1.Advances in studying the differentiation into neurons of mesenchymal stem cells and their application

Mesenchymal stem cells (MSCs) are a population of multipotent cells that can proliferate and differentiate into marrow and non-marrow cell types, such as adipocytes, chondrocytes, myocytes, and so on. In recent years, many researchers have studied whether MSCs are capable of differentiation into neurons in vivo and ex vivo. The result that MSCs-derived neurons express NSE and NF, but don't express GFAP suggests MSCs can differentiate into neurons, some researchers have achieved success in promoting functional recovery in Pakinsons and transactional spinal cord injury rat models by use of MSCs-derived neurons. Therefore, MSCs-derived neurons will play an important role in the therapy for a variety of diseases of the nervous system. [
2.A comparative study on clinical effect of Naukan therapy and health education in patients with alcohol dependence

Objective To explore Naukan therapy and health education combined with drug treatment clinical efficacy in the treatment of alcohol dependence.Methods The 66 cases of male alcohol dependent patients were randomly divided into study group (33 cases) and control group (33 cases),and both of patients were taking generic drugs.The study group was given Naukan treatment (including centralized within concept and dispersion Vipassana),and the control group received health education (including collective health education and individual health education).Duration of treatment was 4 weeks,the total number of hours were 55.5 h,56 h.Obsessive compulsive drinking scale(OCDS),social support rating scale (SSRS),Minnesota multiphasic personality inventory (MMPI) for the two groups before and after treatment; and follow-up the rate of recovery drink three months after hospital discharge.Results ① OCDS score after treatment in the study group was lower than that in the control group,the difference was statistically significant ((48.21 ± 2.05) vs.(54.15 ± 2.01),t =11.885,P<0.01).②After treatment SSRS total score,objective support,subjective support,social utilization factor scores were higher than those in the control group,and differences were statistically significant ((49.05±5.63)vs.(40.15±6.50),t=5.946,P=0.000) ; ((14.32±3.51) vs.(8.72±6.22),t=4.504,P< 0.05) ; (27.02±4.26) vs.(19.45±4.92),t=6.682,P=0.000; (10.02±3.30) vs.(7.52±3.86),t=2.828,P=0.006).③MMPI with schizophrenia (Sc),paranoia (Pa),mental weakness (Pt),psychopathy (Pd) ie 8/6/7/4 model-based in two groups,and Sc,Pa,Pd,Pt dimensions of the original sub in study group were lower than those in the control group after treatment,in addition to outside Pt differences were statistically significant ((25.43 ± 6.76) vs.(29.01 ± 6.72),t =2.158,P=0.035; (14.12±6.01) vs.(18.32±6.42),t=2.744,P=0.008; (18.63±6.85) vs.(23.29±6.12),t=2.342,P=0.022).④Redrinking rate in study group was 39.39% (11/33),and 72.73% (24/33)in the control group,the difference was statistically significant(x2 =7.44,P<0.05).Conclusion The Naukan therapy combined with general drug treatment can solve the problem of patients with alcohol dependenceof alcohol dependence in the psychological and physiological aspects,and its efficacy is superior to general health education combined drug therapy.
3.Bone marrow mesenchymal stem cells modified by adenoviral vector containing CTLA4Ig inhibit immune response ex vivo

AIM: To investigate the feasibility and infection efficiency of MSCs as the target cells of gene delivery mediated by adenoviral vector carrying CTLA4Ig gene, and to study the mechanism of transgenic MSC to inhibit immune response ex vivo. METHODS: The recombinant adenovirus containing CTLA4Ig gene was constructed, by which rat MSCs with various multiplicity of infection (MOI) were conducted. The infection efficiency was analyzed with FACS and fluorescence microscope. The expression of CTLA4Ig protein in transgenic MSCs was detected by FACS and western blot. Co-culturing the transgenic MSCs with mixed lymphocytes, the inhibitory effect of transgenic MSCs on lymphocyte proliferation was also observed. RESULTS: The adenoviral vector delivered CTLA4Ig gene with high efficiency to MSCs. The expression of CTLA4Ig protein was detected in transgenic MSCs. The gene modified MSCs inhibited the proliferation of mixed lymphocytes and maximal inhibition rate was observed on day 4 of MLR. The inhibition induced by CTLA4Ig was donor-specific. CONCLUSION: MSCs is a promising target cell for gene delivery. The expressed CTLA4Ig specifically inhibits the lymphocyte proliferation ex vivo.
4.Preliminary application of MRI-navigation system in assisting the performance of percutaneous biopsy

