Chinese Journal of Rehabilitation Theory and Practice 2008;14(8):754-756
Objective To explore the efficacy and mechanism of exogenous reduced glutathione(GSH)on cardiotoxicity induced by epirubicin(EPI).Methods 60 postoperative patients with breast cancer were randomly divided into the treatment group and control group with 30 cases in each group. The treatment group received 6 cycles of chemotherapy with CEF regimen plus the reduced GSH.The control group also received 6 cycles of chemotherapy only with CEF alone. The cardiotoxicity of all patients were estimated by the ECG,myocardial enzymes, troponin Ⅰ(cTnⅠ),ultrasoundcardiogram and tissue Doppler technique before hemotherapy and after 3 cycles and 6 cycles of chemotherapy. Results After 3 cycles of chemotherapy, there were no significant differences in ECG,cTnⅠ,left ventricle ejection fraction(EF),fraction shortening(FS),E/A,decline time(DT)and myocardial creatine kinase(CK),isoenzyme of creatine kinase containing M and B subunits(CK-MB),lactate dehydrogenase(LDH)between two groups(P>0.05);While,there was a significant difference in Em/Am between two groups(P<0.01).After 6 cycles of chemotherapy, there were also no significant differences in EF,FS,myocardial CK,CK-MB,LDH between two groups(P>0.05);there were significant differences in ECG,cTnI,E/A,DT and Em/Am between two groups(P<0.01).Conclusion The reduced GSH can prevent the cardiotoxicicy induced by EPI,which may be related with the ability of GSH against the oxidant injury and inhibiting calcium overload.

Objective The objective of this study was to explore the expression of miRNA-124(miR-124)and its correlation with E-cadherin and androgen receptor(AR)in triple-negative breast cancer(TNBC).Methods The expression of miR-124 was detected by RT-PCR in TNBC tissues and adjacent normal breast tissues. The expression of E-cadherin and AR in TNBC was detected by immunohistochemistry. Results The expression of miR-124 in TNBC tissues was significantly correlated with histological grade and the expression of E-cadherin(P<0.05),and the expression of miR-124 in TNBC tissues was significantly lower than that in adjacent normal breast tissues. No correlation was found between miR-124 and AR in TNBC tissues(P>0.05).Conclusion In TNBC,miR-124 may play an anti-tumor effect by modulating the expression of E-cadherin.

Objective:To investigate neuroprotective effect of alpinetin on ischemic stroke(IS)rats by regulating Shh/Gli1 signaling pathway. Methods:Fifteen rats were randomly collected as control group, remaining rats were used to construct an IS model.Rats that successfully modeling were randomly grouped into model group, alpinetin group(5 mg/kg),Shh/Gli1 signaling pathway inhibitor cycloparamide group(15 mg/kg)and alpinetin+cycloparamide group(5 mg/kg alpinetin+15 mg/kg cycloparamide),with 15 rats in each group, and administered once a day for two consecutive weeks, control group and model group were given equal amounts of physiological saline.Zea-Longa scoring method was applied to evaluate neural function;ELISA was applied to detect inflammatory factors levels;mass of brain tissue was weighed and water content of brain tissue was calculated;TTC staining was applied to measure volume of cerebral infarction;HE staining was applied to observe nerve cell damage;TUNEL staining was applied to detect neuronal apoptosis;qRT-PCR and Western blot were used to detect mRNA and protein expressions of Shh and Gli1.Results:There were no abnormalities in hippocampal tissue of control group, while hippocampal tissue structure of model group rats was abnor-mal,with disordered cell arrangement and nuclear pyknosis of nerve cells, cell damage rate,Zea-Longa score, infarct volume,IL-1β,IL-6,TNF-α,brain tissue water content, and cell apoptosis rate in model group were obviously higher than control group(P<0.05),mRNA and protein levels of Shh and Gli1 were obviously decreased(P<0.05);compared with model group, cells in alpinetin group were arranged more neatly, and phenomenon of neuronal cell nucleus pyknosis was improved, cell damage rate,Zea-Longa score, infarct volume,IL-1β,IL-6,TNF-α,brain tissue water content, and cell apoptosis rate were obviously decreased(P<0.05),mRNA and protein levels of Shh and Gli1 were obviously increased(P<0.05),trend of above indicators in cyclophosphamide group was opposite, ciclopramide reversed neuroprotective effect of alpinetin on IS rats. Conclusion:Alpinetin may exert neuroprotective effects on IS rats by activating Shh/Gli1 signaling pathway.

Objective:To report a case with frontotemporal dementia (FTD) characterized by involuntary laughter. Methods:The clinical manifestations and imaging characteristics of a patient diagnosed as FTD was analyzed. Then the results of cerebrospinal fluid, positron emission tomography-computed tomography (PET-CT) and single-photon emission computed tomography examinations were collected. Blood samples were tested for related genes of FTD.Results:The patient is a 66 years old woman with insidious onset and progressing symptoms and she was mainly manifested as laughing out loud involuntarily when looking at others, childishness, stubbornness, loss of interest, irritability and other personal changes. Mild motor and language disorders were also manifested as moving slowly and speaking unclearly. The magnetic resonance imaging showed the atrophy of bilateral frontal, temporal lobe and bilateral hippocampal while the image of PET-CT showed the metabolism was reduced in different degrees. Eventually, behavioural variant of FTD was diagnosed. The result of ANXA11 gene sequencing revealed the mutation of c.107C>G(p.P36R).Conclusions:This is the first case in which a heterozygous mutation of ANXA11 gene, which is related to amyotrophic lateral sclerosis (ALS), is found in simple FTD patient, suggesting that ANXA11 gene may play an important role in the pathogenesis of FTD. This further supports the theory that ALS and FTD are spectrum disorders.

Objective:To compare demographic characteristics, clinical characteristics, therapeutic characteris-tics and physiological indicators of patients with bipolar Ⅰ disorder and bipolar Ⅱ disorder. Methods:A total of 381 patients with bipolar disorder(BD)diagnosed by the Diagnostic and Statistical Manual of Mental Disorders 5 th Edi-tion(DSM-5)were selected, including 302 patients with BD-Ⅰ(79.27%),74 patients with BD-Ⅱ(19.42%)and 5 patients with other specific and related disorders(1.31%).Demographic and clinical characteristics were collected with self-designed clinical information questionnaire.Multivariate logistic regression and multivariate linear regres-sion analysis were used for analysis. Results:Compared with patients with BD-Ⅱ,patients with BD-Ⅰ had more risk to have psychotic features(OR=5.75,95%CI:2.82-11.76),longer disease duration, and more repeated transcra-nial magnetic therapy(OR=3.09,95%CI:1.02-9.35),higher uric acid, total cholesterol and high-density lipo-protein.BD-Ⅰ in Han nationality was more common(OR=11.50,95%CI:1.76-75.30),and had lower education level(OR=10.22,95%CI:1.16-89.77),and less family history of psychosis(OR=2.34,95%CI:1.01-5.42).Conclusion:There are significant differences between BD-Ⅰ and BD-Ⅱ in demographic and clinical charac-teristics,treatment status, and physiological indicators, which could provide clues for exploring the pathogenesis of bipolar disorder.
