1.Effects of sevoflurane induced hypotension on cerebral metabolism

Objective To evaluate the efficacy and the effects of sevoflurane induced hypotension on balance of cerebral oxygen supply and consumption and on cerebral energy metabolism. Methods Forty patients undergoing elective procedures were randomly divided into three groups. Group Ⅰ :sevoflurane induced hypotension ; group Ⅱ: sevoflurane maintained normal arterial tension ; group Ⅲ: nitroprusside induced hypotension . In groupⅠ and Ⅲ MAP was decreased to 50%-60% of baseline ,was kept for 40 min either by sevoflurane inhalation or by nitroprusside infusion. MAP ,HR,ECG were monitored continuously, and radial arterial blood samples and jugular blood samples were taken synchronously for measuring blood gas and blood lactate ,SOD and MAD levels. Results In group Ⅰ during induced hypotension Da jvO 2 showed reduction ,but in group Ⅲ Da jvO 2 increased significantly. There were no significant changes in blood lactate level and in the cerebral arteriovenous differences of MAD and SOD in three groups.Conclusions The sevoflurane induced hypotension has no adverse effect on cerebral oxygen balance and the cerebral perfusion . The cerebral energy metabolism is well maintained . Sevoflurane can safely applied to controlled hypotension during cerebral neurosystem.
2.Experimental study of chemolysis with collagenase outside the intervertebral disk

ve To assess the efficacy of chemolysis with collagenase injected into epidural space and the effects upon intervertebral disk and nerve roots. Methods Sixty-four healthy new Zealand rabbits of either sex, weighing 2.5-3.0kg were divided into four groups of sixteen each: group A, B, C and D. In group A and B herniation of disk was produced by injecting normal salin 0.1ml into the intervertebral disk after surgical resection of spine and posterior arch of L4_6, while in group C and D no herniation of disk was produced. In group A and C collagenase 300u( in normal saline 0.5ml) was injected into the epidural space at the level of L4-6, and in group B and D normal saline 0.5ml was injected into epidural space as control. Three animals from each group were sacrificed on the 7th, 15th, and 30th day after operation and intervertebral disk and nerve roots at the operation site were removed and fixed for light and electron microscopic examination. Sciatic nerve conduction velocity (NCV) was measured before operation and on 1st, 7th, 15th and 30th postoperative day. Results In group A the intervertebral disk was dissolved to some extent. In group B nerve roots were injured to different degree. In group A and C NCV was higher than normal on the 7th and 15th day after operation. Conclusions Collagenase may dissolve the herniated intervertebral disk but does not injure the nerve roots. It may increase NCV.

4.Association of femoral bone mineral density with lumbar vertebral fractures in patients with ankylosing spondylitis

OBJECTIVE: Previous opinions consider the secondary diffuse osteoporosis as the very cause of vertebral fractures in patients with ankylosing spondylitis (AS). However, recent studies on bone mineral density (BMD) and vertebral fractures in AS patients reveal that there is no relation between the two. This article aims at investigating the association of lumbar vertebral fractures with clinical, laboratory, and imaging indexes in AS patients.METHODS: A contrast observation was performed between 65 AS patients and 62 healthy physical examinees, whose lumbar vertebral plain radiographs were taken for checking vertebral fractures. Disease activity evaluation indexes included C-reactive protein, erythrocyte sedimentation rate, finger-to-ground distance, Schobar's index score, Bath AS radiology index (BASRI) and syndesmophyte score. Dual energy X-ray absorptiometry technique was used to measure BMD levels of lumbar vertebras and femurs.RESULTS: Out of the total 65 AS patients, 10 ones (15.4%) had lumbar vertebral fractures, with 4 ones with wedge deformities and the other 6 ones with biconcave deformities. BMD levels of Lumbar vertebras and femurs in AS patients were significantly lower than those in controls (P < 0.01). There were significant differences in Schober's index scores, finger-to-ground distance scores, Bath scores, syndesmophyta scores and intertrochanter BMD values between AS patients with and without vertebral fractures (P < 0.01 ). Multiple logistic regression analyses revealed that intertrochanter BMD values were independently associated with lumbar vertebral fractures in AS patients (P =0.043).CONCLUSION: There is a correlation between low femoral BMD levels and the risk of lumbar vertebral fractures in patients with AS, especially at the intertrochanter area.
5.BREEDING OF HIGH PRODUCING STRAIN OF LIPASE USING FAST NEUTRON

