1.The connection between the weight of the new born baby and the age and body mass index of the pregnant woman

Objective: To analyze the connection between the age of the mother and the birth weight of the new born baby as well as the impact of weight index during pregnancy on the birth weight of the new born baby.Method: Analyze the weight of the 1301 new born babies delivered from January to December 2005 in the First Affiliated Hospital of Chongqing University of Medical Sciences;analyze 576 first time lying-in women who had normal vagina delivery in the First Affiliated Hospital of Chongqing University of Medical Sciences from January 1999 to July 2003,among which there were 184 samples of corpulence during early period of pregnancy with BMI equal to or higher than 24 and 152 samples of corpulence during confinement with BMI equal to or higher than 27.Observe the connection between the birth weight and the age and BMI of pregnant women.Results: Excluding gestational diabetes,non-full-month gestation,double and triple embryo gestation,the average birth weight of 1301 new born babies was 3396g with the average birth weight of 3453g for male babies and 3397g for female babies respectively,among which only those women at the age between 20 and 24 had the new born babies with the weight lower than the average.Compared the corpulent during early period of pregnancy with the compared group,the happening rate of huge baby is evident.Compared the corpulent during confinement with the compared group,the happening rate of huge baby is evident.Compared the weight increase equal to or higher than 15kg during early period of pregnancy with the weight increase lower than 15kg,the happening rate of huge baby is evident.Conclusion: Too young the age of the mother has negative impact on the development of fetus.Too much increase of weight during pregnancy results in increasing happening rate of huge baby.

3.The Relationship between the Expression of Bone Morphogenetic Protein-2 and Matrix Metalllopro Teinase-9 in Fetal Membrane and Premature Rupture of Membranes

Objective:To study the correlation between the bone morphogenetic proteins (BMP-2) and matrix metalloproteinase-9(MMP-9) in fetal membrane and premature rupture of membranes. Methods: 18 samples were selected separately from each of the following groups: puerpera with term pregnancy in labor, puerpera with term premature rupture of membranes(PROM), puerpera in preterm labor, puerpera with pre-tenm PROM and puerpera with term pregnancy not in labor (control group). After labor, fetal membranes in the rupture site were taken out. Immunobistochemistry and pathologic image multimedia operation system were adopted to semiquantitative analyze the area integrated optical density (AIOD) of the positive expression of BMP-2 and MMP-9. Results:①The AIOD of BMP-2 and MMP-9 positive expression in fetal membranes was higher in term pregnancy in labor group, term PROM group, preterm labor group and PROM group than that in control group. There was statistic difference ( P < 0. 0071). ②The AIOD of BMP-2 and MMP-9 in term PROM group was higher than that in term pregnancy in labor group(P=0.0065,P=0.0069), which was higher in PROM group than that in preterm labor group (P=0.0069, P = 0.0052) and term PROM group P=0.0037, P =0.0065),③There was obviously positive correlation between BMP-2 and MMP-9 expression in the fetal membranes in five groups. ( r_s =0.76159,P<0.0001). Conclusions: The expression of MMP-9 and BMP-2 is closely relevant to the repture of membranes. There is a correlation between BMP-2 and MMP-9 expression in fetal membranes.
4.Carbapenemase genes and homology of Acinetobacter baumannii in two hospitals of Qingdao

