China Pharmacy 2016;27(10):1374-1377
OBJECTIVE:To study the effects of mifepristone on the expression of inflammatory factors in endometriosis (Ems)model rats. METHODS:SD rats were randomly divided into normal group(normal saline),model group(normal saline), mifepristone low-dose,medium-dose and high-dose groups [0.65,1.30,2.60 mg/(kg·d)],with 10 rats in each group. Except for normal group,Ems model was induced in other groups, and they were given relevant medicine intragastrically for consecutive 4 weeks. The volume of the ectopic focus was compared before and after treatment. The serum contents of TNF-α,IL-6 and IL-8 were detected by ELISA method. The phosphorylation of NF-κB p65 and the expression of COX-2 and PGE2 were detected by West-ern blot. The expression of NF-κB p65 mRNA in ectopic focus was detected by RT-PCR. RESULTS:Compared with normal group, the volume of the ectopic focus, the serum contents of TNF-α,IL-6 and IL-8,the expression of COX-2 and PGE2 protein, the phosphorylation of NF-κB p65 and the expression of NF-κB p65 mRNA in ectopic focus were increased in model group(P<0.01). Compared with model group, the volume of the ectopic focus, the serum contents of TNF-α,IL-6 and IL-8,the expression of COX-2 and PGE2 protein, the phosphorylation of NF-κB p65 and the expression of NF-κB p65 mRNA in ectopic focus were de-creased in mifepristone groups(P<0.01),in dose-dependent manner. CONCLUSIONS:Mifepristone can reduce the volume of the Ems ectopic focus, via blocking NF-κB signaling pathway and reducing the expression of inflammatory factors.

OBJECTIVE:
To study the efficacy of treatment with microsurgery in combination with local injection of type A botulinum toxin for vocal process granuloma.
METHOD:
28 patients with vocal process granuloma received endotracheal intubation under general anesthesia. The lesion was removed with micro-scissor and CO2 laster under a self-retaining laryngoscope and microscope. The incision and mucous membrane surrounding the wound was closed with 8-0 absorbable suture. 4-point injection of botulinum toxin type A 8-15 u was then performed along the thyroarytenoid muscle and arytenoid muscle of the same side. Postoperative medication was administered based on disease causes.
RESULT:
All patients experienced vocal cord dyskinesia of the injected side 2-3 days after surgery. At 1 month after the surgery, wound healing was good in all the 28 patients, and the vocal cord movement was limited at the injected side. At 3 months, movement of the bilateral vocal cords was normal, and the vocal cord process mucosa was smooth. Patients were followed up for more than a year, and only one patient had recurrence in 2 months after surgery. The cure rate was 96. 4%.
CONCLUSION
Combination of laryngeal microsurgery and type A botulinum toxin local injection can shorten the treatment course of vocal process granuloma.

Chinese Journal of Rehabilitation Theory and Practice 2014;(10):995-997
Objective To discuss the impact of the pattern of student-autonomous laboratories opening on the experimental teaching of rehabilitation specialty. Methods Laboratories were opened to sophomore students of rehabilitation specialty. Planned opening pattern is employed on students of 2009 class and autonomous opening pattern is used on students of 2010 class. At the end of each term, results of these opening patterns are compared. Results The total opening hours of planned opening pattern were 89 with 755 person-times involved. The accumulated opening hours of autonomous opening pattern were 323 with 2677 person-times involved. Students of the autonomous opening pattern class achieved better academic results than students of planned opening pattern (P<0.05) in 7 major courses. Opening laboratories to students contributed to the improvement of rehabilitation skills, better understanding of the curricular knowledge, the enhancement of problem-analyzing and problem solving abilities, the stimulation of interest in major study and also the fostering of teamwork spirit. And the autonomous opening pattern achieved more desirable results (P<0.05). Conclusion The student-autonomous lab opening pattern contributes more to the improvement of students' practical skills, innovative spirit as well as comprehensive clinical rehabilitation abilities.

A reporter system for ?C31 integrase was developed in NIH3T3 cells. The reporter plasmid coding green fluorescent protein(GFP) coupled with red fluorescent protein(RFP) was co-transfected with the plasmid coding ?C31 integrase, to show the activity of integrase in the cells.Fluorescence activated cell sorter(FACS) was used to measure the proportion of the cells containing red and green fluorescence. The increment of green cells was positively related to the increase in the transfection with plasmid coding ?C31 integrase.Approximately 90% of green cells were observed under a ratio of plasmid-?C31-integrase/reporter plasmid at 10∶1.This suggests that the ?C31 integrase reporter system provides a probe for the function of ?C31 integrase in cells.

AIM To study the activation of choline on M3-R in heart and observe the hemodynamic changes of rat and rabbit. METHODS A cardiac catheter was inserted into the left ventricular cavity via the right carotid artery, then the HR, LVSP, LVEDP, and ±dp/dt were measured using a polygraph system. RESULTS Choline was shown to decrease the hemodynamic assessments, such as HR, +dp/dt, LVSP and LVEDP. while the M3-R antagonist 4-DAMP (4-diphenylacetoxy-N-methylpiperidine-methiodide) showed little effect on these assessments. It was found to reverse the hemodynamic effects of choline. CONCLUSION M3 receptor agonist can produce negative inotropic and chronotropic effects on the heart of rat and rabbit.

A reporter system for φC31 integrase was developed in NIH3T3 cells. The reporter plasmid coding green fluorescent protein (GFP) coupled with red fluorescent protein (RFP) was eo-transfected with the plasmid coding φC31 integrase, to show the activity of integrase in the cells. Fluorescence activated cell sorter (FACS) was used to measure the proportion of the cells containing red and green fluorescence. The increment of green cells was positively related to the increase in the transfection with plasmid coding φC31 integrase. Approximately 90% of green cells were observed under a ratio of [plasmid-φC31-integrase]/[reporter plasmid] at 10 : 1. This suggests that the φC31 integrase reporter system provides a probe for the function of φC31 integrase in cells.

OBJECTIVE: To screen the anti-tumor fraction of ethanol extracts from Thymus quinquecostatus Celak and investigate its anti-tumor effect on human leukemia cell line. METHODS: Ethyl acetate, n-butanol and acetone fractions were separated from the ethanol extracts of wild Thymus quinquecostatus Celak. Growth inhibiting effects of these extracts on human leukemia cell lines K562 and HL-60 were determined by live cell counting and cell growth curve analysis. The possible anti-tumor mechanism was studied by morphological analysis with norcantharidin as a positive control. RESULTS: Ethyl acetate fraction could significantly inhibit the proliferations of K562 and HL-60 cells, and the inhibiting effect depended on the concentration of ethyl acetate fraction. Ethyl acetate fraction could induce apoptosis of K562 and HL-60 cells. The n-butanol and acetone fractions had no significant inhibiting effect on K562 and HL-60 cells. CONCLUSION: Ethyl acetate fraction is the major anti-tumor fraction in ethanol extracts from Thymus quinquecostatus Celak.