Objective To evaluate the effectiveness and safety of MRI-navigation system EMT-100 in assisting the performance of precise percutaneous puncture biopsy.Methods With the help of MRI-navigation system EMT-100,percutaneous puncture biopsy of thoracic and abdominal lesions was performed in 42 patients.The puncture success rate was used as the main index to evaluate the effectiveness of MRI-navigation system EMT-100.The success rate of puncture biopsy,the total time used for procedure,the average number of puncturing,the average number of scanning,and the incidence of complications were recorded.Results In the 42 patients,the success rates with single and twice puncturing were 86% (36/42) and 14% (6/42) respectively.The success rate of puncture biopsy was 100%.The mean time used for locating puncture site was (11.5±5.5) min;the average number of puncturing was (1.4±0.5) times,and the average number of scanningwas(4.2±0.8) times.Among the 32 patients with pulmonary lesions,bloody sputum occurred in 2 patients (6.2%) and small amount pneumothorax in one patient (3.1%),and no serious complications occurred in other patients.Conclusion In performing percutaneous puncture biopsy of thoracic and abdominal lesions,the use of MRI-navigation system is safe.This technique has certain advantages,such as accurate positioning,high puncture success rate,wide range of application,radiationless,etc.It can help precisely obtain the lesion tissue and get pathological diagnosis.Therefore,it is really a valuable guiding technology.(J Intervent Radiol,2017,26:263-265)
5.Macaca irus bone mesenchymal stem cells differentiate into neuron-like cells induced by cryptotanshinone in vitro

AIM: To determine the optimal protocol and condition in which macaca irus mesenchymal stem cells (MSCs) are induced to differentiate into neuron-like cells by cryptotanshinone in vitro. METHODS: MSCs from macaca irus bone marrow were generated in vitro and induced with cryptotanshinone. The morphological changes of MSCs were evaluated by microscope. The positive percentages of neurofilament (NF), neuron specific enolase (NSE) and glial fibrillary acidic protein (GFAP) expression were measured by immunocytochemistry with ABC staining. RESULTS: The result showed that MSCs were positive for CD29, CD44, CD105, CD166, and negative for CD34, CD71, CD80 and CD86. After induced with cryptotanshinone, MSCs began to display neuronal morphologies, such as contracted multipolar cell body and formed extensive networks. The percentages of positive NSE, NF expression were 68.3%?3.5%, 70.3%?1.5%, respectively. CONCLUSION: Macaca irus MSCs could be induced to differentiate into neuron-like cells in vitro by cryptotanshinone and might be applied in cell transplantation and gene therapy in nervous system disorders.
6. Improvement of preanalytical screening interference on coagulation tests by auto checking system

Objective:
To investigate the potential improvement of sample quality by automatic pre-analysis sample checking system, comparing to visual inspection for coagulation tests routinely.
Methods:
Thirty samples with hemolysis, Icteric and lipemia in different levels were prepared and issued to 13 technicians for visual check, to evaluate the consistency individually. 2 949 blood samples with order for coagulation test were collected in Beijing Chaoyang Hospital in April and May 2018, the quality of all samples was evaluated by both visual check and automatic sample quality checking system before analysis, performance of two measurements detecting hemolysis, lipid, icteric or clot was compared.
Results:
Significant differences were found in visual check among operators. The Kappa coefficients in three randomly selected groups were 0.32, 0.26 and 0.38 respectively, indicating that the consistency of visual check was poor. Among all investigated samples, 3 samples with unacceptable interference were detected visually, including 2 samples with hemolysis and another one with lipemia. On the other hand, 19 unqualified samples were identified by automatic checking system. Five types of interference of unqualified samples were detected as icteric (26.32%,5/19), clot (21.05%,4/19) hemolysis (5.26%,1/19),lipemia (36.84%, 7/19), and hemolysis with lipemia (10.53%,2/19) respectively by automatic checking system. But one case of hemolysis sample rejected by visual check was not rejected by automatic sample quality checking system.7 samples were merely affected on D-dimer by lipemia, which level did not influence the results of prothrombin time(PT) and activated partial thromboplastin time (APTT). Notably, other two samples were interfered with not only tests of PT, APTT and fibrinogen by hemolysis, but also D-dimer by the considerable level of lipemia, which showed the superiority of test-specific quality checking features.
Conclusions
The automatic pre-analysis sample quality checking system can improve the detection rate of unqualified samples and improve the efficiency of routine, helping realization of total quality management.
7.Efficiency of adenoviral vector mediated CTLA4Ig gene delivery into mesenchymal stem cells.