Four methods of breeding high producing strain of Lipase are studied. As a result, the Lipase activity produced by one strain is 93 times as high as befor, and the Lipase activity produced by another is 3.0 times. On these bases, the effects of four methods in breeding are discussed, and the usinng Fast Neutron is more effective method in breeding.
6.Effects of Fasudil on the expression of Nogo-A and NF200 in neonatal hypoxic-ischemic brain damage rats

Objective To study the effects of Fasudil on expression of Nogo-A and NF200 in neonatal hypoxic-ischemic brain damage(HIBD) rats.Methods One hundred and twenty 7-day-old Wistar rats were divided into 3 groups with random number table:Sham operation group (n =40),HIBD group (n =40) and Fasudil group (n =40).Sham group only separated from the common carotid artery,without ligation,direct suture the incision does not do hypoxia; HIBD group were injected with saline; Fasudil group was injected with fasudil(10 mg/kg).The rats were killed at 6 h,12 h,24 h,72 h,7 d,after administration.The pathological changes were observed by means of HE.The expression of Nogo-A and NF200 was studied with immunohistochemical staining.Results 1.Naked eye observation:Sham group bilateral symmetrical cerebral hemispheres; HIBD group of brain edema aggravated,the visible hemisphere focal necrosis; fasudil treatment group of edema than HIBD group ease.2.HE stain:the structure and shape of brain in Sham operation group were normal.In HIBD group,the cells became edema,karyopyknosis,lyse,and the inflammatory cells became more.The number of edema cells and karyopyknosis decreased in Fasudil group.3.Immunohistochemical stain:there were less expressions of Nogo-A in Sham operation group.It increased slightly after 12 h in HIBD group but decreased later.The expression of Nogo-A in Fasudil group was less than the other two groups at any time except 6 h (P <0.01).There was more expression in HIBD and Fasudil group compared with Sham operation group(P <0.01).NF200 was less expression in Sham operation group.NF200 appeared after 6 h and became less after 12 h.The expression of NF200 was at 24 h and later became more.The expression of NF200 in Fasudil group was more with HIBD group at each different time (P < 0.01).The expressions of NF200 in Fasudil and HIBD group were more compared with Sham operation group.Conclusions Fasudil can rehabilitate the damaged axon and promote nerve regeneration through controlling the Rho/Rock and make the expression of NF200 increase.
7.Association of gene polymorphism of signal transducer and activator of transcription 3 with rheumatoid arthritis in the Chinese Han population

BACKGROUND:Signal transducer and activator of transcription 3 (STAT3) is an important cytokine signaling pathway, which plays an important role in inflammatory diseases. However, it is unclear whether gene polymorphism of STAT3 is associated with rheumatoid arthritis in the Chinese Han population. OBJECTIVE: To explore the association of gene polymorphism of STAT3 with rheumatoid arthritis in the Chinese Han population. METHODS:Four tag-single-nucleotide polymorphisms (tag-SNPs) in STAT3 were selected from the Chinese Han population of HapMap database. The study was performed with 228 rheumatoid arthritis cases and 228 normal controls. Four tag-SNPs (rs12601982, rs2293152, rs8078731 and rs9912773) were examined by matrix-assisted laser desorption/ionization-time-of-flight mass spectrometry. RESULTS AND CONCLUSION:The frequency of GG genotype at rs9912773 was 18.9% and 10.5% respectively in rheumatoid arthritis and control groups, and there was a significant difference between the two groups (P < 0.05). These results indicate the possible association between the STAT3 gene polymorphism at rs9912773 and the susceptibility of rheumatoid arthritis in the Chinese Han population.
8.Effects and mechanisms of Rapamycin on renal interstitial fibrosis in rats

