1.Changes of whole hemocyte parameters and hemorrheologic indexes after treatment in patients with cerebral infarction

BACKGROUND: The abnormal changes of hemorrheologic indexes are closely correlated with vascular injury, disorder of microcirculation, thrombosis and its development.OBJECTIVE: To investigate the changes of 25 indexes of hemorheology and whole hemocytes in patients with acute cerebral infarction.DESIGN: A controlled case study.SETTING: Department of Medical Laboratory, Xinxiang Medical College.PARTICIPANTS: Cerebral infarction group: Totally 127 patients with the first attack of acute cerebral infarction were selected from the Department of Neurology, the Second Affiliated Hospital of Xinxiang Medical College from January to December in 2002. The onsetof disease ranged from 4 to 12 hours, and the lesions were confirmed by cranial CT, the diagnostic standard accorded with the diagnostic guidelines for various cerebrovascular diseases formulated by the Fourth National Academic Conference for Cerebrovascular Diseases. Normal control group: Fifty-three healthy physical examinees with normal blood pressure and without diseases of heart,brain and kidney, as well as diabetes mellitus. There were no sigmficant differences in ages and male to femaleratios between the two groups.METHODS: ① Patients in the cerebral infarction group received fibrinogen reduction therapy, they were injected with two pieces of 10 u defibrin on the day of admission, and injected with one piece of 10 u defibrin again on the 3rd, 5th and 7th days respectively. ② Fasting venous blood (5 mL) was drawn with thromboliquine anticoagulated vacuum blood collecting tube,and 2 mL blood was coagulated with EDTA-K2. The whole blood viscosity,whole blood high-sheafing reduction viscosity, whole blood low-shearing reduction viscosity, whole blood high-sheafing relative index, whole blood lowsheafing relative index, equation K value of erythrocyte sedimentation, plasma viscosity, hematocrit, erythrocyte sedimentation, aggregation index of red blood cells (RBC), rigidity index of RBC, deformation index of RBC, counts of while blood cells (WBC) and RBC, mean volume of RBC, mean corpuscular volume (MCV), RBC distribution width (RDW), content of hemoglobin,mean corpuscular hemoglobin content (MCH), mean corpuscular hemoglobin concentration (MCHC), platelet count, mean platelet volume (MPV) and platelet distribution width (PDW) were detected when the shear rate was 200 s-1, 30 s-1, 5 s-1 and 1 s-1 respectively. ③ The above indexes of the patients with cerebral infarction were detected within 24 hours after attack (exacerbation) and at two weeks after treatment (recovery period) respectively.MAIN OUTCOME MEASURES: The hemorrheologic and whole hemocyte indexes were compared between the two groups.RESULTS: All of the 127 cases in the cerebral infarction group entered the analysis of results at exacerbation, and 5 of them were not reexamined because of discharge and 122 cases entered the analysis of results at recovery period. All the 53 cases in the normal control group were involved in the analysis of results. ① Except RDW and MCHC, all the other indexes in the patients with cerebral infarction at exacerbation were significantly different from those in the normal control group (P<0.05, 0.01). As compared with the normal control group, the whole blood low-shearing viscosity, whole blood low-shearing reduction viscosity, whole blood high-sheafing relative index, whole blood low-shearing relative index, plasma viscosity,hematocrit, aggregation index of RBC, WBC count and MCH all returned to normal in the patients with cerebral infarction at recovery period (P>0.05).② As compared with the exacerbation, the RBC count and MPV were obviously decreased in the patients with cerebral infarction at recovery period (P < 0.05), while the other indexes were similar (P > 0.05).CONCLUSION: Abnormalities of hemorrheologic indexes commonly exist in patients with acute cerebral infarction, which indicates that the abnormalities of hemorrheologic and whole hemocyte parameters may be related to the occurrence of acute cerebral infarction.
2.Peripheral blood mononuclear cell transplantation for liver cirrhosis

BACKGROUND:Compared with bone marrow transplantation, peripheral blood stem cel transplantation has its own advantages, including rich resources of stem cel s from the peripheral blood, convenient and easy col ection, without anesthesia, smal trauma, easily accepted, high safety, and easy to restore the patient’s hematopoietic system.
OBJECTIVE:To observe the function and safety of autologous peripheral blood mononuclear cel s in the treatment of patients with decompensated cirrhosis.
METHODS:Four patients with decompensated liver cirrhosis were selected from November 2010 to July 2011 in the First Affiliated Hospital of Dalian Medical University, aged 31-67 (averagely 44 years). Among them, three cases had hepatitis B, and another one had autoimmune liver disease. Peripheral blood stem cel s were col ected after being mobilized by granulocyte colony stimulating factor. Then, autologous peripheral blood stem cel s were transplanted via a hepatic artery catheter.
RESULTS AND CONCLUSION:There were no adverse reactions such as fever, bleeding and nausea after peripheral blood stem cel col ection and hepatic artery transplantation. Symptoms such as fatigue, poor appetite and abdominal distension gradual y improved at 1, 3 and 6 months after transplantation. Liver function and liver fibrosis indexes were improved to some extent after transplantation.
3.Establishment of normal reference intervals of plasma and urine neutrophil gelatinase associated lipocalin in children

