Objective To investigate the influence of intravenous thrombolysis to acute cerebral infarction with cerebral microbleeds (CMB). Methods Eight-nine patients with cerebral infarction in the Neurological Department of the Second Affiliated Hospital of Guangzhou Medical College from January 1st , 2011 to December 31st, 2012 were enrolled in this study. All patients were scanned with MRI and GRE. According to the presence of CMB, the patients were classified into two groups, including CMB group and non-CMB group. Past history of smoking, drinking, hypertension, lacunar infarction, diabetes and leukoaraiosis situation were recorded and risk factors of CMB were explored. All patients were treated with intravenous thrombolysis and rescanned with MRI to compare the total number of CMB 24 hours after thrombolysis. Results In the CMB group, the number of patients with mild CMB decreased and the number of patients with serious CMB increased after thrombolysis. CMB was associated with sex, age, hypertension, lacunar infarction and leukoaraiosis. Conclusions Sex, age, hypertension, lacunar infarction and leukoaraiosis were the risk factors of CMB in acute cerebral infarction. Thrombolysis in acute cerebral infarction augments the incidence of CMB and promotes the hemorrhagic transformation.

Purpose: To study the clinical efficacy, toxicity of preoperative chemotherapy and survival time with Gemicitabine-Cisplatin combination in the treatment of stage Ma( N2) NSCLC. Methods: Thirty patients with stage IIIa( N2) NSCLC were included. Gemicitabine was administered on dl, 8 and 15 at a dose of 1000 mg/m" and Cisplatin at a dose of 100 mg/m on d2. The chemotherapy was repeated every 28days. Results: Thirty patients were evaluable for response. The overall response rate was 70%. Surgical'excision rate after preoperative chemotherapy was 93%. Total surgical excision rate was 70%. Median survival time was 15 months, one year survival rate was 67%. The main toxicity was hematological, thrombocytopenia of grade III-IV appeared in 46% course of treatment, but it did not lead to hemorrhage. Conclusions: Preoperative chemotherapy with Gemicitabine-Cisplatin combination is effective and well-tolerated in the treatment of stage IIIIa( N2) NSCLC. So it is worthy to be further studied and popularized.

Objective: To explore CT characteristics of mycobacterium intracellulare pulmonary diseases compared with mycobacterium kansasii, mycobacterium abscess/chelonei pulmonary diseases. Methods: CT images of 35 patients with mycobacterium intracellulare pulmonary diseases, 18 with mycobacterium kansasii pulmonary disease and 22 with mycobacterium abscess/chelonei pulmonary disease confirmed by clinical data and laboratory tests were retrospectively analyzed, and imaging findings were evaluated and compared. Results: Mycobacterium intracellulare pulmonary disease involved both lungs (33/35, 94.29%), multiple lobes (18/35, 51.43%) or all lobes (16/35, 45.71%). CT characteristics included cord shadow (34/35, 97.14%), air space consolidation (33/35, 94.29%), centrilobular nodules or tree in bud (32/35, 91.43%), thickened pleura (32/35, 91.43%), ground glass opacity (31/35, 88.57%), traction bronchiectasis (30/35, 85.71%), non-traction bronchiectasis (25/35, 71.43%), nodes (24/35, 68.57%), calcification (24/35, 68.57%) and cavity (23/35, 65.71%). Ground glass opacity was more frequently observed in mycobacterium intracellulare than mycobacterium kansasii (P=0.001) and mycobacterium abscess/chelonei pulmonary disease (P<0.001). Walls of the cavity were thicker in mycobacterium intracellulare than mycobacterium kansasii (P=0.019) and mycobacterium abscess/chelonei pulmonary disease (P=0.024). Calcifications were more frequently observed in mycobacterium intracellulare than in mycobacterium kansasii (P=0.014) and mycobacterium abscess/chelonei pulmonary disease (P=0.007). Conclusion: Compared with mycobacterium kansasii and abscess/chelonei pulmonary disease, CT findings of mycobacterium intracellulare have certain characteristics, which may be helpful to differential diagnosis.

Objective To explore the clinical effect and safety of methimazole combined with 131I in treatment of hyperthyroidism.Methods Patients (83 cases) with hyperthyroidism accepted in Xi'an Jiaotong University Affiliated San Er Ling Yi Hospital from June 2012 to June 2015 were selected and divided into observation group with 42 cases and control group with 41 cases.Patients in observation group were given methimazole combined with 131I,and patients in control group were given PTU combined with 131I.Then the clinical effect,thyroid gland serological indexes,adverse reations,and reoccurrence rates of two groups were observed and compared.Results The total clinical efficacy of observation group was 95.24％,which was obviously higher than 70.73％ of control group with statistically significance (P ＜ 0.05).TSH was getting higher after treatment of two groups,and T3,T4,FT3,FT4,TRAb,and TPOAb were getting lower after treatment of two groups.And TSH in the observation group was significantly higher than that in the control group,T3,T4,FT3,and FT4 were significantly lower than those in the control group (P ＜ 0.05).The adverse reaction rate of observation group (14.29％) was obviously lower than that of control group (46.34％) with statistically significance (P ＜ 0.05).The reoccurrence rate of observation group (4.76％) was obviously lower than that of control group (26.83％) with statistically significance(P ＜ 0.05).Conclusion Methimazole combined with 131I has good effect and safety in treatment of hyperthyroidism,which is worth of clinical application.

