1.Effects of subcutaneous transfection of human beta-nerve growth factor gene on expression of substance P in dorsal root ganglion in a rat model of diabetic neuropathic pain

Objective To investigate the effects of subcutaneous transfection of human beta-nerve growth factor (Ad-hNGFβ) gene on the expression of substance P (SP) in the dorsal root ganglion in a rat model of diabetic neuropathic pain (DNP).Methods Male SD rats weighing 180-220 g were used in this study.Ten rats were randomly collected as normal control without DNP (group C).DNP model was made by intraperitoneal injection of streptozocin (STZ) 75 mg/kg.Seventy-five rats with DNP were randomly divided into 3 groups ( n =25 each):DNP group; Ad-hNGF group and Ad-LacZ group.Groups Ad-NGF and Ad-LacZ received subcutaneous transfection of 1.12 × 1010 PFU Ad-hNGFβ 10 μl and 1.12 × 1010 PFU Ad-LacZ 10 μl respectively after pain threshold was measured on 21d after STZ injection.The mechanical and thermal pain threshold was measured before STZ injection (baseline) and at 21,35 and 49 d after STZ injection.The expression of SP in the dorsal root ganglion was determined after the measurement of pain threshold on 49 d after STZ injection.Results Compared with group C,the mechanical and thermal pain threshold was significantly decreased at each time point after STZ injection in groups DNP,Ad-NGF and Ad-LacZ,and the expression of SP in the dorsal root ganglion was signilicantly downregulated in groups DNP and Ad-LacZ (P < 0.05).Compared with group DNP,the thermal pain threshold was significantly increased on 49 d afar STZ injection,and the expression of SP in the dorsal root ganglion was significantly up-regulated in group Ad-NGF ( P < 0.01 ),and no significant change was found in the mechanical and thermal pain threshold and the expression of SP in the dorsal root ganglion at each time point in group Ad-LacZ ( P > 0.05).Conclusion Subcutaneous transfection of Ad-hNGFβ can attenuate DNP to some extent through upregulation of the expression of SP in rat dorsal root ganglion.
2.Chemotherapy after chidamide pretreatment in high-risk and refractory lymphoid malignancy: report of 16 cases

Objective To analyze the therapeutic effects of chemotherapy after chidamide pretreatment in 16 cases of high-risk and refractory lymphoid malignancy. Methods The efficacy and adverse reactions of 16 patients with high-risk and refractory lymphoid malignancy who received chidamide combined with chemotherapy after 3 days pretreatment of chidamide were analyzed. Results Sixteen patients included 6 males and 10 females, and the median age was 49.5 years old (23-88 years old). The median course of previous systemic chemotherapy was 4 (range 0-22). Among 14 patients who received induction chemotherapy, 7 patients achieved complete remission (CR), 7 patients achieved partial remission (PR). Fourteen patients had achieved clinical efficacy, and the overall response rate (ORR) was 100 %. After 2 cases had remission , the patients who entered this regimen for consolidation chemotherapy also had durable CR. The median follow-up time was 13 months (range 2-24 months) until December 2017. Nine cases had overall survival (OS), 7 cases died and 9 cases had progression-free survival. Common adverse effects of the chemotherapy included mild and controllable gastrointestinal reactions after chidamide. Conclusion Chemotherapy after chidamide pretreatment may improve the effect and prognosis of high-risk or refractory lymphoid malignancy.
3.Fingerprint Establishment ,Content Determination and Chemical Pattern Recognition Analysis of Tibetan Medicine Ershiwuwei Luxue Pills

OBJECTIVE:To establish fingerprint of Tibetan medicine Ershi wuwei luxue pills ,and determine the contents of 5 components,and to conduct chemical pattern recognition. METHODS :HPLC method was adopted. Using aesculetin as reference , HPLC fingerprint of 10 batches of Tibetan medicine Ershi wuwei luxue pills were drawn. The similarity evaluation was conducted by using Evaluation System of TCM Chromatogram Fingerprint Similarity (2012 edition),and the common peaks were determined. Same HPLC method was adopted to determine the contents of 5 components in Tibetan medicine Ershi wuwei luxue pills. The cluster analysis and principle component analysis were performed by using SPSS 19.0 software. RESULTS :Totally 11 common peaks were calibrated ,and the similarity was higher than 0.98. Five common peaks were identified ,as aesculetin ,orientin, isovitexin,isoscoparin and ellagic acid. The linear range of aesculetin ,orientin,isovitexin,isoscoparin and ellagic acid were 1.232-11.092 μg/mL(r=0.999 6),2.766-24.893 μg/mL(r=0.999 5),1.400-12.600 μg/mL(r=0.999 8),0.600-5.400 μg/mL(r= 0.999 5),49.447-445.025 μg/mL(r=0.999 4),respectively. RSDs of precision ,stability(24 h)and reproducibility tests were all lower than 2%. The average recoveries were 101.29%(RSD=2.33%,n=3),91.39%(RSD=1.22%,n=3),90.28%(RSD= 1.88%,n=3),98.76%(RSD=2.53%,n=3),101.45%(RSD=2.84%,n=3),100.44%(RSD=1.38%,n=3),100.91% (RSD=1.73%,n=3),97.78%(RSD=2.07%,n=3),99.15%(RSD=1.28%,n=3),100.27%(RSD=1.81%,n=3),98.38% (RSD=1.89% ,n=3),101.92%(RSD=1.17% ,n=3),95.50%(RSD=0.67% ,n=3),99.89%(RSD=0.38% ,n=3), 100.10%(RSD=0.65%,n=3),respectively. Their contents were 0.175-0.310, 0.351-0.632, 0.274-0.395, 0.186-0.278, 61932600 6.956-8.636 mg/g,respectively. Cluster analysis showed that 10 batches of Tibetan medicine Ershi wuwei luxue pills were clustered into two category ,with S1-S4 as one category and S5-S10 as one category. Principal component analysis showed that accumulative contribution rate of two principle components was 89.178%. CONCLUSIONS :Established fingerprint is stable and feasible ,and the method of content determination is simple , accurate and reproducible. They combined with chemical pattern recognition can be used for the quality control of the Tibetan medicine Ershiwuwei luxue pills.