Objective To investigate antimicrobial resistance,distribution,and carriage of carbapenemase genes of Acinetobacterbaumannii(AB)from two hospitals in Qingdao.Methods 145 AB isolates collected from two hospi-tals (78 from hospital A,67 from hospital B)were performed antimicrobial susceptibility testing,carbapenemase genes were amplified by polymerase chain reaction (PCR);homology analysis were conducted with enterobacterial repetitive intergenic consensus (ERIC)-PCR.Results AB from hospital A were generally resistant to 16 commonly used antimicrobial agents,with the lowest resistant rate of 3.85% to cefoperazone/sulbactam,followed by resist-ance rate of 16.67% to minocycline,resistant rates to the other antimicrobial agents were all>73% . AB from hos-pital B were generally resistant to 23 commonly used antimicrobial agents,but the resistance rates to minocycline and tigecycline were both 0,resistance rates to amikacin and levofloxacin were 23.88% and 38.81% respectively, resistant rates to the other antimicrobial agents were all >64% . All strains carried OXA-5 1 gene,the carriage rates of OXA-23 gene in carbapenem-resistant group were 86.76% (59/68)and 56.67% (34/60)in hospital A and B re-spectively,the difference was significant(χ2= 14.53,P<0.001);OXA-58 gene was detected in 3 isolates in hospi-tal A but not detected from hospital B. 145 AB strains were classified into 8 types,the major prevalence types were type A (n= 71)and E(n= 37);the major prevalence types in hospital A were type A (46.15% )and E(41.03% ), hospital B were type A (52.24% )and C (17.91% ).Conclusion Antimicrobial resistance of clinically isolated AB is serious and prevailed in two hospitals. OXA-23 and OXA-51 genes play an important role in AB resistance to car-bapenems.
5.Effects of Xuebijing injection on blood coagulation in patients with sepsis

Objective To observe the effect of Xuebijing injection on blood coagulation in patients with sepsis. Methods 300 patients with sepsis were randomly divided into control group and Xuebijing injection treatment group(each,150 cases). According to the guidelines of treatment for sepsis,the conventional treatment was given to both groups,and additionally Xuebijing injection 50 mL plus normal saline 100 mL intravenous drip was applied in the treatment group,twice a day for consecutive 7 days. Before treatment and after treatment for 3 days and 7 days,platelet count(PLT),four items of blood coagulation〔prothrombin time(PT),thrombin time(TT),fibrinogen (Fib)and activated partial thromboplastin time(APTT)〕,D-dimer levels and disseminated intravascular coagulation (DIC)incidence were observed in the two groups. Results Before treatment,the levels of Fib,PT,TT,APTT, PLT,D-dimer and the incidence of DIC had no significant differences between the two groups(all P>0.05);after treatment,the levels of PT,TT,APTT,D-dimer and the DIC incidence were significantly lower than those before treatment,while Fib and PLT were significantly enhanced after treatment,and on the 7th day after treatment,the changes in Xuebijing group were more obvious〔Fib(g/L):3.02±0.51 vs. 2.53±0.56,PT(s):13.82±1.91 vs. 16.03±1.68,TT(s):13.85±1.94 vs. 16.03±1.65,APTT(s):39.47±4.73 vs. 42.75±5.24,PLT(×109/L):118.10±15.80 vs. 99.77±15.67,D-dimer(mg/L):3.17±0.26 vs. 3.66±0.50,DIC incidence:5.3%(8/150)vs. 14.0%(21/150),P<0.05 or P<0.01〕. Conclusion Xuebijing injection can improve the hypercoagulable state and reduce the incidence of DIC in patients with sepsis.
6.Effects of lentivirus-mediated heat shock protein 70 gene on calcium homeostasis in PC12 cells undergone ischemia and hypoxia