Objective To establish normal reference intervals of plasma and urine neutrophil gelatinase-associated lipocalin (NGAL) in children′s hospital.Methods A total of 183 fresh EDTA anticoagulant samples and 125 fresh urine in healthy children were collected from May 2014 to October 2014.According to the CLSI C28-A2 ,the unilateral upper limit 95% was established the normal reference value in different age group.Results There was significant difference in four groups (P<0.05).The normal reference intervals of plasma NGAL in healthy children:0 to <7 months;<291.28 μg/L;7 months to <5 years old;<150.87 μg/L;5 years old to <9 years old:<127.93 μg/L;9 years old to ≤16 years old:<161.74 μg/L;the normal reference intervals of healthy children urine NGAL:0 to <7 months:<257.31 μg/L;7 months to <5 years old:<201.55 μg/L;5years old to <9 years old:<197.69 μg/L;9 years old to ≤16 years old:<151.46 μg/L.Plasma and urine NGAL results in neonatal group were higher than the other three groups.Conclusion The normal reference intervals of plasma and urine NGAL in children′s hospital is established.this could provide clinical evidence for the diagnosis and treatment of acute renal injury in pediatric patients.
4.Effect of Gemcitabine on viability and apoptosis of non-small cell lung cancer HCC827 in vitro

Objective:To observe the effect of Gemcitabine ( GEM) on the viability and apoptosis of non-small cell lung cancer HCC827 in vitro. Methods:The cell viability,apoptosis and cell cycle of HCC827 cells induced by Gemcitabine were detected with cell counting kit-8 assay (CCK-8),Annexin V-FITC/PI staining and flow cytometry. The expression of Bcl-2 protein of cells treated with GEM was examined by Western blot assay. Results: There was significant inhibition effect on HCC827 cells treated with 0. 1-1 000 ng/ml of GEM,which can promote the occurrence of HCC827 cell apoptosis and arrest cell in the S phrase. The apoptosis induced by GEM was accompanied with the down regulation of Bcl-2 protein. Conclusion: GEM can inhibit the cell viability and induce the HCC827 cell apoptosis and S phrase arrest. Its cell dead type was apoptosis,which was related with the expression of Bcl-2 protein.
5.Evaluation of Artificial Pneumothorax CT Examination in Detection of Pleural Metastatic Tumor

Objective To evaluate the ability of the artificial pneumothorax CT examination in detection and diagnosis of pleural metastatic tumor.Methods A retrospective study of 14 patients with pleural effusion using artificial pneumothorax CT examination was conducted.Results Six of the 14 patients were found to have a nodule or nodules on the pleura.The pleural metastatic tumors of 3 cases were from the lung cancers and another 3 from breast cancers were proved histologically.Conclusion The artificial pneumothorax CT examination is a effective and simple method in detecting of pleural metastatic tumor.
6.Values of kinetic features measured by computer-aided diagnosis for breast MRI

Objective To investigate the value of kinetic features measured by computer-aided diagnosis (CAD)for breast MRI.Methods One hundred and sixty four lesions diagnosed pathologically by operation or biopsy comprised the analysis set.Automated lesion kinetic information from CADStream programs for breast MRI was identified.Three CAD variables were compared for benign and malignant lesions: initial phase peak enhancement (greatest percentage of signal intensity increase on first contrast enhanced sequence),delayed phase enhancement categorized by a single type of kinetics comprising the largest percentage of enhancement (washout,plateau,or persistent),and delayed phase enhancement categorized by single most suspicious type of kinetics (any washout > any plateau > any persistent).Morphological characteristics of breast lesions were described according to breast imaging and reporting data system (BI-RADS).Initial phase peak enhancement mean values between benign and malignant breast lesions were compared by using Wilcoxon rank-sum test,delayed phase enhancement categorized by a single type of kinetics comprising the largest percentage of enhancement or by single most suspicious type of kinetics between benign and malignant breast lesions were compared by using Chi-square test.Results There were 72 benign and 92 malignant breast lesions.A total of 123 (75.0%) mass lesions were identified,and the other 41 (25.0%) lesions showed no mass.Thirty lesions were BI-RADS-MRI 2,68 lesions were BI-RADS-MRI 3,43 lesions were BI-RADS-MRI 4,23 lesions were BI-RADS-MRI 5.Initial phase peak enhancement mean values of benign and malignant lesions were 237% (69% to 629%)and 336% (86% to 793%),respectively.There was no significant difference between benign and malignant lesions in initial peak enhancement mean value (Z =-1.626,P =0.104).Delayed phase enhancement categorized by single most suspicious type of kinetics (any washout > any plateau > any persistent) for benign and malignant lesions were 15,10,47 and 2,3,87 respectively.There was a significant difference between benign and malignant lesions (x2 =23.562,P =0.000).Initial peak enhancement value < 100% or ≥100% were 5 and 67 for benign lesions,3 and 89 for malignant lesions,respectively.There was no significant difference between benign and malignant lesions at 100% threshold (x2 =1.181,P =0.277).Delayed phase enhancement categorized by a single type of kinetics comprising the largest percentage of enhancement (washout,plateau,or persistent) for benign and malignant lesions were 48,6,18 and 47,15,30 respectively.There was no significant difference between benign and malignant lesions (x2 =4.496,P =0.106).Conclusions Of CAD kinetics analyzed,only delayed enhancement categorized by most suspicious type is helpful for the differentiation between benign and malignant lesions.However,there is significant overlap between initial peak enhancement at 100% threshold or delayed kinetics categorized by largest percentage enhancement types of benign and malignant lesions,so lesion morphologic features should be considered.
7.Studies on Skin Permeation in vitro of Phloretin and Ligustilide in Fufang Phloretin Cream