Objective　To determine the expression and the change of vascular endothelial growth factor(VEGF) and it's acceptor Flt-1 during maxillofacial blast injury at early stage,and to evaluate the effect of VEGF on traumatic wound healing.Methods　The rabbit model of maxillofacial blast injury was made by KTY-04 blasting cap. The expression of VEGF and Flt-1 in wound tissue was determined by ABC immunohistochemistry after injury 6h,1d,2d,3d,5d,and 7d. Also,the results were compared with preinjury groups.Results　In the wound tissue of maxillofacial blast injury, the expression of VEGF was rising steadily at the first week after injury. Comparing with normal tissue,it showed different from the first day after injury, and showed very different from the third day(P<0.01).It reached the peak at the seventh day after injury.The expression of Flt-1 showed no difference within 3 days after injury comparing with normal tissue.After injury 5d-7d,the protein expressed strengthly.Conclusion　The stage of the VEGF expression at maxillofacial blast injury is similar to the angiogenic stage during wound healing, and the Flt-1 expression is also occurring at that period.It illustrates that VEGF take part in angiogenic cascades of traumatic wound healing and product auxo-action to the regeneration of blood vessel.

Phospho-histone H2AX(γH2AX) has been widely used in vitro genotoxicity evaluation of ionizing radiation, carcinogenic substances and cigarette smoke as an important biomarker of DNA double stranded breaks ( DSBs) . The study developed an enzyme-linked immunesorbent assay for detection of the content ofγH2AX in cells to evaluate the genotoxicity of cigarette smoke. The study exposed CHO cells with cigarette smoke total particulate matter ( TPM) and cigarette smoke condensate ( CSC) of different dose, dose-effect relationship between smoke exposure and DNA damage and the poisonous difference of cigarette smoke components has been investigated by detecting the content change ofγH2 AX in cells under different exposure time and dose. In addition, reactive oxygen species ( ROS) in cells were also detected to study the mechanism of cigarette smoke exposure induced DSBs. The experiment result showed that the required time for the level ofγH2AX in cells grew to the peak prolonged with increased exposure dose. Once the level of γH2AX in cells grew to the peak value, then it decreased slowly; the level of γH2AX in cells rised with increasing dose of cigarette smoke;The effect of TPM on the content change of γH2AX was more than CSC; Moreover, smoke exposure could induce concentration increase of ROS in cells, and a good correlation of content change of ROS and γH2AX in cells were found, free radicals in cigarette smoke may be one major cause of DSBs.

Aim To develop a simple and rapid method to monitor insulin receptor kinase activity and provide a novel cell-based model for screening anti-diabetes drugs.Methods CHO cells were co-transfected by plasmids which respectively contained insulin receptor gene,STAT5b gene and luciferase gene driven by STAT5 response elements.The expression of exogenous gene in transfected cells was examined by RT-PCR.The transfected cells were treated by insulin,and then the concentration and time-dependent response of luciferase expression to insulin induction was examined.Moreover,the specificity was identified by AG1024 treatment and PTP1B gene transfection.Results Expressions of insulin receptor and STAT5b were detected in the transfected CHO cells.The expression of luciferase in transfected cells was induced by insulin in concentration and time-dependent way.The maximal induction fold was 6.25.Moreover,the inducible expression of luciferase by insulin could be specifically blocked by tyrphostin AG1024,an inhibitor of insulin receptor kinase,or co-transfected PTP1B gene.Conclusions The insulin receptor kinase activity can be detected by expression of reporter gene with high sensitivity and specificity in this cell model,and with potential value in high throughput screening for insulin receptor activators and sensitizers.

The cytoplasmic and nuclear changes during the development of goat oocytes were studied with light and electron microscopy. The oocyte development may be divided into 8 stages in accordance with the number and distribution of follicle cells and the size of the follicle. The results indicated that the Golgi complex, mitochondria, smooth endoplasmic reticulum and cortical granules became well developed and they moved into the cortical region as the oocyte development proceeded. In the oocytes in the follicle of 1.5-3mm in diameter, all the mitochondria became hooded and the number and size of fibrillar centers in the nucleolus reached maximum, but the nucleolar compaction still not occured at this stage. By the time when the follicle reached 3.5-5mm in diameter, the follicular cell processes began to degenerate, the microvilli of the oocyte withdrew from the zona pellueida and the oocytes might be cultured to mature in vitro. In the mature oocytes, Golgi complex and rough endoplasmic reticulum disappeared, the cortical granules arranged themselves in one layer beneath the oolemma, the mitochondria dispersed in the central region of cytoplasm and the eggs were ready for fertilization.

