Apoptosis is a spontaneous process of cell-suicide process triggered in response to physiological and pathological stress stimul, which is to regulate the developmental of body, control cell senescence, maintain a stable internal enviroment in multicellular organisms. The initial and progress of apoptosis is precisely controlled, which is a unique and complex signal system. In this review, it introduced the relationship between apoptosis and bcl-2 gene family, C-myc, tumor suppressor gene p53, Caspase protease family and Fas, and Summaries the detection methods.
2.Curcumin inhibits telomerase activity of HeLa cells

Objective To investigate the inhibitory effect of curcumin on the telomerase activity of HeLa cells.Methods The growth inhibition rate of HeLa cells treated with curcumin was measured with MTT method.The expression of hTERT mRNA was studied using RT-PCR.Telomerse activity was examined by TRAP-ELISA.Results The growth of HeLa cells treated with curcumin was inhibited in concentration dependent manner and the expression of hTERT mRNA and the level of telomerase activity were down-regulated significantly.Conclusion Curcumin can inhibit telomerase activity of HeLa cells.The underlying mechanism might be related to the down-regulation of hTERT transcription.
3.Overexpression of caveolin-1 inhibits the growth of human cervical squamous cell Hela cell line in vitro

Objective To investigate the effects of caveolin-1 overexpressing on the growth of cervical squamous cell cancer Hela cell line. Methods Eukaryotic expression vector of human caveolin-1 gene was introduced into Hela cells by Lipofectamine. The clones stably overexpressed caveolin-1 were identiffed by RT-PCR, immunofluorescence cell staining techniques and Westernblotting. Cells proliferation viabihty was tested by MTT assay, and flow cytometry was used to assay the cell cycle and apoptosis, and the relative phosphorylation level of extracellular regulated protein kinases (Erk1/2) were detected by Westernblotting. Results The clones stably overexpressed caveolin-1 were obtained. Compared with the parental Hela cells, the tranfected cells exhibited a slower rate of growth. FAGS analysis results revealed that overexpression of caveolin-1 resulted in the cell cycle arrest in the G0/G1 [ ( 68. 04 ± 2. 57 ) % vs ( 53.41 ±1.01)%] phase and increased the apoptotic cell fraction[ (19. 18 ±2.20)% vs (5.63 ±0.55)%, P <0. 05 ]. Western blotting results showed that overexpression of caveolin-1 reduced the phosphorylation of Erk1/2(0.28 ±0.05 vs 0.81 ±0.07, P <0.05). Conclusions Overexpression of caveolin-1 suppressed the growth of Hela cells and induced apoptosis, down-regulation of Erk1/2 phosphorylation might be involved in its mechanism.
4.Proteomics analysis of the apoptotic effects of hepatitis B virus large envelope protein on HepG2 cells

Objective To assess the effects of hepatitis B virus(HBV) large envelope protein (LHB) on the apoptosis of HepG2 cells and explore the possible mechanism by proteomic approaches.Methods LHB gene was cloned into pShuttle-IRES-hrGFP-1,and the recombinant adenovirus either barboring LHB(Ad-LHB) or empty vector(Ad-GFP) were separately generated.Annexin V-FITC apoptosis detection kit,JC-1 mitochondrial membrane potential assay kit and propidium iodide(PI) staining kit were employed combined with flow cytometry to detect the apoptotic cells infected with Ad-LHB or control of Ad-GFP.The cellular proteins were collected after infection of HepG2 cells by Ad-LHBs or Ad-GFP,and a total of 600 μg proteins were submitted to two-dimensional gel electrophoresis(2-DE) and stained with R350.The gel images were captured by ImageScanner Ⅱ Imaging System,the differentially expressed proteins were identified by ImageMaster 2D Platinum analysis software and picked up by Ettan Spot Picker.After enzyme digestion,the protein samples were analyzed by MALDI-TOF-TOF MS.Results HepG2 cells infected with Ad-LHB were much more prone to apoptosis.There were thirty nine differentially expressed proteins were determined by 2-DE between HepG2 cells infected with Ad-LHB and Ad-GFP,and they were identified ultimately and categorized into thirty three kinds of proteins by MALDI-TOF-TOF MS.Among these proteins,nine were found to be closely related to cell apoptosis,in which CAPN2,eIF3K and PPP2CB were higher expressed in Ad-LHB infected HepG2 cells,and SERPINH1,LASP1,PRDX1,DHRS2,LDHA and PS-MA4 were lower expressed in Ad-LHB infected HepG2 cells.Conclusion LHB could induce apoptosis of HepG2 cells,and several apoptosis-related proteins participated in this process.
5.Application effect of evidence-based nursing and individual nursing in pregnancy patients with lupus nephritis

