1.Current progress in proteomics research of proliferative vitreoretinopathy

Proliferative vitreoretinopathy (PVR)is a serious causing-blindness disease.It is intimately associated with retinal detachment (RD),proliferative diabetic retinopathy (PDR)and penetrating ocular injury with the complicated course and aggravating outcome.Proteomics is a science about investigating the total proteins in cellular level or organism level.The study of proteomics is helpful for reinforcing the cognition and understanding of pathogenesis and development of PVR.So proteomics study about PVR is becoming a new topic.The study progress in the mechanism of PVR,the study background and technique of proteomics,the application of proteomics study in PVR and relative ocular diseases mentioned above are reviewed in this paper.
2.Sexual psychology changes and analysis of its related factors for ovarian cancer patients after operation

Objective To evaluated the sexual psychology changes and marriage quality for ovarian cancer patients after operation and supply selective psychological nursing intervention for patients and their family members.Methods Self-designed questionnaire was used to investigated ovarian cancer patients under 60 years old one month after operation to understand the sexual psychology changes and its related factors.Nursing intervention was adopted according to different conditions of patients.Results Factors that influenced the sexual psychology of patients included:age,education degree,chemotherapy process,clinical symptom,anxiety and depression status,marital status and degree of nursing intervention.Conclusions Sexual psychology Was influenced by many factors.We should adopt various kinds of nursing intervention to promote the physical and mental health of patients,facihtate the recovery of sexual function and improve their life quality.
3.Influence of foot bath with traditional Chinese medicine combined with foot massage on sleep quality of patients with insomnia

Objective To evaluate the combination of foot bath using tradirional Chinese medicine and foot massage on sleep quality of patients with insomnia. Methods 84 cases of insomnia patients were divided into the treatment group(44 cases) and the control group(40 cases)according to their willingness whether to accept foot bath and foot massage or not. The control group received routine care, the treatment group was given foot bath using traditional Chinese medicine and foot reflex zone massage at bedtime on the basis of the routine care. The treatment effect were evaluated after 10 days of continuous intervention. Pittsburgh Sleep Quality Index Scale (PSQI)was used to assess sleep quality of patients. Results The treatment efficacy and quality of sleep of the treatment group was significantly better than the control group. After treatment, sleep condition in the treatment group significantly improved than before. ConduMons The combination of foot bath using traditional Chinese medicine and foot massage can improve sleep quality in patients with insomnia.
4.Interaction among peroxisome proliferators-activated receptor alpha, cytochrome P450 oxysterol 7α-hydroxylase and estrogen receptor and its association with intrahepatic cholestasis in pregnant rats

