1.Expressions of TNF-αand SOD in cardiac tissues of rats with abdominal aorta champing and LPS attack

Objective To observe the pathomorphological changes and expressions of TNF-α, SOD1 and SOD2 in cardiac tissues of rats with abdominal aorta champing(AAC)and LPS attack.Methods Forty Wistar rats were randomly divided into four groups:sham operated group (Group S),ischemia reperfusion with AAC group(Group I /R),intraperitoneal injection of endotoxin group(Group LPS)and AAC +LPS group(Group I /R +LPS).Animals were sacrificed after 20 minutes of AAC and 8 hours of reperfusion.Cardiac tissues were observed with HE staining for pathmmorphological changes,and detection of TNF-α,SOD1 and SOD2 expressions were tested by immunohistochemistry.Results The mortality rate of Group I /R +LPS was highest.HE staining of the cardiac tissues revealed that the pathological changes of myocardial tissue were not obvious in Group S;myocardial cells were disordered and slightly swollen and red blood cell and inflammatory cells were observed in the intramuscular space of the Group LPS;these injuries were more serious in the Group I /R;the injuries of cardiac tissues were most serious in Group I /R +LPS.The results of immunohistochemical staining revealed that TNF-αexpression level of cardiac tissue increased as followings:Group S,Group LPS,Group I /R and Group I /R +LPS.Significant differences were observed among groups,except for the difference between Group LPS and Group I /R.The expression level of SOD1 decreased as followings:Group S,Group LPS,Group I /R and Group I /R +LPS. Significant differences were observed among groups,except for the difference between Group I /R and Group I /R +LPS.The expression level of SOD2 decreased as followings:Group S,Group LPS,Group I /R +LPS and Group I /R.Significant differences were observed among groups,except for the differences between Group LPS and I /R and Group I /R +LPS.Conclusion The mortality in rats with sepsis after abdominal aorta clamping is high.Obvious myocardial morphological changes and injuries exist either in ischemia reperfusion or sepsis state.At the same time,the expression of TNF-αis up-regulated while SOD1 and SOD2 are down-regulated.
2.Relationship between the international prognosis indexes and the cell-mediated immunity condition in DLBCL patients

ObjectiveTo explore the relationship between the international prognosis indexes(IPT) and the cell-mediated immunity condition in DLBCL patients. Methods52 DLBCL patients were divided into 4 groups and the FCM was used to examine the T lymphocyte subsets,including CD3+、CD4+、CD8+、NK cells.T lymphocyte subsets absolute value and CD4+/CD8+ ratio were examined. Correlation with IPI were compared among groups.ResultsCD3+ cells in high risk group [(1570.9±370.5)/μl]were higher than other IPI groups;CD4+ cells in DLBCL groups were all lower than normal group(751.3±367.4)/μl]; CD8+ cells in high risk group [(1055.9±523.8)/μl] were higher than other groups; CD4+/CD8+ ratio in middle-high risk group and high risk group (1.0±0.2、0.7±1.0)were lower than other IPI groups and normal group;NK cells in middle-high risk group and high risk group were lower than the normal group[(199.5±68.4)/μl、(171.9±126.9)/μl];Age,clinical stage,body state had correlated with the CD3+ 、CD8+ cells and CD4+/CD8+ ratio of the DLBCL patients' peripheralblood T lymphosyte subsets. ConclusionsThe immunity condition in DLBCL patients has correlated with IPI; With increasing IPI value,the immunity depression and disorder become more serious,NK cells function become worse,and the prognosis is bad too.
3.The content difference of T lymphocyte subsets and natural killer cell in diffuse large B-cell lymphoma in different clinical stages and the analysis of their clinical significance

