1.Effects of the zinc finger protein A20 on LPS-TLR4 signaling pathways in severe acute pancreatitis rats

Objective To investigate the effects of the zinc finger protein A20 on LPS-TLR4 signaling pathways in severe acute pancreatitis(SAP) rats.Methods 24 SD rats were randomly divided into 3 groups:group A(the sham operation group),group B (the SAP group),group C (the SAP group treated with LPS).SAP model was induced by retro-injection of intraductal 5% sodium taurocholate into the biliary-pancreatic duct as previously described.The protein expression of A20,TLR4,NF-κBp65 and p38MAPK in pancreatic tissues was evaluated by immunohistochemistry.Results The positive area of A20 in pancreatic tissues was decreased in group B and group C compared with that in group A (t =17.234,19.698,all P < 0.05).On the contrary,the expression of TLR4,NF-κBp65 and p38MAPK in pancreatic tissues were up-regulated(t =15.909,20.432,16.543,18.629,22.105,19.006,all P < 0.05).A20 was decreased in group C than that in group B (t =14.894,P < 0.05),while TLR4、NF-κBp65 and p38MAPK were increased in group C than those in group B (t =14.047,15.582,17.070,all P <0.05).Conclusion The expression of A20 reduced and TLR4,NF-κBp65 and p38MAPK enhanced in the pancreas of rats with SAP,which indicated that A20 inhibited LPS-TLR4 signaling pathways which play important roles in the pathogenesis of SAP.

3.Usage and efficacy of timolol maleate eye drops in treatment of superficial infantile hemangioma

Objective:To determine drug dose and usage of timolol maleate eye drops in the treatment of superficial infantile hemangioma.Methods:A total of 250 superficial hemangioma infants were recruited and assigned into 5 groups (n=50 for each group):an external application group and 4 exterior coating groups (2,4,6,8 times per day).We evaluated the therapeutic effect of different methods for drug application (external application or exterior coating) and the frequency for drug administration on superficial infantile hemangioma.Results:The external application group (twice a day and 0.5 hour per time) showed better effect than that in the exterior coating group with twice a day (P<0.001).The difference in therapeutic effect between the exterior coating group with 6 times a day and exterior coating group with twice a day or with 3 times a day was significant (P<0.001).The differences in drug efficacy were not found among the exterior coating group with 6 times a day,the exterior coating group with 8 times a day,or the external application group with twice a day (All P>0.05).Conclusion:Drug dose may affect the therapeutic effect oftimolol maleate eye drops in superficial hemangioma infants,and exterior coating with 6 times a day may achieve the best curative effect.
4.Association of genetic variants in urotensin Ⅱ gene with polycystic ovary syndrome

Objective To investigate the correlation of single nucleotide polymorphisms( SNP) of rs228648 and rs2890565 in urotensin Ⅱ (UTS2)gene with polycystic ovary syndrome(PCOS). Methods The genotypes of two SNPs in UTS2 gene were determined through polymerase chain reaction Tin-shift genotyping method. 101 Chinese Han families trios in Shandong Province consisting of fathers,mothers,and affected daughters with PCOS and 105 healthy women were recruited. The physiological and biochemical parameters including serum follicular stimulating hormone(FSH),luteinizing hormone( LH),testosterone,fasting plasma glucose,and insulin were also measured. Results The distributions of three genotypes and two alleles of SNP rs228648 in UTS2 gene were similar between PCOS group and control group. The distributions of genotypes of SNP rs2890565 in UTS2 gene differed significantly between PCOS group and control group (P<0. 05). The frequency of A allele of SNP rs2890565 in UTS2 gene was significantly higher in PCOS group than that in control group( P<0.05). Transmission disequilibrium test(TDT) did not show significantly biased transmission of two different alleles from parents to affected daughters at rs228648 locus (P>0. 05) ,but showed an excess transmission of A allele from heterozygous parents to affected offspring at rs2890565 locus (P<0. 05). The carriers with GG genotype of SNP rs228648 had significantly higher HOMA-IR compared to those with AA and AG genotypes. The carriers with AA or AG genotype of SNP rs2890565 had significantly higher fasting plasma glucose and fasting insulin compared to those with GG genotypes. HOMA-IR in cases with AA genotype was significantly higher than that with GG genotype. Conclusions There is no association of SNP rs228648 with PCOS, but exists association with insulin resistance. The genetic polymorphism of UTS2 gene rs2890565 may be associated with PCOS,and the higher frequency of A allele is likely to contribute to PCOS.
5.Experience of application value of collaborative seamless whole-term care model in patients with endoscopic mucosal resection of gastric polyps

Objective To evaluate the application value of the collaborative seamless whole-term care model in patients with endoscopic mucosal resection of gastric polyps. Methods 80 patients with endoscopic mucosal resection of gastric polyps who were treated and given nursing intervention in the hospital from March 2015 to September 2017 were used as the analysis data of this article. The patients were grouped according to different nursing interventions, with 40 cases were in each group. The experimental group was treated with collaborative seamless whole-term care model. And the reference group was treated with general care. The SAS scores, SDS scores, postoperative complication rate and nursing satisfaction total rate between two groups were calculated before and after the intervention. Results There were significant differences in the SAS scores, SDS scores, postoperative complication rate and nursing satisfaction total rate between the experimental group and the reference group after intervention(P<0. 05). Conclusion Collaborative seamless whole-term care model in patients with endoscopic mucosal resection of gastric polyps exhibits good results.
6.Review and progress of drug therapy for infantile hemangioma of skin and soft tissue