Objective To investigate the application effect of evidence-based nursing and individual nurs-ing in pregnancy patients with lupus nephritis. Methods Sixty-five patients were randomly divided into control group and observation group. They were treated with routine care and evidence-based nursing respectively. The complications, pregnancy outcome and satisfaction of patients were compared between the two groups. Results There were not significant difference in renal failure , pregnancy induced hypertension , ecalampsia , heart failure , HELLP syndrome and other complications between observation group and normal group (P > 0.05). The pregnan-cy outcome had no significant difference between two groups (P > 0.05). In observation group, the health score was significantly higher than that of control group(P < 0.05). The mental health score in control group was sig-nificantly higher than that of observation group (P < 0.05). There werenot significant difference in social health and overall health score between the two groups (P > 0.05). Conclusion The application of evidence-based nursing and individual nursing in pregnancy patients with LN not only reduce the incidence of complications , but also reduce the chance of premature birth, stillbirth. And it could improve the cognition on the nursing work of this disease and the satisfaction of patients. But individual nursing is better in improving the patient′s mental health, while the evidence-based nursing is better in improving the health status.
6.Biocompatibility of a magnesium-zinc alloy implanted in rat cecum

BACKGROUND: Magnesium alloy studies in orthopedic field have been carried out,and good biocompatibility has been reported.However magnesium alloys have not yet been researched in the intestine.OBJECTIVE: The biodegradable magnesium-zinc alloy samples are implanted around the rat cecum to investigate the biocompatibility in rat.METHODS: Sprague Dawley rats were randomly divided into magnesium alloy group,medical titanium group and the sham-operated group.Then magnesium-zinc alloy samples with the dimension of 5 mm × 1 mm× 1 mm were embedded in the cecum incision in the magnesium alloy group.The medical titanium was embedded in the medical titanium group,and just suture in the sham-operated group.Prior to surgery and at 7,14,21 and 28 days after operation,the serum glutamic-oxaloacetic transaminase,creatinine and magnesium ion concentration were examined in each group.X-ray film on implanted region.The pathological changes in liver,kidney and cecum were examined.RESULTS AND CONCLUSION: There were no significant differences in serum glutamic-oxaloacetic transaminase,creatinine and magnesium ion concentrations among each group(P > 0.05).Magnesium-zinc alloy degraded gradually during 28 days.The pathology of liver,kidney and cecum was normal.Results suggested that magnesium-zinc alloy had no obvious effect on the cecum.The degradation time to play a fixed function of time was longer than the intestinal healing time,with good biocompatibility.Magnesium-zinc alloy can be used as anastomotic nail for stomach intestine anastomat.
7.Hospital administrators’recognition extent to healthcare-associated infec-tion management staff ’s competency and qualification