Objective To explore the content of peripheral blood T lymphocyte subsets and natural killer (NK) cells in diffuse large B-cell lymphoma (DLBCL) in different clinical stages and analyze their clinical significance.Methods Sixty-two DLBCL patients were divided into 3 groups according to clinical stages:stage Ⅱ group (21 cases),stage Ⅲ group (21 cases ),stage Ⅳ group (20 cases).Thirty healthy examiners were selected as control group.Two ml peripheral blood was taken from every person.Peripheral blood T lymphocyte subsets(CD3+,CD4+,CD8+) and NK cell count were detected by flow cytometry(FCM) and CD4+/CD8+ was calculated.Results CD3+ in stage Ⅱ and Ⅲ groups[(854.6 ± 276.9 ) × 106/L,(823.3 ± 211.5)× 106/L] were lower than those in control group[(1268.8 ±684.0) × 106/L] and stage Ⅳ group [(1332.7 ±571.2) × 106/L](P < 0.05).CD4+ in stage Ⅱ,Ⅲ,Ⅳ groups [(433.5 ± 240.7) × 106/L,(423.8 ± 234.8) ×106/L,(423.0 ± 143.8 ) × 106/L] were lower than those in control group [(751.3 ± 367.4) × 106/L] (P <0.05).CD8+ and CD4+/CD8+ in stageⅣ group [(861.2 ±634.1) × 106/L,0.5 ±0.3] were significantly higher than those in control group [(455.9 ± 334.6) × 106/L,2.1 ± 1.3],stage Ⅱ group [(390.6 ± 54.1 ) × 106/L,1.0 ±0.8] and stageⅢ group [(377.4 ±493) × 106/L,0.9 + 0.6](P<0.05).NK cell in stage Ⅳgroup was obviously lower than that in control group[(210.3 ± 122.0) × 106/L vs.(353.3 ± 284.7) × 106/L,P< 0.05].And the higher clinical stage was,the lower NK cell was.Conclusions With the clinical stage increasing,the immunity depression and disorder of DLBCL patients become more and more serious,the more tumor burden is,the worse NK cell activity is.
4.Th17 cells adoptive immunotherapy impacts tumor growth of diffuse large B-cell lymphoma-bearing mice

Objective To explore the impact of Th17 cells adoptive immunity on tumor growth of diffuse large B cell lymphoma (DLBCL)-bearing mice.Methods The CD4+ CD62L+ na(i)ve T cells purified by Mini MACS immunomagnetic beads were stimulated under the condition of TGF-β, IL-6, anti-IFN-γ, anti-IL-4, IL-23.IL-17 expression was detected by ELISA.SCID mice were inoculated with DLBCL cells line SUDHL-4 to estaldish the model of DLBCL-bearing mice, and then they received Th17 cells adoptive immunotherapy at the 0, 8 th or 18th day (3 groups in total, 5 mice pre-group).The tumor volume and the survival of tumor model were observed.Results Formation of tumor nodules in mice was observed at about the 8th day after the mice received the Th17 SUDHL-4 cells.The tumors volume of DLBCL-bearing mice that had received Th17 cells adoptive immunotherapy was obviously smaller than that of the mice had not (P < 0.05).The survival time of the tumor-bearing mice that had received Th17 cells adoptive immunotherapy was also longer than that of the tumor-bearing mice had not (P < 0.05).Conclusion Th17 cells adoptive immunotherapy displays an anti-tumor effect on DLBCL-bearing mice.
5.Transplantation of non-myeloablative double umbilical cord blood units for severe aplastic anemia in eight adult patients

Eight patients with severe aplastic anemia received unrelated cord blood transplantation at the Guangzhou First People’s Hospital from June 1998 to December 2007. The patients were conditioned with decreased dosage of immunosuppressive agents of cyclophosphamide and antilymphocytic globulin. The median infused donor total nucleated cell were 5.69?10 7/kg of recipient weight, and the CD34+ cell was 4.10?105/kg of recipient weight. Methotrexate and corticoid methylprednisolone were used for prophylaxis of graft versus host disease (GVHD). The time to reach an absolute neutrophil count of 0.5?109/L ranged from 7 to 25 days (median: 17 days) and the time to reach a platelet count of 20?109/L ranged from 13 to 102 days (median: 35 days) after transplantation. DNA finger print map of 7 patients showed donor and recipient chimera. Two patients developed grade I to II acute GVHD, which was controlled. One patients developed grade II chronic GVHD, which was controlled by using methylprednisolone. Five patients had lived for 10-108 months, with no diseases. Taken together, unrelated umbilical cord blood transplantation is effective for adult patients. Partial conditioning regiment could ensure engraft of unrelated umbilical cord blood transplantation.
6.Chemical constituents of mangrove plant Excoecaria agallocha in Hainan Province