Objective: To determine the transdermal absorption of phloretin and ligustilide in Fufang phloretin cream.Methods: In vitro percutaneous absorption test using the Franz intelligent transdermal absorption apparatus was adopted, an HPLC method was used to determine the contents of phloretin and ligustilide in the accepting solution at different time points, and then the cumulative release amount and the total penetration amount were calculated to evaluate the percutaneous absorption behavior.Results: Phloretin and ligustilide showed a good linearity within the range of 0.044 2-4.42 μg·ml-1(r=0.999 9) and 0.048 7-4.87 μg·ml-1(r=0.999 9), respectively, the average recovery was 99.85% and 99.92%, and RSD was 0.67% and 0.66% (n=9), respectively.The in vitro permeation behavior of phloretin and ligustilide through the skin of rats fit Higuchi equations(r>0.9), suggesting a good linear correlation between the cumulatine penetration amount (Qn) and (time).Conclusion: Fufang phloretin cream has a good percutaneous permeation property in vitro.
8.Effect of Finger-acupuncture Massage on Fine Motor Functions of 40 Children with Cerebral Palsy

ObjectiveTo observe the effects of finger-acupuncture massage on the fine motor functions of children with cerebral palsy. Methods40 children with cerebral palsy following diplegia were randomly divided into two groups: observation group(n=20) and control group(n=20). The control group was treated with rehabilitation training, while the observation group received finger-acupuncture massage and the rehabilitation training. All patients were assessed by Peabody Developmental Motor Scale before and after the treatment.ResultsThere was no statistically significant difference in fine motor between the two groups before the treatment (P>0.05). There was a significantly difference in fine motor quotient(FMQ), grasp capacity index, and capacity index of visual-motor integration both in the observation group and the control group between before and after the treatment (P<0.001). There was also a difference in fine motor quotient (FMQ), grasp capacity index, capacity index of visual-motor integration between the observation group and the control group after the treatment(P<0.05).ConclusionFinger-acupuncture massage combined with rehabilitation training can improve fine motor functions of children with cerebral palsy following diplegia.
9.Effect of Ethyl acetate extract of Herba Eclipta on immune function in immunosuppressive mice

Objective:The regulatory effects of ethyl acetate extract of Herba Eclipta EAEE on immune function were studied.Methods: The experimental immunosuppressive models were induced by injection of Cyclophosphamide or hydrocortisonc.Results: The index of thymus gland spleen in immunosuppressive mice was increased significantly. when large and small dose of EAEEP were given. EAEE remarkably enhanced the delayed type hypersensitivity in immunosuppressive mice. Conclusions: EAEE possesses notable immunomodulatory action on immune system in mice.
10.Relationship between ZNF139 and multidrug resistance(MDR) related genes in SGC7901 and SGC7901/ADR cell lines

Background and purpose:It was reported that zinc ifnger protein 139 (ZNF139) was expressed aberrantly in gastric cancer. But the relationship between ZNF139 and multidrug resistance (MDR) of gastric cancer is still not clear. The purpose of this research was to investigate the expressions and signiifcance of ZNF139, MRP-1, MDR1/P-gp, GST-π in human gastric carcinoma cell lines SGC7901 and SGC7901/ADR. Methods: The expressions of ZNF139, MRP-1, MDR1/P-gp, GST-πwere determined with RT-PCR and Western blot in SGC7901 and SGC7901/ADR cell lines. Then siRNA recombinant plasmid of targeting ZNF139 gene was constructed and imported into gastric cancer cell line SGC7901/ADR, and the expressions of MRP-1, MDR1/P-gp, GST-πwere tested simultaneously. Results:The expressions of ZNF139, MRP-1, MDR1/P-gp, GST-πwere higher in SGC7901/ADR than in SGC7901(P<0.05). ZNF139 was inhibited obviously after siRNA-ZNF139 was transfected into SGC7901/ADR, and expression of MRP-1, MDR1/P-gp, GST-πdecreased(P<0.05). Conclusion:ZNF139 may be invovled in multidrug resistance (MDR) of gastric cancer by up-regulating MRP-1, MDR1/P-gp and GST-π.
