1.Conditional Logistic Regression Analysis on Risk Factors of Colorectal Cancer

Objective To investigate the risk factors of color-ectal cancer and to provide scientific basis for prevention of colorectai canc-er. Methods A 1:1 matched case-control study was carried out in Dalian including 200 cases with colorectal cancer and 200 controls. We analyzedthe data with the univariate analysis and mutivariate conditional logistic re-gressiou,theu calculated the Odds Ratio and the 95% confidence. Results Conditional logistic regression showed that history of constipation, family history of other tumor,intake much bloat food ten yeats ago and easily an-gry were the risk factors of colorectal cancer, and intake much fresh vegeta-bles ten years ago was the protective factors of colorectal cancer. One way analysis showed that following factors were the risk factors of coloroctal cancer:there were pollution factor around home 10 years ago, belly CT test,sleep over twelve o'clock at night,family history of coloroctal cancer, history of pries, appendicitis and the operation history of appendicitis, intake much fry and bake food,seashell and animal liver before ten years ago,of-ten feel strew, don't like to communicate with others,pessimism,don't harmonization with colleague. And iutaking much chicken meat, bean prod-ucts,garlic ten years ago,doing exercise and the frequency of exercise,high income were the protective factors of colorectal cancer. Conclusion Coloroctal cancer was the result of many factors. But the incidence of that in Dalian city was related to history of constipation, family history of other tumor, easily angry, intake less fresh vegetables and intake much bloat food ten years ago.
2.Exploring derivation of renocortical prostaglandins in C-BSA nephritis in rats

Objective To explore derivation of renocortieal prostaglandins in C-BSA nephritis. Methods The interfering effect of goat-antiplatelet IgG serum(APS)on this model was studied. Results The levels of renocortical PGE_2、6-keto-PGF_(1?) (PGI_2)、TXA_2 were all reduced in experimental group. The pathological damage of experimental group were less severe in comparison with the model group. Proteinuria of the experimental group was alleviated. Conclusion Platelet may play a pathogenetic role of C-BSA nephritis and renocortical prostaglandins partly derive from platelets.
3.Effect of isoflurane on expression of IL-1β mRNA,IL-6 mRNA and TNF-α mRNA In the hippocamapus of immature rats

Obiective To investigate the effect ofisoflurane on expression of IL-1β mRNA,IL-6 mRNA and TNF-α mRNA in the hippocampus of immature rats.Methods sixty-four 7-clay-old SD rats were randomly assigned into 2 groups(n=32 each):control group(group C)and isoflurane group(group S).group S was exposed to 1.5% isoflurane for 6 h while group C to air.Fore animals were killed before anesthesia(T0,baseline),at 2,4,6 h(T1-3)of isoflurane anesthesia and 4,6,12 and 24 h after anesthesia(T4-7).The hippocampi were immediately removed for determimation of the expression of IL-1β mRNA,IL-6 mRNA and TNF-α mRNA by RT-PCR.Results Compared with group C,the expression of IL-1β mRNA at T1-5,IL-6 mRNA at T2.3 and TNF-α mRNA at T1-6 in the hippocampus was upregulated in group S.Conclusion The expression of IL-1β mRNA,IL-6 mRNA and TNF-β mRNA was elevated in the hippocampus of immature rats after being exposed to isoflurane.
4.EFFECTS OF TWO ACIDIC MUCOPOLYSACCHARIDES FROM SEA CUCUMBERS ON THE INTERACTION OF THROMBIN WITH ENDOTHELIAL CELLS

SJAMP and HLAMP are glucose-containing acidic mucopolysacc-harides isolated from sea cucumbers, Stichopus Japonicus and Holot-huria Lecospilota respectively. Their effects on the interaction of thrombin with endothelial cells were studied. Human vascular endothelial cells isolated from umbilical veins treated with 0.1% collagenasc were cultured with MEM medium. The contents of von Willebrand factor (vWF) and 6-keto-PGFla, stable hydrolysis product of PGI,, in the culture medium were measured with ELISA and immunoradioassay technique. The addition of thrombin to confluent monolayer endothelial cells resulted in a significant increase in the release of vWF and 6 ~ keto-PGFla in the culture medium. When heparin,SJAMP or HLAMP was added, no significant amount of vWF or 6~keto-PGFla could be detected in the medium .Whereas preincubation of thrombin with heparin, SJAMP or HLAMP respectively inhibited the thrombin-induced vWF and 6-keto-PGF 10 release and this effect was well correlated with the dose of heparin, SJAMP or HLAMP, indicating that SJAMP, HLAMP as well as heparin had inhibitory effect on the interaction of thrombin with endothelial cells
5.Expression of coagulation-and fibrinolysis-related molecules of endothelial cells induced by activated complement alternative pathway and intervention

