As an important mediator of allergic inflammation, mast cell tryptase is involved in the induction of hypersensitivity, infiltration of inflammatory cells and tissue remodeling in respiratory tract. The effects of tryptase inhibitors on the actions of tryptase show further the potential of tryptase in the pathogenesis of asthma and its inhibitors in the treatment of asthma.
2.Studies on the Protective Effect of the Mutant of Sj23 DNA Vaccine against Schistosomiasis

Objective To investigate the protective immunity of the vaccine against schistosomiasis,a mutant of Mr 23 000 membrane protein DNA(Sj23DNA) without the homologous sequence of ME491.Methods The mutant of Sj23 DNA with no homologous sequence of ME491 on the cell membrane of human melanoma was obtained by overlap PCR.The mutant was transfected into human embryonic kidney cells of the line HEK293.Indirect fluorescent antibody test(IFAT) was used to detect the expressed protein.Expression of the mutant of Sj23DNA in muscular cells of mice was conducted through vaccinating the mouse with 100 ?g purified plasmids by injecting them into the quadriceps muscle of thigh.Four weeks after the immunization,the quadriceps muscles were taken and cryostat sections were prepared for detecting the expression by IFAT.Forty BALB/c mice were randomly divided into four groups and injected with the mutant of pcDNA3-Sj23 plasmid DNA,pcDNA3-Sj23 plasmid DNA,pcDNA3 blank plasmid(100 ?g per mouse) and sterile saline(30 ?l per mouse) respectively.Four weeks after the immunization,mice were challenged with cercariae(40?2 cercariae per mouse) by abdominal skin penetration.Mice were then killed 6 weeks later,perfusion and squash methods were carried out to collect the adult worms and the number of eggs per gram of liver tissue was calculated.Worm and egg reduction rates were used to evaluate the protective immunity.Results Specific fluorescence was demonstrated in muscular cells of mice vaccinated with the mutant of pcDNA3-Sj23.The worm reduction rate and egg reduction rate were 40.3% and 42.8% respectively in the mutant of pcDNA3-Sj23 group,which were higher than those in the pcDNA3-Sj23 plasmid group(33.1% and 28.9% respectively).The difference between these two groups was significant(P
3.Efficacy of S-1 combined with cisplatin as first-line chemotherapy for advanced AFP positive gastric cancer

Objective:To comparatively analyze the efficacy of S-1 plus cisplatin as first-line chemotherapy between advanced AFP posi-tive and AFP negative gastric cancer. Methods:A total of 89 eligible patients with advanced gastric cancer from January 2010 to June 2013 were enrolled in this retrospective study. The cases were divided into AFP positive group (n=18) and AFP negative group (n=71) based on serum AFP level before treatment. Both groups received S-1 plus cisplatin as first-line treatment. Results:Significant differ-ences were observed in remission rate (66.7% versus 32.4%, P=0.028) and radical surgical resection rate (44.5% versus 18.3%, P=0.029) after chemotherapy between AFP positive group and AFP negative group. In the two groups, neither progression-free survival nor overall survival (OS) showed any significance (P>0.05) in patients without surgery after chemotherapy. However, the disease-free survival of the two groups with radical surgical resection after chemotherapy was significantly different (median:27.0 months versus 15.6 months, P=0.034). The OS of the AFP positive group was evidently longer than that of the AFP negative group (median:16.0 months versus 11.0 months, P=0.005). Conclusion:As first-line chemotherapy, S-1 plus cisplatin was more effective for the advanced AFP positive gastric cancer and prolonged overall survival.
4.Study on the best concentration of miRNA-22 agomir in the treatment of temporal lobe epilepsy by single injection of lateral ventricle

Objective To investigate the optimal initial concentration of microRNA22 agomir in epilepsy model induced by lithium chloride-pilocarpine after single injection of lateral ventricle.Methods 36 rats with acute temporal lobe epilepsy were randomly divided into 6 groups:the control group and the other five groups were the experimental group.All epilepsy rats were selected for right lateral ventricle injection.The control group was given negative control reagent,while the experimental group were given 0.1 mmol/L,2.5 mmol/L,5 mmol/L,10 mmol/L,20 mmol/L different concentrations of miRNA22agomir reagent.6 rats in each group were randomly selected for acute phase experiment after 3 days of administration.The expression of P2X7 in hippocampus of epilepsy rats was determined by Western blot and quantitative real-time polymerase chain reaction (qRT-PCR).Results Compared with control group,the protein and mRNA expression of P2X7 reduced in all of the model group.The protein and mRNA expression level of P2X7 protein in hippocampus of rats injected with 2.5 mmol/L,5 mmol/L and 10 mmol/L in each experimental group were significantly lower than that in the other two groups (P < 0.05).Moreover,the protein and mRNA expression level of P2X7 were the lowest at 2.5 mmol/L injection and 10 mmol/L,and there was no significant difference between the two groups (P > 0.05).Conclusions The optimal onset concentration for unilateral lateral ventricle injection miRNA22 agomir treatment of temporal lobe epilepsy is 2.5 mmol/L.
