Objective:To establish a sensitive and fast HPLC method for determining cucurbitacin IIa in rat plasma and investigate the pharmacokinetic characteristics after intravenous administration. Methods: The analysis was performed on a Dikma Diamonsil C18 column (250 mm × 4. 6 mm, 5 μm) at 35℃ with the mobile phase of acetonitrile-water (45∶ 55, v/v) and the UV detection at 212 nm. The plasma samples were collected after intravenous administration at different time points and measured by HPLC. The pharmaco-kinetic parameters were calculated by DAS 2. 0 software. Results: The calibration plot of cucurbitacin IIa was linear over the range 0. 146-14. 060 μg · ml-1 . The range of method recovery was 99. 02%-104. 22% and that of extraction recovery was 84. 74%-86. 80%. The intra-and inter-day precision were both less than 5%. The stability met the requirements. The main pharmacokinetic pa-rameters were showed as follows:t1/2β(h):0.732 ±0.151, 0.681 ±0.055,0.667 ±0.064;Vd (L·kg-1):0.147 ±0.089, 0.131 ±0.095,0.153±0.047; Cl(L·h-1·kg-1): 0.287±0.031,0.304±0.063,0.318±0.029andAUC0→∞(mg·h·L-1):3. 646 ± 1. 124,4. 916 ± 1. 227,9. 385 ± 1. 419. Conclusion: The validated method is successfully applied in basic pharmacokinetic study in rat plasma after intravenous administration. The plasma concentration-time curves at three dosages are all fitted three-compart-ment model. Within the examined dose range, the pharmacokinetics of cucurbitacin IIa in rat is based on linear pharmacokinetics.

Objective To investigate the characteristics, degradation and biocompatibility of a woven chitosan fiber (CF). Methods Scanning electron microscopic observation, deteronination of density, degree of swelling and tensile strength were performed to characterize the CF. In vitro degradation was observed by immersing CF in buffered aqueous solution of pH 6 containing lysozyme at 37 ℃, while in vivo degradation was studied after intramuscular implantation of CF in rats. Furthermore, rat mesenchymal stem cells were directly cultured on CF to estimate its toxicity. Results The diameter of CF was about 2000?63?m, with a sleek surface. The density and rate of swelling were 1.093?0.27g/cm3 and 2.30?0.21, respectively. The tensile strength was 80.18?1.56MPa. Specific viscosity, measured after the addition of lysozyme to the solution, decreased dramatically within 2 hours compared with that of the control mixture without enzyme. There was obvious inflammatory reaction after intramuscular implantation in the first week after surgery, bat inflammation subsided gradually. Biodegradation was not obvious within 4 weeks, but majority of it was absorbed in twelve weeks. Cells grew well on it with normal morphology, and no inhibition of cell proliferation could be observed. Conclusion Woven CF has a good tensile strength and fairly good biodegradation and biocompatibility, and it can be used in bone and cartilage tissue engineering.

OBJECTIVE:To establish the quality standard of Yanyan granules of concentrated sugar-free type. METHODS:Scrophularia ningpoensis,Menispermum dauricum,Radix et Rhizoma Glycyrrhizae in the granules were identified by TLC,and the content of linarin in Yanyan granules were determined by HPLC.RESULTS:The TLC spots were clear with high resolution. The linear range of linarin was 4.64~69.6 ?g(r=0.999 9) and the average recovery rate was 98.65%(RSD= 0.79%,n=6). CONCLUSION:The established standard is suitable for the quality control of the Yanyan granules of concentrated sugar-free type.

Objective To assess the value of preoperative MRI in predicting the incidence of cement leakage into adjacent discs during percutaneous vertebral augmentation (PVA) for osteoporotic vertebral compression fracture (OVCF).Methods Clinical and radiological characteristics of 127 patients who were treated with PVA for OVCF were analyzed retrospec tively. The following clinical data of these patients were analyzed, including gender, age, location of treated vertebral body and surgical approach. The image features of endplate injury, fracture line extended to the endplate, adjacent intervertebral dick injury and intravertebral cleft were evaluated on the preoperative MRI.The incidence of cement leakage into the adjacent disc were compared for the above factors with statistical methods. Results Totally 127 patients were enrolled in our study, including 179 treated vertebral bodies,358 endplates and 341 adjacent intervertebral discs. The incidence of intradiscal cement leakage was 57.73% (56/97) in endplate injury sign,60.98% (25/41) in fracture line extended to endplate sign,35.91%(51/142) in adjacent discs injury sign and 55.56%(20/36) inintravertebral cleft sign. The differences were statistically significant on preoperative MRI in patients with the above signs compared to those who had not (P<0.05).The incidence of intradiscal cement leakage in percutaneous kyphoplasty (PKP) and percutaneous vertebroplasty (PVP) were 26.67%(16/60) and 42.86%(51/119),respectively,which was significant different (P=0.035).For bone cement volume ≤ 5 ml vertebral bodies, the incidence of intradiscal cement leakage was 31.19 % (34/109),lower than bone cement volume > 5 ml vertebral bodies (47.14% [33/70],P=0.031).There was no statistically significant association between intradiscal cement leakage and age, gender and location of treated vertebral body (P>0.05).Conclusion Preoperative MRI can predict the leakage of bone cement into adjacent discs effectively during PVA.The PKP and the lower volume of bone cement injection can reduce the risk of intradiscal cement leakage.

Objective To investigate the relationship between phosphatidylinositol 3-kinase catalytic subunit α(PIK3CA) expression and the incidence and different pathological grade of gastric cancer, and the mechanism thereof. Meth-ods The expressions of PIK3CA, serine/threonine protein kinase (pAkt) and cell proliferation associated nuclear antigen (ki-67) in gastric carcinoma and adjacent tissues and normal gastric mucosa were detected by immunohistochemical method. The relationship between expressions of PIK3CA, pAkt and ki-67 and tumorigenesis was analyzed. The expressions of PIK3CA, pAkt and ki-67 in different pathological conditions of gastric tissues were analyzed. The relationship between tu-mor pathologic classification and differentiation were analyzed too. Results There were significantly higher expressions of PIK3CA, pAkt and ki-67 in gastric cancer (P<0.05), which were the highest in the poorly differentiated gastric adenocarci-noma (P<0.05). There were a positive correlation between expressions of PIK3CA, pAkt and ki-67 and different pathologi-cal levels of gastric carcinoma and adjacent tissues and normal gastric tissues (P<0.05). Conclusion PIK3CA may be the initiating factor of PI3K/Akt signaling pathway, which induced phosphorylation of Akt and activation PI3K/Akt signaling pathway, promoting the proliferation, invasion and metastasis of gastric adenocarcinoma.

The potential embryotoxicity, developmental toxicity and teratogenic effects of (3 or 8)-(l-methoxyethyl)-8 (or3)-hydroxyethyl-deuteroporphyrin (PsD-044) were investigated in Sprague-Dawley rats with the conventional teratological method in vivo. According to the recommending clinical dosage, PsD-044 was administered intravenously at 20, 10 and 5mg/kg, as compared to the negative control with saline and the positive control with sodium pentachlorophenolate, respectively on the 10th day of the gestation. Eighty-one pregnant rats and 803 fetuses were examined. The results suggest that the maternal toxicity, embryotoxicity and teratogenic effects of PsD-044 were not found, however, the malformation induced by known teratogen was as high as 14.1%.
