1.Progress of Molecular Ecological Research on Marine Anammox Bacteria

Anammox bacteria can perform anaerobic ammonium oxidation,a long missing process which contributes 30%~50% to dinitrogen gas in marine nitrogen cycling.The potential role of anam-mox bacteria coupling with ammonium oxidizing bacteria and archaea will benefit to elaborate the complex mechanism of marine nitrogen cycling.Furthermore,the unique cell and genomic characteris-tics make anammox bacteria an important model microorganism to explore the bacterial evolution.Here we reviewed the current status of molecular ecology of marine anammox bacteria and give a perspective into the future based on our understanding of the literature and our own work.
2.Expression of ER, PR, HER-2, Ki-67 before and after neoadjuvant chemotherapy and the relation between their expression and the curative effects of neoadjuvant chemotherapy in breast cancer

ObjectiveTo investigate the relationship between the expression of ER,PR,HER-2,Ki-67 and the effective rate of neoadjuvant chemotherapy (NAC) and to study the influence of NAC on expression of ER,PR,HER-2 and Ki-67 in breast cancer.Methods41 patients of breast cancer were performed 2 to 6 cycles of CAF chemotherapy.The expression of ER,PR,HER-2 and Ki-67 was detected by SP immunohistochemical method before and after NAC.ResultsThe effective rate of NAC was 73.17% (30/41).The positive and negative efficiency rate was 67.86% (19/28) vs 84.62% ( 11/13 ) for ER,64.29% (9/14) vs 77.78% (21/27 ) for HER-2,70.00% (14/20) vs 76.19% ( 16/21 ) for Ki-67.The difference had no statistical significance (P >0.05).The positive and negative efficiency rate was 66.67% (16/24) vs 82.35% (14/17) for PR.The difference had statistical significance (P < 0.05 ).The expression of ER,PR,HER-2 and Ki-67 before and after NAC changed in 10,8,3 and 9 cases respectively.The difference had no statistical significance (P > 0.05).ConclusionsNAC has no influence on the expression of ER,PR,HER-2 and Ki-67.Patients with PR negative are more sensitive to NAC.The expression state of PR can be used to predict the curative effect of NAC.The expression state of ER,HER-2 and Ki-67 has no significant correlation with NAC.
3.Correlation between Serum Levels of Melatonin TNF-A and EDSS Scores in Multiple Sclerosis Patients

Objective To study the serum levels of melatonin,tumor necrosis factor alpha (TNF-α) and interferon gamma (IFN-γ) in multiple sclerosis (MS) patients and the correlation with disability.Methods Forty patients with multiple sclerosis were collected as MS group and 30 healthy participants were collected as control group.Serum levels of melatonin and cytokines,including IFN-γ and TNF-,were detected in all participants by the enzyme-linked immunosorbent assay (ELISA) method;disability status of patients with MS was evaluated by EDSS scale.The relevant analysis between serum melatonin,TNF-α,IFN-γ levels and EDSS score in patients with MS was conducted.Results The concentration of serum melatonin in MS group was significantly lower than control group (P<0.01).TNF-α levels were higher than control group (P<0.05) and the difference was statistically significant between MS patients and control group.Among MS group and control group,no significant correlation with the serum levels of IFN-γ was seen.The serum melatonin level was inversely correlated with EDSS score in MS patients (r =-0.76,P<0.01),and positively correlated with TNF-α (r =0.83,P<0.01) and as compared to IFN-γ,no significant correlation was found (r =0.17,P>0.05).Conclusion The decrease of melatonin and increase of TNF-α can be a factor in the inflammatory reaction in patients with MS,and is closely related with dysfunction occurring in multiple sclerosis.Serum melatonin and TNF-α can be used as laboratory indicators to monitor clinical curative effect and evaluate prognosis of MS.
4.Mechanism of Polysaccharides from Two Spleen-invigorating Chinese Medicine Prescriptions in Regulating Growth of Bacteroides fragilis in vitro: Based on Differential Expression of Polysaccharide Utilization Loci

ObjectiveTo investigate the effects of using the polysaccharides from two Chinese medicine compound prescriptions as the carbon source on the growth of Bacteroides fragilis and to decipher the mechanism from the perspective of differential expression of polysaccharide utilization loci (PULs) based on transcriptomics. MethodThe media with different carbon sources [20% polysaccharides of Lizhongtang, polysaccharides of Shenling Baizhusan, glucose, and brain heart infusion (BHI) Broth] were used for the anaerobic culture of B. fragile ATCC25285. The effects of different carbon sources on the growth of B. fragilis ATCC25285 were determined by continuous sampling and spectrophotometry. Then, transcriptome sequencing was performed for the cultures obtained with different carbon sources to study the mechanism of different carbon sources in regulating bacterial growth. ResultThe concentration of bacteria with the polysaccharide of Lizhongtang, polysaccharide of Shenling Baizhusan, BHI Broth, and glucose as the carbon sources peaked at 26, 32, 26, 38 h, respectively, and the bacteria in all the four groups achieved robust growth. Gene ontology (GO) enrichment indicated that the differentially expressed genes in the Lizhongtang polysaccharide group and Shenling Baizhusan polysaccharide group were concentrated in the transport and transmembrane transport of dicarboxylic acid. The Shenling Baizhusan polysaccharide and BHI Broth groups showed high expression of PUL 4 and 27, glycoside hydrolase 13 (GH13), and glycosyl transferases 5 (GT5). PUL9 was highly expressed in Shenling Baizhusan polysaccharide group, and PUL 17, 19, and 20, GH3, and GH144 in the BHI Broth group. PUL27 and GT5 were highly expressed in Shenling Baizhusan polysaccharide and glucose groups. PUL 4 and 9 and GH13 were only highly expressed in Shenling Baizhusan polysaccharide group, and PUL 2, 17, and 19 and GH2 in the glucose group. Both Lizhongtang polysaccharide group and BHI group highly expressed PUL 4, 17, 19, 20, and 27, GH3, and GH144. PUL 2, 8, 23, and 27, GH2, and GH57 were highly expressed in Lizhongtang polysaccharide group, while GH13 showed high expression in the BHI group. Both the glucose and Lizhongtang polysacharride groups showed high expression of PUL 4 and 27 and GH2. PUL 4, 8, 20, and 23, GH3, and GH144 were highly expressed in Lizhongtang polysaccharide group, while PUL30 was highly expressed in the glucose group. ConclusionThe in vitro experiments and transcriptome sequencing results confirmed that the expression of PULs and GH may provide benefits or costs to the adaptive growth of Bacteroides fragilis ATCC25285 cultured with different carbon sources, which may be one of the mechanisms by which polysaccharides from Chinese medicine compound prescriptions regulate the growth of B. fragilis ATCC25285. The findings can provide a reference for further research on the relationship between B. fragilis metabolic pathway and polysaccharides of Chinese medicine compound prescriptions.
