1. Correlation between sleep duration and ischemic stroke: A large community study

Objective To investigate the relationship between sleep duration (SD) collected by polysomnography and ischemic stroke in community populations. Methods We recruited 2062 participants, including 724 males and 1338 females. The mean age of the all participants was 61.44 years. In this study, the subjects were divided into three groups according to SD: short SD (<6 h), long SD (>8 h), and normal SD (6-8 h). Logistic regression was used to analyze the independent association between the three different sleep durations and the occurrence of ischemic stroke. Results ① The three groups differed significantly in gender, age, BMI, prevalence of diabetes and serum triglyceride levels (P<0.05). ② The incidence of ischemic stroke was significantly higher in short SD group than in long SD group and normal SD group (the incidence rates were 5.9%, 2.7% and 1.6%, respectively; P=0.044, P<0.001). ③ Univariate logistic regression analysis suggested that short SD was associated with the occurrence of ischemic stroke (OR=3.845, 95% CI: 1.789-8.266, P=0.001). After adjusting potential factors such as gender, age, marital status, alcohol consumption, hypertension, diabetes and triglycerides, multivariate logistic regression analysis showed that short SD was still an independent risk factor for ischemic stroke (OR=2.629, 95% CI: 1.033-6.693, P=0.043). Conclusion The incidence of ischemic stroke differs in people with different SD. Among them, the incidence of ischemic stroke is higher in people with short SD.
2.Expression of vascular endothelial growth factor and its receptor in the epididymal sperm of adolescent male rats

BACKGROUND: Vascular endothelial growth factor (VEGF) and its receptor fms-like tyrosine kinase-1 (Flt-1) have a very important position in the field of male reproduction. However, it is still unclear about their expression meaning and regulatory mechanism in the reproductive system. OBJECTIVE: To study the expression and location of VEGF and Flt-1 in the epididymal sperm of adolescent male rats. METHODS: The expression of VEGF and Flt-1 was detected in 10 adolescent SD rats. The concentration of the sperm was (30-40)×109/L. Immunofluorescent staining was used for VEGF and Flt-1 expression and location in the sperm. RESULTS AND CONCLUSION: Immunofluorescent staining showed that VEGF and Flt-1 were both localized in the acrosome of sperm head, as well as in the neck, middle and principal segment of sperm tail. Specific expression patterns of VEGF and Flt-1 in the sperm of rats display that they may participate in spermiotelcosis, relevant to movement, capacitation and acrosome reaction of the sperm.
3. Echocardiographic diagnosis of bicuspid aortic valve combined with infective endocarditis

Objective: To explore the value of transthoracic echocardiography (TTE) and three-dimensional transesophageal echocardiography (3D-TEE) in diagnosis of bicuspid aortic valve (BAV) combined with infective endocarditis (IE). Methods: Ultrasonic data of 53 BAV combined with IE patients were retrospective reviewed. The ultrasonic findings of BAV combined with IE included vegetation, leaflet perforation, abscess and fistula. The ultrasonic features and diagnostic value of TTE and 3D-TEE were evaluated taken surgical findings as the gold standards. Results: The sensitivity and positive predictive value of 3D-TEE were both higher than those of TTE in detecting vegetation, leaflet perforation, abscess and fistula (100% vs.90.20%, 96.87% vs.90.62%, 92.86% vs.67.86%, 94.12% vs.70.59% and 100% vs.95.83%, 100% vs.93.55%, 96.30% vs.86.36%, 100% vs.85.71%, all P<0.05). Conclusion: The echocardiographic detection rate of BAV combined with IE is very high. 3D-TEE has better visibility and higher spatial resolution than TTE, therefore having higher sensitivity and positive predictive value in diagnosis of BAV combined with IE.
4.The predictive value of metabolic syndrome in patients with acute coronary syndrome after percutaneous coronary intervention