ObjectiveTo investigate the effects of lentivirus-mediated heat shock protein 70 (HSP70) gene on calcium homeostasis in PC12 cells undergone ischemia and hypoxia, and the mechanism involved.Methods PC12 cells at logarithmic phase were collected, and were divided into recombination lentivirus infection group (infected by lentivirus containing HSP70 and fluorescent gene), lentivirus control group (infected by lentivirus containing fluorescin without HSP70 gene) and non-infection group. HSP70 gene and protein expressions in PC12 cells were determined by reverse transcription-polymerase chain reaction (RT-PCR) and Western Blot. After being challenged with ischemia and hypoxia for 4 hours, the viability of cells was detected by methyl thiazolyl tetrazolium (MTT), the levels of lactic acid dehydrogenase (LDH) in cell supernatant were determined by LDH measurement test kit. The concentration of intracelluer calcium ([Ca2+]i) was assayed by flow cytometer. The activities of Na+-K+-ATPase, Ca2+-Mg2+-ATPase and total ATPase were measured by ATPase test kits.Results The expressions of exogenous HSP70 gene and protein were found by RT-PCR and Western Blot in the recombination lentivirus infection group. After being challenged with ischemia and hypoxia, the viability of cells in the recombination lentivirus infection group was increased significantly as compared with the lentivirus control group and non-infection group (A value: 0.575±0.020 vs. 0.395±0.014, 0.363±0.045,t1= 17.996,t2= 10.600, bothP< 0.001), the levels of LDH in culture medium and the concentration of [Ca2+]i were decreased significantly [LDH (U/L): 743.46±23.68 vs. 935.43±34.77, 962.89±26.68,t1= 11.179, t2= 15.044, bothP< 0.001; [Ca2+]i relative fluorescence: (31.60±2.43)% vs. (41.48±3.33)%, (40.40±3.05)%, t1= 5.853,t2= 5.502, bothP< 0.001], and the activities of Na+-K+-ATPase, Ca2+-Mg2+-ATPase and total ATPase were increased significantly [Na+-K+-ATPase (μmol·mg-1·h-1): 8.608±0.307 vs. 6.728±0.173, 6.450±0.091,t1=9.237,P1= 0.001,t2= 11.675,P2< 0.001; Ca2+-Mg2+-ATPase (μmol·mg-1·h-1): 10.523±0.036 vs. 7.910±0.209, 8.064±0.195,t1= 9.718,P1= 0.001,t2= 11.535,P2<0.001; total ATPase (μmol·mg-1·h-1): 17.041±0.324 vs. 14.150±0.182, 13.983±0.085,t1= 16.113,t2= 17.602, bothP<0.001]. There was no statistical difference in above indexes between lentivirus control group and non-infection group.Conclusion HSP70 can maintain the PC12 cells calcium homeostasis, which may be one of the important mechanisms of anti-apoptosis.
7.Percutaneous Radiofrequency Ablation of Pulmonary Metastases in the Patients with Colorectal Carcinoma under CT Guidance

Objective To explore the clinical effects and safety of treatment for colorectal cancer to pulmonary metastases with percutaneous multiple electrodes radiofrequency-2000 (RF-2000) under CT-guided.Methods 26 pulmonary metastatic tumors in 11 cases were treated with RF-2000. The following extents were observed: (1) blood pressure,pulse rate and oxygen saturation during the treatment;(2)the variety of the liver function,the lung function and renal function after treatment;(3) the variety of the volume and low dense shadow of tumors after treatment;(4)the survival rate for 5 years. Results (1) The blood pressure,pulse rate and oxygen saturation were not changed during the treatment;(2) the liver function,the lung function and renal function after treatment were also not changed;(3) the volume of tumors were increased one month after treatment,three months later,23 lesions of 26 tumors were disappeared,2 decreased to more than 80% and 1 to 50%~80% in volume,and the low dense shadow within the tumors appeared as Ⅰtype (less than 50%) in one,Ⅱtype (50%~80%) in 3 and Ⅲ type (more than 80%) in 22;(4)The survival rate for 5 years was 81.82%(9/11). Conclusion The clinical effects and safety of treatment of colorectal cancer to lung metastases with percutaneous radiofrequency-2000 (RF-2000) under CT-guided is excellent.
8.Experiences of surgical treatment with cicatricical laryngotracheal atrisia:report of 48 cases

Objective:In order to improve the treatment methods of laryngotracheal atresia, several improved surgical procedures were used.Method:From 1986,the 48 patients with laryngotracheal atresia accepted surgical treatment in our hospital.Laryngo-fissure with laser cauterization of the glottis, laryngotracheal reconstruction using surgical flaps with or without particles, laryngo-tracheal reconstruction using itself′s cicatrices, tracheal end-to-end anastomosis, two-stage laryngotracheal reconstruction using aqueduct method, bronchotracheal reconstruction through the chest. Result:46/48 cases were cured breathing without tracheal cannula.Following-up survey is 1~12 years.Conclusion:The treatment of cicatriceal laryngotracheal atresia was difficult, but if the surgical methods were selected carefully according to the situation of each patient, we also can get a good result.