This article summarizes the pharmacological treatment of infantile hemangioma in soft tissue.We aim to review the literature and summarize the mechanisms,methods,efficiency and adverse reactions of beta blockers,glucocorticoids,bleomycin and imiquimott,and to analyze the rational options.Beta blockers,as oral and external use,have high efficiency and few side effects,and can be used as a first -line medication for infantile hemangiomas.Imiquimod external use and topical glucocorticoid and bleomycin injection have good efficacy and potential adverse reactions.They can be used as an alternative treatment for infantile hemangioma.
7.Progress in animal models of cell lines and infantile hemangioma

Infantile hemangioma is the most common benign tumors in infancy with predictable disease evolution and duration. At present, the basic understanding of the development, proliferation and regression of infantile hemangioma is not sufficient, an ideal lively human infantile hemangioma model which could provide a standard platform for observation of cellular, molecular basis of infantile hemangioma development and drug mechanism is urgently needed. Up to now, a model that completely mimics the biological behavior of infantile hemangiomas has not been successfully established, but the existing research progress has laid a solid foundation for the pathogenesis of hemangioma and clinical targeted therapy. In this paper, the research progress of cell line and animal model of infantile hemangioma and its clinical value in recent years were summarized, as well as the favorable factors that may be used in the construction of infant hemangioma model in the future, providing the latest reference for the selection of animal model of infantile hemangioma.
8.Role of Transforming Growth Factorβ1 in Improving Renal Pathological Lesions of Rats with Early Chronic Kidney Disease with Danhong

Objective:To explore the efficacy of traditional chinese medicine Danhong on renal pathological lesions of rats with early chronic kidney disease (CKD) and the role of transforming growth factorβ1(TGF‐β1) in it .Methods:Thirty male SD rats were divided into three groups :the early CKD group (model group) ,the sham‐operated group (control group) and Danhong treated group (treated group) ,with 10 in each group .Rats in the three groups were treated by uninephrectomy for building up early CKD model ,sham‐operation (separating tissues to renal capsule without kidney excision) ,and Danhong treatment after building up model ,respectively .Renal pathological lesions ,24 h urine protein excretion and renal function were observed .The concentration of TGF‐β1 in renal cortex and overall expression of TGF‐β1 in renal tissue was examined by enzyme‐linked immune‐sorbent assay (ELISA) and immunohistochemistry ,respectively .Results:Mild to moderate proliferation of glomeruli mesangial cells accompanied by broadened mesangium matrix ,glomeruli capsule wall adhesion and change of capillary wall , interstitial fibrosis and inflammatory monocytes infiltration was found in the model group other than the control group .Renal lesions were alleviated dramatically in treated group ,while compared to that in model group .The gross renal pathological score in model group was higher than that in control group (P<0 .05) ,however ,the score in treated group was lower than that in model group (P<0 .05) .The 24 h urinary protein excretion in model group and treated group was higher than that in control group (P<0 .01) ,while the 24 h urinary protein excretion in treated group was lower than that in model group (P<0 .01) . There was no significant difference among the three groups regarding blood creatinine level and urea level .Serum CysC levels in model group and treated group increased significantly than that in control group (P<0 .05) .The concentration of TGF‐β1 in renal cortex and overall expression of TGF‐β1 in renal tissue were higher in model group than those in control group(P<0 .05) ,while those in treated group were lower than those in model group (P<0 .05) .TGF‐β1 expression level in renal tissues was significantly associated with both the urinary protein excretion and the total renal pathological scores ( P< 0 .01 ) . Conclusions:Danhong alleviated renal pathological lesions by down‐regulating TGF‐β1 expression .

10.Comparative analysis of gut microbiota of Chinese Kunming dog, German Shepherd dog, and Belgian Malinois dog

Objective:
This study examined the gut bacterial communities of dogs from different breeds, all kept under identical domestication conditions.
Methods:
Noninvasive sampling and 16S rRNA high-throughput sequencing were used to compare the composition and function of the gut microbiota of three dog breeds: the Chinese Kunming dog (CKD), German Shepherd dog (GSD), and Belgian Malinois dog (BMD).
Results:
The gut microbiota of the three dog breeds consisted of 257 species across 146 genera, 60 families, 35 orders, 15 classes, and 10 phyla. The dominant bacterial phyla across the three breeds were Firmicutes (57.44%), Fusobacteriota (28.86%), and Bacteroidota (7.63%), while the dominant bacterial genera across the three breeds were Peptostreptococcus (21.08%), Fusobacterium (18.50%), Lactobacillus (12.37%), and Cetobacter (10.29%). Further analysis revealed significant differences in the intestinal flora of the three breeds at the phylum and genus levels. The intestinal flora of BMD was significantly richer than that of CKD and GSD. The functional prediction and Kyoto Encyclopedia of Genes and Genomes analysis showed that the primary functions of the gut microbiota in these breeds were similar, with significant enrichment in various metabolic pathways, including carbohydrate and amino acid metabolism, secondary metabolite biosynthesis, and microbial metabolism in different environments. The intestinal flora of these breeds also played a crucial role in genetic information processing, including transcription, translation, replication, and material transport.
Conclusions
and Relevance: These results provide novel insights into the intestinal flora of intervention dogs and suggest novel methods to improve their health status, which help increase microbial diversity and normalize metabolite production in diseased dogs.