Objective To investigate the predictive value of metabolic syndrome in patients with acute coronary syndrome (ACS) after percutaneous coronary intervention (PCI).Methods A total of 660 patients with ACS admited to cardiovascular department,first affiliated hospital of zhengzhou university were enrolled in this study from January 2009 to June 2010.The enrollment criteria were:the stenosis degree were above 75% in at least one coronary artery by coronary angiography and successful PCI procedure.Exculsion criteria were:liver and renal insufficiency,malignancies and valvular heart diseases.The relevant clinical data and labtory examination were recorded after admission. The patients were followed up by outpatients interview or telephone from March to June 2011 and adverse cardiovascular events were recorded.The patients were divided into MS and non-MS groups,and basic clinical data were compared between two groups.The proportion difference between two groups were tested by chi square. Multivariate logistic regression was established to analyze the factors related to progonosis.The survival ratio was estimated using the Kaplan-Meier method.Statistical significance was established at a P value of less than 0.05.Results ①A total of 606 (91.7%) patients successfully accepted follow-up.Mean follow-up time were ( 14.3 ±1.7 ) months.95 patients experienced adverse cardiovascular events ( 15.7% ).②There were 393 patients (64.96% ) satisfied the definition of metabolic syndrome.The patients in MS group were with higher BMI,SBP,DBP,blood glucose and disordered lipid (all P < 0.05 ),with less fale patients (P =0.016),less current somking (P =0.008 ) and with higher platelet (P =0.037 ). The incidence of adverse cardiovascular events in two groups were 17.81% and 11.79% ( P > 0.05 ). ③ Multivarite logistic regression revealed that the predictors of adverse cardiovascular events were age [ OR =2.628,95% confidence interval (CI) 1.395 ~ 4.954,P =0.003 ],New York Heart Association (NYHA) ≥ 3 grade ( OR =2.310,95% CI 1.095 ~4.870,P =0.028) and left ventricular ejection fraction (LVEF) ( OR =4.328,95% CI 1.955 ~9.580,P < 0.001 ).However,MS was not related with prognosis ( OR =1.170,95% CI 0.583 ~ 2.345,P =0.659 ).④The cumulative survival rates of no adverse cardiovascular events in the two groups were no significant difference ( P > 0.05 ).Conclusions MS is a risk factor with coronary heart disease.Howerer,it has no relationship with adverse cardiovascular events in patients with ACS after PCI.
5.Analysis on risk factors for dyed cornea of orthokeratology and interventions

Objective To investigate the related risk factors for dyed cornea of orthokeratology, and to analyze the related risk factors, and to determine the independent risk factors, Provide evidence for intervention measures. Methods The clinical date were investigated for 990 patients with fitting orthokeratology lens between may.2014 and may.2016 in our hospital,through access to medical records, follow-up visit,questionnaire investigation to find out the cause of orthokeratology adverse reactions, the related risk factors were analyzed by univariate and multivariate Logistic regression analysis, using SPSS17.0 statistical processing. Results The incidence rate of dyed cornea of orthokeratology lens fitting in was 14.55%(144 in 990 patients); the single factor analysis found gender, age, region, refraction, lens position, family members, no significant difference . Eye disease combined (χ2= 28.73, P<0.01), Schirmer I test (χ2=17.68, P<0.01), lens activity (χ2=67.1, P<0.01), Lens deposit(χ2=64.29, P<0.01), lens wearing time (χ2=43.25, P<0.01), health habits (χ2=38.01, P<0.01) and water resources (χ2=3.81, P<0.05), the difference was statistically significant; Logistic Logistic regression analysis found that Schirmer I test ( OR=4.126, P=0.003), lens activity ( OR=1.733, P=0.104), Lens deposit( OR=3.723, P=0.038), lens wearing time ( OR=5.034, P=0.002), health habits ( OR=6.544, P=0.002) and water resources ( OR=7.501, P=0.002) were independent risk factors for adverse reactions of orthokeratology. Conclusions Intervention measures that improve the fitting technology, complete removal of lens depositthe, Control wearing time, improve the health behaviors of the patients with the habit, use saline and professional cleaning are of great significance to reduce the incidence of adverse reactions of orthokeratology.
6.Protein tyrosine phosphatase non-receptor type 12 negatively regulates cardiac HERG channel currents.

OBJECTIVETo study the effect of protein tyrosine phosphatase non-receptor type 12 (PTPN12) in regulating cardiac HERG channel currents.
METHODSThe plasmids pcDNA3.1-PTPN12-RFP and herg mutant constructed by PCR technique were transfected into HEK293 cells via Lipofectamine 2000, and the cells stably expressing PTPN12 selected with G418 were identified by Western blotting with anti-PTPN12 antibody. HERG channel current in cells expressing HERG alone (HEK293/HERG cells), cells overexpressing PTPN12 (HEK293/HERG cells transfected with pCDNA3.1-PTPN12-RFP), PAO-treated cells (PTPN12/HERG cells treated with PAO), and herg mutant cells (HEK293/HERGY327A-Y700A-Y845A cells transfected with pcDNA3.1-PTPN12-RFP) were recorded by patch-clamp technique.
RESULTSThe plasmids pcDNA3.1-PTPN12-RFP and herg mutant were successfully constructed, and the stable expressing cell lines were established. Red fluorescence was obversed in HEK293/HERG cells transfected with pcDNA3.1-PTPN12-RFP, and the protein expression of PTPN12 was detected. Overexpression of PTPN12 significantly decreased HERG current density in HEK293/HERG cells, and this change was significantly weakened in the inhibitor group and herg mutant group.
CONCLUSIONPTPN12 negatively regulates cardiac HERG channel cerrent possibly by decreasing the phosphorylation level of HERG tyrosine residues. This finding provides further insight into the regulatory mechanism of HERG channel and the pathogenesis of long QT syndrome.