Objective To study the chemical constituents from mangrove Excoecaria agallocha.Methods The chemical constituents were separated and purified by chromatographic methods after solventextraction and were identified by spectroscopic analyses(EI-MS,1D NMR).Results Twelve compoundswere isolated from this plant and identified as:14-taraxeren-3-one(Ⅰ),dibutyl phthalate(Ⅱ),?-amyrin(Ⅲ),18-oleanen-3-ol(Ⅳ),18-oleanen-3-one(Ⅴ),phaeophytin A(Ⅵ),betulin(Ⅶ),?-rosasterol(Ⅷ),?-sitosterol(Ⅸ),betulinic aicd(Ⅹ),oleanolic acid(Ⅺ),ursolic acid(Ⅻ).Conclusion CompoundsⅦ,Ⅷ,Ⅹ,andⅫare isolated from this plant for the first time.
7.Application of multi-sliced CT on breast cancer and early lymph node metastasis

Objective To investigate the application of multi-sliced CT on breast cancer and early lymph node metastasis (LNM). Methods 30 cases with infiltrative ductal carcinoma (IDC)confirmed by pathology were collected and set as Study Group,30 fibro-mas as Control Group.128-slice CT plain and 2 phrase enhance scan was undergone.Image signs of mass of two groups were com-pared.Image signs of early lymph node metastasis were analyzed.Results In MSCT images,IDC masses had irregular shape,burr rim.2-phrase MSCI demonstrated obvious enhancement in artery phrase,persistence enhancement in vein phrase and ring shape en-hancement.There were significant different between 2 groups in mass shape,rim and enhancement mode(P<0.05).MSCT demon-strated 22 LNM,but 6 of them had no swelling but obvious enhancement.Conclusion 2-phrase MSCT plays an important role in as-sessing breast cancer and early lymph node metastasis.
8.Electrochemical sensor for acetaminophen based on layer-by-layer self assembly technique.

A novel type of carbon nanotube-coated Au nanoparticle and [bmim]BF4 composite modified glassy carbon electrode was fabricated by a layer-by-layer self-assembly technique. The electrochemical performance of acetaminophen (ACOP) on the modified electrode was investigated by cyclic voltammetry. The Nafion/GNPs/RTIL/MWNTs/GC electrode showed an excellent electrocatalytic activity for the oxidation of ACOP and accelerated electron transfer between the electrode and ACOP. For ACOP, the reversible electrochemical process was observed on the Nafion/GNPs/RTIL/MWNTs/GC electrode, while irreversible electrochemical process occurred on the GC electrode. For the Nafion/GNPs/RTIL/MWNTs/GC electrode, the anodic peak potential of ACOP was moved from 0.562 V to 0.413 V, with a potential drop of 149 mV. At the same time, the reduction peak potential was 0.384 V, and the potential difference was only 29 mV. It was shown that the modified electrode possessed higher electrocatalytic activity and more sensitive effect for the detection of ACOP than both MWNTs/GC electrode and GC electrode. The effects of the different experimental conditions on the electrochemical behaviors of ACOP were explored. Under the optimum conditions of preparation and experimental, the linear calibration curves of ACOP were obtained in a wide range of 2 x 10(-1) to 4.0 x 10(-4) mol x L(-1) with a correlation coefficient 0.999 2 and a detection limit of 2.6 x 10(-8) mol x L(-1) (the ratio of signal to noise, 3:1). The recovery rate was 97.9%-100.8%. This method can be used to determine ACOP in paracetamol tablets with satisfactory results.
9.Genistein and daidzein promote cell proliferation and neurotrophin expression in hippocampal H19-7 cell line