Objective To discuss the intervention effects and mechanisms for Rapamycin on renal interstitial fibrosis in rats.Methods A total of 72 Wistar male rats were randomly divided into 3 groups,normal group(n =24) ,model group(n =24), treatment group (n =24).The model group and treatment group received adenine 200 mg/kg daily,and the treatment group was also given Rapamycin 5 mg/(kg·d) at the 8th day,the normal group was just given the same amount of normal saline for 6 weeks.In the end of the 2nd,4th and 6th week,8 rats in each group were sacrificed respectively.The expression of hepatocyte growth factor(HGF), transforming growth factor-β (TGF-β) and neutrophils gelatinases related apolipoprotein (NGAL) in each group were observed.The software of image analysis system was used for semi-quantitative analysis.Results HE and Masson staining results showed that the renal tubular were progressive swelling, and changed with interstitial fibrosis,atrophy and even necrosis in model group from 2 weeks to 6 weeks.The pathological changes of kidney were more ease in the treatment group compared with those in model group.Immunohistochemical staining results showed that HGF expression levels of renal interstitial tissue in model group and treatment group at the 2nd week were significantly higher than those of normal group(P <0.05), and were significantly decreased at the 4th week and 6th week (P < 0.05);HGF expression levels of renal interstitial tissue in treatment group were significantly higher than those in model group (P < 0.05).NGAL expression levels of renal interstitial tissue in model group and treatment group at the 2nd week were significantly higher than those in normal group(P <0.05) ,and were significantly decreased at the 4th week and 6th week(P < 0.05);NGAL expression levels of renal interstitial tissue in treatment group were significantly higher than those in model group (P < 0.05).TGF-β expression levels of renal interstitial tissue in model group and treatment group at the 2nd,4th,and 6th week were significantly higher than those in normal group (P <0.05) ,while TGF-β expression levels of renal interstitial tissue in treatment group were significantly lower than those in model group (P < 0.05).Conclusion Rapamycin could improve the rat kidney tissue pathology, relieve renal tubular expansion, and slow progression of renal interstitial fibrosis, and has certain protective effect to the kidney.
9.Progress in Gene Logic Networks

As tremendous genomic data avalanches, exploring biological mechanism by data analysis and theory methods has become important for theoretical biology research. This method is significant for the study of complex gene functions and gene networks. Bowers used higher order logic relationships to decipher protein network organization, which is a systemic method called logic analysis of phylogenetic profiles (LAPP). LAPP is a data modeling and different from traditional computational methods. This computational approach identifies logic relationships of the elements (or components) in complex networks through the logic analysis of their expression data. The method can be used to infer functional relationships of two associated proteins to one another. It is important for discovering the new function mechanism of the protein. The clusters of orthologous groups (COGs) involved in a gene network usually are large groups and therefore LAPP is also an approach for complex gene logic networks. After the establishment of the gene logic network, it is convenient for the regulation of gene through the network. The method can used in many fields, such as species evolution, oncologic diagnosis and so on. LAPP was systematically described and analyzed and recent developments in methodologies and applications were highlighted. Some opinions of them were also given.
10.Experimental research of gaits based on young plantar pressure test.

The present paper is to study the center line of the plantar pressure of normal young people, and to find the relation between center line of the plantar pressure and gait stability and balance. The paper gives the testing principle and calculating methods for geometric center of plantar pressure distribution and the center of pressure due to the techniques of footprint frame. The calculating formulas in both x direction and y direction are also deduced in the paper. In the experiments carried out in our laboratory, the gait parameters of 131 young subjects walking as usual speed were acquired, and 14 young subjects of the total were specially analyzed. We then provided reference data for the walking gait database of young people, including time parameters, space parameters and plantar pressure parameters. We also obtained the line of geometry center and pressure center under the foot. We found that the differences existed in normal people's geometric center line and the pressure center line. The center of pressure trajectory revealed foot movement stability. The length and lateral changes of the center line of the plantar pressure could be applied to analysis of the plantar pressure of all kinds of people. The results in this paper are useful in clinical foot disease diagnosis and evaluation of surgical effect.