Objective To investigate the relationship between interaction of peroxisome proliferators-activated receptor alpha (PPARα), cytochrome P450 oxysterol 7α-hydroxylase (CYP7B1) and estrogen receptor (ER) and intrahepatic cholestasis in pregnant rats. Methods Eighty clean SD pregnant rats were selected and divided into four groups randomly with 20 in each. Since the 13th day of pregnancy,rats in the control group was injected subcutaneously with refined vegetable oil 2.0 ml · kg-1 · d -1 , those in the low-dose, moderate-dose and high-dose groups received 17-α-ethynylestradiol (EE) 1.0 mg · kg-1 · d-1,1.25 mg · kg-1 · d-1 and 1.5 mg · kg-1 · d-1, respectively. All rats were sacrificed at the 21at day of pregnancy and maternal hepatic tissues were collected. The serum levels of alanine aminotransferase(ALT), aspartate transaminase (AST), total bile acid (TBA) and bilirubin (BIL) were determined by enzyme linked immunosorbent assay (ELISA). The mRNA expressions of PPARα, CYP7B1, Erα and Erβ in maternal rat livers were examined by real-time PCR. Results (1) Biochemical indicators: the serum levels of ALT,AST, TBA and BIL were significantly lower in the control group than in the rest 3 groups,respectively [ control group: (41.1 ± 2.8 ) U/L, (44.4 ± 3.6) U/L, (26.4 ± 5.6 ) μmol/L and( 2.8 ± 0.2)U/L;low-dose group: (48.2 ±3.4) U/L,(47.9 ±3.7) U/L,(36.4 ±4.2) μmol/L and (4.2 ±0.2) U/L;moderate-dose group: (70.4 ± 5.3 ) U/L, (68.4 ± 5.6) U/L, (64.3 ± 3.8 ) μmol/L and ( 6.2 ± 1.2)U/L; high-dose group: (72.4 ±7.6) U/L, (70.2 ±3.8) U/L, (72.4 ±7.8) μmol/L and (8.2 ±2.2)U/L, P<0.05], and those in the moderate or high-dose groups were higher than in the low-dose group (P<0.05). (2) mRNA expression of Erα and Erβ: the mRNA expression of Erα in pregnant rat livers increased in a dose-dependent manner, which were all significantly higher than that in the control group,respectively ( low-dose group: 0.76 ± 0.02 ); moderate -dose group: ( 0.99 ± 0.04; high-dose group:1.21 ±0.01 ;control group:0.65 ±0.01, P <0.05), but no difference was found among the 4 groups in the mRNA expression of Erβ ( P > 0.05 ). (3) mRNA expression of CYP7B1 and PPARα: the mRNA expression of CYP7B1 in pregnant rat livers increased from the low-dose group to the high-dose group, and were all higher than that of the control group ( low-dose group: 0.93 ± 0.01; moderate-dose group: 0.99 ±0.06; high-dose group: 1.22 ± 0.04; control group: 0.75 ± 0.02, P < 0.05 ). However, the mRNA expression of PPARα decreased from the low-dose group to the high-dose group, and were all lower than that of the control group (low-dose group: 0.83 ± 0.05; moderate-dose group: 0.71 ± 0.02; high-dose group:0.64 ± 0.03; control group: 1.35 ± 0. 05; P < 0.05 ) . Conclusions The down regulated mRNA expression of PPARα, caused by higher dose of estrogen, may increase the expression of CYP7B1 due to the ineffectiveness of the inhibition of PPARα on CYP7B1, which may further stimulate the Erα activity and then induce intrahepatic cholestasis. Abnormal expression of PPARα, CYP7B1 and ER may play a role in the pathogenesis of estrogen-induced intrahepatic cholestasis.
5.BREEDING OF HIGH PRODUCING STRAIN OF LIPASE USING FAST NEUTRON

Four methods of breeding high producing strain of Lipase are studied. As a result, the Lipase activity produced by one strain is 93 times as high as befor, and the Lipase activity produced by another is 3.0 times. On these bases, the effects of four methods in breeding are discussed, and the usinng Fast Neutron is more effective method in breeding.
6.Progress of the relationship between eukaryotic initiation factor 5A and human diseases

Eukaryotic initiation factor 5A (eIF5A),which is ubiquitous in the eukaryote,is the unique protein containing the special lysine hypusine.There are evidences show that eIFSA is involved in the progress of translation,peptide elongation and peptide bond formation,and takes part in the occurrence and development of various human diseases.The activation of eIF5A is essential to its biological function.The active eIFSA is involved in the growth of plasmodium and contributes to tumor proliferation.And it can act as nuclear output protein to integrate special mRNA,and in this way the special mRNA can shuttle between the nucleus and cytoplasm.Basing on these findings,the scientists pay more attention to eIF5A,and deem it as a direction to cure some related human diseases.
7.STUDIES ON THE CLONAL GROWTH AND PASSAGE OF HUMAN EMBRYONIC STEM CELLS

Objective Try to determine the relationship between blastocyst quality,the clonal growth of inner cell mass(ICM)and the establishment of human embryonic cell line. Methods Coculture D 3 discarded embryos with mouse embryonic fibroblast cells(MEFs).Then remove trophoectoderm by immunosurgery after getting different quality blastocysts.Culture ICM and passage these cells on MEFs. Results Human embryonic stem cells derived from good quality blastocysts could be passaged further than that from poor quality blastocysts,and ICMs growing fast could be passaged more quickly and efficiently.Conclusion The establishment of human embryonic stem cells is closely related with blastocyst quality and the original growth of ICM.
8.Choice of smoking cessation methods and its influencing factors in moderate-to-severe nicotine-dependent population