Objective To realize hospital administrators ’recognition extent to healthcare-associated infection (HAI )management staff ’s competency and qualification.Methods The unified questionnaires were filled in by respondents in 173 hospitals of Inner Mongolia autonomous Region,administrators’recognition on the competency and qualification of HAI management staff were surveyed.Results Of 445 hospital administrators,20.09%, 19.89%,and 18.05% considered that staff members in HAI management departments should have the knowledge background of preventive medicine,clinical medicine,and nursing respectively.58.20%,89.44%,and 43.37% of hospital administrators considered that the directors of HAI management departments should have senior profession-al titles,undergraduate course or above,and with 2 -5 working experience,respectively.34.92% and 30.93%considered that the most important professional ability of directors of HAI management department were profession-al and management ability,respectively.Conclusion Hospital administrators are apt to consider that the competen-cy and qualification of HAI management staff are strong professional ability and certain management ability,and are interdisciplinary talents.
8.Study of Cinnamic aldehyde effects on expression of E-cadherin and MMP-9 through SHH signaling pathway in lung adenocarcinoma A549 cells

Objective:To investigate Cinnamic aldehyde effects on expression of E-cadherin and MMP-9 and proliferation of lung adenocarcinoma A549 cells,and explore the possible mechanism of Sonic Hedgehog (SHH) signaling transduction.Methods:After co-cultured with Cinnamic aldehyde at the concentration of 0,10,20 and 40 μg/ml for 24 h,48 h and 72 h respectively,A549 cells were tested for their proliferation by MTT assay;E-cadherin and MMP-9 level in the supernatant by ELISA;expression of E-cadherin and MMP-9 mRNA by realtime-PCR with SYBR GreenⅠ;and protein expression by Western blot.Results: ①Cinnamic adehyde with concentration at 10 μg/ml would inhibited proliferation of A 549 cells after 24 hours′treatment;with concentration at 10, 20 and 40μg/ml can affect the proliferation significantly ( P<0.05 );with concentration of 40μg/ml cinnamic adehyde for 72 h,the re-markably inhibition of proliferation in A 549 cells was observed , the highest inhibitory rate was ( 93.782 ±5.036 )%.②Cinnamic aldehyde also increased migration rate of A 549 cells.③Expression of components on Hedgehog signaling pathway in A 549 was higher than that in human HBE cells.Cinnamic aldehyde could increase further upregulate of components expression in Hedgehog signaling pathway of A549 cells.④Secretion level of E-cadherin,mRNA and protein were decreased in A549 cells co-cultured with Cinnamic al-dehyde,while secretion level of MMP-9,mRNA and protein level in A549 cells co-cultured with cinnamic aldehyde were increased.Pre-treatment with 2 nmol/ml cyclopamine,an increasing of secretion level of E-cadherin ,mRNA and protein level in A549 cells was observed,decreasing of secretion level of E-cadhein,mRNA and protein level was also observed in A 549 cells.Conclusion:Cinnamic aldehyde inhibits the proliferation in a time-and dose-dependent manner and effected expression of E-cadherin and MMP-9 through sonic hedgehog signaling pathway in lung adenocarcinoma A 549 cells.
9.Cloning, Expression and Purification of the Major Surface Antigen P30 of Toxoplasma gondii

Objective To obtain protein of the major surface antigen P30 of Toxoplasma gondii by molecularcloning. Methods The gene of P30 (containing the whole P30 gene sequence, without the gene encoding signalpeptide) was obtained by polymerase chain reaction (PCR) using the primer designed according to the DNA sequence ofP30. The recombinant plasmid was constructed using EcoR Ⅰ, Xho Ⅰ and was then transformed into E. coli Top10. Thepositive clones were identified by restriction enzymes and DNA sequence analysis. The fusion protein was induced by IPTGand purified by affinity chromatography using ProBond~(TM) Resin (a kind of nickel-charged sepharose resin) and was identi-fied by SDS-PAGE and Western blotting. Results The products of PCR, cleavage and link reaction were same as ex-pected and the sequence of inserted fragment in the recombinant plasmid was same as reported except one synonymy muta-tion. A 58 kDa fusion protein was induced by IPTG and was purified by chromatography. Conclusion Fusion proteincontaining Toxoplasma gondii P30 was achieved and was provided as experiment material for further research.

Objective To investigate the relationships between the expression of VEGF,its receptor KDR/flk-1,cNOS mRNA and angiogenesis,cell proliferation,metastasis in human hepatocellular carcinoma(HCC).
