seedlings of P. tenuifolia were obtained by Illunima HiseqTM 2000 150PE sequencing and de novo splicing of Unigene was realized by Trinity software. The GO classification, KOG functional annotation, metabolism of KEGG metabolic pathway, protein function annotation analysis, differential gene analysis and screening were completed based on BLAST. Results: A total of 52.19 Gb clean data were obtained after the transcriptome of P. tenuifolia being assembled by Trinity software, and 54 426 Unigenes were assembled with an average length of 1 604 bp. All Unigenes were annotated in the public databases NR, NT, KEGG, Swissprot, GO, and Pfam. Through differential analysis of genes responding to MeJA, a total of 3 390 differentially expressed genes (DEGs) were found, of which 1 287 were up-regulated and 2 103 were down-regulated. The response of DEGs showed that the total number and up-regulated number of P. tenuifolia seedlings treated by 100 μmol/L MeJA was the highest. KEGG enrichment analysis showed that differentially expressed genes were significantly enriched in metabolic pathways including phenylpropanoid biosynthesis, cysteine and methionine metabolism, starch and sucrose metabolism, carbon fixation in photosynthetic organisms and terpenoid backbone biosynthesis. Furthermore, a total of 59 Unigenes involved in anthraquinones biosynthesis were found according to the assignment of KEGG pathway. Expression analysis showed that AACT, HMGS, HMGR, MK, PMK, MPD, DXS, IDI, FPPS, SQS, SE and β-AS were up-regulated after being induced by MeJA. Conclusion: In this study, the transcriptome of P. tenuifolia seedlings treated with methyl jasmonate was analyzed, and candidate genes related to triterpenoid skeleton biosynthesis of P. tenuifolia were obtained. MeJA can induce the expression of genes related to triterpenoid skeleton synthesis, which provided a wealth of data resources for the molecular biology research and also laid the foundation for the analysis of the secondary metabolic pathways of triterpenoid saponins in P. tenuifolia.

Stroke has been harmful to human health for a long time, and there is no satisfactory treatment strategy because of its complex pathogenesis. Taohechengqi decoction has been effective in the treatment of stroke. In this study, the components were collected by TCMSP, TCMIP, BATMAN-TCM and TCMID databases, the targets were predicted and screened by PharmMapper and BATMAN-TCM databases, and the functional enrichment analysis of the targets was carried out by using R language package clusterProfiler. Finally, the key targets are verified by GEO database and molecular docking. The results showed that 51 active components of Taohechengqi decoction may regulate 15 key targets such as nitric oxide synthase, endothelial (NOS3), prostaglandin G/H synthase 2 (PTGS2), matrix metalloproteinase-9 (MMP9), affecting vascular endothelial growth factor signaling pathway and other pathways to play a role in the prevention of stroke, affecting tumor necrosis factor signaling pathway and other pathways to play a role in the treatment of stroke. GEO data analysis showed that androgen receptor (AR), caspase-8 (CASP8), intercellular adhesion molecule 1 (ICAM1), interleukin-1 beta (IL1B), mitogen-activated protein kinase 14 (MAPK14), MMP9, myeloperoxidase (MPO), peroxisome proliferator-activated receptor gamma (PPARG), PTGS2 and cellular tumor antigen p53 (TP53) were up-regulated genes, while serum albumin (ALB), estrogen receptor 1 (ESR1), NOS3, transcription factor p65 (RELA) and proto-oncogene tyrosine-protein kinase Src (SRC) were down-regulated genes. GEO analysis explained that Taohechengqi decoction may prevent stroke by down-regulating ESR1, NOS3, and treat stroke by up-regulating ICAM1, IL1B, MAPK14, MMP9, PPARG, PTGS2, TP53, and down-regulating RELA and SRC. The study found that in the process of prevention and treatment of stroke, Taohechengqi decoction played a two-way regulation role through multi-genes and multiple ways, which provided a new strategy for the treatment of stroke.

OBJECTIVE
To improve the quality evaluation method of Platycodonis Radix, to study the differences in the quality of three-years-old Platycodonis Radix under different harvesting periods, and to determine the optimal harvesting period of Platycodonis Radix.
METHODS
The leachate, ash, moisture, refractive index and the content of six saponins were used as the quality evaluation indexes. The differences between the herbs of Platycodonis Radix at different harvesting periods were characterized with the help of mathematical and statistical methods. And link the entropy weight method, gray correlation analysis and TOPSIS method were combined to obtain the statistical analysis of the relevant indexes and the quality ranking information of the herbs in different harvesting periods.
RESULTS
There were significant differences between the quality evaluation indexes of three-years-old Platycodonis Radix at different harvesting periods. The added multi-indicator testing had improved the quality evaluation system of Platycodonis Radix and enhanced the "Drug properties-Effectiveness" linkage of the herbs. And the results of the comprehensive quality evaluation model showed that the herbs harvested around October 21 (Frost’s Descent) were ranked best in terms of comprehensive index.
CONCLUSION
In order to ensure the quality of Platycodonis Radix, the best harvesting period for three-years-old Platycodonis Radix is determined around the "Frost’s Descent" season, taking into account the characteristics of the herbs' appearance and the material basis of herbs.