Objective To investigate the interfering efficiency of RNAi technique on the expression of Bcl-2 oncogene in human oral adenoid cystic carcinoma (Acc-3/CDDP).Methods The recombined RNAi plasmids for Bcl-2 oncogene were constituted by the four successive steps-designing of Oligo DNAs, synthesis of Oligo DNAs, transfection of Oligo DNAs into pSUPER.neo+gfp vectors and selection of positive plasmids. In order to silence the expression of Bcl-2 oncogenes, the recombined RNAi plasmids were transfected into Acc-3/CDDP cells by culturing together for about 10 hours, and the interfering efficiency of RNAi for the two oncogenes was evaluated by fluorescence-quantitative RT-PCR.Results The interfering efficiencies for Bcl-2 oncogene were 0,66.20% and 0, respectively in psiB1、siB2、psiB3.Conclusions The recombined RNAi plasmids of psiB2 (CCgggAgATAgTgATgAA) for Bcl-2 oncogene can effectively silence the expression of Bcl-2 oncogenes in Acc-3/CDDP cell line.
10.Correlation between the biofilm-forming ability, biofilm-related genes and antimicrobial resistance of Acinetobacter baumannii

Objective To study the biofilm-forming ability and the distribution of biofilm-related genes in Acinetobacter baumannii clinical isolates as well as antimicrobial resistance,to analyze their relationships with the bacterial resistance phenotype.Methods A prospective study was conducted.Biofilm models of 70 strains Acinetobacter baumannii collected in Chengwu County People's Hospital from October 2012 to October 2013 were constructed using 96-well polystyrene plate.In order to analyze the biofilm-forming ability,a qualitative and quantitative analysis was conduct by crystal violet staining assay.And the antimicrobial resistance of different biofilm-forming ability strains was compared including imipenem,amikacin,meropenem,cefepime,sulbactam cefoperazone,trimethoprim,levofloxacin,gentamicin,ciprofloxacin,cefotaxime,ceftizoxime,aztreonam,piperacillin,ceftriaxone,cefuroxime.In addition,the expressions of biofilm-related gene Bap,bfs and intI1 were tested with polymerase chain reaction (PCR) assay.Results Among 70 strains Acinetobacter baumannii,40 strains were multi-drug resistant (57.14%) and 6 strains were pan-drug resistant (8.57%); 68 strains had biofilm-forming ability (97.14%),14 of which were weakly positive,20 were positive and 34 were strongly positive.The antimicrobial resistant rate of Acinetobacter baumannii to imipenem,amikacin,meropenem and cefepime was decreased,it was 30.00%,32.86%,38.57% and 41.43%,respectively.However,the antimicrobial resistant rates to other commonly used antibiotics were all higher than 50%.The drug resistance of Acinetobacter baumannii to levofloxacin (85.71%,45.00%,38.24%,x2=9.225,P=0.010),cefepime (71.43%,45.00%,29.41%,x2=7.222,P=0.027),gentamicin (78.57%,55.00%,38.24%,x2 =6.601,P=0.037) was significantly decreased when biofilm-forming ability reinforced (weakly positive,positive,hadro-positive).Bap gene positive rate of weakly positive,positive and strong positive biofilm-forming strains Acinetobacter baumannii was 50.00%,65.00% and 79.41% (x2=4.244,P=0.120),respectively.Bfs gene positive rate was 35.71%,65.00% and 88.24%,respectively (x2=13.602,P=0.001) and intI1 gene positive rate was 42.86%,75.00% and 91.18%,respectively (x2 =12.902,P=0.002).Moreover,the antimicrobial resistances of biofilm-related gene positive strains were higher than the negative,of which the drug resistance of intI1 positive group to amikacin was significantly higher than the negative group (40.38% vs.11.11%,x 2=5.194,P=0.023).Conclusions The Acinetobacter baumannii collected from the hospital had strong multi-drug resistance as well as strong biofilm-forming ability.The drug resistance of Acinetobacter baumannii decreased when biofilm-forming ability reinforced.In addition,genes,such as Bap,bfs,and intI1,contributed to biofilm formation.