OBJECTIVETo investigate miR-20a expression in human glioma and normal brain tissues and its effect on the proliferation of glioma cells in vitro.

METHODSThe expression of miR-20a was detected in human normal brain tissues and glioma tissues by real-time RT-PCR. miR-20a mimics were synthesized and transfected into U251 cells via liposome, and the cell proliferation were detected using MTT assay and flow cytometry.

RESULTSThe glioma tissues showed significantly up-regulated expression of miR-20a compared with normal brain tissues (P=0.035). The expression level of miR-20a was higher in high-grade than in low-grade gliomas. miR-20a mimics significantly enhanced the proliferation of U251 cells and the percentage of S-phase cells.

CONCLUSIONmiR-20a shows potent effect in promoting the growth of glioma cells, suggesting its important role in the pathogenesis of human glioma.

Objective: Study of the results and the degree on occupational noise-induced deafness in-to the different high frequency hearing threshold weighted value, in order to provide theoretical basis for the re-vision of diagnostic criteria on occupational noise-induced deafness. Methods: A retrospective study was con-ducted to investigate the cases on the diagnosis of occupational noise-induced deafness in Guangdong province hospital for occupational disease prevention and treatment from January 2016 to January 2017. Based on the re-sults of the 3 hearing test for each test interval greater than 3 days in the hospital, the best threshold of each frequency was obtained, and based on the diagnostic criteria of occupational noise deafness in 2007 edition, Chi square test, t test and variance analysis were used to measure SPSS21.0 data, their differences are tested among the means of speech frequency and the high frequency weighted value into different age group, noise ex-posure group, and diagnostic classification between different dimensions. Results: 1. There were totally 168 cases in accordance with the study plan, male 154 cases, female 14 cases, the average age was 41.18 ±6.07 years old. 2. The diagnosis rate was increased into the weighted value of different high frequency than the mean value of pure speech frequency, the weighted 4 kHz frequency increased by 13.69% (χ²=9.880, P=0.002) , 6 kHz increased by 15.47% (χ²=9.985, P=0.002) and 4 kHz+6 kHz increased by15.47% (χ²=9.985, P=0.002) , the difference was statistically significant. The diagnostic rate of different high threshold had no obvious differ-ence between the genders. 3. The age groups were divided into less than or equal to 40years old group (A group) and 40-50 years old group (group B) , there were higher the diagnostic rate between high frequency weighted 4 kHz (A group χ²=3.380, P=0.050; B group χ²=4.054, P=0.032) , weighted 6 kHz (A group χ²=6.362, P=0.012; B group χ²=4.054, P=0.032) , high frequency weighted 4 kHz+6 kHz (A group χ²=6.362, P=0.012; B group χ²=4.054, P=0.032) than those of speech frequency average value in the same group on oc-cupational noise-induced deafness diagnosis rate, the difference was statistically significant. There was no sig-nificant difference between age groups (χ²=2.265, P=0.944) . 4. The better ear's mean value of pure speech fre-quency and the weighted values into different high frequency of working years on each group were compared, working years more than 10 years group was significantly higher than that of average thresholds of each frequen-cy band in 3-5 group (F=2.271, P=0.001) , 6-10 group (F=1.563, P=0.046) , the difference was statistically significant. The different high frequency weighted values were higher than those of the mean value of pure speech frequency, and the high frequency weighted 4 kHz+6 kHz had the highest frequency difference, with an average increase of 2.83 dB. 5. The diagnostic rate into weighted different high frequency was higher in the mild, moderate and severe grades than in the pure speech frequency. In the comparison of diagnosis for mild occupational noise-induced deafness, in addition to the weighted 3 kHz high frequency (χ²=3.117, P=0.077) had no significant difference, the weighted 4 kHz (χ²=10.835, P=0.001) , 6 kHz (χ²=9.985, P=0.002) , 3 kHz+4 kHz (χ²=6.315, P=0.012) , 3 kHz+6 kHz (χ²=6.315, P=0.012) , 4 kHz+6 kHz (χ²=9.985, P=0.002) , 3 kHz+4 kHz+6 kHz (χ²=7.667, P=0.002) were significantly higher than the diagnosis rate of the mean value of pure speech frequency. There was no significant difference between the two groups in the moderate and se-vere grades (P>0.05) . Conclusion Bring into different high frequency hearing threshold weighted value in-creases the diagnostic rate of occupational noise-induced deafness, the weighted 4 kHz, 6 kHz and 4 kHz+ 6 kHz high frequency value affects the result greatly, and the weighted 4 kHz+6 kHz high frequency hearing threshold value is maximum the effect on occupational noise-induced deafness diagnosis.