Objective To determine the effects of genistein and daidzein on the proliferation and survival of the hippocampal neural cells and underlying mechanism. Methods H19-7/IGF-IR neural cell line was cultured in phenol red free DMEM absented of serum for 72h. Genistein, daidzein or 17β-estradiol was added to the culture at various concentrations. Their proliferation and protective effects on the neuronal cells were determined by BrdU and MTT assay respectively. The effect of phytoestrogens on cell cycle regulation was determined using flow cytometry. The effects of the soy isoflavones on brain-derived neurotrophic factor (BDNF) expression were determined by ELISA and RT-PCR respectively. Results It was observed that, with 72h of treatment, 20nM and 200 nM 17B-estradiol significantly promoted the neuronal cell proliferation at 33% and 36% ;20nM and 200nM genistein significantly promoted the neuronal cell proliferation at 15% and 13% ; 200nM daidzein significantly promoted the neuronal cell proliferation at 11% compared to the control (P<0.05). Genistein and daidzein induced an significantly increase in the S phase arrest at (17.64 ± 0.43) % and (19.48 ± 1.01) % compared to the control (P < 0. 05). Moreover, genistein and daidzein significantly increased the expression of mature BDNF and BDNF mRNA level (P<0.05). The effect of genistein and daidzein on hippocampal neuronal cell proliferation was blocked by K252a, selective inhibitor of tyrosine kinase receptors. Conclusion Genistein and daidzein improved hippocampal neuronal cell proliferation and viability in vitro. The effects might be mediated by increasing in BDNF expression.
10.Influencing factors of osteonecrosis of the femoral head after allogeneic hematopoietic stem cell transplantation in patients with leukemia

Objective To explore the influencing factors and possible mechanism of osteonecrosis of the femoral head(ONFH)in patients with leukemia after allogeneic hematopoietic stem cell transplantation (Allo-HSCT).Methods One hundred and two patients with leukemia who received Allo-HSCT between January 2003 and October 2007 were evaluated for ONFH and graft-versus-host disease(GvHD)within 2years after transplantation,and the dosage of methylprednisolone(MP)in every patient from the first diagnosis of leukemia to 2 years after Allo-HSCT was calculated.For patients who suffered acute GvHD(aGvHD) and chronic GvHD(cGvHD),the serum leveh of soluble ligand of receptor activator of nuclear factor kappa B (sRANKL)which was index for differentiation of osteoclast(OC),tartrate-resistant acid phosphatase-5b(TRAP-5b)and C-terminal telopeptide of collagen Ⅰ(CTP-Ⅰ)which both were indexes for metabolism of OC were detected by enzyme-linked immunosorbent assay in different stages of MP dosage.According to these results.possible factors for ONFH after Allo-HSCT were analyzed.Results Seven in 102 patients after Allo-HSCT had experienced ONFH within 2 years,the incidence was 6.9%(7/102),which was not related to age,gender,types of leukemia and donor.ONFH mainly developed in patients with aGvHD and cGvHD,and the incidence could be achieved to 21.9%(7/32)which Was much higher than that in patients with no GvHD,merely aGvHD or merely cGvHD(P<0.05).In patients with aGvHD and cGvHD,the average dosage of MP in ONFH(+)was(232.7±28.6)mg/kg which was extremely higher than that in ONFH(-)(115.1±16.9)mg/kg(P<0.05).The serum levels of sRANKL,TRAP-5b and CTP-Ⅰ were also higher when ONFH happened than ahead of pretreatment and 28 days after transplantation(-)(P<0.05),and they were closely related to the dosage of MP(correlation coefficient was 0.597,0.664 and 0.682 respectively,P<0.05).Conclusions After Allo-HSCT,ONFH is related to the dosage of MP in treatment of GvHD.MP may be responsible for enhancement of OC differentiation and metabolism in leukemia patients,and ultimately induced the onset of ONFH.