OBJECTIVETo prevent Graft-versus-host disease (GVHD) in rat model, we evaluated the feasibility of mesenchymal stem cells (MSCs) as a gene transfer target and studied the efficiency of recombinant adenovirus mediated gene therapy.
METHODSWe constructed the recombinant adenovirus containing CTLA4Ig gene. Rat MSCs of passages 3-5 were infected by the adenovirus, and the transfection efficiency was monitored by GFP markers. We performed flow cytometric analysis, immunohistochemical and Western blotting analysis to identify the CTLA4Ig expression. The gene transferred MSCs were tested for their ability to inhibit the allogeneic lymphocyte response in vitro and to prevent GVHD in a rat model.
RESULTSRecombinant adenovirus pAd-CTLA4Ig was correctly constructed and confirmed. After MSCs were infected by the adenovirus, the CTLA4Ig protein was detected not only in transgenic MSCs, but also in the culture medium. In a mixed lymphocytes response (MLR) test, the transgenic MSCs could significantly inhibit the allogeneic lymphocyte response compared with the control groups (P < 0.05). A model of GVHD was developed by transplanting bone marrow cells and spleen lymphocytes of F344 rats to lethally irradiated SD rats. The onset of GVHD could be ameliorated or prevented by co-administration of transgenic MSCs. All the rats in the control groups suffered severe acute GVHD. CTLA4Ig expression was observed in the liver, intestine, kidney and spleen 30 days post-transplantation.
CONCLUSIONSOur results indicate that adenoviral vectors could efficiently transfer CTLA4Ig gene into MSCs and sustain long-term stable expression in vitro and in vivo.

8.Effect of group psycho-game therapy on patient with neurosis

Objective To observe the clinical effect of group psycho -game therapy on patient with neurosis .Methods 72 cases of patient with neurosis were randomly divided into the study group and the control group, each with 36 cases.Both groups received drug treatment and psychological health education , and the study group was given 10 weeks of group psycho-game therapy in addition .MMPI, group psycho-therapy factor questionnaire , NOSIE were used to evaluate the effect before and after treatment .Results There was no statistically significant difference of MMPI and NOSIE between two groups before treatment (P>0.05).After treatment, the score of Hs, D, Hy, Pt, Si dimension of MMPI was respectively (10.53 ±4.55),(27.23 ± 5.02),(23.26 ±5.12),(18.25 ±7.21),(35.26 ±5.81)in the study group, which was significantly different from that in the control group (t=2.951,3.261,3.116,3.146,2.116, respectively; P<0.05).There was statistically significant difference of Hs , D, Hy, Pd, Pa, Pt, Sc, Si dimension of MMPI before and after treatment in the study group (P<0.05).There was statistically significant difference of Hs , D, Hy, Pa, Pt dimension of MMPI before and after treatment in the control group (P<0.05).After treatment, the total score of NOSIE was ( 218 .84 ±10 .26 ) in the study group , and ( 195 .30 ±21 .35 ) in the control group , with statistically significant difference (t=5.958,P<0.05).There was statistically significant difference of every dimension of NOSIE except personal cleanliness between two groups after treatment (P<0.05).There was statistically significant difference of all factors except identity and family recurrence of group psycho -therapy factor questionnaire (P <0.05).Conclusions Group psycho-game therapy is effective to control clinical symptoms of patient with neurosis , improve their personality defects and promote their psychological growth .
