1.Influence of insufficient blood specimens volume on the detection results of coagulation tests in SD rats

Objective To explore the best anticoagulant ratio in SD rats .To analyse the influence of insufficient blood specimens volume on coagulation tests .Methods 60 rats were divided into 2 groups.According to the method of vacuum blood, collect abdominal aortic blood after fasting 12 hours.The first group 20 rats were used only for routine blood test.Fully automatic hematology analyzer detected hematocrit and platelet .The second group 40 rats were used for coagulation test .Every rat was collected 2 blood specimens with different anticoagulant ratio [ the proportion of sodium citrate anticoagulation and the whole blood (vlume∶volume)]1∶9(the control group)and 1∶5( the experimental group),1∶8( the experimental group) and 1∶7( the experimental group).Get plasma without platelet through centrifugation.Fully automatic blood coagulation analyzer detected prothrombin time , activated partial thromboplatin time , thrombin time and Fibrinogen.Results HCT (%) and PLT (x 109/L) in SD rats were respectively 41.7 ±2.9 and 1114 ±173.As anticoagulant ratio was increased ,PT, APTT and TT were extended and FIB was decreased .Compared with the control group ,these PT,APTT,TT,FIB four results of 1∶8 group were not statistically different , of 1∶5 group were statistically different, these PT,APTT,FIB three results of 1∶7 group were not statistically different , TT of 1∶7 group was statistically different.Conclusions The detection of coagulation project coagulation test results were affected by the proportion of anticoagulant and blood .1∶9 was the best anticoagulant ratio in SD rats , 1∶8 can also.Rat had its unique physiological characteristics .The results could provide reference for the evaluation of rats .

Objective To explore effect of Xylazine hydrochloride on Bama minipigs under general anesthesia. To emphasize safety consciousness of general anesthesia. To research cardiac main function and structure of normal Bama minipigs in preparation for the subsequent comparative medicine research. Methods 43 Bama minipigs, inject in post aurem muscles of neck with 5 mL of mixed drug conclude Xylazine hydrochloride (2 mL), Atropine Sulfate(1 mL) and Droperidol(2 mL) on each one. Echocardiography after general anesthesia. Observe induction and recovery time of anesthesia, anesthesia maintaining time, total check time and the others. Introduce the method of simple endotracheal intubation. Results Anesthesia, induction period (18 ±3)min, maintaining period (40 ±5)min, recovery period (60 ± 10)min. Echocardiography, LAD (2?54 ± 0?20) cm, LVDd (3?41 ± 0?25) cm, LVDs (2?28 ± 0?23) cm, IVSTd (0?60 ± 0?07) cm, LVPWTd (0?59 ± 0?07) cm, AoD (1?77 ± 0?18) cm, EDV (48?59 ± 8?31) cm, ESV (18?28 ± 4?46) mL, SV ( 39?30 ± 5?16 ) mL, LVEF ( 62?76 ± 5?01 )%. Conclusions Intramuscular injection of xylazine hydrochloride with droperidol and atropine sulfate on bama minipigs for general anesthesia is a highly conserved specie in cardiovascular system and safe. We obtained some information of cardiac main function and structure of normal Bama minipigs which could provide reference for scientific research and veterinarian clinic.
4.The comparative effect of orally or intranasally administered borneol on ketamine-midazolam anesthetized rats′regained consciousness and cognitive functions

Objective To investigate and compare the effect of orally or intranasally administered borneol on ketamine‐mid‐azolam anesthesia anesthetized rats′regained consciousness and cognitive functions .Methods The rats were trained to escape laten‐cy and divided into 6 groups:control group ,model group ,low dose oral borneol group ,high dose group oral borneol group ,low dose intranasal borneol group and high dose intranasal borneol group ,16 rats in each group .All groups were given general anesthesia ,be‐side the control group .The reflex recovery time ,blood gas analysis ,escape latency and space exploration capability were recorded and compared within groups .Results Compared with the control group ,PaCO2 ,P(A‐a)DO2 and latent escape time increased (P<0 .05) ,pH ,PaO2 and times of platform across decreased in the model group (P<0 .05) .Compared with the model group ,the righ‐ting reflex test time ,times of platform across and latent escape time decreased (P<0 .05) ,blood gas analysis improved (P<0 .05) in intranasal borneol groups and oral borneol groups .The righting reflex test time ,blood gas analysis and times of platform across had no significantly differences within borneol oral groups(P> 0 .05) .Compared with low dose oral borneol group ,latent escape time decreased in high dose oral borneol group (P<0 .05) .Compared with borneol oral groups ,the righting reflex test time ,times of platform across and latent escape time decreased (P< 0 .05) ,blood gas analysis improved (P< 0 .05) in intranasal borneol groups .Compared with low dose intranasal borneol group ,the righting reflex test time and latent escape time decreased (P<0 .05) , blood gas analysis improved (P<0 .05) in high dose intranasal borneol group .Conclusion Intranasal borneol could reduce righting reflex test time and improve cognitive function by improving the blood gas analysis .
5.Effect of ?-3 polyunsaturated fatty acids on the serum lipid of ovariectomized rats

Objective: To study the effect of ?-3 polyunsaturated fatty(?-3 PUFA) on the serum lipid of ovariectomized rats.Methods: 60 virgin female Sprague-Dawley rats,3 months of age,were randomized by the stratified weight method into 6 groups: Sham group:deionized water 2 mL/d,OVX group:eionized water 2 mL/d,E2 group: OVX+17?-ethinylestradiol 0.1 mg/(kg?d),A group: OVX+?-3 PUFA 75 mg/(kg?d),B group: OVX+?-3 PUFA 150 mg/(kg?d) and C group: OVX+?-3 PUFA 300 mg/(kg?d).Each group had 10 rats.Experiment period was 90 days.Body weight were measured every week.Serum were collected to measure Cholesterol(TC),Triglyceride(TG),High-density lipoprotein-cholesterol(HDL-C),and Low-density lipoprotein-cholesterol(LDL-C).Results: There were significant difference between groups in the serum lipid.In the OVX group,TC and LDL-C increased.In the E2 group,TC increased compared with that in the Sham group,and HDL-C,LDL-C and HDL-C/TC decreased significantly compared with that in the OVX group.In the A,B and C groups HDL-C/TC increased significantly compared with those in the E2 group.Conclusion: The ovariectomized rats have lipid metabolism disorderd.The ?-3 PUFA and estrogen replacement therapy plays a role in the lipid metabolism.The ?-3 PUFA has the best efficiency in reducing the level of serum lipid.

Transgene silencing is one of two major obstacles in both basic biomedical research for transgene and clinical practice of gene therapy. Based on the model of HT1080 cell clones, which transduced single copy of retroviral vector MGPN2, the mechanism of transgene silencing was explored in this investigation by a serial molecular techniques. In the HT1080 cell clone that absence of GFP protein synthesized, no significant aberration of epigenetic modification was detected, but the transcript size and the sequence changed that resulted in the reading frame shift. In addition to chromosomal position effect leading to transgene silencing, the transcript reading frame shift associated with retroviral vector rearrangements could induce complete silencing of transgene.
