1.Determination of Peoniflorin in Bazhen(Eight Ingredients) Teabag by HPLC

OBJECTIVE: To develop an HPLC method for determination of peoniflorin in Bazhen(eight ingredients) teabag.METHODS:The samples were separated on Diamonsil C18 column(250 mm?4.6 mm,5 ?m) with acetonitrile-water(17∶83) as mobile phase under a detection wavelength of 230 nm.RESULTS: The linear range for peoniflorin was 0.105 3~ 2.106 0 ?g(r=0.999 9) and its average recovery rate was 98.58%(RSD=1.17%,n=9).CONCLUSION: The method is simple,rapid,accurate and applicable for the quality control of Bazhen(eight ingredients) teabag.
2.Screening affinity peptides of transforming growth factor beta type Ⅱreceptor from the phage display peptide library

Objective: To screen high-affinity short peptides of the transforming growth factor beta typeⅡ(TGF-? Ⅱ) receptor from the phage display 12-mer peptide library.Methods: The recombinant solube human TGF-? Ⅱ receptor was used as the target to screen its affinity short peptides from the phage display 12-mer peptide library.The positive phage clones were obtained by 3 rounds of biopanning,followed by single stranded DNA extraction and sequence analysis.Results: Four nucleotide sequences were obtained from 10 positive phage clones with high-affinity TGF-? Ⅱ receptor.Neither conservative nor homologous sequence was found by sequence analysis compared with the transforming growth factor beta 1.Conclusion: Phage short peptides have been screened from the phage display library,which helps pave the ground for further researches on affinity short peptides and their role in inhibiting hepatic fibrosis.
3.MORPHOLOGICAL STUDY OF THE INTERTUBERCULAR GROOVE (BICIPITAL GROOVE) OF HUMERUS

The intertubercular grooves of 212 humerus were studied. The statistical results showed that the depth of intertubercular grooves is highly correlated with the slope of the medial wall of the groove (r=0.116, p=0.05, lineal regression: =50.01+9.04x). It means that the deeper the groove the bigger the slope of the medial wall is, and the shallower the smaller. Their functional relationship has practical significance. The normal surface contour of the intertubercular groove is the important condition that keeps the long head tendon working normally. Therefore internal or external causes, e. g. fracture of the surgical neck, hyperplasia of the bone in aged people, may alter the normal anatomical relation of the intertubercular groove, which precipitated to tendosynovitis of the biceps. By observation of the compactness of the surgical neck cortex in three different levels, we found that it is in a transitional zone of from thin to thick, which agrees with the explanation of why fracture occurs frequently at the surgical neck.
4.Clinical distribution and antimicrobial resistance detection of Acinetobacter baumannii infection during 2012-2014

Objective To understand the clinical distribution of Acinetobacter baumannii and its resistance to commonly used an‐tibiotics so as to provide the theoretical basis for rational clinical use of antibacterial medicines .Methods The clinical distribution and drug resistance situation of 206 strains of clinically isolated Acinetobacter baumannii in the Anhua County People′s during 2012-2014 were analyzed .The bacterial identification was completed by the department of clinical laboratory ,the drug susceptibili‐ty test was performed by adopting the disk diffusion method ,and the detection results were judged according to CLSI 2012 criteria . Results A total of 2 803 strains of bacteria were isolated during 2012-2014 ,including 206 strains of Acinetobacter baumannii .Its constituent ratios during this period were 6 .71% ,6 .82% and 8 .19% ,showing the increasing trend year by year .The samples were mainly isolated from sputum (78 .16% ) ,followed by urine (7 .28% ) ,bronchoaleolar lavage fluid (6 .80% ) ,wound secretion (3 .40% ) and blood(2 .91% ) .The infected patients were mainly distributed in the respiratory department (40 .29% ) and neurosur‐gery department(22 .82% ) .The detection results of Acinetobacter baumannii showed the higher resistance to 20 kinds of commonly used antibacterial drugs ,in which the drugs with most significantly increase in 3‐year drug resistance rate were in turn ceftazidime , cotrimoxazole ,levofloxacin ,piperacillin and ciprofloxacin ,the 3‐year resistance rate to meropenem ,imipenem ,cefoperazone/sulbac‐tam and cefepime was less than 35% .Conclusion Acinetobacter baumannii is the major opportunistic pathogenic bacterium in noso‐comial infection with higher drug resistance rate .It is necessary to strengthen the monitoring of Acinetobacter baumannii drug re‐sistance and rationally use antibacterial medicines in order to stop the drug‐resistant strains colonization and cross expansion in hos‐pital .
5.Correlation between prognosis of patients with severe acute pancreatitis and changes of plasma albumin