Objective·To analyze the choice of smoking cessation methods and its influencing factors in moderate-to-severe nicotine-dependent population.Methods·A retrospective clinical analysis was conducted for170 moderate-to-severe nicotine-dependent smoking-quitters (Fagerstr(o)m scale ≥ 4).Different smoking cessation methods (pharmacotherapy and non-pharmacotherapy) and abstinence rates of the selected population (7 d point prevalence abstinence rates and sustained abstinence rates 1,3,and 6 months after treatment) were compared and possible influencing factors were analyzed.Results·The differences in choosing the smoking cessation method in moderate-to-severe nicotine-dependent population with different ages,levels of education,and employment status were statistically significant (P=0.000).The Logistics regression analysis showed that the level of education was the major factor for choosing the smoking cessation method (P=0.006).The 7 d prevalence abstinence rate was much lower for pharmacotherapy than for non-pharmacotherapy 1 month after treatment (P=0.000) and was much higher for pharmacotherapy than for non-pharmacotherapy 3 and 6 months after treatment (P=0.000).The sustained abstinence rate was much higher for pharmacotherapy than for non-pharmacotherapy 3 and 6 months after treatment (P=0.002,P=0.000).Conclusion·For moderate-to-severe nicotine-dependent smoking-quitters,attention should be paid to the formulation and implementation of personalized strategies.The choice of smoking cessation methods depends on the age,level of education,and employment status.Reasonable pharmacotherapies can significantly increase the abstinence rate.
9.Technology Optimization of Supercritical CO2 Extraction from Zingiberis rhizoma

OBJECTIVE:To optimize the technology of supercritical CO2 extraction from Zingiberis rhizoma. METHODS:With the comprehensive score of the contents of 6-ginger phenol,8-ginger phenol and 10-ginger phenol and the extraction rate of the oil from Z. rhizoma as the index,uniform design method was adopted to investigate the effects of extraction pressure,extrac-tion temperature and extraction duration on the extraction result;verification tests were conducted. RESULTS:The optimal condi-tions were as follows as the extraction pressure of 25 MPa,extraction temperature of 30 ℃ and extraction duration of 2 h. In the verification tests,the average extraction rate of the oil from Z. rhizoma was 3.2%(n=3),and the comprehensive score was 1.874 2 (RSD=0.65%,n=3),with the relative deviation of 0.6% between the measured value and the predicted value. CONCLUSIONS:The optimal extraction technology is stable and feasible,with the advantages of low temperature,short duration.
10.Interleukin-1α induces immunosuppression by mesenchymal stem cells in promoting the growth of prostate cancer cells

Objective To explore the preliminary mechanism of mesenchymal stem cells (MSCs) in promoting prostate cancer proliferation in tumor inflammatory microenvironment.Methods From April 2013 to October 2013,MSCs pretreated with inflammatory cytokine IL-1α (MSCs (IL-1α)) and its culture supernatants mixed with RM-1 cells,which origined from C57BL/6 mice,were subcutaneously administered in the armpit area of C57BL/6 or BALB/c mice to establish homologous or heterologous transplant animal mode and to detect the tumor growth.Meanwhile the influence of MSCs on the proliferation of spleen cells was detected in vitro.Results In homologous transplant model,the relative tumor weight of prostate cancer cells prtreated with MSCs and MSCs (IL-1α) and their culture supernatant were (3.4 ± 0.2),(3.3 ±0.2),(4.9±0.5),and (5.2±0.6) g.The results were statistically significant (P<0.05) compared with the control group (2.4±0.2) g.In heterologous model,the ratio of tumor formation of the pretreated groups were 50%,50%,80% and 80%,respectively,compared with the control group of 0%.The results were statistically significant (P<0.05).In proliferation experiments of spleen cells,the number of spleen cell pretreated with IL-1α were significantly lower than that in control group and unpretreated group (P < 0.05).Conclusions MSCs pretreated with IL-1α could effectively promote the growth of prostate cancer cell in vivo.The reason may be due to inflammatory cytokines induce immune suppression of MSCs and then lead to immune escape of cancer cells.
