1.Influence on Proliferation and Invasion of Human Breast Carcinoma Cell Line MDA-MB-231 by Using MicroRNA Interference Technique to Suppress MTDH Expression

Objective To investigate the effects of RNA interference technique silencing MTDH on proliferation,metastasis and invasion of human breast cancer cell MDA-MB-231.Methods The MTDH miRNA was transfected into Human breast cancer cell MDA-MB-231 by means of lipofectamine 2000.The silencing efficiency of MTDH was examined by RT-PCR and western blot.The influence of MDA-MB-231 cell proliferation,metastasis and invasion ability from inhibition MTDH gene was conducted by using MTT assay,scratch experiments,and Transwell assay.Results MTDH miRNA effectively inhibited MTDH mRNA and protein levels.The best inhibition rate were 79.41% and 80.56% (P < 0.05).With miRNA interferring cells,the ability of cells proliferation was decreased (P < 0.05),and cells invasive and metastasis were depressed (P < 0.05).Conclusion MTDH expression is obviously suppressed after transfected by MTDH miRNA in MDA-MB-231 cells.The inhibition of MTDH expression from microRNA leads to significant suppression of proliferation,invasion and metastasis of breast cancer cell MDA-MB-231.
2.Inhibitory Effect of Octritide Combined 5-Fluorouracil on the Proliferation of Human Colonic Cancer Cell Line SW480

OBJECTIVE:To explore the inhibitory effects of octritide alone or in combination with 5-fluorouracil on the proliferation of human colonic cancer cell line SW480.METHODS:MTT assay was used to detect the absorbance after treatment with octreotide at different concentrations(10-7~10-10 mol?L-1) alone or in combination with 5-fluorouracil on the proliferation of SW480 cells,then the inhibition ratio on SW480 cells was derived.RESULTS:At a concentration of 10-10 mol?L-1,octreotide exhibited a highest inhibition ratio on colonic cancer cell line SW480,at a concentration lower than 10-12 mol?L-1,octreotide had no inhibitory effect on the growth of colonic cancer cell line SW480.The antiproliferative effect of octritide and 5-fluorouracil on SW480 cells was significantly stronger when used in combination than used alone(P

Aim Toexplorewhether1-methylhydan-toin(MH)could inhibit the basal secretion of growth hormone (GH ) and suppress the promoting effect of growth hormone-releasing hormone (GHRH ) in rab-bits.Methods Thirty-sixrabbitswererandomlydi-vided into six experimental groups according to the kind of dosing drugs,namely normal saline group(A), MH group (B ),octreotide group (C ),GHRH group (D),GHRH +MH group(E),GHRH +octreotide group(F),with 6 rabbits in each group.Blood was sampled (1. 0 mL each time)from each rabbit before injecting drugs and 5,15,30,45,60 min after drug administration.Clotting spontaneously,rabbits blood samples were centrifugated for 20 minutes at approxi-mately 1000 ×g and the supernatant was collected. Serum GH concentrations were determined by enzyme linked immunosorbent assay kit(ELISA Kit).Mean-while,the behavior of rabbits in each group after injec-tingdrugswascloselyobserved.Results TheGH level of rabbits in group A at each time point had no significant differences(P>0. 05 ).Group B and group C rabbit GH levels were significantly lower than those of group A (P<0. 05 ),while GH levels in group D were obviously higher than those of group A (P <0. 05 ).Compared with group D,rabbit GH levels in group E and group F decreased markedly(P<0. 05 ). No obvious toxic and side effects had been observed within one week after the experimental rabbits were ad-ministered corresponding drugs by intravenous injec-tion.Conclusions 1-methylhydantoincouldinhibit the basal secretion of GH in rabbits.1-methylhydan-toin could suppress the promoting effect of GHRH in rabbits.

Objective To investigate the expression of BRMS1 in different metastatic breast cancer cells and its relation to HDAC activity.Methods The gene expression of BRMS1 in 3 types breast cancer cell lines(non-metastasis MCF-7(A cell line),low metastasis MDA-MB-453(B cell line)and high metastasis MDA-MB-231(C cell line)) was determied by RT-PCR technique;the protein expression of BRMS1 was measured with Western blot technique and HDAC activity by enzyme linked immunosorbent assay.Results(1) Gene expressive ratio of BRMS1 was 3.1∶2.0∶1.0 in the A、B、C cell lines,respectively.The gene expression of BRMS1 in the C cell line decreased 210% compared to A cell line,and expression of BRMS1 was markedly reduced as the degree of metastasis increased(P

The information and knowledge are generally hidden, unknown, but potentially useful, which can be expressed as concepts, rules, laws, modes or other forms. This article introduces the background and concepts of data mining, and reviews its application in medicine and the diagnosis of breast cancer for the most recent years. We also discuss the significance and current problems of this application in the diagnosis of breast cancer.