Objective To investigate the correlation between prognosis of patients with severe acute pancreatitis and changes of plasma albumin. Methods A total of 235 patients with severe pancreatitis who were admitted into hospital from September 2008 to October 2013 were selected as research object. According to the prognosis, they were divided into the survival group (143 cases who were recovered and still alive) and the death group (92 cases who died of the deterioration of diseases). Retrospectively analyzed the basic situation of the pa-tients and the changes of serum albumin levels in the two groups. And the correlation between prognosis of patients with severe acute pancrea-titis and changes of plasma albumin were evaluated by the logistic regression analysis. Results Compared with the death group,there were significant differences (P<0. 05) in terms of age,APCHEⅡ score,Ranson score,SOFA score,length of hospital stay,duration of mechani-cal ventilation,continuous renal replacement therapy,average serum albumin level,decrease of serum albumin within 24 hours,and variation of serum albumin. And ROC curves suggested the best cutoff value of decrease of serum albumin within 24 hours to predict death in patients within 24 hours was 4. 24 g/L,and the best cutoff value of variation of serum albumin to predict death was 13. 6%. Logistic regression analy-sis showed that the major risk factors of death in severe acute pancreatitis were decrease of serum albumin within 24 hours,variation of serum albumin within 24 hours,average serum albumin level,and the APCHEⅡscore. Conclusion Changes in plasma albumin can reflect the se-verity of severe acute pancreatitis,and the sharp decrease of serum albumin within 24 hours can be used as a risk factor for death.
6.Effects of Suppressor of Cytokine Signaling-1 Silencing on the Proliferation and Interferon Gamma Susceptibility of Human Pancreatic Cancer Cells

Objective To detect the changes of cell proliferation and IFN-γsusceptibility of human pancreatic can-cer cells after suppressor of cytokine signaling-1 (SOCS1) gene silencing, and to explore the SOCS1 as the target of anti-tu-mor therapy through enhancing the function of IFN-γ. Methods Western blot assay, PCR and real-time PCR were used to verify the down regulation of SOCS1 in human pancreatic cancer cell (PANC1) after transfection;subsequently, PANC1 was stimulated with IFN-γ. Western blot assay was also used to detect the expression of signal transducers and activators of tran-scription (STAT)1 and phosphorylation STAT(pSTAT)1;and the change of IFN-γsusceptibility was detected by MTT assay. Real-time PCR was used to detect the mRNA of interferon regulatory factor-1(IRF-1). Flow cytometry was used to detect the cell cycle. Results The expression levels of SOCS1 mRNA and protein were significantly decreased in small hairpin SCOS1 (shSOCS1) transfected PANC1 cells. After the silence of SOCS1, the expression levels of IRF-1 and pSTAT1 in-creased significantly (P<0.05), and the median inhibitory concentration(IC50)of IFN-γfor PANC1 cells decreased signifi-cantly (P<0.01). The cell count of shSOCS1 cells dropped significantly compared with that of control group after the SOCS1 silencing for 72 hours (P<0.05). The cell cycle arrest was promoted at the G0/G1 phase, but the percentage of cells in S phase and G2/M decreased compared to that of control groups (P<0.05). Conclusion After the inhibition of SOCS1 gene expression, the proliferation ability of human pancreatic cancer cell line PANC1 decreased, and the sensitivity of PANC1 cells to IFN-γwas enhanced.
7.Study on the influencing factors of surgical treatment of patients with laryngeal cancer

Objective To investigate the influencing factors of surgical treatment of patients with laryngeal cancer.It could provide the better way to treat laryngeal cancer.Methods A total of 80 patients with laryngeal cancer from May 2003 to May 2008 in our hospital were studied.All patients were treated by surgery.The effect of surgery would be observed.The influencing factors and the effect of surgery were analyzed by Logistic.Results A total of 80 patients in the 5-year survival rates were 98.75% (79/80),76.25% (61/80),and 51.25% (41/80).Univariate results showed that age,primary site of the tumor,lymph node metastasis,tumor size,tumor node metastasis(TNM) stage,and treatment modality significantly impacted on the prognosis of patients (P <0.01).Multivariate Logistic regression analysis showed that the primary site of the tumor,lymph node metastasis,TNM staging and therapy were independent prognostic risk factors with a statistically significant difference(P < 0.05).Conclusions The prognosis of laryngeal cancer is affected by many factors,the primary sites of tumor,TNM staging,lymph node metastasis,and treatment methods were independent prognostic factors.Laryngeal function preservation surgery is an ideal method in the treatment of laryngeal cancer.
8.Subjective Quality and Related Factors of child Patients

Objective:To explore the subjective quality of life of pediatric patients.Methods:42 children with acute diseases, 38 with chronic diseases and 57 normal children were investigated with Children's Subjective Quality of Life Questionnaire. Results: The subjective life satisfaction were similar in acute disease group (159.9?13.9) and in chronic group (160.0?16.9), which were lower than normal control (165.8?17.6). Multiple regression showed that girls, from urban area, late onset, longer duration, less activities out of class, lack of support from relatives, poor mental health of parents were risk factors. Conclusion: Children with acute or chronic diseases have poorer life quality than normal control.

10.Biopanning of HCV antigen epitopes from a random 12-peptide library by anti-hepatitis C virus polyclonal antibody

Objective To analyze the epitopes of anti-hepatitis C virus(HCV)antibodies by peptide library biopanning. Methods Phage random peptide library of 12 amino acids was immunoscreened with purified IgG from the sera of hepatitis C patients. Positive clones which were obtained after 3 rounds of biopanning were detected by ELISA and 4 of them were sequenced. Results After 3 rounds of screening, the radio of output to input increased from (4.6×10~(-4))% to (5.3×10~(-2))%, meaning the enrichment was effective. At the third round of screening, all the selected clones proved to specifically react with the sera for immunoscreening. Four positive phage clones were sequenced, which shared a very conservative sequence and was named as C1. Its inserting sequence in the coat protein III was deduced to be GSMSPYVRWYTP, and the positive rate of C1 reacted with 20 cases of HCV patients was 85%.Conclusion The antigen-mimic peptide C1 is successfully screened from 12 random phage peptide library and it has some diagnostic value.