7.Joint effects of parent-child relationship and environmental sensitivity on adolescent mental health

【Objective】 To examine the joint effects of parent-child relationship and environmental sensitivity (ES) on adolescent mental health from the perspective of sensitivity X family conflict interaction. 【Methods】 In 2023, we investigated 7,010 adolescents from six junior high schools in Shaanxi Province, China. Self-reported questionnaires were used to assess parent-child relationship, ES, and adolescent mental health. ES was assessed by Highly Sensitive Child Scale (HSC); Patient Health Questionnaire-9 (PHQ-9), Strengths and Difficulties Questionnaire (SDQ), and Happiness index were used to measure depression, externalizing problems, and subjective well-being. 【Results】 ① Girls’ ES scores were significantly higher than boys’ (t=-7.504, P<0.01). ② When predicting depression and externalizing problems, the main effects of parent-child relationship and ES in adolescents were significant (b=-0.263, 0.237, 0.332, P<0.001; b=0.201, 0.220, P<0.001), and the interaction between family conflicts and ES was significant (b=-0.072, P<0.05; b=0.092, P<0.05). (3) Widaman’s confirmatory and competitive approach results showed the significant interaction of sensitivity X family conflict with depression and externalizing problems, which were consistent with the differential susceptibility and diathesis-stress framework, respectively (BIC=2 170.98, 2 162.6). 【Conclusion】 Adolescents with high environmental sensitivity suffer more from high family conflict and benefit more from low family conflicts. These sensitive adolescents can be described as having developmentally susceptibility rather than vulnerability.
8.Childhood maltreatment on psychiatric symptoms and drug efficacy in patients with schizophrenia

【Objective】 To explore the effect of childhood maltreatment on clinical symptoms and early efficacy of antipsychotics in patients with schizophrenia. 【Methods】 Totally 73 schizophrenic patients were divided into mild maltreatment group(n=42) and severe maltreatment group(n=31) according to the Child Trauma Questionnaire(CTQ). The Positive and Negative Symptom Scale(PANSS) and Clinical Global Impression(CGI) were measured and compared between the two groups at baseline and 3 weeks after antipsychotic treatment to analyze the correlation between child maltreatment experience and mental symptoms and the response to early treatment of antipsychotics. 【Results】 PANSS positive factor score(P=0.026) and cognitive deficit factor score (P=0.042) were significantly higher in severe abuse group than in mild abuse group. The positive factor was significantly positively correlated with emotional abuse factor in CTQ score(r=0.257, P=0.028), and the cognitive deficit factor was significantly positively correlated with emotional neglect factor(r=0.283, P=0.015). After antipsychotic treatment, the reduction rate of PANSS negative factor in severe abuse group was significantly lower than that in mild abuse group(P=0.035), and had the highest correlation with CTQ physical abuse factor(r=-0.302, P=0.011). 【Conclusion】 The severity of childhood maltreatment experienced by schizophrenic patients is more related to positive symptoms and cognitive deficits, and more childhood maltreatment experience will affect the improvement of negative symptoms by antipsychotics, suggesting a poor prognosis.
9.Correlation of serum inflammatory factors with clinical symptoms and cognitive functions in schizophrenia

【Objective】 To study the correlation of clinical symptoms and cognitive functions with serum inflammatory factors in schizophrenia. 【Methods】 A total of 42 SCz patients (case group) and 47 healthy controls (control group) were enrolled in this study. We used enzyme-linked immunosorbent assay (ELSA) to determine six inflammatory factors in serum. PANSS was used to assess clinical symptoms and MCCB was used to assess the patients’ cognitive functions. 【Results】 ① Inflammatory factors: The serum levels of IL-1β, IL-4, IL-6 and IL-8 were significantly higher in case group than in control group (P<0.01). ② Cognitive functions: The scores of Trail Making Test, Hopkins Verbal Learning Test, Symbol Coding, Spatial Span, Brief Visuospatial Memory Test, Assessment Battery-Mazes, Category Fluency and Test-Managing Emotions of case group were significantly lower than those of control group (P<0.05). ③ Correlation between serum inflammatory factors and clinical symptoms: There was no correlation between serum inflammatory factors and psychiatric symptoms in schizophrenia. ④ Correlation between serum inflammatory factors and cognitive functions: The levels of IL-6 (r

【Objective】 To elucidate the possible role of arginine and histidine in the pathogenesis of schizophrenia by exploring the serum levels of semi-essential amino acids (arginine and histidine) in patients with schizophrenia and their correlation with psychiatric symptoms. 【Methods】 We selected 72 inpatients with schizophrenia admitted to The First Affiliated Hospital of Xi’an Jiaotong University from March 2021 to October 2022 and 72 healthy volunteers enrolled in Yanta Community during the same period as the research subjects. Serum arginine and histidine levels were measured in patients with schizophrenia and healthy controls using serum liquid chromatography-mass spectrometry (LC-MS). We used the Positive and Negative Symptom Scale (PANSS) to evaluate the mental symptoms of patients with schizophrenia and analyzed the correlation of serum arginine and histidine levels with disease course, frequency of onset, and PANSS score. 【Results】 The levels of serum arginine (P<0.001) and histidine (P=0.011) in the schizophrenia group were significantly lower than those in the control group.