Aim To investigate the change of molecu-lar expression related to coagulation and fibrinolysis in human microvascular endothelial cells ( HMEC ) in-duced by activated complement alternative pathway and effect of pyrrolidine dithiocarbamate ( PDTC ) and res-veratrol on intervention. Methods Normal human se-rum was activated by cobra venom factor ( CVF) . After exposure of HMEC to activated complement for various times, supernatant was removed and assayed for ex-pressions of P-selectin, VWF, t-PA, PAI-1, TF, TM, and NO by using reagent kits. The expressions of the above molecules in HMEC pretreated with PDTC and resveratrol were also investigated. Results P-selectin and VWF were rapidly released by endothelial cells and the expression reached the peak at the time point of 15 min. The expressions of t-PA, PAI-1, and TF were continuously upregulated, whereas NO and TM were decreased. PDTC and resveratrol inhibited the upregulation of P-selectin, VWF, t-PA, PAI-1 and TF, and intervened the downregulation of NO. Res-veratrol further downregulated the expression of TM. Conclusion Activated complement alternative path-way can influence the expression of molecules related to coagulation and fibrinolysis in HMEC, and PDTC and resveratrol can affect this change.
6.The optimized condition for delivery gtfB antisense oligonucleotide into Streptococcus mutans

0.05).Conclusion:So-fastTM can greatly improve the penetration of PS-ODN into S.mutans and can be used as appropriate delivery system of PS-ODN for S.mutans. No matter what approaches were adopted, the uptake rate reached the maximum at 5 ?mol/L and 2 h-exposure. The penetration can not be enhanced by increasing the PS-ODN concentration and the transformation time.
7.Determination of Cucurbitacin IIa in Rat Plasma by HPLC and its Pharmacokinetic Study

Objective:To establish a sensitive and fast HPLC method for determining cucurbitacin IIa in rat plasma and investigate the pharmacokinetic characteristics after intravenous administration. Methods: The analysis was performed on a Dikma Diamonsil C18 column (250 mm × 4. 6 mm, 5 μm) at 35℃ with the mobile phase of acetonitrile-water (45∶ 55, v/v) and the UV detection at 212 nm. The plasma samples were collected after intravenous administration at different time points and measured by HPLC. The pharmaco-kinetic parameters were calculated by DAS 2. 0 software. Results: The calibration plot of cucurbitacin IIa was linear over the range 0. 146-14. 060 μg · ml-1 . The range of method recovery was 99. 02%-104. 22% and that of extraction recovery was 84. 74%-86. 80%. The intra-and inter-day precision were both less than 5%. The stability met the requirements. The main pharmacokinetic pa-rameters were showed as follows:t1/2β(h):0.732 ±0.151, 0.681 ±0.055,0.667 ±0.064;Vd (L·kg-1):0.147 ±0.089, 0.131 ±0.095,0.153±0.047; Cl(L·h-1·kg-1): 0.287±0.031,0.304±0.063,0.318±0.029andAUC0→∞(mg·h·L-1):3. 646 ± 1. 124,4. 916 ± 1. 227,9. 385 ± 1. 419. Conclusion: The validated method is successfully applied in basic pharmacokinetic study in rat plasma after intravenous administration. The plasma concentration-time curves at three dosages are all fitted three-compart-ment model. Within the examined dose range, the pharmacokinetics of cucurbitacin IIa in rat is based on linear pharmacokinetics.

9.On conversions to open surgery during laparoscopic cholecystectomy:An analysis of 25 cases

Objective To study preventive measures against conversions to open surgery during laparoscopic cholecystectomy(LC).Methods The study was based on 25 cases of conversions to laparotomy out of 852 cases of LC(2.93%)from December 1999 to October 2005 in this hospital.The clinical data revealed a bile duct injury in 3 cases,bleeding in 1 case,severe adhesion in abdominal cavity or between the gallbladder and neighboring tissues in 9 cases,disturbed anatomy and adhesion in the region of the Calot's triangle in 5 cases,cholecystoduodenal fistula in 2 cases,and Mirizzi syndrome(type Ⅱ)in 5 cases.Results The conversion to open surgery was successfully completed in all the 25 cases.No complication was encountered.Follow-up examinations for 1~2 years showed no bile duct stenosis.Conclusions Proper preoperative patient selection and use of intraoperative preventive measures are helpful to minimize the possibility of conversion to open surgery.
10.Diagnosis and treatment of heterotopic pancreas: a report of 21 cases

Objective To explore the diagnosis and treatment of heterotopic pancreas. Methods A total of 21 histologically confirmed cases of heterotopic pancreas were treated in our Department from 1986 to 1998. We retrospectively analyzed the major clinical symptoms, pathogenesis, diagnostic procedure, results of operation and pathological findings. Results Among all the patients, 9 presenting with severe symptoms were surgically treated and the prognosis was fine. Of the 9 patients, 5 had epigastric pain, 2 obstructive and 2 massive digestive hemorrhage. The other 12 asymptomatic cases were followed up for 1.8 to 12 years. It was difficult to diagnose the disease by clinical situation, observation and imaging examination and only pathological examination could provide the final diagnosis. Conclusions Cases of heterotopic pancreas with symptoms should be surgically managed and the results of resection is satisfactory. Long term follow up needs to be conducted for those asymptomatic cases.
