1.Analysis of epileptic electroencephalogram based on multi-variable state space reconstruction

Based on the high-dimension chaos characteristics of electroencephalogram(EEG) and the analysis by the multi-variable state space reconstruction,better estimation of the correlative dimension can be obtained by dividing the brain into two parts and using eight EEG channels as the reconstruction samples.Lorenz system was firstly tested by request of the amount of data to test the feasibility of the algorithms.After Comparing the high-dimension data of the EEG of epileptic patients with the results of control subjects,it is indicated that the multi-variable state space reconstruction is applicable in short time noise-containing time sequences to obtain reliable results and can free the researchers from the hard choice of delay time and embedding dimension.
2.Depressive effect on proliferation of vascular smooth muscle cells by tetrandrine in hypertensive rats

To analyse the effect of tetrandrine(Tet) on proliferation of aortic vascular smooth muscle cells ( AVSMC), AVSMC were isolated and cultured from sham-operated rats(Sham), renovascular hypertensive rats〔RHR, 18 weeks after two kidney one clip(2K1C) operation〕, and Tet (50 mg*kg-1*d-1 po for 9 weeks from week 9 after 2K1C operation)treated RHR. The proliferation of AVSMC was detected by MTT method, and the DNA synthesis was evaluated by [3H]-thymidine incorporation. The results showed that ①The ultrastructure of aorta suggested that AVSMC in RHR had transferred from contractile phenotype to synthetic phenotype; ②Compared to Sham, AVSMC from RHR showed a higher proliferative property with a higher cell number and an increased growth rate stimulated by norepinephrine(NE) or angiotensinⅡ(AngⅡ); ③Compared to untreated RHR, AVSMC from Tet treated RHR showed a reduced reactivity to NE- or AngⅡ-stimulated proliferation and growth rate; ④Tet(0.1-10 μmol*L-1) treated in vitro induced a concentration-dependent depression in [3H] thymidine-incorporation stimulated by NE or AngⅡ in AVSMC from either RHR or Sham. This study provides an evidence of increased reactivity to NE or AngⅡ in AVSMC of RHR. Tet inhibits the proliferation and DNA synthesis in AVSMC, depresses the susceptibility of AVSMC to AngⅡ and NE, both contribute to the regression effect on hypertensive vascular remodeling.

4.Comprehensive Evaluation of Water Environment in the Eling Lake Area

Objective To know the situation of the water environment in the Eling Lake Area and provide the theoretical basis for protecting the biosystem of this area. Methods The water samples were collected from the sites 0.5 meters below the water surface of input and output water area, the water area near the east and west bank of the Eling Lake, the center of the lake, and also from the sites 0.5 meters above the bottom of the lake .The water sampling was carried out in August 2005, October 2005 and May 2006. Country standard method was used to determine the physical and chemical characteristics of water samples. The sampling, determination and evaluation in the investigation were according to related standard regulations. Results The water temperature was 5.6 ℃, pH value was 8.49, the total amount of Ca2+ and Mg2+, suspended matter, chlorides, sulfate, DO, COD, BOD5, Cu2+ and Zn2+ were 95.4 mg/L, 87.0 mg/L, 45.0 mg/L, 80.0 mg/L, 7.1mg/L, 11.0 mg/L, 2.0 mg/L, 0.002 mg/L and 0.009 mg/L respectively. Conclusion All of the physical and chemical characteristics of the water samples meet the water quality criterion of the class ruled by GB 3838-2002, and the water can be used as source water.
5.Cloning of Squalene Synthase Gene from Saccharomyces cerevisiae and Construction of Yeast Expression Vector

Objective To lay the foundation for studying the synthesis of artemisinin in microorganism,squalene synthase(SS) gene,a key enzyme gene from Saccharomyces cerevisiae,was cloned and a yeast expression vector was constructed.Methods After amplification of SS gene by polymerase chain reaction(PCR),ligation to T-vector and analysis of the cloned sequence,enzyme digestion and reconfirmation of the target gene,the antisense yeast expression vector was constructed by inverted insertion of the target gene into a yeast expression vector,pGAPZ?A,and digested with two restriction enzymes for vertifying the recombinant.Results The length of SS gene was 1335bp.The preliminary sequence data indicated that SS gene obtained from the experiment had a high sequence homology with that from GenBank,except for a few base pairs.The antisense yeast expression vector has been constructed and vertified by digesting with two enzymes.Conclusion SS gene from Saccharomyces cerevisiae has been successfully cloned and sequenced.An antisense yeast expression vector has been also constructed.
6.Effects of tetrandrine on hydroxyproline content and myosin ATPase activity of hypertrophied myocardium in renovascular hypertensive rats

To study the effects of tetrandrine (Tet) on hypertrophied myocardial hydroxyproline content and myosin ATPase activity, left ventricular hypertrophy(LVH) was induced by renovascular hypertension (two-kidney, one-clip) in rats. Eight weeks after operation Tet 50 mg*kg-1*d-1 and enalapril(Ena) 6 mg*kg-1*d-1 were given by gavage for 8 weeks. The results showed that hydroxyproline content in LVH group was much higher than that of sham-operated one 〔(5.9±0.3) vs (3.6±0.4) mg*g-1 dry weight〕, and was decreased by 28.2% and 39.0% in Tet and Ena groups, respectively. Myosin ATPase activity in LVH group was much lower than that of sham-operated group 〔(0.43±0.09) vs (0.97±0.06) mmol Pi*min-1*g-1 protein, P<0.01〕. In Tet and Ena groups they were 60.5% and 118.6% higher than that of LVH group, respectively. The results suggest that Tet or Ena partially reduce the hydroxyproline content and elevate myosin ATPase activity of hypertrophied myocardium in renovascular hypertensive rats.
7.Ultrasound-mediated microbubble destruction accompanied with cationic liposome enhanced gene transfection in vitro

