Objective To detect the expression of Beclin1 and Bcl2 in invasive pituitary adenomas and to explore the relationship of Beclin1 and Bci2 in invasive pituitary adenomas and the relativity between the 2 genes. Methods 61 specimens were classified into invasive group (32 cases) and non-invasive group (29 cases) according to the comprehensive evaluation of invasive pituitary adenomas.lmmunofluorescence analysis and RT-PCR were adopted respectively to detect the protein and mRNA expressions of Beclinl and Bcl2.The difference and relativity of Beclin1 and Bcl2 expression in invasive group and non-invasive group were analyzed. Results 32 specimens of pituitary adenoma were invasive and 29 were non-invasive.Beclin1 protein and mRNA expressions were lower in the invasive group than in the non-invasive group (P <0.01 ).Bcl2 protein and mRNA expressions were higher in the invasive group than in the non-invasive group (P <0.01 ).Pearson related analysis showed that Beclin1 mRNA expression was negtively correlated with Bcl2 mRNA expression in the invasive group ( r =-0.42,P =0.028 ).Conclusions Beclinl expression is decreased in invasive pituitary adenomas. The invasiveness of pituitary adenoma is closely related to the high expression of Bcl2 protein and mRNA,and the low expression of Beclin1 protein and mRNA.The inhibition of the autophagy may lead to the enhancement of the invasiveness of pituitary adenomas and that inhibition may come from the interaction of Beclin1 and Bcl2.

In this study, the effect of heparin-derived oligosaccharide(HDO)on lipopolysaccharides(LPS)-induced inflammation in human umbilical vein endothelial cells(HUVECs)and the molecular mechanisms were investigated. The generation of intracellular reactive oxygen species(ROS)was detected by 20, 70-dichlorofluorescein diacetate(DCFH-DA). Experiment is divided into blank group(0. 5% serum medium), model group(LPS+0. 5% serum medium)and HDO dosing group(LPS+0. 5% serum medium +0. 01, 0. 1, 1mol/L HDO). The intracellular reactive oxygen species level was detected by reactive oxygen species experiment, the level of key regulatory proteins p38 and p-p38 in MAPK pathways and VCAM-1 were determined by Western blot. The results showed that HDO at 0. 01, 0. 1 and 1 μmol/L could inhibit the expression of VCAM-1 in HUVECs induced by 100 μg/mL LPS, and reduce the expression of key regulatory proteins p38 and p-p38, but could not obviously affect NF-κB nuclear translocation. The results all above showed that HDO could decrease the key regulatory proteins expression, and suppress the transcription of VCAM-1, resulting in inhibiting inflammation.

Chinese Journal of Blood Transfusion 2022;35(9):973-976
【Objective】 To explore the influencing factors and characteristics of blood donation-related vasovagal reactions in whole blood donation in Hefei city, and to put forward specific prevention measures. 【Methods】 Whole blood donors with vasovagal reactions related to blood donation in Hefei area from January 2021 to December 2021 were selected as the study subjects, and the related factors such as gender, age, blood donation month, initial or multiple blood donation, blood donation volume, blood donation location, and organization type of blood donation were retrospectively analyzed. 【Results】 The incidence of blood donation-related vasovagal reactions was 10.46‰ in the whole blood donation in Hefei in 2021, among which 71.89% occurred during blood collection. There were statistically significant differences in blood donation-related vasovagal reactions by blood donation age, blood donation history, blood collection month, organization type of blood donation and blood donation volume(P<0.05), but not by gender and location of blood collection(P>0.05). After balancing the age factors, there was no statistically significant difference in the incidence of DRVR between donors aged 18~22 and 23~29 years by gender(P>0.05), while there were significant differences in blood donors aged 30~39, 40~49, and 50~60 years by gender(P<0.05). Among blood donors of different ages, there were statistically different incidences of DRVR among first-time and repeated donors (P<0.05). 【Conclusion】 In the process of whole blood donation, attention should be paid to blood donors first-time, young (18~30 years old) and student, which can help prevent the occurrence of blood donation-related vasovagal reactions and better retain blood donors.

Objective:
To explore the value of quantitative CT radiomics features in predicting the anaplastic lymphoma kinase (ALK) mutation status in lung adenocarcinoma patients.
Methods:
This retrospective study reviewed one hundred and ninety-five lung adenocarcinoma patients (including 60 patients with ALK mutation) whose ALK genetic test results were available from Nov 2015 to May 2018 in PUMCH. VOIs were labeled by an automatic pulmonary nodule detection and segmentation algorithm and were later revised and confirmed by two senior radiologists. The PyRadiomics tools were used to resample the labeled regions, followed by image pre-processing (Wavelet filter or Laplacian of Gaussian (LoG) filter) and feature extraction. Normalized features were selected based on their representativeness on Dr. Wise research platform. Multivariate logistic regression was performed to develop prediction models of ALK mutation gene based on different image pre-processing techniques and different radiomics feature types. The results were validated by ten runs of five-fold cross validation. ROC curve analysis and Delong test were used to compare the predictive performance among models.
Results:
Fifteen radiomics features with the highest representativeness were selected from the original 1 232 features. The prediction model based on these radiomics features showed good performance (AUC=0.88 in the training set and 0.78 in the validation set) and was not significantly different from the prediction models based on radiomics features of different pre-processing images (AUC=0.76,