Objective To investigate whether ultrasound-mediated microbubble destruction(UMD) could enhance cationic liposome (CL) induced plasmid DNA delivery or not,and optimize the transfection conditions.Methods Multiple parameters were explored to obtain the optimal transgene efficiency by means of with or without serum in culture medium,various CL or nano-liposomal bubble(NB) concentrations,different time point of ultrasonic irradiation.The transfection efficiency was assessed by fluorescence microscopy and flow cytometer,and cell viability was evaluated by Cell Counting Kit-8 (CCK-8) assay.Results The serum could protect the cells but show little impact on transfection efficiency induced by CL.CL and plasmid DNA at a weight ratio of 4 ∶ 1 exhibited high transfection efficiency of (17.71-± 0.79)% and high cell viability of (91.28 ± 0.76) %.CL combining with ultrasonic irradiation at the time point of 1 hour could increase the transfection efficiency to (24.85 ± 0.78)% (P <0.01).Higher transfection rate (32.47 ± 4.01) % was obtained by adding NB at the concentration of 10 % (P <0.05).Conclusions UMD accompanied with CL could enhance gene delivery effectively,which would provide a new method for gene therapy.
8.Enhanced gene transfection to HepG2 cells mediated by self-made cationic nanobubbles

Objective To observe self-made cationic nanobubbles as non-viral gene carrier to transfer green fluorescent protein reporter gene into HepG2 cell in vitro.Methods Cationic nanobubbles(PNB) were prepared by sonicating liposomes、polyethylenimine and perfluoropropan.The surface potential and the size of nanobubbles were assessed by laser particle analyzer.HepG2 cells were incubated with DNA,nanobubbles with or without ultrasound exposure.The transfection efficiency was evaluated by flow cytometer and the cell viability by cell counting Kit-8.Results The mean diameter of PNB was (834.57 ± 6.4) nm and the surface charge was (4.15± 0.98)mV.The PNB-DNA complexes,which blocked by the Agarose gel electrophoresis,could effectively transfer HepG2 cells,and the ultrasound exposure could enhance the transfection efficiency further significantly (P < 0.05).Conclusions The new PNB could effectively combine with pDNA to enhance gene delivery and ultrasound exposure could improve its efficiency further in HepG2 cell in vitro.
9.Upregulating the renin-angiotensin system in bone marrow mesenchymal stem cells by hypoxia

Objective The renin-angiotensin system ( RAS) is involved in myocardial anoxic injury .This study aimed to in-vestigate the expressions of AT 1-R, AT2-R, and angiotensin-converting enzyme ( ACE ) in bone marrow mesenchymal stem cells (MSCs) under hypoxia. Methods Rat MSCs were isolated, cultured, and identified with CD29 and CD11b/c antibodies.The is-chemic injury model was established by exposing the MSCs to hypoxia and serum deprivation ( Hypoxia/SD) for 24 hours, while the control cells were cultured in L-DMEM with 10%FBS.The vitality and apoptosis of the cells were detected by trypan blue staining , CCK8 assay, and Annexin V-FITC staining.The mRNA and protein expressions of AT 1-R, AT2-R, and ACE were determined by real-time quantitative PCR and Western blot , respectively. Results The positive rate of CD29 was >97%and that of CD11b/c was <1% in the MSCs.Compared with the control group, Hypoxia/SD significantly increased the rate of cell apoptosis ([6.73 ±0.78]%vs [19.93 ±4.92]%, P<0.01), decreased the rate of cell viability ([78.49 ±4.94]%vs [37.33 ±2.91]%, P<0.01), and up-regulated the mRNA and protein expressions of AT 1-R, AT2-R, and ACE. Conclusion Hypoxia/SD activates the RAS in MSCs and improves the protective function of the cells against myocardial anoxic injury .
10.Fundamental study on the characteristics and ultrasound contrast enhancing effect of a self-made nanoliposomal bubble

Objective To made a kind of nano-liposomal bubbles(NB) and observe its base physical properties and ultrasound contrast enhanced effect,and compared with SonoVue in vitro and in vivo.Methods The liposome was made with the method of reverse phase evaporation,and then the liposome was sonieated to prepare NB.After the physical properties of NB such as its morphous,average particle size,surface potential and concentration were observed and calculated,200 μl of saline and two contrast agents (NB and SonoVue) were injected into degassed PBS (10 ml) respectively to observe their enhancing effects.For in vivo study,a healthy rabbit heart and liver were imaged before and after intravenous injection of 2.0 ml/kg of saline and two contrast agents in succession to compare their dynamic enhancing effects in the grey scale imaging subjectively.Results The self-made NB distributed uniformly and its size ranged from 133.1 ~199.5 nm with a mean diameter of (171.60 ± 30.82) nm.Its surface potential and concentration were -(1.92± 0.65)mV and (3.8 ~ 5.6) × 108/ml individually.These basic characteristics were not observed changing dramatically after placement one week to one month under room temperature.The NB and SonoVue all displayed significantly enhancing effect comparing with saline no matter in vitro or in vivo.Conclusions The self-made NB are stable and distribute uniformly,which display the same contrast enhancing effect as SonoVue in vitro and in vivo.The self made NB would have a more potential use in ultrasound molecular imaging and gone or drug delivery.
